DHA induces adipocyte lipolysis through endoplasmic reticulum stress and the cAMP/PKA signaling pathway in grass carp(Ctenopharyngodon idella)

Docosahexaenoic acid(DHA)is a biologically active fatty acid that reduces the accumulation of lipids.However,the molecular mechanism underlying this process,particularly in fish,is not well understood.Recent studies show that endoplasmic reticulum(ER)stress triggers the activation of the unfolded protein response,which has been revealed to play an essential role in lipid metabolism.In this study,we explored the effect of DHA on ER stress and investigated the potential molecular mechanisms underlying DHA-induced adipocyte lipolysis in grass carp(Ctenopharyngodon idella)both in vivo and in vitro.We found that DHA remarkably reduced the triglyceride content,increased the secretion of glycerol,pro-moted lipolysis in adipocytes and evoked ER stress,whereas inhibiting ER stress using 4-phenyl butyric acid(4-PBA)inhibited the effects of DHA(P<0.05).These results implied that ER stress potentially participates in DHA-induced adipocyte lipolysis.Additionally,STF-083010,a specific inositol-requiring enzyme 1a(IRE1a)-inhibitor,attenuated the effects of DHA on lipolysis,demonstrating that IRE1a and X-box binding protein 1 potentially participate in DHA-induced lipolysis.DHA also activated the cyclic adenosine monophosphate(cAMP)-dependent protein kinase A(PKA)pathway by increasing the level of cAMP and activating the PKA enzyme(P<0.05).Nevertheless,H89,a PKA inhibitor,weakened DHA-induced lipolysis by inhibiting the cAMP/PKA signaling pathway.Furthermore,inhibiting ER stress us-ing 4-PBA also inhibited lipolysis and alleviated DHA-induced activation of the cAMP/PKA signaling pathway,suggesting that ER stress may participate in DHA-induced lipolysis through the activation of the cAMP/PKA signaling pathway.Our data illustrate that DHA supplementation can be a promising nutritional strategy for ameliorating lipid accumulation in grass carp.The present study elucidated the molecular mechanism for DHA-induced lipolysis in grass carp adipocytes and emphasized the impor-tance of ER stress and the cAMP/PKA pathway in DHA-induced lip

Aiming at the poor stability of attitude control of mobile robot mechanism, a kind of attitude stabilitycontrol system of mobile robot mechanism based on nanodisplacement sensor is designed.In the hardware part, a hydraulic drive is used to control the action posture of the mobile robot,a nanodisplacement sensor is used to collect the walking data of the robot, and serial communicationof the upper computer is used to convert the data into electrical signals to realise therobot posture control. In the software part, the mathematical coordinate system of robot walkingis constructed, and the rotating posture of the robot is controlled by the Euler angle. Theexperimental results show that the control performance of the designed system is stable andthe control precision is high, which can realise the attitude stabilisation control of the mobilerobot.

Digital twins for wide-areas(DT-WA)can model and predict the physical world with high fidelity by incorporating an artificial intelligence(AI)model.However,the AI model requires an energy-consuming updating process to keep pace with the dynamic environment,where studies are still in infancy.To reduce the updating energy,this paper proposes a distributed edge cooperation and data collection scheme.The AI model is partitioned into multiple sub-models deployed on different edge servers(ESs)co-located with access points across wide-area,to update distributively using local sensor data.To reduce the updating energy,ESs can choose to become either updating helpers or recipients of their neighboring ESs,based on sensor quantities and basic updating convergencies.Helpers would share their updated sub-model parameters with neighboring recipients,so as to reduce the latter updating workload.To minimize system energy under updating convergency and latency constraints,we further propose an algorithm to let ESs distributively optimize their cooperation identities,collect sensor data,and allocate wireless and computing resources.It comprises several constraint-release approaches,where two child optimization problems are solved,and designs a largescale multi-agent deep reinforcement learning algorithm.Simulation shows that the proposed scheme can efficiently reduce updating energy compared with the baselines.

In order to find viable alternative protein sources for aquaculture,we evaluated the effect of partial or complete replacement of dietary soybean meal with yellow mealworm(TM)on the flesh quality of grass carp.In this study,180 grass carp(511.85±0.25 g)were fed 3 experimental diets in which 0%(CN),30%(YM30)and 100%(YM100)dietary soybean meal was replaced by TM for 90 d.The results showed that growth performance,biological parameters and serum antioxidant capacity of grass carp were not affected by dietary TM(P>0.05).Both muscle and whole body crude protein were obviously promoted with the increase of dietary TM(P0.05),indicating that food safety was not influenced by TM.Dietary TM improved muscle textural characteristics by elevating adhesiveness,springiness and chewiness in YM100(P<0.05).In addition,the muscle tenderness was significantly increased by declining the shear force(P<0.05).The muscle fiber density in YM30&YM100 and length of dark bands and sarcomeres in YM100 were obviously increased(P<0.05).The expression of myf5,myog and myhc exhibited a significant upward trend with the increase of dietary TM(P<0.05),which promoted fiber density,length of sarcomere and texture of grass carp muscle.According to the results of metabolomics,the arachidonate(ARA)and eicosapentaenoic acid(EPA)were notably elevated in YM30 and YM100,which indicated that the improvement of flesh quality of grass carp may contribute to the dietary TM influence on muscle lipid metabolism,especially the polyunsaturated fatty acids.In conclusion,TM can completely replace dietary soybean meal and improve the nutritional value of grass carp.

Adaptive bitrate video streaming(ABR)has become a critical technique for mobile video streaming to cope with time-varying network conditions and different user preferences.However,there are still many problems in achieving high-quality ABR video streaming over cellular networks.Mobile Edge Computing(MEC)is a promising paradigm to overcome the above problems by providing video transcoding capability and caching the ABR video streaming within the radio access network(RAN).In this paper,we propose a flexible transcoding strategy to provide viewers with low-latency video streaming services in the MEC networks under the limited storage,computing,and spectrum resources.According to the information collected from users,the MEC server acts as a controlling component to adjust the transcoding strategy flexibly based on optimizing the video caching placement strategy.Specifically,we cache the proper bitrate version of the video segments at the edge servers and select the appropriate bitrate version of the video segments to perform transcoding under jointly considering access control,resource allocation,and user preferences.We formulate this problem as a nonconvex optimization and mixed combinatorial problem.Moreover,the simulation results indicate that our proposed algorithm can ensure a low-latency viewing experience for users.

A 90-day feeding trial was conducted to assess the effects of black soldier fly larvae meal(BSFLM)as a replacement for soybean meal(SM)on growth performance and flesh quality of grass carp.A total of 420grass carp(299.93±0.85 g)were randomly divided into 7 groups(triplicate)and fed 7 diets with SM substitution of 0%(SM,control),15%(BSFLM15),30%(BSFLM30),45%(BSFLM45),60%(BSFLM60),75%(BSFLM75)and 100%(BSFLM100)by BSFLM.The growth performance of grass carp in the BSFLM75 and BSFLM100 groups were significantly lower compared to other groups(P<0.05).The mid-gut villus height was the lowest in the BSFLM100 group(P<0.05).Muscle nutritional value was improved due to increased DHA(docosahexaenoic acid),EPA(eicosapentaenoic acid),total HUFA(highly unsaturated fatty acids)and glycine levels,and reached the optimum in the BSFLM100 group(P<0.05).According to the results of principal component analysis and weight analysis of muscle texture and body color,all the BSFLM diets except BSFLM15 could improve muscle texture and body color and reached the optimum level in the BSFLM100 group.Muscle drip loss and hypoxanthine content were the lowest and muscle antioxidant capacity was the highest in the BSFLM75 group,and water-and salt-soluble protein contents reached the optimum level in the BSFLM60 group(P<0.05).Dietary BSFLM significantly reduced muscle fiber area and diameter,and increased muscle fiber density and the proportion of small fiber(diameter<20μm)(P<0.05).Additionally,sarcomere lengths in the BSFLM75 and BSFLM100 groups were significantly higher than that in the SM group(P<0.05).The m RNA relative expression levels of Myo D,Myf5,My HC and FGF6b were remarkably up-regulated at an appropriate dietary BSFLM level(P<0.05).In conclusion,BSFLM could replace up to 60%SM without an adverse effect on growth performance and improve the flesh quality of grass carp.The optimum levels of dietary BSFLM were 71.0 and 69.1 g/kg diet based on the final body weight and feed conversion ratio.The flesh quality was optimal when dietary SM was

Due to its unique electronic properties,the difluoromethylene group(CF2)has served as a valuable unity in the design of biologically active molecules.Sinceγ-lactones display a broad range of biological properties,α,α-difluoro-γ-lactones may exhibit unexpected biological activities,and thus their synthesis has received increasing attention.Traditional synthetic methods suffer from tedious multi-step processes,and very few effective methods have been reported recently.Herein,we describe the difunctionalization of alkenes with BrCF_(2)CO_(2)K under photoredox catalysis with the use of a boron-Lewis acid for the access toα,α-difluoro-γ-lactones.In this transformation,the alkene substrates and the used reagents,including BrCF_(2)CO_(2)K and the boron-Lewis acid,PhB(OH)2 or BF3·THF,are cheap and widely available.High efficiency and atom economy may make this protocol attractive.

We used Takydromus septentrionalis,a sexually size-monomorphic lacertid lizard,as a model system to test the hypothesis that sexual size monomorphism may evolve in lizards where reproductive performance is maximized at a similar body size for both sexes.We allowed lizards housed in laboratory enclosures to lay as many clutches(for females)as they could or to mate as many times(for males)as they could in a breeding season.Size-assortative mating was weak but evident in T.septentrionalis,as revealed by the fact that male and female snout–vent lengths(SVLs)in mating pairs were significantly and positively correlated.Mating frequency(indicative of male reproductive performance)varied from 1 to 8 per breeding season,generally increasing as SVL increased in adult males smaller than 67.4 mm SVL.Clutch frequency varied from 1 to 7 per breeding season,with female reproductive performance(determined by clutch frequency,annual fecundity,and annual reproductive output)maximized in females with a SVL of 68.0 mm.Accordingly to our hypothesis,the reproductive performance was maximized in the intermediate sized rather than the largest individuals in both sexes,and the body size maximizing reproductive performance was similar for both sexes.Future work could usefully investigate other lineages of lizards with sexually monomorphic species in a phylogenetic context to corroborate the hypothesis of this study.

Substrate specificity is a hallmark of enzymatic catalysis.In this work,the biomimetic catalytic oxidation of styrene and cyclohexanone by iron(III)porphyrins and molecular oxygen was carried out,and remarkable differences in efficiency were observed.The specificity of the substrates for biomimetic catalytic oxidation was investigated by kinetics and mechanistic studies.Kinetics studies revealed that the oxidation of styrene followed Michaelis-Menten kinetics with KM=8.99 mol L^(-1),but the oxidation of cyclohexanone followed first-order kinetics with kobs=1.46×10^(-4) s^(-1),indicating that the styrene epoxidation by metalloporphyrins exhibited characteristics of enzyme-like catalysis,while the oxidation of cyclohexanone was in agreement with the general rules of chemical catalysis.Different catalytic mechanisms for the two substrates were discussed by operando electron paramagnetic resonance spectroscopy,operando UV-vis spectroscopy,and KI/starch experiments.Substrate specificity was concluded to be attributed to the stability of high-valence species and oxygen transfer rate.
