“Warming yang and invigorating qi” acupuncture alters acetylcholine receptor expression in the neuromuscular junction of rats with experimental autoimmune myasthenia gravis

Myasthenia gravis is an autoimmune disorder in which antibodies have been shown to form against the nicotinic acetylcholine nicotinic postsynaptic receptors located at the neuromuscular junction.＂Warming yang and invigorating qi＂ acupuncture treatment has been shown to reduce serum inflammatory cytokine expression and increase transforming growth factor beta expression in rats with experimental autoimmune myasthenia gravis.However,few studies have addressed the effects of this type of acupuncture on the acetylcholine receptors at the neuromuscular junction.Here,we used confocal laser scanning microscopy to examine the area and density of immunoreactivity for an antibody to the nicotinic acetylcholine receptor at the neuromuscular junction in the phrenic nerve of rats with experimental autoimmune myasthenia gravis following ＂warming yang and invigorating qi＂ acupuncture therapy.Needles were inserted at acupressure points Shousanli（LI10）,Zusanli（ST36）,Pishu（BL20）,and Shenshu（BL23） once daily for 7 consecutive days.The treatment was repeated after 1 day of rest.We found that area and the integrated optical density of the immunoreactivity for the acetylcholine receptor at the neuromuscular junction of the phrenic nerve was significantly increased following acupuncture treatment.This outcome of the acupuncture therapy was similar to that of the cholinesterase inhibitor pyridostigmine bromide.These findings suggest that ＂warming yang and invigorating qi＂ acupuncture treatment increases acetylcholine receptor expression at the neuromuscular junction in a rat model of autoimmune myasthenia gravis.

Radio frequency interference(RFI)is an important challenge in radio astronomy.RFI comes from various sources and increasingly impacts astronomical observation as telescopes become more sensitive.In this study,we propose a fast and effective method for removing RFI in pulsar data.We use pseudo-inverse learning to train a single hidden layer auto-encoder(AE).We demonstrate that the AE can quickly learn the RFI signatures and then remove them from fast-sampled spectra,leaving real pulsar signals.This method has the advantage over traditional threshold-based filter method in that it does not completely remove contaminated channels,which could also contain useful astronomical information.

INFLUENCE OF “WARMING YANG AND NOURISHING QI” ACUPUNCTURE TREATMENT ON SERUM IL-12 AND IL-18 EXPRESSION LEVELS IN RATS WITH EXPERIMENTAL AUTOIMMUNE MYASTHENIA GRAVIS AND ITS MECHANISM

Objective: To observe the influence of the "Warming yang and Nourishing qi" acupuncture treatment on the serum interleukin-12(IL-12) and interleukin-18(IL-18) expression levels in rats with experimental autoimmune myasthenia gravis(EAMG) and to analyze the mechanism of acupuncture in treatment of myasthenia gravis(MG).Methods: AchRα1 129-145 peptide fragments were used to immune the female Lewis rats to establish EAMG rat models.Thirty EAMG model rats were randomly divided into acupuncture treatment group, drug control group and model control group(n=10); the unmodeled 10 rats purchased at the same period were used as the blank control group.The rats in acupuncture treatment group were treated by "Warming yang and Nourishing qi" acupuncture treatment, 30 min, once a day, 7 d for a period, 1 d rest between two periods, and two periods of continuous treatment.The rats in drug control group were treated by the pyridostigmine lavaged treatment, 18.5 mg/(kg·d), continuous treatment for 15 d.The rats in the other two groups had no any special treatments.After treatment, the blood samples from the vein of tail root of the rats in 4 groups were collected, and the serum expression levels of IL-12 and IL-18 were detected with ELISA method.Results: After treatment, compared with the model control group, the serum IL-12 and IL-18 expression levels of the EAMG rats in the acupuncture treatment group and the drugcontrol group were significantly decreased(P0.05).Conclusion: "Warming yang and Nourishing qi" acupuncture treatment can reduce the serum IL-12 and IL-18 expression levels of EAMG rats.For the treatment of MG, the effect of acupuncture was similar to pyridostigmine.

Effect of grouped ST36 or RN12 on gastric motility and sensation

Objective: To investigate the effects of paired ST36 and RN12 acupuncture stimulation on gastric motility and the firing rate of responsible neurons in anesthetized rats.Methods: Using electrophysiological methods, we measured the effects of acupuncture at RN12, ST36, or RN12+ST36 on gastric motility as well as nerve discharges from the gastric sympathetic, vagal nerve, and wide dynamic range(WDR) neurons in the spinal dorsal horn T7-9 in anesthetized rats.Results: 1) Acupuncture at RN12 inhibited gastric motility(83.84±4.49)% and vagal nerve discharge(65.64±5.67)%, but promoted sympathetic nerve activity(243.70±40.67)%; acupuncture at ST36 induced opposing effects on gastric motility(111.31±2.01)% and vagal nerve discharge(166.98±15.92)%. Interestingly, acupuncture at RN12+ST36 together yielded effects similar to that of acupuncture at RN12 alone(93.59±2.05)%,(54.52±7.12)%. 2) WDR neuron discharges induced by gastric distension were inhibited by acupuncture at RN12(194.56 ±18.79)%. However, acupuncture at ST36 induced a more significant inhibitory effect.Conclusion: Acupuncture at RN12 inhibits gastric motility by activation of the sympathetic reflex at the spinal level; acupuncture at ST36 promotes gastric motility by activation of the parasympathetic reflex at the supraspinal level, and acupuncture at RN12+ST36 produces a similar decrease in gastric motility as RN12 stimulation alone. Both acupoints decrease the effect of noxious gastric distention on WDR neuron activity, but paired RN12+ST36 stimulation does not significantly affect WDR neuron discharge.

Global and regional trends in the incidence and mortality burden of endometrial cancer,1990–2019:Updated results from the Global Burden of Disease Study,2019

Background:The disease burdens for endometrial cancer(EC)vary across different countries and geographical regions and change every year.Herein,we reported the updated results of the Global Burden of Disease Study 2019 on EC with respect to age-standardized incidence and mortality from 1990 to 2019.Methods:The annual percentage change(APC)of incidence and mortality was evaluated using joinpoint regression analysis to examine the temporal trends during the same timeframe in terms of the global landscape,different sociodemographic indices(SDI),and geographic regions.The relationship between Human Development Index(HDI)and incidence and mortality was additionally explored.Results:The age-standardized incidence rates(ASIRs)revealed a significant average global elevation by 0.5%per year(95%confidence interval[CI],0.3–0.7;P<0.001).The age-standardized mortality rates(ASMRs),in contrast,fell by an average of 0.8%per year(95%CI,−1.0 to−0.7;P<0.001)worldwide.The ASIRs and ASMRs for EC varied across different SDIs and geographical regions.We noted four temporal trends and a significant reduction by 0.5%per year since 2010 in the ASIR,whereas we detected six consecutively decreasing temporal trends in ASMR during the entire period.Notably,the estimated APCs were significantly positively correlated with HDIs(ρ=0.22;95%CI,0.07–0.35;P=0.003)with regard to incident cases in 2019.Conclusions:Incidence rates for EC reflected a significant increase overall(although we observed a decline since 2010),and the death rates declined consecutively from 1990 to 2019.We posit that more precise strategies can be tailored and then implemented based on the distinct age-standardized incidence and mortality burden in different geographical areas.

GLI1 and PTTG1 expression in colorectal carcinoma patients undergoing radical surgery and their correlation with lymph node metastasis

BACKGROUND Few studies have investigated the expression of GLI1 and PTTG1 in patients undergoing radical surgery for colorectal carcinoma(CRC)and their association with lymph node metastasis(LNM).Therefore,more relevant studies and analyses need to be conducted.AIM To explore GLI1 and PTTG1 expression in patients undergoing radical surgery for CRC and their correlation with LNM.METHODS This study selected 103 patients with CRC admitted to our hospital between April 2020 and April 2023.Sample specimens of CRC and adjacent tissues were collected to determine the positive rates and expression levels of GLI1 and PTTG1.The correlation of the two genes with patients’clinicopathological data(e.g.,LNM)was explored,and differences in GLI1 and PTTG1 expression between patients with LNM and those without were analyzed.Receiver operating characteristic(ROC)curves were plotted to evaluate the predictive potential of the two genes for LNM in patients with CRC.RESULTS Significantly higher positive rates and expression levels of GLI1 and PTTG1 wereobserved in CRC tissue samples compared with adjacent tissues.GLI1 and PTTG1 were strongly linked to LNM in patients undergoing radical surgery for CRC,with higher GLI1 and PTTG1 levels found in patients with LNM than in those without.The areas under the ROC curve of GLI1 and PTTG1 in assessing LNM in patients with CRC were 0.824 and 0.811,respectively.CONCLUSION GLI1 and PTTG1 expression was upregulated in patients undergoing radical surgery for CRC and are significantly related to LNM in these patients.Moreover,high GLI1 and PTTG1 expression can indicate LNM in patients with CRC undergoing radical surgery.The expression of both genes has certain diagnostic and therapeutic significance.

Risk Factors of Depression Screened by Two-Sample Mendelian Randomization Analysis:A Systematic Review

Objective This study explored the potentially modifiable factors for depression and major depressive disorder(MDD)from the MR-Base database and further evaluated the associations between drug targets with MDD.Methods We analyzed two-sample of Mendelian randomization(2SMR)using genetic variant depression(n=113,154)and MDD(n=208,811)from Genome-Wide Association Studies(GWAS).Separate calculations were performed with modifiable risk factors from MR-Base for 1,001 genomes.The MR analysis was performed by screening drug targets with MDD in the DrugBank database to explore the therapeutic targets for MDD.Inverse variance weighted(IVW),fixed-effect inverse variance weighted(FE-IVW),MR-Egger,weighted median,and weighted mode were used for complementary calculation.Results The potential causal relationship between modifiable risk factors and depression contained 459 results for depression and 424 for MDD.Also,the associations between drug targets and MDD showed that SLC6A4,GRIN2A,GRIN2C,SCN10A,and IL1B expression are associated with an increased risk of depression.In contrast,ADRB1,CHRNA3,HTR3A,GSTP1,and GABRG2 genes are candidate protective factors against depression.Conclusion This study identified the risk factors causally associated with depression and MDD,and estimated 10 drug targets with significant impact on MDD,providing essential information for formulating strategies to prevent and treat depression.

Atomization characteristics of pyrolysis oil derived from waste tires

The atomization characteristics play a key role in the highly efficient combustion of pyrolysis oil derived from waste tires.In this study,the fuel properties of tire pyrolysis oil(TPO)were initially studied,and then a high-speed camera and a phase Doppler particle analyzer were employed to characterize the atomization feature of TPO.The influence of pressure and nozzle orifice diameter on atomization characteristics such as spray angle,droplet velocity,and droplet size distribution was investigated.The results showed that TPO had a high calorific value of about 43.6 MJ/kg and a low viscosity of 3.84×10^(–6)m^(2)/s at 40℃,which made it have the potential to be used as an alternative fuel.Higher pressure expanded the spray angle and extended the spray in both the axial and radial directions.With increasing pressure,spray angle and droplet velocity raised,and the increase in crushing effect of air reduced the Sauter mean diameter(SMD)of the droplets.To obtain proper atomization quality for combustion,the pressure is expected to be higher than 1.25 MPa.With increasing nozzle orifice diameter,droplet velocity increased,and the SMD of the droplets increased as well due to weakened crushing effect of the orifice.Therefore,the pressure must be increased to maintain the atomization quality when using a nozzle with a larger orifice.Due to the lower viscosity,the velocity and particle size distribution of TPO droplets after atomization were smaller than those of diesel droplets.The extremely small carbon black contained in TPO also contributed to the breaking of droplets and played a certain role in the size reduction of the oil droplets,but it may cause the risk of nozzle blockage.In summary,TPO showed great atomization characteristics for alternative fuel applications.

Microstructural Evolutions and Mechanical Properties of Energetic Al_(1) (TiZrNbTaMoCr)_(15) High-Entropy Alloys

The present study focuses on investigating the microstructural evolutions and mechanical properties of energetic Al_(1)(TiZrNbTaMoCr)_(15) refractory high-entropy alloys with the different heat treatments at low Al content state. It is found that even with a reduction in the Al content, the strength of these alloys remains unaffected at room temperature and high temperature, while the plasticity improves significantly. In particular, the coherent BCC/B_(2) microstructure with needle-like B_(2) nanoprecipitates dispersed into the BCC matrix is formed in 873 K-aged Al_(1)Ti_(6)Zr_(2)Nb_(3)Ta_(3)Mo_(0.5)Cr_(0.5) alloy. Therefore, this alloy exhibits the highest compression yield strength (σ_(YS) = 1333 and 717 MPa) at room temperature and 1073 K, respectively. After 973 K-aged, the coherent BCC/B_(2) microstructure underwent destabilization, and the B_(2) phase transforms into the brittle Zr5Al3 phase which then coarsens and dominates the microstructure of S3-AlTa_(3) alloy after 1073 K-aged. Moreover, these current alloys exhibit exceptionally high theoretical exothermic enthalpy (ΔH), surpassing 11606 J·g^(−1), which highlights their significant potential as innovative high-performance energetic structural materials.
