Lower respiratory tract samples are reliable for severe acute respiratory syndrome coronavirus 2 nucleic acid diagnosis and animal model study

Severe acute respiratory syndrome coronavirus 2(SARS-Co V-2) and coronavirus disease 2019(COVID-19) continue to impact countries worldwide.At present, inadequate diagnosis and unreliable evaluation systems hinder the implementation and development of effective prevention and treatment strategies. Here, we conducted a horizontal and longitudinal study comparing the detection rates of SARS-Co V-2 nucleic acid in different types of samples collected from COVID-19 patients and SARS-Co V-2-infected monkeys. We also detected anti-SARS-Co V-2 antibodies in the above clinical and animal model samples to identify a reliable approach for the accurate diagnosis of SARS-Co V-2 infection. Results showed that, regardless of clinical symptoms, the highest detection levels of viral nucleic acid were found in sputum and tracheal brush samples, resulting in a high and stable diagnosis rate. Anti-SARS-Co V-2 immunoglobulin M(Ig M) and G(Ig G) antibodies were not detected in6.90% of COVID-19 patients. Furthermore,integration of nucleic acid detection results from the various sample types did not improve the diagnosis rate. Moreover, dynamic changes in SARS-Co V-2 viral load were more obvious in sputum and tracheal brushes than in nasal and throat swabs. Thus,SARS-Co V-2 nucleic acid detection in sputum and tracheal brushes was the least affected by infection route, disease progression, and individual differences. Therefore, SARS-Co V-2 nucleic acid detection using lower respiratory tract samples alone is reliable for COVID-19 diagnosis and study.

Branched alkanes are ubiquitous in the troposphere and play an important role in the chemical processes.In this work,the rate constants and products for the reaction of Cl atoms with 3-methylhexane and 2-methylheptane were measured at room temperature(298±0.2K)and atmospheric pressure using a conventional relative rate method.The rate constants of 3-methylhexane and 2-methylheptane in units of cm~3/(mol·sec)are(3.09±0.31)×10^(-10)and(3.67±0.40)×10^(-10),respectively.Furthermore,the corresponding atmospheric lifetime of the studied branched alkanes with Cl was 6.92-89.90 hours and 5.82-75.69 hours,respectively.The estimated atmospheric lifetimes indicated that the reaction with Cl atoms could be the most important atmospheric degradation pathway for 3-methylhexane and2-methylheptane.Primary gas-phase products of the reactions were identified and quantified,and particle-phase products were also obtained.The atmosphere oxidation mechanism of Cl atoms with 3-methylhexane and 2-methylheptane is proposed.The SOA yields of 3-methylhexane and 2-methylheptane from the reaction of Cl atoms were determined to be7.96%±0.89%and 13.35%±1.50%respectively.Overall,the results reveal that the primary loss process of branched alkanes is the reaction with Cl atoms,which impacts its degradation on a regional scale.

Prediction and interpretation of photocatalytic NO removal on g-C_(3)N_(4)-based catalysts using machine learning

Predictive modeling of photocatalytic NO removal is highly desirable for efficient air pollution abatement.However,great challenges remain in precisely predicting photocatalytic performance and understanding interactions of diverse features in the catalytic systems.Herein,a dataset of g-C_(3) N_(4)-based catalysts with 255 data points was collected from peer-reviewed publications and machine learning(ML)model was proposed to predict the NO removal rate.The result shows that the Gradient Boosting Decision Tree(GBDT)demonstrated the greatest prediction accuracy with R 2 of 0.999 and 0.907 on the training and test data,respectively.The SHAP value and feature importance analysis revealed that the empirical categories for NO removal rate,in the order of importance,were catalyst characteristics>reaction process>preparation conditions.Moreover,the partial dependence plots broke the ML black box to further quantify the marginal contributions of the input features(e.g.,doping ratio,flow rate,and pore volume)to the model output outcomes.This ML approach presents a pure data-driven,interpretable framework,which provides new insights into the influence of catalyst characteristics,reaction process,and preparation conditions on NO removal.

Non-alcoholic fatty liver disease(NAFLD)poses a significant health challenge in modern societies due to shifts in lifestyle and dietary habits.Its complexity stems from genetic predisposition,environmental influences,and metabolic factors.Epigenetic processes govern various cellular functions such as transcription,chromatin structure,and cell division.In NAFLD,these epigenetic tendencies,especially the process of histone methylation,are intricately intertwined with fat accumulation in the liver.Histone methylation is regulated by different enzymes like methyltransferases and demethylases and influences the expression of genes related to adipogenesis.While early-stage NAFLD is reversible,its progression to severe stages becomes almost irreversible.Therefore,early detection and intervention in NAFLD are crucial,and understanding the precise role of histone methylation in the early stages of NAFLD could be vital in halting or potentially reversing the progression of this disease.

Early changes in corneal densitometry after FS-LASIK combined with accelerated corneal cross-linking for correction of high myopia

AIM:To observe the effects of femtosecond laserassisted excimer laser in situ keratomileusis combined with accelerated corneal cross-linking(FS-LASIK Xtra)on corneal densitometry after correcting for high myopia.METHODS:In this prospectively study,130 patients underwent FS-LASIK or FS-LASIK Xtra for high myopia.Their right eyes were selected for inclusion in the study,of which 65 cases of 65 eyes in the FS-LASIK group,65 patients with 65 eyes in the FS-LASIK Xtra group.Patients were evaluated for corneal densitometry at 1,3,and 6mo postoperatively using Pentacam Scheimpflug imaging.RESULTS:Preoperative differences in corneal densitometry between the FS-LASIK and FS-LASIK Xtra groups in different ranges were not statistically significant(P>0.05).Layer-by-layer analysis revealed statistically significant differences in the anterior(120μm),central,and total layer corneal densitometry between the FS-LASIK and FS-LASIK Xtra groups at 1 and 3mo postoperatively(all P0.05).CONCLUSION:There is an increase in internal corneal densitometry during the early postoperative period after FS-LASIK Xtra for correction of high myopia.However,the densitometry values decreased to the level of conventional FS-LASIK at 6mo after surgery,with the most significant changes observed in the superficial central zone.

Constructing extrinsic oxygen vacancy on the surface of photocatalyst as CO_(2)and electrons reservoirs to improve photocatalytic CO_(2)reduction activity

Constructing own oxygen vacancies in the photocatalysts is a very promising method to improve their photocatalytic CO_(2)reduction activity.However,some catalysts have excellent stabilities,making it difficult for them to construct their own oxygen vacancies.To simplify the above difficulty of stable photocatalysts,constructing extrinsic oxygen vacancies on their surface as a novel idea is proposed.Here,a stable TiO_(2)nanosheet is chosen as a research object,we uniformly deposited BiOCl quantum dots on their surface via a simple adsorption-deposition method.It is found that BiOCl quantum dots are able to simultaneously self-transform into defective BiOCl with many oxygen vacancies when the photocatalyst is performed photocatalytic CO_(2)reduction.These extrinsic oxygen vacancies can act as“CO_(2)and photo-generated electrons reservoirs”to improve CO_(2)capture and accelerate the separation of photogenerated electrons and holes.For the above reasons,the modified TiO_(2)showed obvious enhancement of photocatalytic CO_(2)reduction compared to pristine TiO_(2)and BiOCl.This work may open a new avenue to broaden the use of oxygen vacancies in the process of photocatalytic CO_(2)reduction.

Identification of overlay differentially expressed genes in both rats and goats with blast lung injury through comparative transcriptomics

Purpose:To identify the potential target genes of blast lung injury(Bli)for the diagnosis and treatment.Methods:This is an experimental study.The Bli models in rats and goats were established by conducting a fuel-air explosive power test in an unobstructed environment,which was subsequently validated through hematoxylin-eosin staining.Transcriptome sequencing was performed on lung tissues from both goats and rats.Differentially expressed genes were identified using the criteria ofq≤0.05 and|log2 fold change|≥1.Following that,enrichment analyses were conducted for gene ontology and the Kyoto Encyclopedia of Genes and Genomes pathways.The potential target genes were further confirmed through quantitative real-time polymerase chain reaction and enzyme linked immunosorbent assay.Results:Observations through microscopy unveiled the presence of reddish edema fluid,erythrocytes,and instances of focal or patchy bleeding within the alveolar cavity.Transcriptome sequencing analysis identified a total of 83 differentially expressed genes in both rats and goats.Notably,49 genes exhibited a consistent expression pattern,with 38 genes displaying up-regulation and 11 genes demonstrating down-regulation.Enrichment analysis highlighted the potential involvement of the interleukin-17 signaling pathway and vascular smooth muscle contraction pathway in the underlying mechanism of Bli.Furthermore,the experimental findings in both goats and rats demonstrated a strong association between Bli and several key genes,including anterior gradient 2,ankyrin repeat domain 65,bactericidal/permeability-increasing fold containing family A member 1,bactericidal/permeability-increasing fold containing family B member 1,and keratin 4,which exhibited up-regulation.Conclusions:Anterior gradient 2,ankyrin repeat domain 65,bactericidal/permeability-increasing fold containing family A member 1,bactericidal/permeability-increasing fold containing family B member 1,and keratin 4 hold potential as target genes for the prognosis,diagnosis,and treatment of BL

Association between the cumulative triglyceride-glucose index and the recurrence of atrial fibrillation after radiofrequency catheter ablation

BACKGROUND Triglyceride-glucose(TyG)index values are a new surrogate marker for insulin resistance.This study aimed to explore the relationship between cumulative TyG index values and atrial fibrillation(AF)recurrence after radiofrequency catheter ablation(RFCA).METHODS A total of 576 patients with AF who underwent RFCA at the Second Affiliated Hospital of Xi'an Jiaotong University were included in this study.The participants were grouped based on cumulative TyG index values tertiles within 3 months after ablation.Cox regression and restricted cubic spline analyses were used to determine the relationship between cumulative TyG index values and AF recurrence.The predictive value of all risk factors was assessed by receiver operating curve analysis.RESULTS There were 375 patients completed the study(age:63.23±10.73 years,64.27%male).The risk of AF recurrence increased with increasing cumulative TyG index values tertiles.After adjusting for potential confounders,patients in the medium cumulative TyG index group[hazard ratio(HR)=4.949,95%CI:1.778–13.778,P=0.002]and the high cumulative TyG index group(HR=8.716,95%CI:3.371–22.536,Pline regression model also showed an increased risk of AF recurrence with increasing cumulative TyG index values.When considering cumulative TyG index values,left atrial diameter,and lactate dehydrogenase levels as a comprehensive factor,the model could effectively predict AF recurrence after RFCA[area under the curve(AUC)=0.847,95%CI:0.797–0.897,P<0.001].CONCLUSIONS Cumulative TyG index values were a risk factor for AF recurrence after RFCA.Monitoring longitudinal TyG index values may assist with optimized for risk stratification and outcome prediction for AF recurrence.

In-situ coating and surface partial protonation co-promoting performance of single-crystal nickel-rich cathode in all-solid-state batteries

The poor electrochemical performance of all-solid-state batteries(ASSBs),which is assemblied by Ni-rich cathode and poly(ethylene oxide)(PEO)-based electrolytes,can be attributed to unstable cathodic interface and poor crystal structure stability of Ni-rich cathode.Several coating strategies are previously employed to enhance the stability of the cathodic interface and crystal structure for Ni-rich cathode.However,these methods can hardly achieve simplicity and high efficiency simultaneously.In this work,polyacrylic acid(PAA)replaced traditional PVDF as a binder for cathode,which can achieve a uniform PAA-li(lixPAA(0liNi_(0.83)Co_(0.12)Mn_(0.05)O_(2)(SC-NCM83)due to H^(+)/li^(+)exchange reaction during the initial charging-discharging process.The formation of PAA-li coating layer on cathode can promote interfacial li^(+)transport and enhance the stability of the cathodic interface.Furthermore,the partially-protonated surface of SC-NCM83 casued by H^(+)/li^(+)exchange reaction can restrict Ni ions transport to enhance the crystal structure stability.The proposed SC-NCM83-PAA exhibits superior cycling performance with a retention of 92%compared with that(57.3%)of SC-NCM83-polyvinylidene difluoride(PVDF)after 200 cycles.This work provides a practical strategy to construct high-performance cathodes for ASSBs.

Impact of non-ideal UE hardware on cell-free massive MIMO network with centralized operation

This paper investigates the impact of non-ideal user equipment(UE) hardware on a cell-free(CF) massive MIMO(mMIMO) network with centralized operation under spatially correlated channels. The minimum mean-squared error(MMSE) estimator can be derived with the help of the generic non-ideal UE hardware model. It is demonstrated that even if the effective signal-to-noise ratio approaches infinity, pilot contamination and imperfect hardware can cause a non-zero estimation error floor. After that, a lower bound is determined for the ergodic uplink capacity of the centralized CF mMIMO network under non-ideal UE hardware. Moreover, the optimal receive combining vector is obtained to maximize the uplink spectral efficiency(SE). The maximum ratio(MR) and regularized zero-forcing(RZF) combining schemes are offered as alternatives in light of the computational complexity of the MMSE receiver. Comparing the RZF to the MMSE scheme under different levels of hardware impairments, our findings indicate that the RZF receiver suffers a negligible loss in total SE. For MR combining, a novel closed-form uplink achievable SE expression is obtained based on the MMSE estimator and the use-and-then-forget bounding technique. This expression gives vital insights into the achievable uplink performance with UE hardware impairments. Besides, for various hardware impairment factors, the impact of pilot sequence length on average sum SE is disclosed for different receive combining schemes. To increase the overall SE of the max-min fairness scheme, a heuristic fractional power control scheme with UE hardware impairments is developed, which can essentially avoid sacrificing the SE of other UEs while maximizing the SE of the unluckiest UE in the whole network. Finally,our theoretical performance analysis and power control algorithm are validated by simulation results, and fundamental design guidelines are provided for selecting hardware satisfying the practical UE requirements.
