High-speed impulsive stimulated Brillouin microscopy

Brillouin microscopy, which maps the elastic modulus from the frequency shift of scattered light, has evolved to a faster speed for the investigation of rapid biomechanical changes. Impulsive stimulated Brillouin scattering (ISBS)spectroscopy has the potential to speed up measurement through the resonant amplification interaction from pulsed excitation and time-domain continuous detection. However, significant progress has not been achieved due to the limitation in signal-to-noise ratio (SNR) and the corresponding need for excessive averaging to maintain high spectral precision. Moreover, the limited spatial resolution also hinders its application in mechanical imaging. Here, by scrutinizing the SNR model, we design a high-speed ISBS microscope through multi-parameter optimization including phase, reference power, and acquisition time. Leveraging this, with the further assistance of the Matrix Pencil method for data processing, three-dimensional mechanical images are mapped under multiple contrast mechanisms for a millimeter-scale polydimethylsiloxane pattern immersed in methanol, enabling the identification of these two transparent materials without any contact or labeling. Our experimental results demonstrate the capability to maintain high spectral precision and resolution at a sub-millisecond integration time for one pixel. With a two-order improvement in the speed and a tenfold improvement in the spatial resolution over the state-of-the-art systems, this method makes it possible for ISBS microscopes to sensitively investigate rapid mechanical changes in time and space.

The fall armyworm,Spodoptera frugiperda(S.frugiperda),represents the most resistant insect species and poses serious threat to grain yield.Chlorantraniliprole(CHL),which targets the ryanodine receptors(RyRs)in insects,has demonstrated the efficacy in controlling S.frugiperda.Nevertheless,this has led to emerging resistance in several countries.To counter this resistance,a viable approach involves the development of novel compounds that bind against RyRs via distinct binding sites or modes.In this study,a series of 22 novel anthranilic diamide derivatives was designed and synthesized,and their insecticidal activities were evaluated.Most of these derivatives showed moderate to good insecticidal activity against S.frugiperda and Mythimna separata.Time-lapse fluorescence measurements of endoplasmic reticulum luminal calcium revealed that most derivatives elicited cellular responses similar as CHL when assessed on HEK293 cells expressing S.frugiperda ryanodine receptors(SfRyRs).The mode of action of compound 13a was studied and verified on the isolated neurons by calcium imaging technique.Finally,molecular docking analysis was employed to predict the binding mechanism of compound 13a against SfRyRs.Overall,these novel diamide derivatives hold promise as a valuable resource for guiding the future design of insecticidal compounds targeting RyRs.

Extraction mechanism and separation behaviors of low-concentration Nd^(3+) and Al^(3+) in P507-H_(2)SO_(4) system

In order to clarify the solvent extraction and separation behaviors of rare earths and impurity of Al during the extraction and enrichment of low-concentration leach solution of ion-adsorption rare earth ore,the extraction mechanism and separation behaviors of Nd^(3+)and Al^(3+)in the Nd_(2)(SO_(4))_(3)-AI_(2)(SO_(4))_(3) mixed solution using P507 were studied in this work.The extraction of Nd^(3+)and Al^(3+)follows the cation exchange mechanism.With the increase of the equilibrium pH,β_(Nd/Al) in the extraction of the Nd_(2)(SO_(4))_(3)-Al_(2)(SO_(4))_(3) mixed solution using P507 is always higher than that in the extraction of single Nd_(2)(SO_(4))_(3) and Al_(2)(SO_(4))_(3) solutions.It can be attributed to the fact that the extraction of Nd^(3+)using P507 is much faster than that of Al^(3+),and Al^(3+)is more prone to be hydrolyzed at lower pH.β_(Nd/Al) in the extraction of the Nd_(2)(SO_(4))_(3)-Al_(2)(SO_(4))_(3) mixed solution decreases gradually with the increase of mixing time within the equilibrium pH range of 1.5-1.9.The extraction of Nd^(3+)using P507 is much faster than that of Al^(3+),but the stability of Al^(3+)-Ioaded organic phase is better than that of Nd^(3+)-loaded organic phase,thus Nd^(3+)in the Nd^(3+)-loaded organic phase is gradually replaced by Al^(3+)in the aqueous phase with the increase of mixing time.

A Three-Dimensional Model for the Formation Pressure in Wellbores under Uncertainty

Formation pressure is the key parameter for the analysis of wellbore safety.With increasing drilling depth,how-ever,the behavior of this variable becomes increasingly complex.In this work,a 3D model of the formation pres-sure under uncertainty is presented.Moreover a relevant algorithm is elaborated.First,the logging data of regional key drilling wells are collected and a one-dimensional formation pressure proﬁle along the well depth is determined.Then,a 3D model of regional formation pressure of the hierarchical group layer is deﬁned by using the Kriging interpolation algorithm relying on a support vector machine(SVM)and the formation pressure of the drilled wells.To validate the method,the formation pressure of one pre-drilled well is compared with the well logging results.The comparison reveals that the maximum relative error is less than 4.5%.The software based on this model is complemented by a computer visualization technology,which provides a relevant tool for under-standing and analyzing the 3D formation pressure.The outcomes of this study are intended to support the char-acterization of areas with missing or poor 3D seismic data and provide more accurate information for the analysis of wellbore integrity.

PAM-Expanded Streptococcus thermophilus Cas9 C-to-T and C-to-G Base Editors for Programmable Base Editing in Mycobacteria

New therapeutic strategies for the rapid and effective treatment of drug-resistant tuberculosis are highly desirable,and their development can be drastically accelerated by facile genetic manipulation methods in Mycobacterium tuberculosis(M.tuberculosis).Clustered regularly interspaced short palindromic repeat(CRISPR)base editors allow for rapid,robust,and programmed single-base substitutions and gene inactivation,yet no such systems are currently available in M.tuberculosis.By screening distinct CRISPR base editors,we discovered that only the unusual Streptococcus thermophilus CRISPR associated protein 9(St1Cas9)cytosine base editor(CBE)-but not the widely used Streptococcus pyogenes Cas9(SpCas9)or Lachnospiraceae bacterium Cpf1(LbCpf1)CBEs-is active in mycobacteria.Despite the notable C-to-T conversions,a high proportion of undesired byproducts exists with St1Cas9 CBE.We therefore engineered St1Cas9 CBE by means of uracil DNA glycosylase inhibitor(UGI)or uracil DNA glycosylase(UNG)fusion,yielding two new base editors(CTBE and CGBE)capable of C-to-T or C-to-G conversions with dramatically enhanced editing product purity and multiplexed editing capacity in Mycobacterium smegmatis(M.smegmatis).Because wild-type St1Cas9 recognizes a relatively strict protospacer adjacent motif(PAM)sequence for DNA targeting,we engineered a PAM-expanded St1Cas9 variant by means of structureguided protein engineering for the base editors,substantially broadening the targeting scope.We first developed and characterized CTBE and CGBE in M.smegmatis,and then applied CTBE for genome editing in M.tuberculosis.Our approaches significantly reduce the efforts and time needed for precise genetic manipulation and will facilitate functional genomics,antibiotic-resistant mechanism study,and drugtarget exploration in M.tuberculosis and related organisms.

Multi-sensor monitoring of Ulva prolifera blooms in the Yellow Sea using different methods

The massive Ulva （U.）prolifera bloom in the Yellow Sea was first observed and reported in summer of 2008. After that, the green tide event occurred every year and influenced coastal areas of Jiangsu and Shandong provinces of China. Satellite remote sensing plays an important role in monitoring the floating macroalgae. In this paper, U. prolifera patches are detected from quasi- synchronous satellite images with different spatial resolu- tion, i.e., Aqua MODIS （Moderate Resolution Imaging Spectroradiometer）, HJ-1A/B （China Small Satellite Con- stellation for Environment and Disaster Monitoring and Forecasting）, CCD （Charge-Coupled Device）, Landsat 8 OLI （Operational Land Imager）, and ENVISAT （Environ- mental Satellite） ASAR （Advanced Synthetic Aperture Radar） images. Two comparative experiments are per- formed to explore the U. prolifera monitoring abilities by different data using detection methods such as NDVI （Normalized Difference Vegetation Index） with different thresholds. Results demonstrate that spatial resolution is an important factor affecting the extracted area of the floating macroalgae. Due to the complexity of Case II sea water characteristics in the Yellow Sea, a fixed threshold NDVI method is not suitable for U. prolifera monitoring. A method with adaptive ability in time and space, e.g., the threshold selection method proposed by Otsu （1979）, is needed here to obtain accurate information on the floating macroalgae.

Overcome Debye Length Limitations for Biomolecule Sensing Based on Field Effective Transistors

Biosensors based on field effective transistor(FET)have aroused tremendous attention in the past few years owning to their huge application in drug discovery,disease diagnosis and environmental monitoring.The FET biosensors possess small volume,high sensitivity at ultra-low concentration,considerable mechanical strength,as well as excellent stability in solution,which plays a vital role in the point of care testing(POCT)systems.Recent advances have summarized some progress involved in the improvement of morphology and structure of channel materials,the functionalization of organic molecule,the influence of device operation and sensing environment on the detecting performance.However,for FET biosensors,the charge screening phenomena were inevitable in the solution,which seriously degrade the device performance.In this article,we summarize recent advances to overcome debye length limitations for biomolecule sensing based on FET.We will firstly describe the charge screening mechanism,then focous on the strategy to overcome charge screening,including synthesizing special channel materials with crumpled morphology,designing aptamer binding mode,and modulating device measurement.Finally,we discuss the major challenges and perspectives about overcoming debye length limitations of FET biosensors.These summaries provide further insights to realize real-time,lable-free,high-sensitivity FET sensors for medical healthcare.

Transcriptome based high-throughput SSRs and SNPs discovery in the medicinal plant Lagenaria siceraria

Lagenaria siceraria(Molina)Standley has unique biological characteristics with high nutritional and medicinal values.It is an important pharmaceutical plant with various biologically active ingredients.Genetic improvement and deeper genomic studies require a rich resource of molecular markers.The application of next-generation sequencing technology,especially for transcriptome profiling,has greatly facilitated high throughput single nucleotide polymorphism(SNP)and simple sequence repeat(SSR)discovery.In this study,we sequenced the transcriptome of three major cultivars of L.siceraria and obtained 64.88 GB of clean data.The assembled high-quality reads were clustered into 89,347 unigenes,which were annotated by non-redundant protein database,Swiss-Port,Eukaryotic Ortholog Groups,Kyoto Encyclopedia of Genes and Genomes,and gene ontology databases.A total of 8,891 SSR and 35,873 SNP markers were predicted from unigenes by MISA and SAM tools,respectively.Characterization of the predicted markers in L.siceraria showed that the SSR and SNP densities were 60 and 243 markers per Mb of genome,respectively,and the estimated ratio of transition to transversion of SNP was 2.016.These markers will be very useful for genetic studies in L.siceraria,especially for the high-density linkage map construction and genome-wide association studies.Further genomic studies based on these results will facilitate the identification of novel genes or alleles of pharmaceutical importance.

Diagnosis of disease-specific proteins in cerebrospinal fluid of children infected with tuberculous meningitis

BACKGROUND： Isolated mycobacterium tuberculosis from cerebrospinal fluid （CSF） is regarded as the ＂gold standard＂ for diagnosis of tuberculous meningitis （TBM）. However, culture of CSF specimens is time-consuming and lacks sensitivity. There is a strong need to determine complementary disease-specific markers, which are essential for increasing early diagnosis and improving prognosis in patients with TBM OBJECTIVE： To establish proteomic profiles of CSF in TBM and normal children using two-dimensional polyacrylamide gel electrophoresis, and to screen for disease-specific proteins. DESIGN, TIME AND SETTING： The case-control study was conducted at the Department of Pediatrics, Xiangya Hospital of Central South University and the Key Laboratory of Cancer Proteomics of Ministry of Public Health of China between January 2008 and January 2009. PARTICIPANTS： The TBM group included three patients with a strongly positive tuberculin skin test, as well as positive CSF mycobacterial staining and culture, who were admitted to the Department of Pediatrics, Xiangya Hospital from January 2008 to January 2009. Three healthy, age- and gender-matched children served as the control group. METHODS： CSF proteins were separated using two-dimensional polyacrylamide gel electrophoresis in both groups. Gels were scanned using Image scanner and LabScan software. Differentially expressed proteins were analyzed using PDQuest 7.0 software. The clearly discernible spots, which were expressed only in the TBM group, were chosen to perform matrix-assisted laser desorption/ionization time-of-flight mass spectrometry analysis. MAIN OUTCOME MEASURES： Differentially expressed spots on CSF profiles of TBM and normal children were measured. RESULTS： Following comparison of two-dimensional polyacrylamide gel electrophoresis maps between TBM and control groups, 546 and 533 spots were detected, respectively. A total of 64 differentially expressed proteins were observed between the groups, including 15 upregulated spots, eight downregulated spots, 27 spot

患者,女性,年龄45岁,身高155 cm,体质量57 kg。因"大便时肛门处肿物脱出,便后不能还纳"入院。既往甲状腺功能减退病史7年,口服左甲状腺素钠片治疗,病情控制良好;剖腹产手术史20余年,自诉曾因腰部疼痛行腰部牵引治疗,近十来年无明显腰腿部不适。诊断为混合痔、便秘、甲状腺功能减退,拟在椎管内阻滞下行吻合器痔上黏膜环切加痔外剥内扎术。ASA分级Ⅱ级,心电图及各项实验室检查未见明显异常。
