Characteristic analysis of skin keratinocytes in patients with type 2 diabetes based on the single-cell levels

Background: Keratinocytes play an important role in wound healing;however, less is known about skin keratinocytes in patients with type 2 diabetes mellitus (T2DM). Therefore, this study aimed to search for the transcriptional characteristics of keratinocytes at the single-cell level from T2DM patients, and to provide experimental data for identifying the pathological mechanisms of keratinocytes under pathological ***: We performed single-cell RNA sequencing on the skin tissue from two T2DM patients and one patient without diabetes-induced trauma using the BD Rhapsody^(TM) Single-Cell Analysis System. With the help of bioinformatics R-based single-cell analysis software, we analyzed the results of single-cell sequencing to identify the single-cell subsets and transcriptional characteristics of keratinocytes at the single-cell level, including Kyoto Encyclopedia of Genes and Genomes and Gene Ontology ***: In this study, we found specific highly expressed signature keratinocyte-related genes. We analyzed the transcriptome of keratinocytes from experimental and control groups and screened a total of 356 differential genes, which were subject to bioinformatics analysis. Enriched pathways included oxidative phosphorylation, antigen processing and presentation, prion and Huntingtons’ diseases, bacterial invasion of epithelial cells, thermogenesis, vasopressin-regulated water reabsorption, and protein processing in the endoplasmic ***: This study revealed the characteristics of keratinocytes at the single-cell level and screened a group of differentially expressed genes related to T2DM-associated keratinocytes, oxidative phosphorylation, cytokine receptor interactions, prion diseases, and other signaling pathways.

The Persistence and Zonal Scale of Atmospheric Dipolar Modes

This study investigates the relationship between the persistence and the zonal scale of atmospheric dipolar modes(DMs). Results from the daily data of ERA5 and the long-term output of an idealized atmospheric model show that the atmospheric DMs with a broader(narrower) zonal scale dipolar structure possess a longer(shorter) persistence. A detailed vorticity budget analysis indicates that the persistence of a hemispheric-scale DM(1/1 DM) and a regional or sectoral DM(1/8 DM) in the model both largely rely on the persistence of the nonlinear eddy forcing. Linear terms can indirectly reduce the persistence of the anomalous nonlinear eddy forcing in a 1/8 DM by modifying the baroclinicity via the arousal of anomalous vertical motions. Therefore, the atmospheric DMs with a broader(narrower) zonal scale possess a longer(shorter) persistence because the effects of the linear terms are less(more) pronounced when the atmospheric DMs have better(worse) zonal symmetry. Further analyses show that the positive eddy feedback effect is weak or even absent in a 1/8DM and the high-frequency eddy forcing acts more like a concomitant phenomenon rather than a leading driving factor for a 1/8 DM. Thus, the hemispheric-scale DM and the regional or sectoral DMs are different, not only in their persistence but also in their dynamics.

The burgeoning demand for modern electronic devices and electric vehicles has driven the development of efficient,reliable,and environmentally friendly batteries[1,2].Lithium–sulfur(Li–S)batteries with high theoretical capacity(1672 mA h g1)and energy density(2600 W h kg1),have garnered significant interest in both academic and industrial research[3,4].However,the widespread production and commercialization of Li–S batteries are impeded by the inherent characteristics of sulfur and lithium,along with their complex electrochemical ***,challenges such as the polysulfide shuttle effect and the slow kinetics of the sulfur nucleation/decomposition reaction hinder capacity utilization and cycling *** overcome these challenges,innovative electrocatalyst strategies aimed at enhancing activity have been explored.

The learning curve for transurethral enucleation with bipolar energy for benign prostate hyperplasia:a single-surgeon experience of 494 patients

This study was performed to investigate the learning curve of transurethral enucleation with bipolar energy(TUEB)for benignprostatic *** study involved 494 consecutive patients who underwent TUEB for benign prostatic hyperplasia fromAugust 2018 to March 2022 by one surgeon(SJJ,Seoul National University Bundang Hospital,Seongnam,Korea).The patientswere followed up at 1 week,1 month,3 months,and 6 months *** evaluate the learning curve of TUEB,perioperativeparameters including the enucleation ratio(enucleated tissue weight/transitional zone volume),TUEB efficiency(enucleatedtissue weight/operation time),and enucleation efficiency(enucleated tissue weight/enucleation time)were *** and postoperative complications were also assessed,including the International Prostate Symptom Score(IPSS),IPSSquality-of-life(QoL)score,and uroflowmetry *** patients’median age was 72(interquartile range[IQR]:66–78)years,and the estimated prostate volume and transitional zone volume were 63.0(IQR:46.0–90.6)ml and 37.1(IQR:24.0–60.0)ml,*** enucleation ratio,TUEB efficiency,and enucleation efficiency were 0.60(IQR:0.46–0.54)g ml−1,0.33(IQR:0.22–0.46)g min−1,and 0.50(IQR:0.35–0.72)g min−1,respectively,plateauing after 70 *** functional outcomes,including total IPSS,IPSS QoL score,and uroflowmetry outcomes,significantly improved at 6 months after TUEB(all P<0.05),but without significant differences over the learning ***-five(13.2%)patients developed complications after TUEB,21.5%of whom experienced major complications(Clavien–Dindo grade≥3).The rate of major complications declined as the number ofTUEB cases increased(P=0.013).Our results suggest that the efficiency of TUEB stabilized within 70 procedures.

Ulva prolifera is the causative species of the annually occurring large-scale green tides in China since *** specific biological features on reproductivity strategies,as well as intra-species genetic diversity,are still largely unknown,especially at the genome level,despite their importance in understanding the formation and outbreak of massive green *** the present study,the restriction site-associated DNA genotyping approach(2b-RAD)was adopted to identify the genome-wide single-nucleotide polymorphisms(SNPs)of 54 individual thalli including samples collected from Subei Shoal in 2019 and Qingdao coast from 2019 to *** genotype results revealed that most of the thalli in 2019 and 2020 were haploid gametophytes,while only half of the thalli were gametophytes in 2021,indicating flexibility in the reproductive strategies for the formation of the green tides among different years and the dominance of asexual and vegetative reproductive mode for the floating ***,population analysis was conducted,and it revealed a very low genetic diversity among samples from Subei Shoal and the Qingdao coast in the same year and a higher divergence among samples in different *** results showed the efficiency of 2b-RAD in the exploration of SNPs in *** and provided the first genome-wide scale evidence for the origin of the large-scale green tides on the Qingdao *** study improved our understanding of the reproductive strategy and genetic diversity of the green tide causative species and will help further reveal the biological causes of the green tide in China.
