Safety,immunogenicity and protective effect of sequential vaccination with inactivated and recombinant protein COVID-19 vaccine in the elderly:a prospective longitudinal study

The safety and efficacy of COVID-19 vaccines in the elderly,a high-risk group for severe COVID-19 infection,have not been fully *** clarify these issues,this prospective study followed up 157 elderly and 73 young participants for 16 months and compared the safety,immunogenicity,and efficacy of two doses of the inactivated vaccine BBIBP-CorV followed by a booster dose of the recombinant protein vaccine *** results showed that this vaccination protocol was safe and tolerable in the *** administering two doses of the BBIBP-CorV,the positivity rates and titers of neutralizing and anti-RBD antibodies in the elderly were significantly lower than those in the young *** the ZF2001 booster dose,the antibody-positive rates in the elderly were comparable to those in the young;however,the antibody titers remained ***,age,and underlying diseases were independently associated with vaccine immunogenicity in elderly *** pseudovirus neutralization assay showed that,compared with those after receiving two doses of BBIBP-CorV priming,some participants obtained immunological protection against BA.5 and BF.7 after receiving the ZF2001 *** infection symptoms last longer in the infected elderly and pre-infection antibody titers were negatively associated with the severity of post-infection *** antibody levels in the elderly increased significantly after breakthrough infection but were still lower than those in the *** data suggest that multiple booster vaccinations at short intervals to maintain high antibody levels may be an effective strategy for protecting the elderly against COVID-19.

Zhang and collaborators [1] demonstrated that bees of the genus Ctenoplectra collect oil from the newly reported oilreward orchids of the genera Dendrobium and Galeola in Asia, and that the majority of the bees that carry out this act and pollinate the orchids are male. These are new discoveries with significant implications for biodiversity conservation.

Synthesis and characterization of SBA-15 macrospheres with hierarchical pore structure

The new macroscopic-scale SBA-15 spheres with diameter of 0.2-0.5 mm are prepared by a sol-gel method, in which the tetrabutylorthosilicate （TBOS） is used as silicon source and triblock copolymer （P123） and polyoxyethylene octylphenol ether （OP- 10） are used as templates. The resulting spherical samples are characterized by XRD, N2 adsorption-desorption, TEM and SEM methods. The results show that the inner structure of SBA-15 macrospheres has macroporous channels （0.5 um） and mesoporous skeleton （4-5 nm）. These SBA-15 macrospheres with the hierarchical pore structure demonstrate high absorption capacity for Emodin molecules.

Enhancement of CTLs induced by DCs loaded with ubiquitinated hepatitis B virus core antigen

AIM： To investigate whether hepatitis B virus （HBV） could induce a hepatitis B virus core antigen （HBcAg）specific cytotoxic T lymphocyte （CTL） response in vitro by dendritic cells （DCs） transduced with lentiviral vector-encoding ubiquitinated hepatitis B virus core antigen （LV-Ub-HBcAg）.METHODS： Recombinant LV-Ub-HBcAg were transfected into highly susceptible 293 T cells to obtain high virus titres, Bone marrow-derived DCs isolated from BALB/c mice were cultured with recombinant granulocyte-macrophage colony-stimulating factor and recombinant interleukin （IL）-4. LV-Ub-HBcAg, lentiviral vector-encoding hepatitis B virus core antigen （LV-HBcAg）, lentiviral vector （LV） or lipopolysaccharide were added to induce DC maturation, and the DC phenotypes were analyzed by flow cytometry. The level of IL-12 in the supernatant was detected by enzyme-linked immunosorbent assay （ELISA）. T lymphocytes were proliferated using Cell Counting Kit-8. DCs were cultured and induced to mature using different LVs, and co-cultured with allogeneic T cells to detect the secretion levels of IL-2, IL-4, IL-10and interferon-γ in the supernatants of T cells by ELISA. Intracellular cytokines of proliferative T cells were analyzed by flow cytometry, and specific CTL activity was measured by a lactate dehydrogenase release assay.RESULTS： LV-Ub-HBcAg-induced DCs secreted more IL-12 and upregulated the expression of CD80, CD86 and major histocompatibility class ]I, DCs sensitised by different LVs effectively promoted cytokine secretion; the levels of IL-2 and interferon-y induced by LV-Ub- HBcAg were higher than those induced by LV-HBcAg, Compared with LV-HBcAg-transduced DCs, LV-Ub- HBcAg-transduced DCs more efficiently stimulated the proliferation of T lymphocytes and generated HBcAgspecific cytotoxic T lymphocytes.CONCLUSION： LV-Ub-HBcAg effectively induced DC maturation. The mature DCs efficiently induced T cell polarisation to Thl and generated HBcAg-specific CTLs.

Optimized the vanadium electrolyte with sulfate-phosphoric mixed acids to enhance the stable operation at high-temperature

Herein, the influence of the concentration design and comprehensive performance of the sulfate-phosphoric mixed acid system electrolyte is investigated to realize an electrolyte that maintains high energy density and stable operation at high temperatures. Static stability tests have shown that VOPO4 precipitation occurs only with vanadium(V) electrolyte. The concentration of vanadium ion of 2.0–2.2 mol·L^(–1), phosphoric acid of 0.10–0.15 mol·L^(–1), and sulfuric acid of 2.5–3.0 mol·L^(–1) are suitable for a vanadium redox flow battery in the temperature range from –20 to 50 ℃. The equations for predicting the viscosity and conductivity of electrolytes are obtained by the response surface method. The optimized electrolyte overcomes precipitation generation. It has 2.8 times higher energy density than the non-phosphate electrolyte, and a coulomb efficiency of 94.0% at 50 ℃. The sulfate-phosphoric mixed acid system electrolyte promotes the electrode reaction process, increases the current density, and reduces the resistance. This work systematically optimizes the concentrations of composition of positive and negative vanadium electrolytes with mixed sulfate-phosphoric acid. It provides a basis for the different valence states and comprehensive properties of sulfate-phosphoric mixed acid system vanadium electrolytes under extreme environments, guiding engineering applications.

ROLE OF B LYMPHOCYTE AND ITS SUBPOPULATIONS IN PATHOGENESIS OF IMMUNORELATED PANCYTOPENIA

Objective To measure the quantities and apoptosis-related protein levels of B lymphocyte in the patients with immunorelated pancytopenia （IRP） and explore the action of B lymphocyte in the pathogenic mechanism of IRP. Methods Quantifies of whole B lymphocytes and CD5^＋ B lymphocytes as well as the expressions of Fas and Bcl-2 in B lymphocytes in 35 patients with untreated IRP, 15 IRP patients in complete remission （CR）, and 10 normal controls were assayed by flow cytometry. The percentages of B lymphocyte and CD5^＋ B lymphocyte were significantly higher in untreated IRP patients than in CR IRP patients and normal controls （ P 〈 0. 05 ）, and there was no significant difference between the latter two groups （ P 〉 0. 05 ）. There was no significant difference of Fas expression in B lymphocyte among three groups （ P 〉 0. 05）. The expression of Bcl-2 in B lymphocyte was significantly higher in untreated patients than in CR patients or normal controls （ P 〈 0. 01 ）, and significantly higher in CR patients than in normal controls （ P 〈 0. 01 ）. The apoptosis. related index was significantly lower in untreated patients than in CR patients or normal controls （ P 〈 0. 05 ）, and signif. icantly lower in CR patients than in normal controls （ P 〈 0. 05 ）. The percentage of B lymphocyte was positively correlated with post-treated response time （ r = 0. 53, P 〈 0. 01 ）. Conclusion The production of auto-antibodies in IRP patients probably has some relationship with the abnormal quantifies of B lymphocyte and its subpopulations as well as with the inhibition of B lymphocyte apoptosis.

A positive nematic liquid crystal(5CB)sample is confined in cylindrical cells under strong or weak axial anchoring boundary conditions when a radial nonuniform low-frequency electric field is applied and the flexoelectric effect is taken into *** on the Frank elastic free energy,the surface energy of the Rapini-Papoular approximation,the polarization free energy and the flexoelectric free energy caused by electric field,we obtain the free energy density of the nematic and solve the corresponding Euler-Lagrange equation *** investigate the director distribution,the critical voltage and the critical exponent of nematic liquid crystal in cylindrical *** follows that the critical exponent is the classical *** is also shown that the critical voltage in the system is affected by the flexoelectric effect,the geometric effect and radial weak anchoring effect on the cylindrical surfaces.A new type of director transition caused by the flexoelectric effect,the dielectric coupling effect and the radial weak anchoring effect is found.

Nuclear Factor-κB Signaling Pathway Constitutively Activated in Esophageal Squamous Cell Carcinoma Cell Lines and Inhibition of Growth of Cells by Small Interfering RNA

Although constitutive nuclear factor(NF)-kB activation has been reported in many humantumors,the role of the NF-kB pathway in esophageal squamous cell carcinoma(ESCC)has not been *** this study,NF-kB pathway in two ESCC cell lines was investigated using immunocytochemistry,Westernblot and reverse transciiption-polymerase chain *** activation of NF-kB DNA binding was determinedby electrophoretic mobility-shift *** interference was used to specifically inhibit the expression *** of cells Was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide *** results showed that p50,p65,IkBα,p-IkBα and IkB kinase β were expressed and mainly localized in *** transcription-polymerase chain reaction results showed the constitutive expressions ofp50,p65 and IkBα mRNA in the two ESCC cell ***,the nuclear extracts revealed that p50and p65 translocated to the nucleus had DNA-binding ***,small interfering RNA of p65 decreasedthe expression of p65,and the viability of ceils transfected with p65 small interfering RNA was significantlysuppressed at the same concentration of 5-fluorouracil(P<0.05)compared to untransfected *** resultsof this study showed that there was the constitutively activated NF-kB signaling pathway in the ESCC *** interference targeting at p65 increased the sensitivity of the ESCC cell lines to 5-fluorouracil,suggesting that NF-kB might be a good target for cancer treatment.

A Degree Condition for Graphs Having All(a,b)-parity Factors

Let a and b be positive integers such that a≤b and a≡b(mod 2).We say that G has all(a,b)-parity factors if G has an h-factor for every function h:V(G)→{a,a+2,…,b-2,b} with b|V(G)| even and h(v)≡b(mod 2) for all v∈V(G).In this paper,we prove that every graph G with n≥2(b+1)(a+b) vertices has all(a,b)-parity factors if δ(G)≥(b^(2)-b)/a,and for any two nonadjacent vertices u,v ∈V(G),max{d_(G)(u),d_(G)(v)}≥bn/a+***,we show that this result is best possible in some sense.
