Mesenchymal stem cell(MSC)-mediated immunomodulation has been harnessed for the treatment of human diseases,but its underlying mechanism has not been fully *** cells,including apoptotic cells have immunomodulatory *** has been repeatedly reported that the proportion of nonviable MSCs in a MSC therapeutic preparation varied from 5-50%in the ongoing clinical *** is conceivable that the nonviable cells in a MSC therapeutic preparation may play a role in the therapeutic effects of *** found that the MSC therapeutic preparation in the present study had about 5%dead MSCs(DMSCs),characterized by apoptotic ***,1×10^(6) MSCs in the preparation contained about 5×10^(4) *** found that the treatment with even 5×10^(4) DMSCs alone had the equal therapeutic effects as with 1×10^(6) *** protective effect of the dead MSCs alone was confirmed in four mouse models,including concanavalin A(ConA)-and carbon tetrachloride(CCI4)-induced acute liver injury,LPS-induced lung injury and spinal cord *** also found that the infused MSCs died by apoptosis in ***,the therapeutic effect was attributed to the elevated level of phosphatidylserine(PS)upon the injection of MSCs or *** direct administration of PS liposomes(PSLs)mimic apoptotic cell fragments also exerted the protective effects as MSCs and *** Mer tyrosine kinase(MerTK)deficiency or the knockout of chemokine receptor C-C motif chemokine receptor 2(CCR2)reversed these protective effects of MSCs or *** results revealed that DMSCs alone in the therapeutic stem cell preparation or the apoptotic cells induced in vivo may exert the same immunomodulatory property as the'living MSCs preparation"through releasing PS,which was further recognized by MerTK and participated in modulating immune cells.

Cancer and impaired tissue wound healing with ageing are closely related to the quality of life of the elderly *** the increased incidence of cancer and the population ageing trend globally,it is very important to explore how ageing impairs tissue wound healing and spontaneous *** a murine model of DSS-induced acute colitis and AOM/DSS-induced colitis-associated cancer(CAC),we found ageing significantly decreases intestinal wound healing and simultaneous CAC initiation,although ageing does not affect the incidence of AOM-induced,sporadic non-inflammatory ***,reduced fibroblasts were observed in the colitis microenvironment of ageing *** conditional lineage tracing,an important source of fibroblasts potentially derived from intestinal smooth muscle cells(ISMCs)was identified orchestrating intestinal wound healing and CAC initiation in young ***,the number of transformed fibroblasts from ISMCs significantly decreased in ageing mice,accompanied by decreased intestinal wound healing and decreased CAC ***-fibroblasts transformation in young mice and reduction of this transformation in ageing mice were also confirmed by ex-vivo intestinal muscular layer culture *** further found that activation of YAP/TAZ in ISMCs is required for the transformation of ISMCs into ***,the reduction of YAP/TAZ activation in ISMCs during intestinal wound healing was observed in ageing *** knockdown of YAP/TAZ in ISMCs of young mice results in reduced fibroblasts in the colitis microenvironment,decreased intestinal wound healing and decreased CAC initiation,similar to the phenotype of ageing *** addition,the data from intestine samples derived from inflammatory bowel disease(IBD)patients show that activation of YAP/TAZ also occurs in ISMCs from these ***,our work reveals an important role of the ageing stromal microenvironment in intestinal wound healing and CAC ini

Tumor escape from immune-mediated destruction has been associated with immunosuppressive mechanisms that inhibit T-cell activation.A promising strategy for cancer immunotherapy is to disrupt key pathways regulating immune tolerance,such as program death-1(PD-1/PD-L1)pathway in the tumor ***,the determinants of response to anti-PD-1 monoclonal antibodies(mAbs)treatment remain incompletely *** murine models,PD-1 blockade alone fails to induce effective immune responses to poorly immunogenic tumors,but is successful when combined with additional interventions,such as cancer *** cancer vaccines combined with antibody may offer promising control of cancer development and *** this investigation,we generated a novel tumor cell vaccine simultaneously expressing anti-PD-1 mAbs and granulocyte-macrophage colony stimulating factor(GM-CSF)in CT26 colon cancer and B16-F10 *** antitumor effect of the vaccine was verified by therapeutic and adoptive animal experiments in *** antitumor mechanism was analyzed using Flow cytometry,Elispot and in vivo intervention *** results showed that tumor cell vaccine secreting PD-1 neutralizing antibodies and GM-CSF induced remarkable antitumor immune effects and prolonged the survival of tumor-bearing animals compared with animals treated with either PD-1 mAbs or GM-CSF *** effects and prolonged survival correlated with strong antigen-specific T-cell responses by analyzing CD11c^(+)CD86^(+)DC,CD11b^(+)F4/80^(+)MΦcells,increased ratio of Teff/Treg in the tumor microenvironment,and higher secretion levels of Th1 proinflammatory cytokines in ***,the results of ELISPOT and in vivo blocking strategies further confirmed that the antitumor immune response is acquired by CD4 and CD8 T immune responses,primarily dependent on CD4 Th1 immune response,not NK innate immune *** combination of PD-1 blockade with GM-CSF secretion potency creates a novel

近年来,随着对肿瘤抗原和抗肿瘤免疫反应机制的深入认识,肿瘤治疗性疫苗发展迅速,有望成为临床肿瘤治疗的重要手段。同时,随着新抗原的筛选、疫苗设计、疫苗递送系统、佐剂等关键技术的突破,进一步加速了这一领域的发展。在产品研发方面,众多国际大型的医药企业和新兴的生物科技公司正在布局不同的肿瘤治疗性疫苗项目,多个肿瘤治疗性疫苗获批上市,但临床效果欠佳。尽管目前肿瘤治疗性疫苗大多处于临床前和临床试验阶段,但展现出了良好的临床应用前景和市场价值。本文论述了国内外肿瘤治疗性疫苗的研发现状(主要聚焦当前发展迅速的个性化新抗原疫苗、DC疫苗和mRNA疫苗),总结了目前所面临的挑战并展望了其发展前景,为未来肿瘤治疗性疫苗的研究和产品研发提供了新思路。

To solve the challenges of connecting and coordinating multiple platforms in the automotive industry and to enhance collaboration among different participants,this research focuses on addressing the complex supply relationships in the automotive market,improving data sharing and interactions across various platforms,and achieving more detailed integration of data and *** propose a trust evaluation permission delegation method based on the automotive industry *** proposed method combines smart contracts with trust evaluation mechanisms,dynamically calculating the trust value of users based on the historical behavior of the delegated entity,network environment,and other factors to avoid malicious node attacks during the permission delegation *** also introduce strict control over the cross-domain permission granting and revocation mechanisms to manage the delegation path,prevent information leakage caused by malicious node interception,and effectively protect data integrity and *** analysis shows that this method meets the realtime requirements of collaborative interaction in the automotive industry chain and provides a feasible solution to permission delegation issues in the automotive industry chain,offering dynamic flexibility in authorization and scalability compared to most existing solutions.
