Secondary cell walls(SCWs)in stem cells provide mechanical strength and structural support for *** thickening varies under different light *** previous study revealed that blue light enhances SCW thickening through the redundant function of MYC2 and MYC4 directed by CRYPTOCHROME1(CRY1)signaling in fiber cells of the Arabidopsis inflorescence *** this study,we find that the Arabidopsis PHYTOCHROME B mutant phyB displays thinner SCWs in stem fibers,but thicker SCWs are deposited in the PHYTOCHROME INTERACTING FACTOR(PIF)quadruple mutant pif1pif3pif4pif5(pifq).The shaded light condition with a low ratio of red to far-red light inhibits stem SCW ***4 interacts with MYC2 and MYC4 to affect their localization in nuclei,and this interaction results in inhibition of the MYCs’transactivation activity on the NST1 *** evidence shows that regulation of SCW thickening by PIFs is dependent on MYC2/MYC4 ***,the results of this study reveal a PHYB-PIF4-MYC2/MYC4 module that inhibits SCW thickening in fiber cells of the Arabidopsis stem.

Cryptochromes(CRY) are blue light photoreceptors that mediate a variety of blue light-induced responses in plants and animals. Cryptochromes typically have an N-terminal PHR(photolyase related) domain that shares sequence similarity with photolyase, a family of flavoproteins that catalyzes the repair of UV light-damaged DNA, and a distinguishing C-terminal domain that is absent in photolyase and has no strong sequence similarity with any known protein domain. It is demonstrated that one of the mechanisms by which Arabidopsis CRY regulates light responses is via its physical interaction with COP1, a RING motif–containing E3 ligase, which interacts with the bZIP transcriptional factor HY5, and promotes its ubiquitination and degradation. We have characterized both the biochemical and genetic interactions between CRY1 and SPA1. Both CRY1 and CRY2 physically interact with SPA1 in yeast cells in a blue light-dependent manner, colocalize in nuclear bodies(NBs) in vivo, and CRY1 associates with SPA1 in plant cells specifically under blue light. Furthermore, the blue light-induced CRY1–SPA1 interaction promotes the dissociation of COP1 from SPA1 in both yeast and plant cells. These results indicate that blue light-triggered CRY1–SPA1 interaction may negatively regulate COP1, at least in part, by promoting the dissociation of COP1 from SPA1. This interaction and consequent dissociation defines a dynamic photosensory signalling mechanism. We have also characterized the signalling mechanism by which CRY regulates photoperiodic flowering. Under long days(LDs), the transcription of FLOWERING LOCUS T(FT) is activated by CONSTANS(CO) in leaf, and the FT protein moves to the shoot apex to induce flowering. CO protein is degraded in darkness, whereas it is stabilized by the CRY-mediated signal. We demonstrate that CO acts genetically downstream of COP1 and CRY to regulate photoperiodic flowering. 研究目的:铁路隧道作为重要的铁路设施,在长期运营期间可能发生形变灾害而影响安全,为杜绝此类灾害发生,隧道限界侵限检测是隧道检测的一项重要内容。随着我国铁路隧道里程不断增加,提高隧道限界侵限检测效率成为提升铁路工务部门工作质量的一项工作,高效快捷的检测方法必不可少。基于此,本文重点研究基于激光扫描的隧道限界侵限检测技术,旨在利用激光扫描能密集、连续地自动获取大量数据的特点,完成隧道限界侵限的检测及侵限值的计算。研究结论:(1)激光扫描技术可以有效地采集铁路隧道内壁轮廓,作为隧道限界侵限的检测依据,进而判断是否存在限界侵限问题;(2)试验证明,利用激光扫描方法检测隧道内壁轮廓的测量精度达到±4 mm;(3)本研究成果可以有效针对铁路隧道限界侵限问题进行检测,用于指导维修养护工作,可为铁路的安全运营提供帮助。
