Electricity Production in Microbial Fuel Cell Subjected to Different Operational Modes

The effects of inoculum species, substrate concentration, temperature, and cathodic electron acceptors on electricity production of microbial fuel cells （MFCs） were investigated in terms of start-up time and power output. When inoculated with aeration tank sludge, this MFC outperformed the cell that was inoculated with anaerobic sludge in terms of start-up time and power output. After running for a certain time period, the dominant populations of the two MFCs varied significantly. Within the tested range of substrate concentration （200-1800 mg L-l）, the voltage output increased and the time span of the electricity generation lengthened with increasing substrate concentration. As the temperature declined from 35 to 10 ℃, the maximum power density reduced from 2.229 to 1.620 W m-3, and anodic polarization resistance correspondingly dropped from 118 to 98 Ω. The voltage output of MFC-Cu2＋ was 0.447 V, which is slightly lower than that achieved with MFC-[Fe（CN）6]3- （0.492 V）, thereby indicating that MFCs could be used to treat wastewater con- taining Cu2＋ pollutant in the cathode chamber with removal of organics in anode chamber and simultaneous electricity generation.

In this study, a ‘‘direct-starting'' procedure was used to activate a single-chamber biocathode microbial electrolysis cell(MEC) and the development of a biocathode was studied through output current curves and cyclic voltammograms. It only took 163 h for a successful start-up, and a current density of 14.75 A/m^2 was obtained. In the formal hydrogen-production stage, it was found that the biocathode MEC was comparable with the Pt/C cathode MEC in terms of current density and energy efficiency, and the hydrogen recovery, cathodic hydrogen recovery, and hydrogen production rate of the biocathode MEC were 71.22% ± 8.98%, 79.42% ± 5.94%, and 0.428 ± 0.054 m^3 H_2/m^3 days, respectively, which were slightly higher than those obtained with the Pt/C cathode MEC. Besides, under the effect of applied voltage, the microbial populations in the anodophilic biofilm of MEC(MECan) and the cathodophilic biofilm of MEC(MECca) were less diverse than those of the original aerobic activated sludge(AAS) and the anodophilic biofilm of MEC(MECan). Furthermore, the microbial community structures evidently differed between MECan/MECca and AAS/MFC.

The conserved domains of reverse transcriptase （RT） genes of Tyl-copia and Ty3-gypsy groups of long terminal repeat （LTR） retrotransposons were isolated from the Malus domestica genome using degenerate oligonucleotide primers. Sequence analysis showed that 45% of Ty1-copia and 63% of Ty3-gypsy RT sequences contained premature stop codons and/or indels disrupting the reading frame. High heterogeneity among RT sequences of both Ty1-copia and Ty3-gypsy group retrotransposons was observed, but Ty3-gypsy group retrotransposons in the apple genome are less heterogeneous than Ty1-copia elements. Retrotransposon copy number was estimated by dot blot hybridizations for Ty1-copia （-5000） and Ty3-gypsy （-26000）. All elements of the two types of LTR retrotransposons comprise approximately 38% of the M. domestica genome, with the Ty3-gypsy group contribution being higher （33.5%） than the Tyl.copia one （4.6%）. Transcription was not detected by reverse transcription-polymerase chain reaction for either Ty1-copia or Ty3-gypsy retrotransposons in the leaves of plants in vitro or in leaf explants cultured on medium supplemented with high concentration benzylaminopurine. This research reveals the differences in heterogeneity and copy number between Ty1-copia and Ty3-gypsy retrotransposons in the apple genome. Ty1-copia retrotransposon has higher heterogeneity than Ty3-gypsy retrotransposon, but the latter has a higher copy number, which implies that Ty3-gypsy retrotransposons may play a more important role in the apple genome evolution.

Background： Angiotensin II （Ang ll） is a major contributor to the development of heart failure. However, the molecular and cellular mechanisms that underlie this process remain elusive. Inadequate angiogenesis in the myocardium leads to a transition from cardiac hypertrophy to dysfunction, and our previous study showed that Ang II significantly impaired the angiogenesis response. The current study was designed to examine the role of Jaggedl-Notch signaling in the effect of Ang II during impaired angiogenesis and cardiac hypertrophy. Methods： Ang II was subcutaneously infused into 8-week-old male C57BL/6 mice at a dose of 200 ng·kg^-1·min^-1 for 2 weeks using Alzet micro-osmotic pumps. N-[N-（3, 5-difluorophenacetyl）-L-alanyl]-S-phenylglycine tert-butyl ester （DAPT）, a y-secretase inhibitor, was injected subcutaneously during Ang II infusion at a dose of 10.0 mg.kg^-1·d^-1. Forty mice were divided into four groups （n = 10 per group）： control group; Ang 11 group, treated with Ang I1; DAPT group, treated with DAPT; and Ang II ＋ DAPT group, treated with both Ang I1 and DAPT. At the end of experiments, myocardial （left ventricle [LV]） tissue from each experimental group was evaluated using immunohistochemistry, Western blotting, and real-time polymerase chain reaction, Data were analyzed using one-way analysis of variance test followed by the least significant difference method or independent samples t-test. Results： Ang II treatment significantly induced cardiac hypertrophy and impaired the angiogenesis response compared to controls, as shown by hematoxylin and eosin （HE） staining and immunohistochemistry fbr CD31, a vascular marker （P 〈 0.05 for both）. Meanwhile, Jaggedl protein was significantly increased, but gene expression for both Jagl and Heyl was decreased in the LV following Ang II treatment, compared to that in controls （relative ratio for Jag1 gene： 0.45 ± 0.13 vs. 0.84± 0.15; relative ratio for Heyl gene： 0.51 ± 0.08 vs. 0.91 ± 0.09; P 〈 0.05）. Objective: To investigate the correlation and clinical significance between the serum Fractalkine (FKN) and interleukin-6 (IL-6), C-reactive protein (CRP), BNP, CK, CK-MB, TC, TG, LDL-C, HDL-C levels in patients with coronary atherosclerotic heart disease (CHD). Methods: A total of 95 cases of patients with coronary heart disease were admitted to the Department of Cardiology, the First Affiliated Hospital of Xinjiang Medical University, March 2016 to December 2016, including 52 patients with stable angina pectoris (SAP) and 43 patients with Acute coronary syndrome (ACS), 45 matched healthy subjects were collected. The Serum levels of FKN were detected by ELISA. The correlation and significance of FKN and clinical indexes were analyzed. Results: (1) the levels of serum FKN, IL-6, CRP, BNP, CK, CK-MB, TC, TG, LDL-C in the SAP group and ACS group were significantly higher than those in the control group, HDL-C were significantly decreased. Compared with SAP group, serum FKN level was significantly increased and IL-6, CRP, BNP, CK and CK-MB levels were significantly increased in ACS group, TC, TG, LDL-C, HDL-C levels were not significant difference between SAP group and ACS group. (2) Correlation analyses showed that there was a significant positive correlation between FKN levels and IL-6, CRP, BNP, CK, CK-MB, LDL-C, significant Negative correlation between FKN levels and HDL-C, weakly positive correlation between FKN levels and TC, and no correlation between FKN levels and TG. Conclusions: The serum levels of FKN in patients with CHD are significantly increased, which are closely related to the inflammatory reaction and can be used as biomarker of CHD for early diagnosis and prognosis.

Mainstream partial nitritation-anammox(PNA)process easily suffers from performance instability and even reactor collapse in application.Thus,it is of great significance to unveil the characteristic of performance recovery,understand the intrinsic mechanism and then propose operational strategy.In this study,we combined long-term reactor operation,batch tests,and metagenomics to reveal the succession of microbial community and functional metabolism variation from system collapse to recovery.Proper aeration control(0.10-0.25mg O_(2)/L)was critical for performance recovery.It was also found that Candidatus Brocadia became the dominant flora and its abundance increased from 3.5%to 11.0%.Significant enhancements in carbon metabolism and phospholipid biosynthesis were observed during system recovery,and the genes abundance related to signal transduction was dramatically increased.The up-regulation of sdh and suc genes showed the processes of succinate dehydrogenation and succinyl-CoA synthesis might stimulate the production of amino acids and the synthesis of proteins,thereby possibly improving the activity and abundance of AnAOB,which was conducive to the performance recovery.Moreover,the increase in abundance of hzs and hdh genes suggested the enhancement of the anammox process.Changes in the abundance of key genes involved in nitrogen metabolism indicated that nitrogen removal pathway was more diverse after system recovery.The achievement of performance recovery was driven by anammox,nitrification and denitrification coupled with dissimilatory nitrate reduction to ammonium.These results provide deeper insights into the recovery mechanism of PNA system and also provide a potential regulation strategy for the stable operation of the mainstream PNA process.

The welding arc,as a carrier for the conversion of electrical energy to thermal energy,has a direct impact on the quality of welding by its properties and states.In the tungsten inert gas(TIG)welding process under the condition of Ar-He alternating gas supply,the arc is alternately converted between Ar arc and He arc with an alternating gas supply cycle,which has obvious arc change characteristics.The FLUENT software was used to numerically simulate the characteristics of the TIG arc under the condition of alternating gas supply,and the arc temperature field,arc pressure,electric potential and current density distribution under the condition of alternating gas supply were obtained.Combined with the real-time data of arc pressure measured by the water-cooled copper plate with holes,it is proved that the TIG arc has obvious dynamic characteristics under the condition of Ar-He alternating gas supply.This unique dynamic TIG arc acts on the 5A06 aluminum alloy weld,causing the molten pool to stir,resulting in uniform microstructure and grain refinement at the weld,and thereby improving the mechanical properties of the welded joint.

BACKGROUND Both N6-methyladenosine(m6A)methylation and autophagy are considered relevant to the pathogenesis of ulcerative colitis(UC).However,a systematic exploration of the role of the com-bination of m6A methylation and autophagy in UC remains to be performed.AIM To elucidate the autophagy-related genes of m6A with a diagnostic value for UC.METHODS The correlation between m6A-related genes and autophagy-related genes(ARGs)was analyzed.Finally,gene set enrichment analysis(GSEA)was performed on the characteristic genes.Additionally,the expression levels of four characteristic genes were verified in dextran sulfate sodium(DSS)-induced colitis in mice.RESULTS GSEA indicated that BAG3,P4HB and TP53INP2 were involved in the inflammatory response and TNF-αsignalling via nuclear factor kappa-B.Furthermore,polymerase chain reaction results showed significantly higher mRNA levels of BAG3 and P4HB and lower mRNA levels of FMR1 and TP53INP2 in the DSS group compared to the control group.CONCLUSION This study identified four m6A-ARGs that predict the occurrence of UC,thus providing a scientific reference for further studies on the pathogenesis of UC.
