Efficacy and Safety of Yanggan Jian in Hepatitis B Virusrelated Decompensated Cirrhosis:A Randomized,Doubleblind,Controlled Trial

Background and Aims:The aim was to evaluate the efficacy and safety of Yanggan Jian(YGJ)in HBV-infected patients with decompensated ***:This randomized,double-blind controlled trial enrolled 160 patients with HBV-related decompensated cirrhosis who were already receiving or about to start antiviral *** were randomly assigned to receive YGJ or placebo for 24 weeks,and were followed-up to 36 *** primary outcome was the proportion of patients with a≥2 point reduction in Child-Turcotte-Pugh(CTP)score from baseline at week *** outcomes were CTP class and score,serum liver function indices,mortality,incidence of hepatocellular carcinoma and variceal ***:The proportion of patients with a CTP score reduction≥2 was significantly greater in the YGJ than in the placebo group(p=0.009);the percentage of patients with CTP class C was significantly less than that in the placebo group(pline in CTP score at week 24(p=0.034).The improvement in measured values and change from baseline of prothrombin time,serum albumin,platelets,cholinesterase,international normalized ratio,and activated partial thromboplastin time were significantly better with YGJ than with ***-group differences in cumulative rates of variceal bleeding,hepatocellular carcinoma,death,or the frequency of any adverse event(AE),AEs related to treatment,or discontinuation because of AEs were not ***:YGJ significantly improved CTP scores and hepatic synthetic and reserve function in patients with HBV-related decompensated cirrhosis,and was safe and well tolerated.

The neutron total cross section data of^(9)Be are essential in the nuclear structure model research of light nuclei and nuclear power *** neutron total cross section of^(9)Be in the 0.3 eV−120 MeV energy region has been measured using time-of-flight and transmission methods with the Neutron Total Cross Sectional Spectrometer(NTOX)based on the multi-cell fast fission chamber at the China Spallation Neutron Source(CSNS)-Back-n white neutron source(Back-n).The fission count-neutron energy distributions of ^(235)U and ^(238)U without samples and with Be samples with three thicknesses were measured in the double-bunch operation mode for a beam power of 100 *** Bayesian method was used to eliminate the influence of the double-bunch problem on neutron measurement in the energy region above 10 *** neutron total cross section of^(9)Be results was consistent with ENDF/B-VIII.0 evaluation library data in the 0.3 eV−20 MeV energy *** the energy ranges of 0.3 eV to 10 keV and 0.01 to 20 MeV,the deviations between our results and the evaluation results of ENDF/B-VIII.0 were within 2.5%and 15%,*** the resonance energy region,the measured resonance energies in our experiment were 0.63,0.82,and 2.8 MeV,*** results showed that the total cross section uncertainties of three Be samples were within 2.2%in the energy region below 1 *** total cross section uncertainty of 30 mm Be from ^(235)U was the smallest and less than 5%in the energy region of 0.3 eV−120 *** results of this experiment can provide technical support for further data analysis and related nuclear data evaluation.

Diabetic nephropathy(DN)is one of the most common complications of diabetes mellitus,which is characterized in renal tubulointerstitial fibrosis(TIF).The current study was designed to investigate the protective effect of Jujuboside A(Ju A)on TIF in type 2 diabetes(T2DM)mice,and explore its underlying anti-fibrosis mechanism.A mouse T2DM model was established using high fat diet(HFD)feeding combined with intraperitoneal injection of streptozotocin(STZ).Then,diabetic mice were treated with Ju A(10,20 and 40 mg·kg^(−1)·d^(−1),i.g.)for 12 *** showed that administration of Ju A not only down-regulated fasting blood glucose(FBG)levels,but also improved hyperlipidemia and renal function in diabetic ***,the reduced ECM accumulation was observed in the renal cortex of Ju A treated diabetic mice,while the TIF progression was also attenuated by Ju A through blocking the epithelial-to-mesenchymal transition(EMT)of renal tubular epithelial cells(RTECs).Further mechanism studies showed that Ju A treatment effectively down-regulated the protein expression and subsequent nuclear translocation of Yin Yang 1(YY1)in the renal cortex of diabetic mice,and reduced the levels of transforming growth factor-β1(TGF-β1)in the serum and renal cortex of Ju A treated *** to in vitro studies,the up-regulated YY1/TGF-β1 signaling pathway was restored by Ju A in high glucose(HG)cultured HK-2 *** together,these findings demonstrated that Ju A can ameliorate the TIF of DN through down-regulating the YY1/TGF-β1 signaling pathway.

Background The hematopoietic microenvironment （HM） plays a critical role in malignant cell growth, patient survival, and response to chemotherapy in hematologic malignancies. However, mechanisms associated with this environmental influence remain unclear. In this study, we investigated the role of bone marrow derived mesenchymal stem cells （MSCs） in U937 cell line, the MSCs. to find out the relations between leukemia drug resistance and Methods U937 cells were cultured in suspension or grew adherently with MSCs. The cell growth curve was drawn and the cell cycle was measured by flow cytometry. Apoptosis and sensitivity of U937 to daunoblastina （DNR） were quantified by DNA ladder detection and trypan blue exclusion assays, respectively. The gene expression profile chip technology was used to determine and analyze the changes in apoptosis-related gene expression after adherent culture and the expression of MDR 1 mRNA was assessed by reverse transcriptional polymerase chain reaction （RT-PCR） at the same time. Results In the adherent culture, the proliferation of the U937 cells was inhibited, the G0/G1 phase cells increased （F=64.9726, P〈0.0001）, G2/M phase cells were decreased （F=98.1361, P〈0.0001） and the natural apoptosis rate was decreased （F=24.0866, P〈0.0001） compared with those in the suspended culture. U937 cell viability was enhanced and cell apoptosis was blocked during DNR treatment in adherent culture with MSCs. Thirty-nine differently expressed genes were screened from the 487 apoptosis related genes in the adherent culture U937 cells. Among the 37 upregulated genes, Bcl-XL was upregulated most significantly. Two genes were downregulated. Adherent culture did not induce MDR1 mRNA expression in U937 cells. Conclusions MSCs play a role in modulating the proliferation of U937 cells and response of U937 cells to DNR, and Bcl-XL apoptosis-inhibiting gene may be most important in determining the sensitivity of leukemic cells to treatment, which is not related to

Objective Human Lyme Borreliosis （LB）, which is caused by Borrefia burgdorferi sensu lato （B. burgdorferi）, has been identified as a major arthropod-borne infectious disease in China. We aimed to develop a multiple locus variable-number tandem repeat （VNTR） analysis （MLVA） assay for the genotyping of Borrelia burgdorJ：eri strains detected in China. Methods B. garinii PBi complete 904.246 kb chromosome and two plasmids （cp26 and Ip54） were screened by using Tandem Repeats Finder program for getting potential VNTR loci, the potential VNTR loci were analyzed and identified with PCR and the VNTR loci data were analyzed and MLVA clustering tree were constrcted by using the categorical coefficient and the unweighted pair-group method with arithmetic means （UPGMA）. Results We identified 5 new VNTR loci through analyzing 47 potential VNTR loci. We used the MLVA protocol to analyse 101 B. burgdorferi strains detected in China and finally identified 51 unique genotypes in 4 major clusters including B. burgdorferi sensu stricto （B.b.s.s）, B. garinii, B. a[zelii, and B. valaisiana, consistent with the current MLSA phylogeny studies. The allele numbers of VNTR-1, VNTR-2, VNTR-3, VNTR-4, and VNTR-5 were 7, 3, 9, 7, and 6. The Hunter-Gaston index （HGI） of five VNTR loci were 0.79, 0.22, 0.77, 0.71, and 0.67, respectively. The combined HGI of five VNTR loci was 0.96. Clustering of the strains of Xinjiang, Inner Mongolia and Heilongjiang was confirmed, and this situation was consistent with the close geographical distribution of those provinces. Conclusion The MLVA protocol esytablished in this study is easy and can show strains＇ phylogenetic relationships to distinguish the strains of Borrelia species. It is useful for further phylogenetic and epidemiological analyses of Borrelia strains.

Effects of Mg and La on the evolution of inclusions and microstructure in Ca-Ti treated steel

The evolution of inclusions and the formation of acicular ferrite(AF)in Ca-Ti treated steel was systematically investigated after Mg and La *** inclusions in the molten steel were Ca-Al-O,Ca-Al-Mg-O,and La-Mg-Ca-Al-O after Ca,Mg,and La addition,*** type of oxide inclusion in the final quenched samples was the same as that in the molten ***,unlike those in molten steel,the inclusions were Ca-Al-Ti-O+MnS,Ca-Mg-Al-Ti-O+MnS,and La-Ca-Mg-Al-Ti-O+MnS in Mg-free,Mg-containing,and La-containing samples,*** inclusions distributed dispersedly in the La-containing *** addition,the average size of the inclusions in the La-containing sample was the smallest,while the number density of inclusions was the *** size of effective inclusions(nucleus of AF formation)was mainly in the range of 1-3μ*** addition,the content of ferrite side plates(FSP)decreased,while the percentage of AF increased by 16.2%due to the increase in the number of effective inclusions in the La-containing sample in this study.
