Trilogy of drug repurposing for developing cancer and chemotherapy-induced heart failure co-therapy agent

Chemotherapy-induced complications,particularly lethal cardiovascular diseases,pose significant challenges for cancer *** intertwined adverse effects,brought by cancer and its complication,further complicate anticancer therapy and lead to diminished clinical *** supplementation of cardioprotective agents falls short in addressing these *** bifunctional co-therapy agents provided another potential solution to consolidate the chemotherapy and reduce cardiac events *** repurposing was naturally endowed with co-therapeutic potential of two indications,implying a unique chance in the development of bi-functional ***,we further proposed a novel“trilogy of drug repurposing”strategy that comprises function-based,targetfocused,and scaffold-driven repurposing approaches,aiming to systematically elucidate the advantages of repurposed drugs in rationally developing bi-functional *** function-based repurposing,a cardioprotective agent,carvedilol(CAR),was identified as a potential neddylation inhibitor to suppress lung cancer *** target-focused SAR studies and scaffold-driven drug design,we synthesized 44 CAR derivatives to achieve a balance between anticancer and ***,optimal derivative 43 displayed promising bi-functional effects,especially in various self-established heart failure mice models with and without ***,the present study validated the practicability of the“trilogy of drug repurposing”strategy in the development of bi-functional cotherapy agents.

Spectrum analysis of interval process model and its application in uncertain vibration analysis

In recent years, the authors have extended the traditional interval method into the time dimension to develop a new mathematical tool called the “interval process model” for quantifying time-varying or dynamic uncertainties. This model employs upper and lower bounds instead of precise probability distributions to quantify uncertainty in a parameter at any given time point. It is anticipated to complement the conventional stochastic process model in the coming years owing to its relatively low dependence on experimental samples and ease of understanding for engineers. Building on our previous work, this paper proposes a spectrum analysis method to describe the frequency domain characteristics of an interval process, further strengthening the theoretical foundation of the interval process model and enhancing its applicability for complex engineering problems. In this approach, we first define the zero midpoint function interval process and its auto/cross-power spectral density(PSD) functions. We also deduce the relationship between the auto-PSD function and the auto-covariance function of the stationary zero midpoint function interval process. Next, the auto/cross-PSD function matrices of a general interval process are defined, followed by the introduction of the concepts of PSD function matrix and cross-PSD function matrix for interval process vectors. The spectrum analysis method is then applied to random vibration problems, leading to the creation of a spectrum-analysis-based interval vibration analysis method that determines the PSD function for the system displacement response under stationary interval process excitations. Finally, the effectiveness of the formulated spectrum-analysis-based interval vibration analysis approach is verified through two numerical examples.

A deep learning model enabled multi-event recognition for distributed optical fiber sensing

Fiber optic sensors that utilize backscattered light offer distributed real-time measurements and have been seen tremendous improvements in sensing distance and spatial resolution over the last ***, these improvements in sensor capabilities lead to a significant increase in the amount of data that needs to be processed. Traditional processing schemes are no longer adequate, so the development of novel signal processing methods is critical. Phase-sensitive optical time domain reflectometry(Φ-OTDR) is now applied in various applications for multi-event recognition, and it would usually be difficult, sometimes even unrealistic to label all the acquired samples due to its real-time and seamless monitoring nature. To fully take advantage of the information contained within the large number of unlabeled samples, which were formerly not utilized and hence wasted, we propose a semi-supervised model to boost the event classification performance of Φ-OTDR. The model extracts respectively the temporal features and the spatial bidirectional features together with a dual attention mechanism. Its classification accuracy has been improved up to 96.9%with only 1230 labeled samples. In addition, our model shows significant advantages when the number of labeled samples is reduced. Importantly, our method improves the accuracy of multi-event classification without any modification to the optical setup.

Full-field three-dimensional(3D)measurement technology based on phase information has become an indispensable part of geometric dimension measurement in modern scientific research and engineering *** field has been developing and evolving for the study of highly reflective phenomena,diffuse reflections,and specular surfaces,and many novel methods have emerged to increase the speed of measurements,enhance data accuracy,and broaden the robustness of the ***,we will discuss the latest research progress in full-field 3D shape measurement based on phase information systematically and ***,the fundamentals of 3D shape measurement based on phase information are introduced,namely,phase-shifting and transform-based ***,recent technological innovations are highlighted,including increases in measurement speed and automation and improvements in robustness in complex *** particular,the challenges faced by these technological advances in solving highly dynamic,composite surface measurement problems are presented,i.e.,with multiexposure techniques proposed for high dynamics that extend the dynamic range of the camera to reduce the effects of overexposure but increase the cost of time and have high hardware requirements,fringe adaptive techniques that overcome light variations but are computationally complex,and multipolarized camera techniques that reduce the effects of light variations but are sensitive to the light ***,the phase-shifting method combined with coding is proposed to improve the measurement speed,but the accuracy is slightly *** learning techniques are proposed to cope with measurements in complex environments,but the dataset computation process is ***,future research directions are suggested,and the challenges are ***,this work provides a reference for researchers and engineers.

Identification of the lysine and histidine transporter family in Camellia sinensis and the characterizations in nitrogen utilization

In plants,the lysine and histidine transporter(LHT)family represent a class of proteins that mediate the uptake,translocation,and utilization of amino *** tea plant(Camellia sinensis)is a perennial evergreen with a relatively high level of amino ***,systematic identification and molecular characterization of the LHT gene family has rarely been reported in tea *** this study,22 CsLHTs were identified from the‘Shuchazao’genome and classified into two *** modeled three-dimensional structure and the conserved domains presented a high similarity among the LHTs ***,it was predicted that a few genes were conserved through the analysis of the physiochemical characters,structures and cis-elements in *** expression patterns in tea plants revealed that CsLHT7 was mainly expressed in the roots,and CsLHT4 and CsLHT11 exhibited relatively high expression in both the roots and ***,the expression of all three genes could be induced by organic ***,heterogeneous expression of CsLHT4,CsLHT7 and CsLHT11 in Arabidopsis thaliana decreased the aerial parts biomass compared with that in WT plants while significantly increased the rosette biomass only for CsLHT11transgenic plants versus WT ***,our results provide fundamental information about CsLHTs and potential genes in N utilization for further analysis in tea plants.

Comparative experiments and theoretical analysis of the surface chemistry changes of goethite(GT)and goethite containing ni(GTN)in the lattice in the presence of salicylhydroxamic acid(SA)were *** was revealed that in the presence of 100 g·t^(-1)of SA,the flotation recovery of GTN and GT increased with increasing pH,achieving a maximum recovery of 98.9%for both minerals at p H 8.3 and decreasing beyond that pH,with GTN having a slightly higher recovery than GT,except at pH *** was further confirmed by the higher complexation energies of GTN···SA(-883.87 kJ·mol^(-1))compared with GT···SA(-604.23 kJ·mol^(-1))resulting from covalent,closed-shell,and conventional hydrogen *** higher adsorption of SA onto GTN relative to GT was due to the formation of aπ-hole in GTN,thereby promoting a higher interaction of the collector with the ***,the presence of ni in the GT lattice improves and decreases the adsorption and desorption of SA onto and from the mineral,respectively,compared with those onto and from GT.
