TRIM14 promotes endothelial activation via activating NF-κB signaling pathway

Endothelial activation by proinflammatory cytokines is closely associated to the pathogenesis of atherosclerosis and other vascular diseases;however, the molecular mechanisms controlling endothelial activation are not fully understood. Here we identify TRIM14 as a new positive regulator of endothelial activation via activating NF-κB signal pathway. TRIM14 is highly expressed in human vascular endothelial cells (ECs) and markedly induced by inflammatory stimuli such as TNF-α, IL-1β, and LPS. Overexpression of TRIM14 significantly increased the expression of adhesion molecules such as VCAM-1, ICAM-1, E-selectin, and cytokines such as CCL2, IL-8, CXCL-1, and TNF-α in activated ECs and by which it facilitated monocyte adhesion to ECs. Conversely, knockdown of TRIM14 has opposite effect on endothelial activation. Upon TNF-α stimulation, TRIM14 is recruited to IKK complex via directly binding to NEMO and promotes the phosphorylation of IκBα and p65, which is dependent on its K63-linked ubiquitination. Meanwhile, p65 can directly bind to the promoter regions of human TRIM14 gene and control its mRNA transcription. Finally, TRIM14 protein level is significantly upregulated in mouse and human atheroma compared to normal arteries. Taken together, these results indicate that TRIM14-NF-κB forms a positive feedback loop to enhance EC activation and TRIM14 may be a potential therapeutic target for vascular inflammatory diseases such as atherosclerosis.

The solution set of the Sun-perturbed optimal two-impulse trans-lunar orbit is helpful for overall optimization of the lunar exploration mission.A model for computing the two-impulse trans-lunar orbit,which strictly satisfies the boundary constraints,is established.The solution set is computed first with a circular restricted three-body model using a generalized local gradient optimization algorithm and the strategy of design variable initial continuation.By taking the solution set of a circular restricted three-body model as the initial values of the design variables,the Sun-perturbed solution set is calculated based on the dynamic model continuation theory and traversal search methodology.A comparative analysis shows that the fuel cost may be reduced to some extent by considering the Sun’s perturbation and choosing an appropriate transfer window.Moreover,there are several optimal two-impulse trans-lunar methods for supporting a lunar mission to select a scenario with a certain ground measurement and to control the time cost.A fitted linear dependence relationship between the Sun’s befitting phase and the trans-lunar duration could thus provide a reference to select a low-fuel-cost trans-lunar injection window in an engineering project.

Voronoi diagrams on triangulated surfaces based on the geodesic metric play a key role in many applications of computer graphics.Previous methods of constructing such Voronoi diagrams generally depended on having an exact geodesic metric.However,exact geodesic computation is time-consuming and has high memory usage,limiting wider application of geodesic Voronoi diagrams(GVDs).In order to overcome this issue,instead of using exact methods,we reformulate a graph method based on Steiner point insertion,as an effective way to obtain geodesic distances.Further,since a bisector comprises hyperbolic and line segments,we utilize Apollonius diagrams to encode complicated structures,enabling Voronoi diagrams to encode a medial-axis surface for a dense set of boundary samples.Based on these strategies,we present an approximation algorithm for efficient Voronoi diagram construction on triangulated surfaces.We also suggest a measure for evaluating similarity of our results to the exact GVD.Although our GVD results are constructed using approximate geodesic distances,we can get GVD results similar to exact results by inserting Steiner points on triangle edges.Experimental results on many 3D models indicate the improved speed and memory requirements compared to previous leading methods.

Deep learning is widely used for seismic impedance inversion,but few work provides in-depth research and analysis on designing the architectures of deep neural networks and choosing the network hyperparameters.This paper is dedicated to comprehensively studying on the significant aspects of deep neural networks that affect the inversion results.We experimentally reveal how network hyperparameters and architectures affect the inversion performance,and develop a series of methods which are proven to be effective in reconstructing high-frequency information in the estimated impedance model.Experiments demonstrate that the proposed multi-scale architecture is helpful to reconstruct more high-frequency details than a conventional network.Besides,the reconstruction of high-frequency information can be further promoted by introducing a perceptual loss and a generative adversarial network from the computer vision perspective.More importantly,the experimental results provide valuable references for designing proper network architectures in the seismic inversion problem.

The biomineralization of CaCO_(3)often involves the transformation of amorphous precursors into crystalline phases,which is regulated by various proteins and inorganic ions such as Mg^(2+)ions.While the effects of Mg^(2+)ions on the polymorph and shape of the crystalline CaCO_(3)have been observed and studied,the interplay between Mg^(2+)ions and CaCO_(3)during the mineralization remains unclear.This work focuses on the mechanism of Mg^(2+)ion-regulated mineralization of CaCO_(3).By tracing the Mg isotope fractionation,the different mineralization pathways of CaCO_(3)under different Mg^(2+)ion concentrations had been clarified.Detailed regulatory role of Mg^(2+)ions at the different stages of mineralization had been proposed through combining the fractionation data with the analyses of the CaCO_(3)polymorph and shape evolution.These results provide a clear view of the Mg-mediated crystallization process of amorphous CaCO_(3),which can be used to finely control the phase of the crystalline products according to different needs.

Shock-driven hydrodynamic instabilities in a plasma usually lead to interfacial mixing and the generation of electromagnetic fields,which are nonequilibrium processes coupling kinetics with meso-and macroscopic dynamics.The understanding and modeling of these physical processes are very challenging tasks for single-fluid hydrodynamic codes.This work presents a new framework that incorporates both kinetics and hydrodynamics to simulate shock waves and hydrodynamic instabilities in high-density plasmas.In this hybrid code,ions are modeled using the standard particle-in-cell method together with a Monte Carlo description of collisions while electrons are modeled as a massless fluid,with the electron heat flux and fluid–particle energy exchange being considered in the electron pressure equation.In high-density plasmas,Maxwell’s equations are solved using Ohm’s law instead of Ampere’s law.This hybrid algorithm retains ion kinetic effects and their consequences for plasma interpenetration,shock wave propagation,and hydrodynamic instability.Furthermore,we investigate the shock-induced(or gravity-induced)turbulent mixing between a light and a heavy plasma,where hydrodynamic instabilities are initiated by a shock wave(or gravity).This study reveals that self-generated electromagnetic fields play a role in the formation of baroclinic vorticity along the interface and in late-time mixing of the plasmas.Our results confirm the ability of the proposed method to describe shock-driven hydrodynamic instabilities in a plasma,in particular,nonequilibrium processes that involve mixing and electromagnetic fields at the interface.

The selective aerobic oxidation of benzyl alcohol to benzaldehyde has attracted considerable attention because benzaldehyde is a high value-added product. The rate of this typical gas–liquid reaction is significantly affected by mass transfer. In this study, CoTPP-mediated(CoTPP: cobalt(II) mesotetraphenylporphyrin) selective benzyl alcohol oxidation with oxygen was conducted in a membrane microchannel(MMC) reactor and a bubble column(BC) reactor, respectively. We observed that 83% benzyl alcohol was converted within 6.5 min in the MMC reactor, but only less than 10% benzyl alcohol was converted in the BC reactor. Hydrodynamic characteristics and gas–liquid mass transfer performances were compared for the MMC and BC reactors. The MMC reactor was assumed to be a plug flow reactor,and the dimensionless variance was 0.29. Compared to the BC reactor, the gas–liquid mass transfer was intensified significantly in MMC reactor. It could be ascribed to the high gas holdup(2.9 times higher than that of BC reactor), liquid film mass transfer coefficient(8.2 times higher than that of BC reactor), and mass transfer coefficient per unit interfacial area(3.8 times higher than that of BC reactor). Moreover,the Hatta number for the MMC reactor reached up to 0.61, which was about 15 times higher than that of the BC reactor. The computational fluid dynamics calculations for mass fractions in both liquid and gas phases were consistent with the experimental data.

A numerical model of an iron bath smelting reduction furnace with side-bottom combined blowing was established to study the influence of blowing arrangements on the stirring effect of the molten pool,and the accuracy of numerical simulation was verified by water model experiment.By comparing the flow field of molten pool with single nozzle,double nozzles(symmetrical and asymmetrical),and four nozzles(symmetrical and asymmetrical),the proportion of dead zone,average turbulent kinetic energy,and mixing time,the results show that asymmetrical bottom blowing is better than symmetrical bottom blowing,and the effect of double nozzles bottom blowing was better than that of four nozzles bottom blowing.The mixing effect is the worst under the condition of single nozzle.When the bottom blowing is asymmetrical with double nozzles,the mixing time is the shortest.Under the condition of double nozzles asymmetrical bottom blowing,when the insertion angle and depth of side lance are larger and deeper,the velocity streamline of molten slag layer is denser and the value is larger;meanwhile,the reflux of molten iron layer is larger,the proportion of dead zone is smaller,and the whole molten pool is fully stirred.When the insertion depth of the side lance is deeper,the gas holdup in the molten pool is greater and the stirring of the molten pool is more intense,while the insertion angle has little effect on the gas holdup.By comparing the influence of different side blowing conditions on the slag layer,it is found that the slag layer is divided into two layers by double-layer side lance,with the critical surface of the slag layer at about 200-260 mm from the bottom,and the insertion depth of the lower side lance has a greater influence on the layering of the slag.
