Axonal regeneration following surgical nerve repair is slow and often incomplete,resulting in poor functional recovery which sometimes contributes to lifelong ***,there are no FDA-approved therapies available to promote nerve *** accelerates axonal regeneration,but systemic side effects presently outweigh its potential benefits for peripheral nerve *** authors describe herein a biodegradable polyurethane-based drug delivery system for the sustained local release of tacrolimus at the nerve repair site,with suitable properties for scalable production and clinical application,aiming to promote nerve regeneration and functional recovery with minimal systemic drug *** is encapsulated into co-axially electrospun polycarbonate-urethane nanofibers to generate an implantable nerve wrap that releases therapeutic doses of bioactive tacrolimus over 31 *** and drug loading are adjustable for applications in small and large caliber nerves,and the wrap degrades within 120 days into biocompatible *** released from the nerve wrap promotes axon elongation in vitro and accelerates nerve regeneration and functional recovery in preclinical nerve repair models while off-target systemic drug exposure is reduced by 80%compared with systemic *** its surgical suitability and preclinical efficacy and safety,this system may provide a readily translatable approach to support axonal regeneration and recovery in patients undergoing nerve surgery.

Information about 255 children and adults with Pompe disease was gathered thr ough a questionnaire. Disease severity was associated with disease duration and not with age; an early manifestation of the disease implied earlier wheelchair o r ventilator dependency. The patient group under age 15 included a subgroup with a more severe and rapid course of the disease. They require more intensive foll ow- up and early intervention, before irreversible damage has occurred.

As environmental concerns from fossil fuel consumption intensify, large-scale energy storage becomes imperative for the integration of renewable sources like wind, hydro, and solar with the electrical *** flow batteries, particularly those employing organic molecules, are positioned as a key technology for this purpose. This review explores the growing field of symmetric organic redox flow batteries(ORFBs) within this context. Unlike traditional asymmetric designs based on unique active materials for each electrode, symmetric ORFBs involve a single bipolar species for both electrodes. This review highlights the benefits of a symmetric design, and categorizes five distinct classes of organic bipolar molecules used in both aqueous and non-aqueous solvents. By providing a comprehensive overview of their cell cycling and performance characteristics, the strengths and weaknesses of the diverse categories of bipolar molecules are highlighted for both solvent systems, as are opportunities for future development. This should guide new research directions and advance the development of practical symmetric ORFBs.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. we then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.

weakly consolidated reservoirs are prone to sand production problem,which can lead to equipment damages and environmental *** conditions for sand production depend on stresses and properties of rock and *** sand volume estimation is,however,still a challenging issue,especially for reservoirs in weak *** weak reservoirs containing viscous or heavy oil are mainly discovered in shallow depths in Kazakhstan,with moderate temperature and *** prediction models developed for open-hole completions where the reservoir materials usually possess certain strength are not applicable for the local reservoirs where the materials are significantly weaker even if casing is used to support the wellbore with oil produced through the perforation *** this context,a prediction model was proposed where the volume of the produced sand was estimated as the volume of the plastic zone of the failed materials surrounding the perforation *** model assumes an evolving truncated conical shape for the damage zone and takes into account stress distributions and shear failure in this ***,the proposed model was used to estimate sand volumes in 20 wells during oil production with sequential increase of flow *** predictions match well with the measured sand volumes in a local oil ***,a sensitivity analysis was conducted on the model *** shows that the permeability of the plastic zone was the most significant controlling factor in the prediction results.

Sand production is often characterized as a two-stage process,in which material failure occurs near the cavity,leading to the formation of a plastic zone,from which particles are detached and transported out because of continuous hydrodynamic erosion under the effects of the produced fluid *** plastic zone porosity is affected by coupled processes,while the plastic zone permeability has a significant impact on the performance of sand production prediction,especially in weak sandstone ***-scale sand production experiments were conducted using a customized high-pressure consolidation *** results show that specific stress-fluid pressure conditions may create a plastic zone around a hole,which has lower permeability than the intact *** plastic zone comprises two subzones:a high-permeability shear band zone and a low-permeability compaction *** sand production,sand migrates from the compaction zone through the shear band zone to the perforation ***,sand production is associated with the increased permeability and porosity in the compaction *** sand prediction models were modified according to the new findings,resulting in a modified model with improved *** modified model was validated using the sanding data from a weak sandstone reservoir in Kazakhstan.

Sand production is,a challenging issue in petroleum industry,mainly associated with weak unconsolidated formations.A novel testing procedure and a new apparatus were developed to conduct an integrated experiment of diagenesis,perforation and sand production on a single large cylindrical artificial sandstone specimen,where solid and fluid pressures can be independently controlled such that realistic reservoir historical conditions can be well simulated in the *** injection can be performed in both radial and vertical directions,where both single-and two-phase flows can be implemented for study of sand production behaviors at different reservoir’s maturity *** equipment consists of an intensive instrumentation system to monitor pressures,displacements and material states *** produced sand particles were filtered and monitored in real-time for the study of time-dependent *** experimental results showed similar patterns to that observed in the field and provided valuable insight for the development of prediction methods for sand production of similar materials.

To study the temperature distribution and thermal-stress field in different service stages, a two-dimensional model of a turbine blade with thermal barrier coatings is developed, in which the conjugate heat transfer analysis and the decoupled thermal-stress calculation method are adopted. Based on the simulation results, it is found that a non-uniform distribution of temperature appears in different positions of the blade surface, which has directly impacted on stress field. The maximum temperature with a value of 1030 ℃ occurs at the leading edge. During the steady stage, the maximum stress of thermally grown oxide （TGO） appears in the middle of the suction side, reaching 3.75 GPa. At the end stage of cooling, the maximum compressive stress of TGO with a value of-3.5 GPa occurs at the leading edge. Thus, it can be predicted that during the steady stage the dangerous regions may locate at the suction side, while the leadine edge mav be more Drone to failure on cooling.
