Targeting LTA4H facilitates the reshaping of the immune microenvironment mediated by CCL5 and sensitizes ovarian cancer to Cisplatin

Ovarian cancer is the most lethal and aggressive gynecological cancer with a high recurrence rate and is often diagnosed late.In ovarian cancer,multiple metabolic enzymes of lipid metabolism are abnormally expressed,resulting in metabolism disorder.As a characteristic pathway in polyunsaturated fatty acid(PUFA)metabolism,arachidonic acid(AA)metabolism is disturbed in ovarian cancer.Therefore,we established a 10-gene signature model to evaluate the prognostic risk of PUFA-related genes.This 10-gene signature has strong robustness and can play a stable predictive role in datasets of various platforms(TCGA,ICGC,and GSE17260).The high association between the risk subgroups and clinical characteristics indicated a good performance of the model.Our data further indicated that the high expression of LTA4H was positively correlated with poor prognosis in ovarian cancer.Deficiency of LTA4H enhanced sensitivity to Cisplatin and modified the characteristics of immune cell infiltration in ovarian cancer.Additionally,our results indicate that CCL5 was involved in the aberrant metabolism of the AA/LTA4H axis,which contributes to the reduction of tumor-infiltrating CD8+T cells and immune escape in ovarian cancer.These findings provide new insights into the prognosis and potential target of LTA4H/CCL5 in treating ovarian cancer.

Electrode particulate materials for advanced rechargeable batteries:A review

The demand for large-scale energy storage is increasing due to the decreasing non-renewable resources and deteriorating environmental pollution.Developing rechargeable batteries with high energy density and long cycle performance is an ideal choice to meet the demand of energy storage system.The development of excellent electrode particles is of great significance in the commercialization of nextgeneration batteries.The ideal electrode particles should balance raw material reserves,electrochemical performance,price and environmental protection.Among them,the development of electrode particulate materials with excellent electrochemical properties is the top priority at present.In this review,the typical researches of electrode materials are summarized in terms of crystal structure,morphology,pore structure,surface and interface regulation.Firstly,the structural characteristics and improvement methods of transition metal oxides,polyanionic compounds,Prussian blue and their analogues are introduced.Then,the different effects of particulate morphology,pore,surface and interface structure on the performance of electrode materials are discussed.For designing high-performance electrode materials,preparation route should be set according to the particle properties of the materials and the synergistic effect of various optimization methods should be adopted.At the same time,in addition to the electrode materials,other components of the rechargeable batteries,such as current collector,separator and electrolytes,should be optimized to improve the overall performance of the batteries.This review would provide important guiding principle for designing high-performance electrode particulate materials.

A Comprehensive Overview of CFN From a Commonsense Perspective

Chinese FrameNet(CFN)is a scenario commonsense knowledge base(CKB)that plays an important role in research on Chinese language understanding.It is based on the theory of frame semantics and English FrameNet(FN).The CFN knowledge base contains a wealth of scenario commonsense knowledge,including frames,frame elements,and frame relations,as well as annotated instances with rich scenario-related labels on Chinese sentences and discourses.In this paper,we conduct a comprehensive overview of CFN from a commonsense perspective,covering topics such as scenario commonsense representation,CFN resources,and its applications.We also summarize recent breakthroughs and identify future research directions.First,we introduce the concept of scenario commonsense,including its definitions,examples,and representation methods,with a focus on the relationship between scenario commonsense and the frame concept in CFN.In addition,we provide a comprehensive overview of CFN resources and their applications,highlighting the newly proposed frame-based discourse representation and a human-machine collaboration framework for expanding the CFN corpus.Furthermore,we explore emerging topics such as expanding the CFN resource,improving the interpretability of machine reading comprehension,and using scenario CKBs for text generation.

A Type-2 Fuzzy Approach to Driver-Automation Shared Driving Lane Keeping Control of Semi-Autonomous Vehicles Under Imprecise Premise Variable

The driver-automation shared driving is a transition to fully-autonomous driving,in which human driver and vehicular controller cooperatively share the control authority.This paper investigates the shared steering control of semi-autonomous vehicles with uncertainty from imprecise parameter.By considering driver’s lane-keeping behavior on the vehicle system,a driver-automation shared driving model is introduced for control purpose.Based on the interval type-2(IT2)fuzzy theory,moreover,the driver-automation shared driving model with uncertainty from imprecise parameter is described using an IT2 fuzzy model.After that,the corresponding IT2 fuzzy controller is designed and a direct Lyapunov method is applied to analyze the system stability.In this work,sufficient design conditions in terms of linear matrix inequalities are derived,to guarantee the closed-loop stability of the driver-automation shared control system.In addition,an H∞performance is studied to ensure the robustness of control system.Finally,simulation-based results are provided to demonstrate the performance of proposed control method.Furthermore,an existing type-1 fuzzy controller is introduced as comparison to verify the superiority of the proposed IT2 fuzzy controller.

Protein crystallization with DNA templates

Protein crystallization plays a significant role in three-dimensional structural analysis and protein pu-rification.It is important to increase the crystallization efficiency,which is possible by adding hetero-geneous templates in crystallization systems.DNA is biologically compatible and artificially designable polymer,which is easy to extract.In this study,single-and double-stranded DNA of precise sequences were designed and used as templates to promote protein crystallization of lysozyme and catalase.In-fluence of DNA,single-stranded DNA with 10,20,40 bases and double-stranded DNA with 10,20,40 base pairs,were investigated.The success rate of obtaining crystals of lysozyme and catalase in equal period was significantly improved with the addition of DNA comparing without templates added.Double-stranded DNA led to higher nucleation rate than that with single-stranded DNA.The promotion of nucleation was more obvious at low concentration of protein solution and with longer chain DNA templates.Crystal number and crystallization rate was enhanced with addition of long double-stranded DNA templates.All the results confirm that DNA is an effective polymer additive to enhance protein crystallization,especially for the application of the scarce protein crystallization.

A Bifunctional-Blocker-Aided Hybridization Chain Reaction Lighting-Up Self-calibrating Nanocluster Fluorescence for Reliable Nucleic Acid Detection

In this work,we proposed a ratiometric silver nanoclusters(AgNCs)fluorescent assay by designing a bifunctional-blockeraided hybridization chain reaction(HCR).Hairpin probe 1(HP1)containing two special DNA fragments(5′-CAC CGC T-3′and 5′-ATT TGC CTT TTG GGG ACG GATA-3′)at two terminals creates a red-emitting AgNC nucleation sequence(rNS,5′-CAC CGC TAT TTG CCT TTT GGG GAC GGATA-3′).We found that the presence of a toehold fragment(5′-TGCCC-3′)in HP1 could silence the rNS.Upon the addition of a target nucleic acid,HCR of HP1 and hairpin probe 2(HP2)could be initiated,resulting in the formation of long chain of DNA duplexes with multibranched rNS.As the toehold fragment in HP1participated in generating duplexes,a strong emission of rNS-templated AgNCs was observed at 670 nm.More significantly,a bifunctional blocker was introduced not only to reduce the background red-emitting fluorescence but also to play as an internal green-emitting AgNCs nucleation sequence.On the one hand,the blocker could increase the signal-to-noise-ratio of the constructed biosensor,and on the other hand,the blocker also helped to prepare ratiometric HCR-AgNCs assay with self-calibrating ability to strengthen its reproducibility.Compared with the traditional HCR-AgNCs sensors,the developed ratiometric assay based on the bifunctional-blocker-aided HCR has higher reliability,which is important for the fabrication of biosensors in various fields for practical biosensing applications.

The application and perspective of NMR and MS based strategies for functional compounds mining in medicinal and dietary plants

Medicinal and dietary plants provide numerous nutritional and functional compounds and also have various potential health benefits to humanity. The specific and efficient techniques for accurate identification of nutritional compounds and functional metabolites is crucial for the development of functional foods from medicinal and dietary plants. Nuclear magnetic resonance (NMR) and mass spectrometry (MS) are indispensable and essential technologies that provide an unsurpassed wealth of untargeted identification， quantitative and qualitative analysis， and structural information in the study of food and plant products. In the past decade， the rapid development of modern analytical technology has led to the emergence of new approaches and strategies for natural products discovery. Especially the application of novel NMR- and MS-based identification and dereplication strategies aided by artificial intelligence and machine learning algorithms have brought about a significant shift in the natural products discovery process. These developments and changes in the natural products filed have given us insights into how to accurately target and mining nutritional， functional， and bioactive compounds. Thus， we have summarized recent research on novel NMR and MS based strategies and methods focusing on functional compounds， accurate identification and efficient discovery mainly in medicinal and dietary plants. This review could provide a comprehensive perspective for a better understanding of novel strategies and methods based on NMR and MS technologies， which could provide valuable insights and ideas for functional compounds mining.

Dendrocalamus latiflorus and its component rutin exhibit glucose-lowering activities by inhibiting hepatic glucose production via AKT activation

The potential medicinal value of Ma bamboo(Dendrocalamus latiflorus), one of the most popular and economically important bamboo species in China, has been underestimated. In the present study, we found that D. latiflorus leaf extract(DLE) reduced fasting blood glucose levels, body weight,and low-density lipoprotein cholesterol with low liver toxicity in db/db mice. In addition, gene expression profiling was performed and pathway enrichment analysis showed that DLE affected metabolic pathways.Importantly, DLE activated the AKT signaling pathway and reduced glucose production by downregulating glucose-6-phosphatase(G6 PC) and phosphoenolpyruvate carboxykinase 1(PCK1) expression. Moreover, network pharmacology analysis identified rutin as an active component in DLE through targeting insulin growth factor 1 receptor(IGF1 R), an upstream signaling transducer of AKT. Due to its hypoglycemic effects and low toxicity, DLE may be considered an adjuvant treatment option for type 2 diabetes patients.

Mitochondrial energy metabolism changes during aging-mouse cranial nerve cells treated with various doses and forms of Fructus schizandrae

BACKGROUND： During the cellular aging process, the number of mitochondria, generation of adenosine triphosphate （ATP）, activity of respiratory chain enzyme complex 1 and 4, and oxidation decrease. OBJECTIVE： To observe the effects of aqueous and spirituous extract, as well as polysaccharides from Fructus schizandrae （Magnolia Vine） on energy metabolism and mitochondrial anti-oxidation in cranial nerve cells of a D-gal-induced aging mouse model. DESIGN, TIME AND SETTING： A randomized, controlled, animal study. The experiment was conducted at the Department of Biochemistry, Qiqihar Medical College between March and July 2006. MATERIALS： Fifty healthy, Kunming mice of both sexes, aged 2 3 months old and weighing 18 22 g, were used for the present study. Fructus schizandrae was purchased from the Medical College of Jiamusi University. Aqueous extracts, spirituous extracts, and polysaccharides from Fructus schizandrae were prepared. D-galactose （D-gal） is a product of the Second Reagent Factory, Shanghai City, China. Mn-superoxide dismutase （Mn-SOD） kit, malonaldehyde （MDA） kit, protein quantification kit, and inorganic phosphorus testing kit were purchased from Jian Cheng Bioeng. Co., China. METHODS： Fifty mice were randomly divided into five groups, with 10 mice in each group： young control, aging model, aqueous Fructus schizandrae extract, spirituous Fructus schizandrae extract, and Fructus schizandrae polysaccharides. Over a course of 30 days, mice in aging model, aqueous Fructus schizandrae extract, spirituous Fructus schizandrae extract, and Fructus schizandrae polysaccharides groups were injected subcutaneously with D-gal （100 mg/kg） into the nape of the neck daily, and administered intragastrically with an equal volume of sterile, warm water （aging model）, aqueous Fructus schizandrae extract （2 g/kg）, spirituous Fructus schizandrae extract （2 g/kg）, or Fructus schizandrae polysaccharides （0.2 g/kg）, respectively. Establishment of an optimized CTC detection model consisting of EpCAM,MUC1 and WT1 in epithelial ovarian cancer and its correlation with clinical characteristics

Objective:Emerging studies have demonstrated the promising clinical value of circulating tumor cells(CTCs)for diagnosis,disease assessment,treatment monitoring and prognosis in epithelial ovarian cancer.However,the clinical application of CTC remains restricted due to diverse detection techniques with variable sensitivity and specificity and a lack of common standards.Methods:We enrolled 160 patients with epithelial ovarian cancer as the experimental group,and 90 patients including 50 patients with benign ovarian tumor and 40 healthy females as the control group.We enriched CTCs with immunomagnetic beads targeting two epithelial cell surface antigens(EpCAM and MUC1),and used multiple reverse transcription-polymerase chain reaction(RT-PCR)detecting three markers(EpCAM,MUC1 and WT1)for quantification.And then we used a binary logistic regression analysis and focused on EpCAM,MUC1 and WT1 to establish an optimized CTC detection model.Results:The sensitivity and specificity of the optimized model is 79.4%and 92.2%,respectively.The specificity of the CTC detection model is significantly higher than CA125(92.2%vs.82.2%,P=0.044),and the detection rate of CTCs was higher than the positive rate of CA125(74.5%vs.58.2%,P=0.069)in early-stage patients(stage I and II).The detection rate of CTCs was significantly higher in patients with ascitic volume≥500 mL,suboptimal cytoreductive surgery and elevated serum CA125 level after 2 courses of chemotherapy(P<0.05).The detection rate of CTC;and CTC;was significantly higher in chemo-resistant patients(26.3%vs.11.9%;26.4%vs.13.4%,P<0.05).The median progression-free survival time for CTC;patients trended to be longer than CTC;patients,and overall survival was shorter in CTC;patients(P=0.043).Conclusions:Our study presents an optimized detection model for CTCs,which consists of the expression levels of three markers(EpCAM,MUC1 and WT1).In comparison with CA125,our model has high specificity and demonstrates better diagnostic values,especially for early-stage ovarian cancer.Detection of
