AIM To study cancer hotspot mutations by next-generation sequencing(NGS) in stool DNA from patients with different gastrointestinal tract(GIT) neoplasms. METHODS Stool samples were collected from 87 Finnish patients diagnosed with various gastric and colorectal neoplasms, including benign tumors, and from 14 healthy controls. DNA was isolated from stools by usingthe PSP~? Spin Stool DNA Plus Kit. For each sample, 20 ng of DNA was used to construct sequencing libraries using the Ion AmpliS eq Cancer Hotspot Panel v2 or Ion AmpliS eq Colon and Lung Cancer panel v2. Sequencing was performed on Ion PGM. Torrent Suite Software v.5.2.2 was used for variant calling and data *** NGS was successful in assaying 72 GIT samples and 13 healthy controls, with success rates of the assay being78% for stomach neoplasia and 87% for colorectal tumors. In stool specimens from patients with gastric neoplasia, five hotspot mutations were found in APC,CDKN2 A and EGFR genes, in addition to seven novel mutations. From colorectal patients, 20 mutations were detected in AKT1, APC, ERBB2, FBXW7, KIT, KRAS,NRAS, SMARCB1, SMO, STK11 and TP53. Healthy controls did not exhibit any hotspot mutations, except for two novel ones. APC and TP53 were the most frequently mutated genes in colorectal neoplasms, with five mutations, followed by KRAS with two *** was the most commonly mutated gene in stools of patients with premalignant/benign GIT *** Our results show that in addition to colorectal neoplasms,mutations can also be assayed from stool specimens of patients with gastric neoplasms.

Objective:To investigate the hormonal,histopathological toxicity and sperm quality of ciprofloxacin in male genital system in the rabbit ***:Twenty adult New Zealand male rabbits were randomly divided into the treatment and control groups,and respectively received 30 mg/kg/day ciprofloxacin and normal saline intraperitoneally for 14 *** samples and testicular tissues were taken for testosterone by enzyme linked immunosorbent assay method,and measurement of histopathologic and histomorphometric investigations was made on the 14th day of the experiment and 56 days after the last dose of ***,epididymis sperm sample was collected for quality ***:On day 14 of the experiment,histopathologic examination revealed severe degenerative changes in the epithelium of seminiferous tubules and loss of all germ cells in most tubes in the treatment *** the histomorphometric study,significant reductions in the seminiferous tubules diameter,epithelium height,and Johnsen’score were *** day 56 after the last dose of ciprofloxacin,the seminiferous tubules revealed regeneration in the treatment group and no significant difference was observed in the spermatogenesis parameters except epithelial height parameter between the control group and the treatment *** sperm quality parameters were significantly decreased on day 56 after the last dose of *** levels did not significantly change during the follow-ups ***:There is potential reversible testicular toxicity for ciprofloxacin in the rabbit model in according to relative regeneration of spermatogenic epithelium after 56 days of last dose of *** quality will be improved with a delay after complete regeneration of seminiferous tubules.

A new pyridine derivative ligand, (E)-(Pyridine-2-ylmethylidene) ({2-(E)-(Pyridine-2-ylmethylidene) amino] ethyl} has been synthesized and kf value of it’s complexes with Cu2+, Ni2+, Cd2+, Zn2+, Co2+, Hg2+ and Ag+ has been determined spectrophometrically. The stability of the complexes to vary in acetonitrile solvent was in the order of Cu2+ > Ni2+ > Cd2+ > Zn2+ > Co2+ > Hg2+ > Ag+, thus because this ligand have good selectivity to copper ion, a simple, reliable and rapid method for preconcentration and determination of the ultratrace amount of copper using octadecyl silica membrane disk modified by this ligand, and determination by flame atomic absorption has been presented. Various parameters including pH of aqueous solution, flow rates, the amount of ligand and the type of stripping reagent were optimized. Under optimum experimental conditions, the breakthrough volume is greater than 2000 ml with an enrichment factor of more than 400 and 0.054 μg×L?1 detection limit. The capacity of the membrane disks modified by 6 mg of the ligand has been found to be330.17 gof copper. The effects of various cationic interferences on the percent recovery of copper ion were studied. The method has been successfully applied for the determination of copper ion in different water samples.

Background: A genital outflow tract obstruction is an uncommon cause of primary amenorrhea. If ovulation occurs, menstrual bleeding is prevented. Patients typically present with abdominal/pelvic pain due to hematocolpos. Absence of significant hematocolpos could indicate a secondary source of primary amenorrhea and be challenging to the clinical diagnosis. Case: 17 year-old patient with primary amenorrhea, appropriate Tanner staging secondary sex characteristics, and transverse vaginal septum presents with virtual absence of hematocolpos. After vaginal septum resection, the patient began menstruating, although only evidenced by two cycles of vaginal spotting. Conclusion: Significant hematocolpos is an expected sequella of distal outlet obstruction when collated with secondary sexual characteristics. Absence of such along with suboptimal return of menstruation reflects pathophysiology which may be attributed to a coexistent disorder of the hypothalamus or higher central nervous system function.

An important factor in the course of daily medical diagnosis and treatment is understanding patients’ emotional states by the caregiver physicians. However, patients usually avoid speaking out their emotions when expressing their somatic symptoms and complaints to their non-psychiatrist doctor. On the other hand, clinicians usually lack the required expertise(or time) and have a deficit in mining various verbal and non-verbal emotional signals of the patients. As a result, in many cases, there is an emotion recognition barrier between the clinician and the patients making all patients seem the same except for their different somatic symptoms. In particular, we aim to identify and combine three major disciplines(psychology, linguistics, and data science) approaches for detecting emotions from verbal communication and propose an integrated solution for emotion recognition support. Such a platform may give emotional guides and indices to the clinician based on verbal communication at the consultation time.
