BACKGROUND In vitro expansion to increase numbers of hematopoietic stem cells(HSCs)in cord blood could improve clinical efficacy of this vital resource.nicotinamide(NAM)can promote HSC expansion ex vivo,but its effect on hematopoietic stem and progenitor cells(HSPCs,CD34^(+)CD38)and functional subtypes of HSCs-shortterm repopulating HSCs(ST-HSCs,CD34^(+)CD38CD45RACD49f^(+))and long-term repopulating HSCs(LT-HSCs,CD34^(+)CD38CD45RACD49f^(+)CD90^(+))is not yet known.As a sirtuin 1(SIRT1)inhibitor,NAM participates in regulating cell adhesion,polarity,migration,proliferation,and differentiation.However,SIRT1 exhibits dual effects by promoting or inhibiting differentiation in different tissues or cells.We propose that the concentration of NAM may influence proliferation,differentiation,and SIRT1 signaling of HSCs.AIM To evaluate the effects and underlying mechanisms of action of different concentrations of NAM on HSC proliferation and differentiation.METHODS CD34^(+)cells were purified from umbilical cord blood using MacsCD34 beads,and cultured for 10-12 d in a serum-free medium supplemented with cytokines,with different concentrations of NAM added according to experimental requirements.Flow cytometry was used to detect phenotype,cell cycle distribution,and apoptosis of the cultured cells.Real-time polymerase chain reaction was used to detect the transcription levels of target genes encoding stemness-related factors,che mokines,components of hypoxia pathways,and antioxidant enzymes.Dichloro-dihydro-fluorescein diacetate probes were used to evaluate intracellular production of reactive oxygen species(ROS).Determination of the effect of different culture conditions on the balance of cytokine by cytometric bead array.RESULTS Compared with the control group,the proportion and expansion folds of HSPCs(CD34^(+)CD38)incubated with 5 mmol/L or 10 mmol/L NAM were significantly increased(all Pnificantly higher than those of the control and 10 mmol/L N

Adriamycin(ADR),also known as doxorubicin,is an anthracycline anticancer drug and a chemotherapeutic drug commonly used in breast cancer treatments.1 However,breast cancer patients can gradually become tolerant to chemotherapy.Therefore,improving the curative effect of ADR remains an urgent problem to be solved.Autophagy is a complex catabolic process;normal living cells break down damaged organelles or aggregated molecules and absorb energy to maintain homeostasis through autophagy.

Objective:Complete resection of malignant gliomas is often challenging.Our previous study indicated that intraoperative contrast-enhanced ultrasound(ICEUS)could aid in the detection of residual tumor remnants and the total removal of brain lesions.This study aimed to investigate the survival rates of patients undergoing resection with or without the use of ICEUS and to assess the impact of ICEUS on the prognosis of patients with malignant glioma.Methods:A total of 64 patients diagnosed with malignant glioma(WHO grade HI and IV)who underwent surgery between 2012 and 2018 were included.Among them,29 patients received ICEUS.The effects of ICEUS on overall survival(OS)and progression-free survival(PFS)of patients were evaluated.A quantitative analysis was performed to compare ICEUS parameters between gliomas and the surrounding tissues.Results:The ICEUS group showed better survival rates both in OS and PFS than the control group.The univariate analysis revealed that age,pathology and ICEUS were significant prognostic factors for PFS,with only age being a significant prognostic factor for OS.In multivariate analysis,age and ICEUS were significant prognostic factors for both OS and PFS.The quantitative analysis showed that the intensity and transit time of microbubbles reaching the tumors were significantly different from those of microbubbles reaching the surrounding tissue.Conclusion:ICEUS facilitates the identification of residual tumors.Age and ICEUS are prognostic factors for malignant glioma surgery,and use of ICEUS offers a better prognosis for patients with malignant glioma.

Esophageal cancer ranks among the most prevalent malignant tumors globally,primarily due to its highly aggressive nature and poor survival rates.According to the 2020 global cancer statistics,there were approximately 604000 new cases of esophageal cancer,resulting in 544000 deaths.The 5-year survival rate hovers around a mere 15%-25%.Notably,distinct variations exist in the risk factors associated with the two primary histological types,influencing their worldwide incidence and distribution.Squamous cell carcinoma displays a high incidence in specific regions,such as certain areas in China,where it meets the cost-effect-iveness criteria for widespread endoscopy-based early diagnosis within the local population.Conversely,adenocarcinoma(EAC)represents the most common histological subtype of esophageal cancer in Europe and the United States.The role of early diagnosis in cases of EAC originating from Barrett's esophagus(BE)remains a subject of controversy.The effectiveness of early detection for EAC,particularly those arising from BE,continues to be a debated topic.The variations in how early-stage esophageal carcinoma is treated in different regions are largely due to the differing rates of early-stage cancer diagnoses.In areas with higher incidences,such as China and Japan,early diagnosis is more common,which has led to the advancement of endoscopic methods as definitive treatments.These techniques have demonstrated remarkable efficacy with minimal complications while preserving esophageal functionality.Early screening,prompt diagnosis,and timely treatment are key strategies that can significantly lower both the occurrence and death rates associated with esophageal cancer.

BACKGROUND The detection rate of peptic ulcer in children is improving,with development of diagnostic procedures.Gastroscopy is the gold standard for the diagnosis of peptic ulcer,but it is an invasive procedure.Gastrointestinal contrast-enhanced ultrasonography(CEUS)has the advantages of being painless,noninvasive,nonradioactive,easy to use,and safe.AIM To investigate the clinical value of CEUS for diagnosis and treatment of peptic ulcer in children.METHODS We investigated 43 children with digestive tract symptoms in our hospital from January 2021 to June 2022.All children were examined by routine ultrasound,gastrointestinal CEUS,and gastroscopy.The pathological results of gastroscopy were taken as the gold standard.Routine ultrasonography was performed before gastrointestinal CEUS.Conventional ultrasound showed the thickness of the gastroduodenal wall,gastric peristalsis,and the adjacent organs and tissues around the abdominal cavity.Gastrointestinal CEUS recorded the thickness of the gastroduodenal wall;the size,location and shape of the ulcer;gastric peristalsis;and adjacent organs and tissues around the abdominal cavity.The results of routine ultrasound and gastrointestinal ultrasound were compared with those of gastroscopy to evaluate the diagnostic results and coincidence rate of routine ultrasound and gastrointestinal CEUS.All children received informed consent from their guardians for CEUS.This study was reviewed and approved by the hospital medical ethics committee.RESULTS Among the 43 children,17(15 male,2 female)were diagnosed with peptic ulcer by gastroscopy.There were 26 children with nonpeptic ulcer.There were eight cases of peptic ulcer and 35 of nonpeptic ulcer diagnosed by conventional ultrasound.The diagnostic coincidence rate of peptic ulcer in children diagnosed by conventional ultrasound was 79.1%(34/43),which was significantly different from that of gastroscopy(P=0.033).It indicates that the coincidence rate of gastrointestinal contrast-enhanced ultrasound and gastroscope is low.Fifteen cas

We reported alpine oak fossil leaves ( the Heterobalanus group, Quercus section Ilex, Fagaceae) rediscovered on Mount Shisha Pangma. According to twice field investigations, the fossil site is located at the front of the No. 5 glacier tongue on the south side of the Dasuopu Glacier at an altitude of about 5800m on the north slope of Shisha Pangma Peak. These fossils are from the siltstone interlayer dominated by sandy conglomerate in the lower part of the Yebokangjiale Group. These newly discovered fossil leaves have typical characteristics of alpine oaks, including oval or oblong leaf, thick and short petiole, entire or toothed leaf margin, and 5 similar to 6 pairs of secondary veins, which is completely consistent with the leaf morphology of alpine oaks from Mount Shisha Pangma reported previously. Through morphological comparison with living and fossil species in this group, as well as quantitative analysis combined with geometric morphometry, these fossil leaves from Mount Shisha Pangma were assigned to Quercus preguajavifolia tao. Alpine oaks may originate in southeastern margin of the Qinghai -Tibetan Plateau, then thrive locally and spread into regions nearby accompanying by the uplift of the plateau. The implication of alpine oak fossil leaves from Mount Shisha Pangma on paleoelevation needs to be further investigated due to the present wide range and the uncertainty of these fossils. The rediscovery of alpine oak fossils from Mount Shisha Pangma not only helps to clarify the specific layers bearing these fossils, but also further enriches the morphological characteristics of this fossil record and its systematic position. This discovery would provide important basis for better revealing the evolution of biodiversity and environmental changes in the middle part of the Himalaya during the Cenozoic.

Neonatal hypoxic-ischemic brain injury is the main cause of hypoxic-ischemic encephalopathy and cerebral palsy.Currently,there are few effective clinical treatments for neonatal hypoxic-ischemic brain injury.Here,we investigated the neuroprotective and molecular mechanisms of exogenous nicotinamide adenine dinucleotide,which can protect against hypoxic injury in adulthood,in a mouse model of neonatal hypoxic-ischemic brain injury.In this study,nicotinamide adenine dinucleotide(5 mg/kg)was intraperitoneally administered 30 minutes befo re surgery and every 24 hours thereafter.The results showed that nicotinamide adenine dinucleotide treatment improved body weight,brain structure,adenosine triphosphate levels,oxidative damage,neurobehavioral test outcomes,and seizure threshold in experimental mice.Tandem mass tag proteomics revealed that numerous proteins were altered after nicotinamide adenine dinucleotide treatment in hypoxic-ischemic brain injury mice.Parallel reaction monitoring and western blotting confirmed changes in the expression levels of proteins including serine(or cysteine)peptidase inhibitor,clade A,member 3N,fibronectin 1,5'-nucleotidase,cytosolic IA,microtubule associated protein 2,and complexin 2.Proteomics analyses showed that nicotinamide adenine dinucleotide ameliorated hypoxic-ischemic injury through inflammation-related signaling pathways(e.g.,nuclear factor-kappa B,mitogen-activated protein kinase,and phosphatidylinositol 3 kinase/protein kinase B).These findings suggest that nicotinamide adenine dinucleotide treatment can improve neurobehavioral phenotypes in hypoxic-ischemic brain injury mice through inflammation-related pathways.

Aqueous rechargeable batteries are the promising energy storge technology due to their safety,low cost,and environmental friendliness.Ammonium ion(NH_(4)^(+))is an ideal charge carrier for such batteries because of its small hydration radius and low molar mass.In this study,VO_(2)·xH_(2)O with rich oxygen defects(d-HVO)is designed and synthesized,and it exhibits unique nanoarray structure and good electrochemical performances for NH_(4)^(+)storge.Experimental and calculation results indicate that oxygen defects in d-HVO can enhance the conductivity and diffusion rate of NH_(4)^(+),leading to improved electrochemical performances.The most significant improvement is observed in d-HVO with 2 mmol thiourea(d-HVO-2)(220 mAh·g^(-1) at 0.1 A·g^(-1)),which has a moderate defect content.A full cell is assembled using d-HVO-2 as the anode and polyaniline(PAni)as the cathode,which shows excellent cycling stability with a capacity retention rate of 80%after 1000 cycles and outstanding power density up to 4540 W·kg^(-1).Moreover,the flexible d-HVO-2||PAni battery,based on quasi-solid electrolyte,shows excellent flexibility under different bending conditions.This study provides a new approach for designing and developing high-performance NH_(4)^(+)storage electrode materials.

Evolution hydrothermal aging resistance mechanism study of zirconium and manganese doped CeO2 catalysts in soot catalytic combustion based on low Miller indices crystal surface effect

The thermogravimetric analysis（TGA） experiments were carried out to reveal the mechanism of Zr and Mn doping on catalytic activity of CeO2catalyst both fresh and after hydrothermal aging,and the lattice morphology and valence changes were characterized by means of Brunauer-Emmett-Teller（BET）method,X-ray diffraction（XRD）,X-ray photoelectron spectroscopy（XPS）,and H2-temperature programmed reduction（H2-TPR）.Density functional theory（DFT） and molecular thermodynamics calculations were applied to investigate the change in catalytic activity,crystal surface energy and crystal morphology caused by hydrothermal aging.The maximum reaction rate temperature of fresh Mn/CeO2（389℃） is similar to that of CeO2（371℃） and lower than that of Zr/CeO2（447℃）,but the catalytic performance of CeO2decreases more severely after hydrothermal aging.The catalyst crystals show different degrees of crystal surface migration after hydrothermal aging,which leads to the reduction of Ce3+/Ce4+ratio and the active sites shift.DFT calculations indicate that the doping of Zr and Mn reduces the surface energy of the low Miller indices surface and increases the oxygen vacancy formation energy,leading to better thermal stability and lower catalytic activity.The Zr and Mn doping also changes the adsorption energy and Gibbs free energy of H2O,which dominates the migration of（1 1 1） to（1 1 0） and（1 0 0） in the vapor environment.The crystal surface migration mechanism of CeO2catalysts doped with Zr and Mn induced by H2O molecules at high temperature obtained in this study can provide a valuable addition to the regeneration of CeO2catalysts in the after-treatment systems of diesel engines.
