BACKGROUND Gastrointestinal stromal tumors(GISTs)are the most common mesenchymal tumors of the gastrointestinal(GI)tract,and cases of GISTs tend to be of the disseminated type,with a global incidence of 10 to 15 cases/million each *** rarer familial GISTs,which often represent a population,differ in screening,diagnosis,and *** GISTs include primary familial GISTs with predominantly KIT/PDGFRA mutations and wild-type ***,whether the same genetic family has different phenotypes has not been *** SUMMARY We report two cases of rare GISTs in the same family:A male patient with the V561D mutation in exon 12 of the PDGFRA gene,who has been taking the targeted drug imatinib since undergoing surgery,and a female patient diagnosed with wild-type GIST,who has been taking imatinib for 3 years since undergoing *** favorable prognosis of these patients during the 7-year follow-up period validates the accuracy of our treatment strategy,and we have refined the entire process of diagnosis and treatment of familial GISTs in order to better manage this rare familial *** Different mutation types of familial GISTs in the same family are very rare,thus it is very important to make the correct diagnosis and treatment strategies according to the results of molecular detection for the management of familial GISTs.

Chlorajaponins A—Q,Lindenane-Related Sesquiterpenoid Dimers from Chloranthus japonicus and Their Biological Activities

Seventeen undescribed lindenane-related sesquiterpenoid dimers,chlorajaponins A—Q(1—17),and 13 reported analogs(18—30)were isolated from Chloranthus japonicus *** 1 possesses an unprecedented 3/5/7/5/5/6/5/3 fused octacyclic scaffold,featuring a 6(5→4)-abeo-lindenane monomer,while 2 exhibits a 3/5/6/6/5/6/5/3 fused octacyclic *** structures were determined through a combination of spectroscopic analyses and X-ray *** 1,2,and 18 demonstrated significant inhibitory effects on lipid accumulation and effectively reduced the levels of triglycerides and total cholesterol,as well as the levels of aspartate aminotransferase and alanine aminotransferase in a HepG2 cell *** addition,compounds 1,2,and 18 significantly suppressed the protein expression of the fatty acid synthase(FASN)and the sterol regulatory element-binding protein 1(SREBP1).Moreover,the anti-inflammatory assay showed that compounds 19—22 and 25 inhibited the NO production induced by lipopolysaccharide in RAW 264.7 macrophages with IC50 values of 7.89±0.44,6.25±0.46,2.98±0.29,10.77±0.60,and 3.60±0.28μmol/L.

Widespread and active piezotolerant microorganisms mediate phenolic compound degradation under high hydrostatic pressure in hadal trenches

Effect of different bottom blowing elements on stirring characteristics of molten bath in converter

The bottom blowing element is the key equipment to ensure the bottom blowing effect of the *** types of bottom blowing elements,dispersive type(D1),double circular seam(D2)and straight cylinder type(D3),were built,and the effects of bottom blowing element type on molten bath flow,wall erosion and furnace bottom erosion were *** was found that when the bottom blowing elements of dispersive type(D1)and double circular seam(D2)were used,the dead zone area in the lower part of the molten bath was smaller,and the high-speed zone area was larger;therefore,the stirring effect on the bottom melt was *** the straight cylinder type(D3)bottom blowing element was used,the gas penetrated the molten bath at a faster rate to reach the surface of molten bath and failed to disperse in the bottom molten bath,and the wall shear stress near the nozzle outlet was *** argon was blown by three different bottom blowing elements,the area of the wall shear stress greater than 3 Pa was 4.8,5.6 and 8.7 cm2,respectively,within 0.2 m of the bottom blowing nozzle outlet.

Measuring drug similarity using drug–drug interactions

Natural killer cell membrane doped supramolecular nanoplatform with immuno-modulatory functions for immuno-enhanced tumor phototherapy

Photodynamic therapy(PDT) has garnered significant attention as a promising approach to cancer therapy, harnessing the combined benefits of localized light treatment and the accompanying host immune response. In this study, we engineered an immuno-enhanced PDT nanoplatform, denoted as HM@p-MOF(hybrid membrane@porphyrin-metal organic framework). The core porphyrin-MOF was cloaked with a hybrid membrane derived from B16F10 cancer cells and NK cells, resulting in enhanced stability. In both in vitro and in vivo experiments, our finding demonstrated that the hybrid membrane conferred dual targeting capabilities to the nanoplatform, leveraging the unique properties of the B16F10 membrane and NK membrane to augment immunogenic cell death(ICD) induced by photodynamic effects. Additionally,in conjunction with the immunomodulatory functions of the NK cell membrane, we observed an expansion of in situ immune infiltration leading to a systemic immune response. The HM@p-MOF nanoplatform exhibited the capacity to not only inhibit the growth of mouse melanoma but also suppress metastasis. This innovative HM@p-MOF nanoplatform present a viable strategy to enhance phototherapeutic efficacy for both localized and metastatic tumors. It provides a direction for the fabrication of biomimetic nanomedicines possessing immuno-modulatory function.
