Objective To investigate the protective effects and mechanism of Chinese herbal compound Tongxinluo Capsule(通心络胶囊,TXL)on the Parkin-mediated mitophagy and the ubiquitin-proteasome system in a rat model of myocardial ischemia-reperfusion injury(MIRI).Methods Seventy adult male Sprague-Dawley rats were randomly divided into 7 groups:sham group,MIRI group,low-and high-dose TXL(0.5 and 1 g·kg^(-1)·d^(-1),respectively)groups,atorvastatin(ATV)group(7.2 g·kg^(-1)·d^(-1)),chloroquine(CQ)group(10 g·kg^(-1)·d^(-1)),and highdose TXL+CQ group.After pharmacological administration for 7 days,rats underwent left anterior descending artery ligation surgery to establish the MIRI models with 50 min ischemia followed by 4 h reperfusion.Blood was taken for cardiac troponin I(cTnI)detection and hearts were harvested for infarct staining and apoptosis detection.The autophagy or mitophagy proteins and ubiquitinated proteins were detected by Western blotting.Results Compared with the sham group,the MIRI group exhibited a larger infarcted area(27.13%±0.01%,P<0.01),a higher apoptotic index(34.33%±2.03%vs.1.81%±0.03%,P<0.01),and higher cTnI expression(14.18±1.01 vs.7.96±0.32,P<0.01).The mitochondrial integrity was damaged in the MIRI group,while TXL and ATV alleviated the damage of MIRI.More autophagosomes were observed in the high-dose TXL group than in the MIRI group(7.00±0.58 vs.4.33±1.15,P<0.05).More amounts of PTEN-induced putative kinase protein 1(PINK1)and Parkin translocated onto the mitochondria were detected in the high-dose TXL group than in the MIRI group(P<0.05).The ubiquitin response was signifificantly downregulated in the high-dose TXL group relative to the MIRI group(P<0.05).CQ administration abolished the activation of autophagy flux and the PINK1/Parkin pathway induced by high-dose of TXL.Conclusions TXL ameliorates MIRI via activating Parkin-mediated mitophagy in rats.The downregulation of the ubiquitin-proteasome system is also involved.

Objective:To explore the relationships between Chinese nurses’knowledge,perceptions,and attitudes and their behavior and actual implementation of safety measures when handling antineoplastic drugs(ADs)in their daily work.Methods:This was a multisite study conducted in 8 public hospitals in China.A self-administered questionnaire was sent to par ticipants querying the degree of contact with ADs.The hypothesized relations were explored using structural equation modelling via the bootstrap method.Mediation analysis was applied to explore the mediating role of protective knowledge regarding AD exposure on the associations among protective training,using warning labels,and using protective masks.Results:A total of 305 nurses were enrolled.The average age of all par ticipants was 30.2(standard deviation[SD]:6.2)years.Nurses who had received protective training for AD exposure were more likely to use labels for ADs after age,body mass index(BMI),length of service,marital status,education,and department were controlled as covariates.The bias-corrected bootstrap of 95%confidence interval(CI)indicated that protective knowledge significantly mediated(23.4%)the association between protective training and using labels(indirect effect=0.202,95%CI:0.009,0.495);the proportion of mediation was 23.4%.Protective knowledge significantly mediated the association between protective training and using protective masks(indirect effect=0.157,95%CI:0.048,0.325);the propor tion of mediation was 27.2%.Conclusions:The findings of this study have provided baseline information on the current state of Chinese nurses’perceptions,knowledge,and preventive behaviors toward ADs as the crisis is happening.Training is also recommended to improve nurses’perceptions of the risks associated with ADs.

Although bone marrow mesenchymal stem cells(BMSCs)might have therapeutic potency in ischemic stroke,the benefits are limited.The current study investigated the effects of BMSCs engineered to overexpress vascular endothelial growth factor(VEGF)on behavioral defects in a rat model of transient cerebral ischemia,which was induced by middle cerebral artery occlusion.VEGF-BMSCs or control grafts were injected into the left striatum of the infarcted hemisphere 24 hours after stroke.We found that compared with the stroke-only group and the vehicle-and BMSCs-control groups,the VEGF-BMSCs treated animals displayed the largest benefits,as evidenced by attenuated behavioral defects and smaller infarct volume 7 days after stroke.Additionally,VEGF-BMSCs greatly inhibited destruction of the blood-brain barrier,increased the regeneration of blood vessels in the region of ischemic penumbra,and reducedneuronal degeneration surrounding the infarct core.Further mechanistic studies showed that among all transplant groups,VEGF-BMSCs transplantation induced the highest level of brain-derived neurotrophic factor.These results suggest that BMSCs transplantation with vascular endothelial growth factor has the potential to treat ischemic stroke with better results than are currently available.

Frequent occurrence of harmful algal blooms has already threatened aquatic life and human health.In the present study,floating BiOCl_(0.6)I_(0.4)/ZnO photocatalyst was synthesized in situ by water bath method,and and applied in inactivation of Microcystis aeruginosa under visible light.The composition,morphology,chemical states,optical properties of the photocatalyst were also characterized.The results showed that BiOCl_(0.6)I_(0.4)exhibited laminated nanosheet structure with regular shape,and the light response range of the composite BZ/EP-3 (BiOCl_(0.6)I_(0.4)/ZnO/EP-3) was tuned from 582 to 638 nm.The results of photocatalytic experiments indicated that BZ/EP-3 composite had stronger photocatalytic activity than a single BiOCl_(0.6)I_(0.4)and ZnO,and the removal rate of chlorophyll a was 89.28%after 6 hr of photocatalytic reaction.The photosynthetic system was destroyed and cell membrane of algae ruptured under photocatalysis,resulting in the decrease of phycobiliprotein components and the release of a large number of ions (K^(+),Ca^(2+)and Mg^(2+)).Furthermore,active species trapping experiment determined that holes (h+) and superoxide radicals (·O_(2)-) were the main active substance for the inactivation of algae,and the p-n mechanism of photocatalyst was proposed.Overall,BZ/EP-3 showed excellent algal removal ability under visible light,providing fundamental theories for practical algae pollution control.

DNA analysis is the core of biotechnology applied in petroleum resources and engineering. Traditionally accurate determination of DNA purity and concentration by spectrometer is the first and critical step for downstream molecular biology research. In this study, three different spectrophotometry methods, BPM, NDTT and NPMTTZ were compared for their performance in determining DNA concentration and purity in 32 oil samples, and molecule methods like quantitative real-time PCR (qPCR) and high-throughput sequence were also performed to help assess the accuracy of the three methods in determining DNA concentration and purity. For ordinary heavy oil (OHO), extra heavy oil (EHO) and super heavy oil (SHO), the characteristics of high viscosity (η), density (ρ) and resin plus asphaltene content will affect the DNA extraction and UV determination. The DNA concentration was decreased as density increased: OHO (11.46 ± 18.34 ng/μL), EHO (6.68 ± 9.67 ng/μL) and SHO (6.20 ± 7.83 ng/μL), and the DNA purity was on the reverse: OHO (1.31 ± 0.27), EHO (1.54 ± 0.20), and SHO (1.83 ± 0.32). The results suggest that spectrophotometry such as BPM and NPMTTZ are qualitatively favorite methods as the quick non-consumable methods in determining DNA concentration and purity of medium oil and heavy oil.

The purpose of this paper is to study the critical sand starting velocity and transformation law of flow pattern based on gas-water-sand three-phase flow in an inclined pipe.Firstly,the indoor simulation experiment system of gas-water-sand three-phase flow was used to test the conversion law of flow pattern based upon the different gas void fraction.Secondly,the influence of slug bubbles on sand migration was investigated according to distinctive hole deviation angles,gas void fraction and sand concentration.Finally,the critical sand starting velocity was tested based on dissimilar hole deviation angles,gas void fraction,sand concentration and sand particle size,and then the influence of the abovementioned key parameters on the sand starting velocity was debated based on the force analysis of the sand particles.The experimental results illustrated that when the gas void fraction was less than 5%,it was bubbly flow.When it increased from 5%to 30%,the bubbly flow and slug flow coexisted.When it was between 30%and 50%,the slug flow and agitated flow coexisted.When it reached 50%,it was agitated flow.Providing that the hole deviation angle was 90°,the phenomenon of overall migration and wavelike migration on the surface of sand bed was observed.On the contrary,the phenomenon of rolling and jumping migration was recognized.The critical sand starting velocity was positively correlated with the hole deviation angle and sand particle size,but negatively associated with the gas void fraction and sand concentration.This research can provide a certain reference for sand-starting production in the field of petroleum engineering.

In this work,we firstly synthesized a CeO_(2)/C_(3)N_(4) photocatalyst with Z-scheme heterojunction by a facile LiC-KCI molten salt method.The synthesized catalyst has an excellent quality for removing organic pollution of dyes and antibiotics in wastewater.As an example,the CeCN-1:5 prepared with a mass ratio of Ce_(2)(CO_(3))_(3)·xH_(2)O:C_(3)H_(3)N_(6)=1:5 exhibits a methylene blue(MB)removal capacity of 100%within 90 min and tetracycline(TC)removal capacity of 94.6%.After 4 cycles,the CeCN-1:5 keeps a removal efficiency of nearly 100%in 150 min for MB and 85.7%for TC.The kinetics study reveals that the MB removal process with the CeCN-1:5 fits the modified Elovich model with strong adsorption while TC removal fits the first-order model.The large surface area(238 m^(2)/g)and negative zeta potential(-39.3 mV)of CeCN-1:5 contribute to superior adsorption capacity to MB.However,the adsorption of TC is restricted due to the positive surface/pore potential in acidic ***-1:5has combined Z-scheme heterojunction and exhibits a low recombination rate of electrons(e^(-))/holes(h^(+))and the photo-generated active radicals of·OH/·O_(2)^(-)that promotes the photocatalytic performance.This novel CeO_(2)/C_(3)N_(4) photocatalyst with an excellent photocatalysis removal activity has an enormous potential for photocatalytic applications.

Correlation between serum vitamin B12 level and peripheral neuropathy in atrophic gastritis

AIM To explore the correlation between serum vitamin B12 level and peripheral neuropathy in patients with chronic atrophic gastritis(CAG). METHODS A total of 593 patients diagnosed with chronic gastritis by gastroscopy and pathological examination fromSeptember 2013 to September 2016 were selected for this study. The age of these patients ranged within 18-to 75-years-old. Blood pressure, height and weight were measured in each patient, and the body mass index value was calculated. Furthermore, gastric acid, serum gastrin, serum vitamin and serum creatinine tests were performed, and peripheral nerve conduction velocity and Helicobacter pylori(H. pylori) were detected. In addition, the type of gastritis was determined by gastroscopy. The above factors were used as independent variables to analyze chronic gastritis with peripheral neuropathy and vitamin B12 deficiency risk factors, and to analyze the relationship between vitamin B12 levels and peripheral nerve conduction velocity. In addition, in the treatment of CAG on the basis of vitamin B12, patients with peripheral neuropathy were observed. RESULTS Age, H. pylori infection, CAG, vitamin B9 and vitamin B12 were risk factors for the occurrence of peripheral nerve degeneration.Furthermore,CAG and H. pylori infection were risk factors for chronic gastritis associated with vitamin B12 deficiency. Serum vitamin B12 level was positively correlated with sensory nerve conduction velocity in the tibial nerve(R = 0.463). After vitamin B12 supplementation, patients with peripheral neuropathy improved. CONCLUSION Serum vitamin B12 levels in patients with chronic gastritis significantly decreased, and the occurrence of peripheral neuropathy had a certain correlation. CAG and H. pylori infection are risk factors for vitamin B12 deficiency and peripheral neuropathy. When treating CAG, vitamin B12 supplementation can significantly reduce peripheral nervous system lesions. Therefore, the occurrence of peripheral neuropathy associated with vitamin B12 deficiency may be considered
