A novel PIK3CD C896T mutation detected in bilateral sudden sensorineural hearing loss using next generation sequencing:An indication of primary immunodeficiency

Objective:To investigate immune-related genetic background in bilateral sudden sensorineural hearing loss (SSNHL). Case report and methods: The case is a 45-year-old man presenting with a 7-year history of bilateral profound SSNHL. Blood biochemical testing demonstrated increased levels of total cholesterol (5.88 mmol/L). Tests for hepatitis B showed a positive antibody against the hepatitis B core antigen. Complement C3 was below the normal value, and complement C4 and IgG were in the lower range of normal values. CT images showed a normal inner ear and vestibular aqueduct but round window membranous ossification on both sides. A total number of 232 immune-associated genes were sequenced using the next generation sequencing technique. Results: Mutations were detected in 5 genes, including the phosphoinositide 3-kinase catalytic subunit delta (PIK3CD), caspase recruitment domain-containing protein 9 (CARD9), complement factor H-related (CFHR2), immunoglobulin lambda-like polypeptide 1 Protein (IGLL1), and transmembrane channel-like gene family 8 (TMC8). In the PIK3CD gene, a C896T substitute in exon 7 was detected. This mutation causes primary immunodeficiency and is an autosomal dominant disease. Conclusion: The PIK3CD C896T mutation responsible for primary immunodeficiency may contribute to the onset of bilateral SSNHL with subsequent rapid progression.

This study was to investigate the structure and rat fecal microbial fermentation properties of a polysaccharide fraction(PHP2)isolated from the red marine alga Porphyra haitanensis.PHP2 was characterized as a sulfated glucogalactan,with a hypothetical backbone structure of→4)Gα(1→6)G4 Sβ(1→4)Glc(1→and a side chain of Man(1→6)Glc.PHP2 had an irregular spherical chain conformation.The 16 S r RNA sequence analysis revealed that PHP2 modulated the rat fecal micro-flora composition,with a similar effect to inulin,changing the dominant genus(Lactobacillus and Escherichia-Shigella)and promoting the growth of organisms that degrade sulfur-containing polysaccharides,such as Desulfovibrio,Ruminococcaceae_UCG-005,and Ruminococcus_2.PHP2 can promote production of acetic,propionic and butyric acid by rat fecal micro-flora.Prediction of metabolic function suggested that PHP2 could modulate cholesterol metabolism.The sulfated glucogalactan fermentation behavior may be associated with its monosaccharide composition,chain branching and chain conformation.PHP2 appeared to have considerable potential as functional food,and was associated with sulfur-containing polysaccharides in general.

The simultaneous high-capacity and fast removal of radioactive barium ion(Ba^(2+))from nuclear wastewater is necessary but still faces large challenge.In this study,a novel adsorbent containing sulfonic group,MOF-808-SO_(3)H,was prepared based on the oxidation of-SH group.Characterizations confirm the successful introduction of-SO_(3)H,good porosity,and stability of the material.The adsorbent exhibits excellent capture ability for Ba^(2+)due to strong hard acid-base and electrostatic interactions,with the adsorption capacity of 152.0 mg g^(-1)and fast equilibrium adsorption time,superior to other reported materials.Optimal pH was found to be almost neutral and temperature cannot affect the adsorption largely.Besides,the irreversible adsorption was demonstrated and thereby the secondary pollution may be avoided.Therefore,our work provides an efficient adsorbent for capturing Ba^(2+)irreversibly,and proposes a feasible strategy for constructing adsorbents with simultaneous high adsorption capacity and fast kinetics.

Effect of polysaccharide fractions from Fortunella margarita on the fecal microbiota of mice and SCFA production in vitro

Fortunella margarita polysaccharides(FMPS)are composed of 4 polysaccharide fragments,namely mannogalactoglucan,galactoglucan,glucan and arabinoglucan.This study investigated the in vitro fermentation of FMPS fractions and their effect on the fecal microbiota of mice and short chain fatty acids(SCFAs)production.The results showed that the structure of the fecal microbiota was changed after FMPS fermentation.At the phylum level,the polysaccharide fractions reduced the relative abundance of Bacteroidetes compared with the inulin group.At the genus level,the polysaccharide fractions increased the abundance of Lactobacillus,and decreased the abundance of Granulicatella,Proteus and Rummeliibacillus.Polysaccharide fractions could promote the proliferation of beneficial bacteria(Lactobacillus),and decreased harmful bacteria(Granulicatella,Proteus,Rummeliibacillus and Serratia).Furthermore,there was a significant distinction in the genus level flora processed by different polysaccharide fractions,especially mannogalactoglucan.Polysaccharide fractions showed potential prebiotic effects,with mannogalactoglucan,in particular,promoting proliferation of SCFA-producing bacteria.SCFAs were regulated by monosaccharide composition,molecular weight of the FMPS fractions,and the composition of the fecal bacteria.Polysaccharide fractions could modulate metabolic function of the fecal microbiota,altering the levels of SCFAs.These results suggested that FMPS are important functional components in gut health,especially mannogalactoglucan.

Calculation of oil saturation in clay-rich shale reservoirs:A case study of Qing 1 Member of Cretaceous Qingshankou Formation in Gulong Sag,Songliao Basin,NE China

The targeted reservoir,which is referred as the first member of Cretaceous Qingshankou Formation in Gulong Sag,Songliao Basin,NE China,is characterized by the enrichment of clay and lamellation fractures.Aiming at the technical challenge of determining oil saturation of such reservoir,nano-pores were accurately described and located through focused ion beam scanning electron microscopy and quantitative evaluation of minerals by scanning electron microscopy based on Simandoux model,to construct a 4D digital core frame.Electrical parameters of the shale reservoir were determined by finite element simulation,and the oil saturation calculation method suitable for shale was proposed.Comparison between the results from this method with that from real core test and 2D nuclear magnetic log shows that the absolute errors meet the requirements of the current reserve specification in China for clay-rich shale reservoir.Comparison analysis of multiple wells shows that the oil saturation values calculated by this method of several points vertically in single wells and multiple wells on the plane are in agreement with the test results of core samples and the regional deposition pattern,proving the accuracy and applicability of the method model.

A Novel Tele-Operated Flexible Robot Targeted for Minimally Invasive Robotic Surgery

In this paper, a novel flexible robot system with a constrained tendon-driven serpentine manipulator(CTSM) is presented. The CTSM gives the robot a larger workspace, more dexterous manipulation, and controllable stiffness compared with the da Vinci surgical robot and traditional flexible robots. The robot is tele-operated using the Novint Falcon haptic device. Two control modes are implemented, direct mapping and incremental mode. In each mode, the robot can be manipulated using either the highest stiffness scheme or the minimal movement scheme. The advantages of the CTSM are shown by simulation and experimental results.

Air pollutant emissions represent a critical challenge in the green development of the non-ferrous metallurgy industry.This work studied the emission characteristics,formation mechanisms,phase transformation and separation of typical air pollutants,such as heavy metal particles,mercury,sulfur oxides and fluoride,during non-ferrous smelting.A series of purification technologies,including optimization of the furnace throat and hightemperature discharge,were developed to collaboratively control and recover fine particles from the flue gas of heavy metal smelting processes,including copper,lead and zinc.Significant improvements have been realized in wet scrubbing technology for removing mercury,fluoride and SO_(2)from flue gas.Gas-liquid sulfidation technology by applying H_(2)S was invented to recycle the acid scrubbing wastewater more efficiently and in an eco-friendly manner.Based on digital technology,a source reduction method was designed for sulfur and fluoride control during the whole aluminum electrolysis process.New desulfurization technologies were developed for catalytic reduction of the sulfur content in petroleum coke at low temperature and catalytic reduction of SO_(2)to elemental sulfur.This work has established the technology for coupling multi-pollutant control and resource recovery from the flue gas from non-ferrous metallurgy,which provides the scientific theoretical basis and application technology for the treatment of air pollutants in the non-ferrous metallurgy industry.

Elite sd1 alleles in japonica rice and their breeding applications in northeast China

The Green Revolution gene sd1 has been used extensively in modern rice breeding,especially in indica cultivars.However,elite sd1 alleles and related germplasm resources used for japonica rice breeding have not been identified,and extensive efforts are needed for japonica rice breeding to obtain new dwarfing sources.Data from MBKbase-Rice revealed seven sd1 haplotypes in indica and four in japonica rice.Two new sd1 alleles were identified in indica rice.In 295 japonica accessions from northeast Asia,except for the weak functional allele SD1-EQ,sd1-r was the major allele,reducing plant height in comparison with SD1-EQ.Japonica germplasm resources carrying reported sd1 alleles were identified by genotype searching and further verified by literature search,genealogical analysis,and d Caps markers.Pedigrees and geographic distribution showed that sd1-r is an excellent allele widely used in northern China and Tohoku in Japan,and sd1-j is commonly used in east China and Kyushu in Japan.Dongnong-and Xiushui-series cultivars carrying sd1-r and sd1-j,respectively,are essential branches of the backbone parents of Chinese japonica rice,Akihikari and Ce21,with the largest number of descendants and derived generations.In semi-dwarf japonica rice breeding,sd1-d was introgressed into Daohuaxiang 2(DHX2).Dwarf and semi-dwarf lines carrying sd1-d were selected and designated as 1279 and 1280,respectively,after withstanding typhoon-induced strong winds and heavy rains in 2020,and are anticipated to become useful intermediate materials for future genetic research and breeding.This work will facilitate the introduction,parental selection,and marker-assisted breeding,and provide a material basis for the next step in identifying favorable genes that selected together with the sd1 alleles in japonica backbone parents.

Phenotype and Function of Monocyte-Derived Dendritic Cells from Chinese Rhesus Macaques

Dendritic cells （DCs） play a pivotal role in linking the innate immunity and acquired immunity in responses to pathogen. Non-human primates such as Chinese Rhesus Macaque （CRM） are the favorable models for preclinical study of potential therapeutic drugs, vaccines and mechanisms of human diseases. However, the phenotypical characterization of monocyte-derived dendritic cells （MDDCs） from CRM has not been elucidated. Monocytes from CRM were cultured with GM-CSF and IL-4 in RPMI-1640. Six days later, these cells were differentiated with typical dendritical morphology. CDllc and DC-SIGN were highly expressed. The immature MDDCs expressed the low levels of CD25, CD80, CD83, moderate CD40, CD86, and high MHC. After stimulation, the mature MDDCs increased expression of mature molecules CD25 and CD83, co-stimulatory molecules such as CD80, CD86 and CD40, and kept a high level of MHC. The capacity of endocytosis decreased with maturation. The mature MDDCs have strong ability of inducing allogeneic T cell proliferation and producing IL-12. In conclusion, we have characterized the phenotype and ultimate function of MDDCs from CRM for the first time.
