Fritillaria cirrhosa D.Don(Liliaceae)is an endangered perennial bulbous plant and its dry bulb is a valuable med-icinal material with antitussive and expectorant effects.Nevertheless,lack of resources and expensive prices make it difﬁcult to meet clinical needs.This study presents a regeneration system aimed at overcoming the challenge of inadequate supply in F.cirrhosa,focusing on:(1)callus induction,(2)bulblets and adventitious bud induction,and(3)artiﬁcial seed production.Callus development was achieved in 84.93%on Murashige and Skoog(MS)medium fortiﬁed with 1.0 mg·L^(-1) picloram.The optimal medium for callus differentiation into regenerated bulb-lets was MS medium supplemented with 1.0 mg·L^(-1)6-benzyladenine(6-BA)and 0.2 mg·L^(-1)α-naphthaleneacetic acid(NAA).Subsequently,bulblets and adventitious buds were induced from regenerated bulblet sections cul-tured on MS medium fortiﬁed with 0.3 mg·L^(-1)6-BA+1.0 mg·L^(-1)2,4-dichlorophenoxyacetic acid(2,4-D),2.0 mg·L^(-1)6-BA+0.5 mg·L^(-1),and the induction rates were 88.17%and 84.24%,respectively.The regenerated bulblets were transplanted into a substrate of humus soil,river sand,and pearlite(1:1:1)after low-temperature treatment.The germination rate was 42.80%after culture for 30 days.Regenerated bulblets were used for encap-sulations in liquid MS medium containing 3%sucrose(w/v)+0.5 mg·L^(-1) NAA+2.0 mg·L^(-1)6-BA+3%sodium alginate(w/v)with a 10 min exposure to 2%CaCl_(2).Under non-aseptic conditions,the germination rate reached 81.67%,while the rooting rate was 20.56%after 45 days.The capsule added 1.0 g·L^(-1) carbendazim and 1.0 g·L^(-1) activated carbon was the best component of artiﬁcial seeds.This study successfully established an efﬁcient regen-eration system for the rapid propagation of F.cirrhosa,involving in vitro bulblet regeneration and artiﬁcial seed production.This method introduces a novel approach for efﬁcient breeding and germplasm preservation,making it suitable for large-scale industrial resource production.

Immune checkpoint inhibitors(ICIs)therapy targeting programmed cell death ligand 1(PD-L1)and programmed death protein 1(PD-1)had exhibited significant clinical benefits for cancer treatment such as triple negative breast cancer(TNBC).However,the relatively low anti-tumor immune response rate and ICIs drug resistance highlight the necessity of developing ICIs combination therapy strategies to improve the anti-tumor effect of immunotherapy.Herein,the immunomodulator epigallocatechin gallate palmitate(PEGCG)and the immunoadjuvant metformin(MET)self-assembled into tumor-targeted micelles via hydrogen bond and electrostatic interaction,which encapsulated the therapeutic agents doxorubicin(DOX)-loaded PEGCG-MET micelles(PMD)and combined with ICIs(anti-PD-1 antibody)as therapeutic strategy to reduce the endogenous expression of PD-L1 and improve the tumor immunosuppressive microenvironment.The results presented that PMD integrated chemotherapy and immunotherapy to enhance antitumor efficacy in vitro and in vivo,compared with DOX or anti-PD-1 antibody for the therapy of TNBC.PMD micelles might be a potential candidate,which could remedy the shortcomings of antibody-based ICIs and provide synergistic effect to enhance the antitumor effects of ICIs in tumor therapy.

We developed a magnetic solid-phase extraction method using Fe_(3)O_(4)@polydopamine(Fe_(3)O_(4)@PDA)as adsorbents,coupled with liquid chromatography-tandem mass spectrometry(LC-MS/MS),for the enrichment and quantification of trace matrine and oxymatrine pesticide residues in tea.The adsorbent was easily synthesized at room temperature with high yields(500 mg).We investigated and optimized several factors that influence the efficiency of the method,including matrix pH,adsorbent amount,adsorption time,type of elution solvent.

Dear Editor,Traffic driving is a dynamic and complicated task in which drivers are required to pay close attention to the important targets or regions to maintain safe margins.Rainy weather conditions make it more challenging with factors such as low visibility,raindrops,pedestrian with umbrellas,wipers,etc.Studies showed that rainy condition affects driving safety significantly[1],[2].It is reported that,in raining weather condition,the odds for a fatal accident are 3.340 times higher on highways than on streets[3].An investigation of the relationship between rainfall and fatal crashes in Texas from 1994 to 2018 on fatality analysis reporting system(FARS)database illustrated that rain-related fatal crashes represented about 6.8%of the total fatal crashes on average,moreover,the proportion showed high variability at the annual,monthly,and hourly time scales[4].Therefore,raining is a complex and critical factor for road safety planning and management.In fact,the traffic environment is a dynamic scene with multiple sources of information,including important targets that are highly relevant to the current driving task as well as irrelevant targets that may distract the driving task[5].

A naturally derived small molecule compound suppresses tumor growth and metastasis in mice by relieving p53-dependent repression of CDK2/Rb signaling and the Snail-driven EMT

The tumor suppressor protein p53 is central to cancer biology,with its pathway reactivation emerging as a promising therapeutic strategy in oncology.This study introduced LZ22,a novel compound that selectively inhibits the growth,migration,and metastasis of tumor cells expressing wild-type p53,demonstrating ineffectiveness in cells devoid of p53 or those expressing mutant p53.LZ22’s mechanism of action involves a high-affinity interaction with the histidine-96 pocket of the MDM2 protein.This interaction disrupted the MDM2-p53 binding,consequently stabilizing p53 by shielding it from proteasomal degradation.LZ22 impeded cell cycle progression and diminished cell proliferation by reinstating the p53-dependent suppression of the CDK2/Rb signaling pathway.Moreover,LZ22 alleviated the p53-dependent repression of Snail transcription factor expression and its consequent EMT,effectively reducing tumor cell migration and distal metastasis.Importantly,LZ22 administration in tumor-bearing mice did not manifest notable side effects.The findings position LZ22 as a structurally unique reactivator of p53,offering therapeutic promise for the management of human cancers with wild-type TP53.

Morphological alterations in dendritic spines have been linked to changes in functional communication between neurons that affect learning and memory.Kinesin-4 KIF21A helps organize the microtubule-actin network at the cell cortex by interacting with KANK1;however,whether KIF21A modulates dendritic structure and function in neurons remains unknown.In this study,we found that KIF21A was distributed in a subset of dendritic spines,and that these KIF21A-positive spines were larger and more structurally plastic than KIF21A-negative spines.Furthermore,the interaction between KIF21A and KANK1 was found to be critical for dendritic spine morphogenesis and synaptic plasticity.Knockdown of either KIF21A or KANK1 inhibited dendritic spine morphogenesis and dendritic branching,and these deficits were fully rescued by coexpressing full-length KIF21A or KANK1,but not by proteins with mutations disrupting direct binding between KIF21A and KANK1 or binding between KANK1 and talin1.Knocking down KIF21A in the hippocampus of rats inhibited the amplitudes of long-term potentiation induced by high-frequency stimulation and negatively impacted the animals’cognitive abilities.Taken together,our findings demonstrate the function of KIF21A in modulating spine morphology and provide insight into its role in synaptic function.

在以多维性为贫困重要属性的后脱贫时代,高等教育减贫相较于其他扶贫方式具有显著优势,因此有必要对高等教育减贫效果进行评估。本文基于2018年中国家庭追踪调查数据,利用各省扩招政策强度差异和样本出生队列信息构造双重差分模型,考察高校扩招对个体陷入多维相对贫困概率的影响。研究发现,高等教育减贫的效果显著,高校扩招后直接受到政策影响的队列陷入多维相对贫困的概率降低17.6%。在改变关键变量度量方式后,研究结果显示高等教育减贫效果依然稳健。高校扩招过程中为个体提供的高等教育机会提高了个体的非认知能力、非农就业的概率和社会资本,从而降低个体陷入多维相对贫困的概率。但是,高校扩招的减贫效应在不同群体之间存在差异,高校扩招对非农村人口、女性、父辈教育水平高的人群的减贫效应更大。因此,需更加注重不同群体在获得高质量高等教育方面的公平,以更好地发挥高等教育的减贫效应。

Contextual influence on the tilt after-effect in foveal and parafoveal vision

A sensory stimulus can only be properly interpreted in light of the stimuli that surround it in space and time. The tilt illusion （TI） and tilt after-effect （TAE） provide good evidence that the perception of a target depends strongly on both its spatial and temporal context. In previous studies, the TI and TAE have typically been investigated separately, so little is known about their co-effects on visual perception and information processing mechanisms. Here, we considered the influence of the spatial context and the temporal effect together and asked how center- surround context affects the TAE in foveal and para- foveal vision. Our results showed that different center-surround spatial patterns significantly affected the TAE for both foveal and para-foveal vision. In the fovea, the TAE was mainly produced by central adaptive gratings. Cross-oriented surroundings significantly inhibited the TAE, and iso-oriented surroundings slightly facilitated it; surround inhibition was much stronger than surround facilitation. In the para-fovea, the TAE was mainly decided by the surrounding patches. Likewise, a cross-oriented central patch inhibited the TAE, and an iso-oriented one facilitated it, but there was no significant difference between inhibition and facilitation. Our findings demonstrated, at the perceptual level, that our visual system adopts different mechanisms to process consistent or inconsistent central-surround orientation information and that the unequalmagnitude of surround inhibition and facilitation is vitally important for the visual system to improve the detectability or discriminability of novel or incongruent stimuli.
