BACKGROUND Acute liver failure(ALF)has a high mortality with widespread hepatocyte death involving ferroptosis and pyroptosis.The silent information regulator sirtuin 1(SIRT1)-mediated deacetylation affects multiple biological processes,including cellular senescence,apoptosis,sugar and lipid metabolism,oxidative stress,and inflammation.AIM To investigate the association between ferroptosis and pyroptosis and the upstream regulatory mechanisms.METHODS This study included 30 patients with ALF and 30 healthy individuals who underwent serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)testing.C57BL/6 mice were also intraperitoneally pretreated with SIRT1,p53,or glutathione peroxidase 4(GPX4)inducers and inhibitors and injected with lipopolysaccharide(LPS)/D-galactosamine(D-GalN)to induce ALF.Gasdermin D(GSDMD)^(-/-)mice were used as an experimental group.Histological changes in liver tissue were monitored by hematoxylin and eosin staining.ALT,AST,glutathione,reactive oxygen species,and iron levels were measured using commercial kits.Ferroptosis-and pyroptosis-related protein and mRNA expression was detected by western blot and quantitative real-time polymerase chain reaction.SIRT1,p53,and GSDMD were assessed by immunofluorescence analysis.RESULTS Serum AST and ALT levels were elevated in patients with ALF.SIRT1,solute carrier family 7a member 11(SLC7A11),and GPX4 protein expression was decreased and acetylated p5,p53,GSDMD,and acyl-CoA synthetase long-chain family member 4(ACSL4)protein levels were elevated in human ALF liver tissue.In the p53 and ferroptosis inhibitor-treated and GSDMD^(-/-)groups,serum interleukin(IL)-1β,tumour necrosis factor alpha,IL-6,IL-2 and C-C motif ligand 2 levels were decreased and hepatic impairment was mitigated.In mice with GSDMD knockout,p53 was reduced,GPX4 was increased,and ferroptotic events(depletion of SLC7A11,elevation of ACSL4,and iron accumulation)were detected.In vitro,knockdown of p53 and overexpression of GPX4 reduced AST and ALT levels,the cytostatic rate

Thermoplastic polyurethane(PU)elastomers have attracted significant attention because of their many important industrial applications.However,the creation of fire-retardant and anti-dripping PU elastomers has remained a grant challenge due to the lack of crosslinking and weak interchain interactions.Herein,we report a mechanically robust,biodegradable,fire-retardant,and anti-dripping biobased PU elastomer with excellent biodegradability using an abietic acid-based compound as hard segments and polycaprolactone diol(PCL)as soft segments,followed by physically crosslinking with cellulose nanocrystals(CNC)through dynamic hydrogen-bonding.The resultant elastomer shows the balanced mechanical and fire-retardant properties,e.g.,a tensile strength and break strain of 9.1 MPa and 560%,a self-extinguishing ability(V-0 rating in UL-94 testing),and an anti-dripping behavior.Moreover,the as-developed PU can be completely degraded in 1.0 wt.%lipase solution at 37℃ in 60 days,arising from the catalytic and wicking effect of CNC on PU chains.This work provides an innovative and versatile strategy for constructing robust,fire-retardant,anti-dripping,and biodegradable PU elastomers,which hold great promise for practical applications in electronic and automobile sectors.

Dear Editor,The novel coronavirus SARS-CoV-2 rapidly evolutes to increase its infectivity and transmissibility,facilitates its immune escape,impairs vaccine efficacy,and currently causes repeated infections in human,which is mainly determined by the accumulated mutations in spike(S).1 Transmembrane S is a heavily glycosylated protein on the surface of the virion as homotrimer,responsible for binding to human angiotensin-converting enzyme 2(hACE2)receptor.2 While the architecture of S trimers has been well studied,how the stability of S is regulated by potential host factors remains unknown.

Assessing recent recurrence after hepatectomy for hepatitis Brelated hepatocellular carcinoma by a predictive model based on sarcopenia

BACKGROUND Sarcopenia may be associated with hepatocellular carcinoma(HCC)following hepatectomy.But traditional single clinical variables are still insufficient to predict recurrence.We still lack effective prediction models for recent recurrence(time to recurrence5 cm(HR=2.222,95%CI:1.285-3.842,P<0.05),and hepatitis B virus DNA level≥2000 IU/mL(HR=2.1,95%CI:1.407-3.135,P<0.05)were independent risk factors associated with postoperative recurrence of HCC.Based on the sarcopenia to assess the RFS model of hepatectomy with hepatitis B-related liver cancer disease(SAMD)was established combined with other the above risk factors.The area under the curve of the SAMD model was 0.782(95%CI:0.705-0.858)in the training cohort(sensitivity 81%,specificity 63%)and 0.773(95%CI:0.707-0.838)in the validation cohort.Besides,a SAMD score≥110 was better to distinguish the high-risk group of postoperative recurrence of HCC.CONCLUSION Sarcopenia is associated with recent recurrence after hepatectomy for hepatitis B-related HCC.A nutritional status-based prediction m

BACKGROUND Intraductal papillary neoplasm of the bile duct(IPNB)is a premalignant biliarytype epithelial neoplasm with intraductal papillary or villous growth.Currently reported local palliative therapeutic modalities,including endoscopic nasobiliary drainage,stenting and biliary curettage,endoscopic biliary polypectomy,percutaneous biliary drainage,laser ablation,argon plasma coagulation,photodynamic therapy,and radiofrequency ablation to relieve mechanical obstruction are limited with weaknesses and disadvantages.We have applied percutaneous transhepatic cholangioscopy(PTCS)-assisted biliary polypectomy(PTCS-BP)technique for the management of IPNB including mucin-hypersecreting cast-like and polypoid type tumors since 2010.AIM To assess the technical feasibility,efficacy,and safety of PTCS-BP for local palliative treatment of IPNB.METHODS Patients with mucin-hypersecreting cast-like or polypoid type IPNB and receiving PTCS-BP between September 2010 and December 2019 were included.PTCS-BP was performed by using a half-moon type snare with a soft stainless-steel wire,and the tumor was snared and resected with electrocautery.The primary outcome was its feasibility,indicated by technical success.The secondary outcomes were efficacy,including therapeutic success,curative resection,and clinical success,and safety.RESULTS Five patients(four with mucin-hypersecreting cast-like type and one with polypoid type IPNB)were included.Low-and high-grade intraepithelial neoplasia(HGIN)and recurrent IPNB with invasive carcinoma were observed in one,two,and two patients,respectively.Repeated cholangitis and/or obstructive jaundice were presented in all four patients with mucin-hypersecreting cast-like type IPNB.All five patients achieved technical success of PTCS-BP.Four patients(three with mucin-hypersecreting cast-like type and one with polypoid type IPNB)obtained therapeutic success;one with mucin-hypersecreting cast-like type tumors in the intrahepatic small bile duct and HGIN had residual tumors.All four patients with mucin-

Beamforming and fronthaul compression design for intelligent reflecting surface aided cloud radio access networks

Owing to the inherent central information processing and resource management ability,the cloud radio access network(C-RAN)is a promising network structure for an intelligent and simplified sixth-generation(6G)wireless network.Nevertheless,to further enhance the capacity and coverage,more radio remote heads(RRHs)as well as high-fidelity and low-latency fronthaul links are required,which may lead to high implementation cost.To address this issue,we propose to exploit the intelligent reflecting surface(IRS)as an alternative way to enhance the C-RAN,which is a low-cost and energy-efficient option.Specifically,we consider the uplink transmission where multi-antenna users communicate with the baseband unit(BBU)pool through multi-antenna RRHs and multiple IRSs are deployed between the users and RRHs.RRHs can conduct either point-to-point(P2P)compression or Wyner-Ziv coding to compress the received signals,which are then forwarded to the BBU pool through fronthaul links.We investigate the joint design and optimization of user transmit beamformers,IRS passive beamformers,and fronthaul compression noise covariance matrices to maximize the uplink sum rate subject to fronthaul capacity constraints under P2P compression and Wyner-Ziv coding.By exploiting the Arimoto-Blahut algorithm and semi-definite relaxation(SDR),we propose a successive convex approximation approach to solve non-convex problems,and two iterative algorithms corresponding to P2P compression and Wyner-Ziv coding are provided.Numerical results verify the performance gain brought about by deploying IRS in C-RAN and the superiority of the proposed joint design.

Phase engineering activation of low-cost iron-containing sulfide minerals for advanced electrocatalysis

Sustainable energy conversion and storage provide feasible approaches towards green energy solutions and carbon neutralization.The high cost and complex fabrication process of advanced energy nanomaterials,however,has impeded the practical application of emerging sustainable technologies.The direct use of earth-abundant natural minerals which contain active elements for effective catalysis and energy storage should be a promising approach to achieve affordable sustainable energy supply and green fuel generations.Herein,as typical examples of activating natural minerals for electrocatalysis,two common minerals,pyrite and chalcopyrite,are activated via a one-step phase transformation strategy.Through a facile thermal reduction process,the minerals are completely transformed into active pyrrhotite(FeS)and haycockite(Cu_(4)Fe_(5)S_(8))phases.The thermal reduction resulting phase transformation can lead to significant surface disordering and can contribute to the catalytic activity by offering favourable electronic structure for intermediates adsorption,abundant surficial active centres,and substantial surface redox pairs.The activated minerals are examined for hydrogen evolution reaction(HER)and oxygen evolution reaction(OER)catalysis.The obtained haycockite phase delivers the best performance towards acidic HER and alkaline OER.Further phase optimization is performed via loading a low percentage of iridium nanoclusters on the haycockite phase deposited onto a carbon cloth substrate,through which an overpotential as low as 310 mV for achieving 10 mA cmand a small Tafel slope of 55.6 m V dec-1are recorded for alkaline OER.This work demonstrates the feasibility of the direct use of cost-effective natural resources for addressing the current energy-related issues and paves a way to reach affordable practical emerging sustainable technologies.

Predicting disease progression in cirrhotic patients after transjugular intrahepatic portosystemic shunt implantation:A sexstratified analysis

BACKGROUND Transjugular intrahepatic portosystemic shunt(TIPS)has been extensively used to treat portal hypertension-associated complications,including cirrhosis.The prediction of post-TIPS prognosis is important for cirrhotic patients,as more aggressive liver transplantation is needed when the post-TIPS prognosis is poor.AIM To construct a nutrition-based model that could predict the disease progression of cirrhotic patients after TIPS implantation in a sex-dependent manner.METHODS This study retrospectively recruited cirrhotic patients undergoing TIPS implantation for analysis.Muscle quality was assessed by measuring the skeletal muscle index(SMI)by computed tomography.Multivariate Cox proportional hazard models were utilized to determine the association between SMI and disease progression in cirrhotic patients after TIPS implantation.RESULTS This study eventually included 186 cirrhotic patients receiving TIPS who were followed up for 30.5±18.8 mo.For male patients,the 30-mo survival rate was significantly lower and the probability of progressive events was higher(3.257-fold)in the low-level SMI group than in the high-level SMI group.According to the multivariate Cox analysis of male patients,SMI<32.8 was an independent risk factor for long-term adverse outcomes after TIPS implantation.A model was constructed,which involved creatinine,plasma ammonia,SMI,and acute-on-chronic liver failure and hepatic encephalopathy occurring within half a year after surgery.This model had an area under the receiver operating characteristic curve of 0.852,sensitivity of 0.926,and specificity of 0.652.According to the results of the DeLong test,this model outperformed other models(Child-Turcotte-Pugh,Model for End-Stage Liver Disease,and Freiburg index of post-TIPS survival)(P<0.05).CONCLUSION SMI is strongly associated with poor long-term outcomes in male patients with cirrhosis who underwent TIPS implantation.

戊型肝炎病毒(HEV)是导致全球急性病毒性肝炎的重要病因之一,且发病率有逐年上升趋势。部分特殊人群如孕妇、老年人、慢性肝脏疾病如慢性乙型肝炎和肝硬化等及免疫功能低下患者感染HEV后可导致急性、亚急性或慢加急性肝衰竭,病死率高。现从HEV感染的临床特点、HEV相关肝衰竭的发病机制以及诊治进展三个方面进行阐述,提升临床医师对HEV相关肝衰竭的识别和预防能力,改善其临床结局。

西安地区洞桩法地铁车站基础结构选型

结合西安地层特点,依托5座在建洞桩法地铁车站实体工程,分析了不同基础型式的结构承载特性和最不利工况,选取地基承载力、水平抗剪切、抗倾覆、基底抗隆起、结构允许水平位移作为评价基础结构强度及刚度安全性的基本准则,构建了洞桩法基础结构的力学模型,推导了不同破坏模式下结构的安全系数计算公式,提出了适用于西安地区洞桩法基础结构选型理论体系中各项安全系数取值建议,给出了不同地层及埋深条件下单排长桩基础和柱下条形基础的适用范围。研究结果表明:西安地区洞桩法基础结构按地基承载力、水平抗剪切、抗倾覆、基底抗隆起及结构允许水平位移等基本准则计算时,安全系数应分别不小于1.2、1.3、1.3、1.6、1.4;洞桩法的中柱基础结构选型主要受地基竖向承载力准则的影响,采用相同柱跨型式时,柱下条形基础在砂土地层中的适用性优于其在粉质黏土-砂互层和粉质黏土地层中的适用性,而单排长桩基础可通过增加桩长等方式提高结构承载力,理论上在各类地层条件下的适用性均优于柱下条形基础;边桩基础结构需要采用复合准则综合分析其适用性,柱下条形基础结构的适用性受地层条件影响较大,在粉质黏土和粉质黏土-砂互层条件下结构抗倾覆安全性难以满足要求,而单排长桩基础在各项准则下主要受自身结构强度和刚度的影响,在各类地层中的适用埋深无明显差异;西安地区洞桩法地铁车站采用单排长桩基础型式的适用性较好,而柱下条形基础可通过扩大基础宽度等方式来满足结构安全性要求。
