Blockade of CD300A enhances the ability of human NK cells to lyse hematologic malignancies

Objective: The human cluster of differentiation(CD)300A, a type-I transmembrane protein with immunoreceptor tyrosine-based inhibitory motifs, was investigated as a potential immune checkpoint for human natural killer(NK) cells targeting hematologic malignancies(HMs).Methods: We implemented a stimulation system involving the CD300A ligand, phosphatidylserine(PS), exposed to the outer surface of malignant cells. Additionally, we utilized CD300A overexpression, a CD300A blocking system, and a xenotransplantation model to evaluate the impact of CD300A on NK cell efficacy against HMs in in vitro and in vivo settings. Furthermore, we explored the association between CD300A and HM progression in ***: Our findings indicated that PS hampers the function of NK cells. Increased CD300A expression inhibited HM lysis by NK cells. CD300A overexpression shortened the survival of HM-xenografted mice by impairing transplanted NK cells. Blocking PS–CD300A signals with antibodies significantly amplified the expression of lysis function-related proteins and effector cytokines in NK cells, thereby augmenting the ability to lyse HMs. Clinically, heightened CD300A expression correlated with shorter survival and an “exhausted” phenotype of intratumoral NK cells in patients with HMs or solid ***: These results propose CD300A as a potential target for invigorating NK cell-based treatments against HMs.

In underground engineering,the detection of structural cracks on tunnel surfaces stands as a pivotal task in ensuring the health and reliability of tunnel ***,the dim and dusty environment inherent to under-ground engineering poses considerable challenges to crack *** paper proposes a crack segmentation algorithm termed as Focused Detection for Subsurface Cracks YOLOv8(FDSC-YOLOv8)specifically designed for underground engineering structural ***,to improve the extraction of multi-layer convolutional features,the fixed convolutional module is replaced with a deformable convolutional ***,the model’s receptive field is enhanced by introducing a multi-branch convolutional module,improving the extraction of shallow features for small ***,the Dynamic Snake Convolution module is incorporated to enhance the extraction capability for slender and weak ***,the Convolutional Block Attention Module(CBAM)module is employed to achieve better target *** FDSC-YOLOv8s algorithm’s maP50 and maP50-95 reach 96.5%and 66.4%,according to the testing data.

Although lymphocytes are known to circulate throughout lymphoid tissues and blood,they also establish residency in nonlymphoid organs,most prominently in barrier tissues,such as the *** adaptation of T lymphocytes to intestinal environments requires constant discrimination between natural stimulation from commensal flora and food and pathogens that need to be *** variations that cause a defective defense or a break in tolerance along with environmental cues,such as infection or imbalances in the gut microbiota known as dysbiosis,can trigger several immune disorders via the activation of T lymphocytes in the *** of the immune mechanisms that distinguish between commensal flora and pathogenic organisms may reveal therapeutic targets for the prevention or modulation of inflammatory diseases and boost the efficacy of cancer *** this review,we discuss the development and adaptation of T lymphocytes in the intestine,how these cells protect the host against pathogenic infections while tolerating food antigens and commensal microbiota,and the potential implications of targeting these cells for disease management and therapeutics.

Background/Aim: Primary biliary cirrhosis(PBC) is a typical autoimmune liver disease with a high occurrence in middle-aged women. Histologically, it is characterized by progressive bile duct destruction, with portal lymphocyte infiltration, granulomas, and liver fibrosis. Serologically, 90%-95% PBC patients are positive for anti-mitochondrial antibodies. Clinically, most PBC patients would miss the most proper diagnosis time because of the asymptomatic stage at first. Accordingly, a comprehensive mechanism of PBC is in need urgently. However, patient samples are hard to get, researchers have been dedicated on making an ideal mouse model for easier investigation. Several mouse models are now used to clarify the pathogenesis of PBC but few centered on how the immune tolerance is broken down. Methods: we got a new mouse model of PBC by immunizing female mice with congenic bile duct protein. Results: We found congenic bile duct antigens would efficiently break down the immune tolerance of recipient mice, leading to a phenotype similar with PBC patients. First, our mouse model showed bile duct damage, with inflammation focused of portal tracts. Moreover, flow cytometry analysis demonstrated higher number of CD4+ and CD8+ T cells in the liver and spleen, and these T cells exhibited an activated phenotype. Furthermore, the IFN-γ secreting ability of these T cells was more potent in these immunized mice. Then, we found these mice had greater germinal center reaction ability. At last, we analyzed the anti-mitochondrial antibodies in these mice, and found they are 100% positive for Ama. Conclusion: we defined a novel mouse model of PBC, which would provide help in elucidating the detailed mechanism of this complex disease.

Autoimmune diseases often induce dysregulated hematopoiesis with altered number and function of hematopoietic stem and progenitor cells(HSPCs).However,there are limited studies on the direct regulation of HSPCs on T cells,which are often detrimental to *** we found that in a murine model of Concanavalin A induced autoimmune hepatitis,LSK(Lineage’Sca-1c-Kit)-like cells accumulated in liver,spleen and bone marrow,which were myeloid progenitors(Lineage"Sca-1c-Kit) that upregulated Sca-1 expression upon T cell derived IFN-γ ***,bone marrow LSK-like cells from mice induced by Con A to develop autoimmune hepatitis or alternatively myeloid progenitors from wild type mice possessed strong in vitro suppressive *** suppressive function depended on T cell derived IFN-γ in a paracrine fashion,which induced STAT1 phosphorylation,inducible Nitric Oxide Synthase expression and nitric oxide *** IFN-γ/IFN-γ receptor interaction,knockout of STAT1 or iNOS inhibition abrogated their suppressive *** addition,the suppressive function was independent of differentiation;mitomycin C treated myeloid progenitors maintained T cell suppressive ability in *** data demonstrate a mechanism of inflammation induced suppressive function of myeloid progenitors which may participate directly in suppressing T cell mediated immunopathology.

关于五水和三水硝酸铥的热分解机理,还未见文献报道,而且对于三水合硝酸铥的制备亦未曾报道。关于五水和四水硝酸铥的制备虽有文献报道,但不详细。本文采用TG—DTG法研究了五水和三水硝酸铥的热分解行为,较详细地研究了这两种化合物的制备及存在条件,并对它们进行了表征。通过IR对反应物,中间产物及最终产物进行了鉴别。还用Kissinger法计算了Tm(NO_3)_3·5 H_2O部分脱水过程的表观活化能值。

Effect of Compression on the Crystallization Behavior and Corrosion Resistance of Al_(86)Ni_9La_5 Amorphous Alloy

The effect of compression on the crystallization behavior and corrosion resistance of Al（86）Ni9La5 amorphous ribbons was investigated by X-ray diffraction （XRD）, transmission electron microscopy （TEM）, scanning elec-tron microscopy （SEM） and electrochemistry test. The XRD and TEM results reveal that the compressed Al（86）Ni9La5 ribbons spun with the circumferential speed （R） of 29.3 m/s are in fully amorphous state; however, the compressed ribbons spun with R=14.7 m/s have crystalline phases embedded in the amorphous matrix. The SEM images indicate that after compression, the toughness of the ribbons increases. Electrochemical results show that the compression decreases the stability of the passive film of the Al（86）Ni9La5 amorphous ribbons, because of the compression-introduced free volume, shear bands and crystalline phases; meanwhile, with R=14.7 m/s, the compression-induced crystalline phases in the Al（86）Ni9La5 ribbons increase the corrosion potential.

Crystallization and Corrosion Resistance of (Fe_(0.78)Si_(0.09)B_(0.13))100_(-x)Ni_x (x=0,2 and 5) Glassy Alloys

The crystallization of the （Fe0.78Si0.09B0.13）100-xNix glassy alloys （x=O, 2 and 5） has been investigated by X-ray diffraction （XRD） and differential scanning calorimeter （DSC）. Corrosion resistance analyses have been carried out using electrochemical measurements and corrosion products have been analyzed by scanning electron microscopy （SEM）. The experimental results show that the addition of Ni can promote the nucleation of c^-Fe, retard the decomposition of the metastable borides, and alter the crystallization mode of the present Fe-based glassy alloys. The lattice constant （ao^e） of c^-Fe in the annealed samples shows a decreasing trend with increasing annealing time. The Ni addition can improve the corrosion resistance of the as-quenched Fe-based glassy alloys in H2S04, NaCI and NaOH solutions. The results indicate that Ni can promote the diffusion of Si atoms during quenching and annealing processes.

Fabrication and properties of high quality InGaN-based LEDs with highly reflective nanoporous GaN mirrors

Distributed Bragg reflectors (DBRs) are essential components for the development of optoelectronic *** this paper, we first report the use of the nanoporous GaN (NP-GaN) DBR as a template for regrowth of InGaN-based light-emitting diodes (LEDs). The wafer-scale NP-GaN DBR, which is fabricated by electrochemical etching in a neutral solution, has a smooth surface, high reflectivity (>99.5%), and wide spectral stop band width (>70 nm). The chemical composition of the regrown LED thin film is similar to that of the reference LED, but the photoluminescence (PL) lifetime, PL intensity, and electroluminescence intensity of the LED with the DBR are enhanced several times compared to those of the reference LED. The intensity enhancement is attributed to the light reflection effect of the NP-GaN DBR and improved crystalline quality as a result of the etching scheme, whereas the enhancement of PL lifetime is attributable to the latter.
