Maternal resveratrol improves the intestinal health and weight gain of suckling piglets during high summer temperatures: The involvement of exosome-derived microRNAs and immunoglobin in colostrum

Previous studies have shown that maternal resveratrol improved growth performance and altered the microbial composition of suckling piglets under hot summer conditions. However, it remains unclear how maternal resveratrol improves growth performance of suckling piglets during high summer temperatures. A total of 2. sows(Landrace × Large White; three parity) were randomly assigned to 2.groups(with or without 300 mg/kg resveratrol) from d 75 of gestation to d 2. of lactation during high ambient temperatures(from 2. to 30℃). The results showed that maternal resveratrol supplementation increased total daily weight gain of piglets under hot summer conditions, which is consistent with previous studies. Furthermore, we found that maternal resveratrol improved the intestinal morphology and intestinal epithelial proliferation in suckling piglets. Dietary resveratrol supplementation affected the characteristics of exosome-derived microRNAs(miRNAs) in sow colostrum, as well as the genes targeted by differentially produced miRNAs. MiRNAs are concentrated in the tight junction pathway. As a result, the expression of intestinal tight junction proteins was increased in suckling piglets(P < 0.05).Notably, maternal resveratrol increased the intestinal secretory immunoglobulin A(sIgA) levels of suckling piglets via colostrum immunoglobin(P < 0.05), which could increase the abundance of beneficial microbiota to further increase the concentration of short chain fatty acids(SCFA) in suckling piglets' intestine(P < 0.05). Finally, our correlation analysis further demonstrated the positive associations between significantly differential intestinal microbiota, intestinal sIgA production and SCFA concentrations,as well as the positive relation between total daily weight gain and intestinal health of suckling *** together, our findings suggested that maternal resveratrol could promote intestinal health to improve piglet growth during high summer temperatures, which might be associa

Engineering Cell-Selective Charge-Altering Lipid Nanoparticles for Efficient siRNA Delivery in Vivo

Lipid nanoparticles(LNPs)have emerged as a powerful platform for RNA delivery;the chemical engineering of LNP for efficient delivery of RNA into cytosol remains critical but *** promising strategy is the use of permanently positively charged lipids,which have been shown to enhance the stability and delivery efficiency of ***,the resulting strong electrostatic interactions reduced the RNA release capacity from the ***,we engineered a hydrogen peroxide(H_(2.O_(2.)-triggered charge-altering LNP(CALNP)for efficient small interfering RNA(siRNA)delivery and tumor therapy in *** incorporation of phenylboronic acid(PBA)into ionizable lipids generated permanently positively charged lipids.A CALNP with optimal lipid formulations was identified,exhibiting enhanced transfection efficiency with effective lysosomal escape through dual effects of electrostatic interaction and ligand-receptor binding.H_(2.O_(2.-triggered removal of PBA groups regenerated ionizable LNP with reduced positive charges at physiological pH,allowing cell-selective siRNA release in the *** results demonstrated that CALNPs exhibited improved siRNA transfection and gene silencing *** also showed potent CALNP activity against the polo-like kinase 1(Plk1)gene by effectively silencing Plk1 mRNA and subsequent suppression of tumor ***,these findings highlighted the potential of CALNP as an efficient platform for RNA delivery and tumor therapeutics.

Success rate of current human-derived gastric cancer organoids establishment and influencing factors:A systematic review and meta-analysis

BACKGROUND Human-derived gastric cancer organoids(GCOs)are widely used in gastric cancer research;however,the culture success rate is generally *** To explore the potential influencing factors,and the literature on successful culture rates of GCOs was reviewed using *** PubMed,Web of Science,and EMBASE were searched for *** trained researchers selected the studies and extracted *** 17.0 software was used for meta-analysis of the incidence of each outcome *** adjusted Methodological Index for Non-Randomized Studies scale was used to assess the quality of the included *** plots and Egger’s test were used to detect publication *** analyses were conducted for sex,tissue source,histo-logical classification,and the pathological tumor-node-metastasis(pTNM)cancer staging *** Eight studies with a pooled success rate of 66.6%were *** derived from women and men had success rates of 67%and 46.7%,*** from surgery or biopsy/endoscopic submucosal dissection showed success rates of 70.9%and 53.7%,*** of poorly-differentiated,moderately-differentiated and signet-ring cell cancer showed success rates of 64.6%,31%,and 32.7%,*** with pTNM stages I-II and III-IV showed success rates of 38.3%and 65.2.,respectively.Y-2.632.and non-Y-2.632.use showed success rates of 58.2.and 70%,*** generated with collagenase were more successful than those constructed with Liberase TH and TrypLE(72.1%vs 71%,respectively).EDTA digestion showed a 50%lower success rate than other methods(P=0.04).CONCLUSION GCO establishment rate is low and varies by sex,tissue source,histological type,and pTNM *** Y-2.632.and using Liberase TH,TrypLE,or collagenase yields greater success than EDTA.
