Beneficial Effect of Berberis amurensis Rupr. on Penile Erection

Objective： To investigate whether the methanol extract of Berberis amurensis Rupr. （BAR） augments penile erection using in vitro and in vivo experiments. Methods： The ex vivo study used corpus cavemosum strips prepared from adult male New Zealand White rabbits. In in vivo studies for intracavernous pressure （ICP）, blood pressure, mean arterial pressure （MAP）, and increase of peak ICP were continuously monitored during electrical stimulation of Sprague-Dawley rats. Results： Preconstricted with phenylephrine （PE） in isolated endothelium- intact rabbit corus cavernosum, BAR relaxed penile smooth muscle in a dose-dependent manner, which was inhibited by pretreatment with NG-nitro-L-arginine methyl ester （L-NAME）, a nitric oxide synthase inhibitor, and 1H-[1,2,4]-oxadiazole-[4,3-α]-quinoxalin-l-one, a soluble guanylyl cclase inhibitor. BAR significantly relaxed penile smooth muscles dose-dependently in ex vivo, and this was inhibited by pretreatment with L-NAME 1H-[1,2,4]- oxadiazole-[4,3-α ]-quinoxalin-l-one. BAR-induced relaxation was significantly attenuated by pretreatment with tetraethylammonium （TEA, P〈0.01）, a nonselective K＋ channel blocker, 4-aminopyridine （4-AP, P〈0.01）, a voltage-dependent K＋ channel blocker, and charybdotoxin （P〈0.01）, a large and intermediate conductance Ca2. sensitive-K＋ channel blocker, respectively. BAR induced an increase in peak ICP, ICP/MAP ratio and area under the curve dose dependently. Conclusion： BAR augments penile erection via the nitric oxide/cyclic guanosine monophosphate system and Ca2. sensitive-K＋（BK＋ and IK＋） channels in the corpus cavemosum.

AIM: To examine the potency of 1,2,3,4,6-penta-O-galloyl-β-D-glucose (PGG) as a hepatic heme oxygen-ase-1(ho-1) inducer and its regulation in HepG2 cells. METhoDS: Expression of ho-1 and NF-E2-related factor 2 (Nrf2) and activation of mitogen-activated protein (MAP) kinases were analyzed by Western blot, immuno-fluorescence assay, and flow cytometry. Transfections of ho-1 gene, small interfering RNAs for ho-1 and Nrf2, and dominant-negative gene for MAP/extracellular signal-regulated kinase (ERK) were carried out to dissect the signaling pathways leading to ho-1 expression in HepG 2 cells. RESULTS: PGG up-regulated ho-1 expression and this expression conferred cytoprotection against oxidative injury induced by t-butyl hydroperoxide. Moreover, PGG induced Nrf2 nuclear translocation, which was found to be an upstream step of PGG-induced ho-1 expression, and ERK activation, of which pathway was involved in PGG-induced Nrf2 nuclear translocation, ho-1 expression and cytoprotection. CONCLUSION: PGG up-regulates ho-1 expression by stimulating Nrf2 nuclear translocation in an ERK-depen-dent manner, and ho-1 expression by PGG may serve as one of the important mechanisms for its hepatoprotective effects.

Hwangryunhaedoktang exerts anti-inflammation on LPS-induced NO production by suppressing MAPK and NF-κB activation in RAW264.7 macrophages

OBJECTIVE： This study aimed to evaluate whether Hwangryunhaedoktang （HHT）, a herbal compound, has an inhibitory effect on lipopolysaccharide （LPS）-induced inflammation in RAW264.7 macrophages. METhoDS： The effects of HHT were evaluated by confirming nitric oxide （NO） production and expression of inducible NO synthase （iNOS） and mitogen-activated protein kinases （MAPKs） in LPS-stimulated RAW264.7 macrophages via the Griess assay, Western blotting, and real-time reverse transcription quantitative polymerase chain reaction. Western blot analyses and luciferase assays were used to evaluate whether HHT has an effect on the phosphorylation and translocation of nuclear factor-κB （NF-κB）. The secretion and expression of tumor necrosis factor-α （TNF-α） and interleukin-6 （IL-6） were determined via enzyme-linked immunosorbent assay and Western blot analyses. RESULTS： HHT suppressed LPS-induced NO production and expression of iNOS in a dose-dependent manner. Additionally, MAPKs activation was also attenuated via inhibition of phosphorylation of extracellular signal-regulated kinases 1/2, c-Jun N-terminal kinase and p38 which were related to inflammatory pathway. Furthermore, HHT also effectively attenuated NF-κB activation and its translocation to the nucleus, a process that is closely linked to inflammation. LPS normally induced the expression of inflammatory cytokines such as TNF-α and IL-6, but the secretion and expression of TNF-α and IL-6 were significantly attenuated by pretreating the cells with HHT. CONCLUSION： HHT suppressed LPS-induced NO production by blocking the activation of NF-κB and MAPK signaling pathways in RAW264.7 macrophages. Furthermore, HHT may have an anti-inflammatory effect by suppressing the LPS-induced secretion of TNF-α and IL-6. Therefore, the traditional herbal formula HHT might be a useful potential therapeutic agent for inflammation.

Involvement of heme oxygenase-1 induction in anti-vascular inflammation effects of Xanthoceras sorbifolia in human umbilical vein endothelial cells

OBJECTIVE: To define the effects of Xanthoceras sorbifolia(EXS) on vascular inflammation and the mechanisms in endothelial ***: Vascular protective effects of an ethanol extract of seeds from EXS(1-50 μg/mL) against tumor necrosis factor-α(TNF-α)-induced vascularinflammation were examined in human umbilical vein endothelial cells(HUVECs).RESULTS: EXS significantly decreased TNF-α-induced expression of cell adhesion molecules, such as intracellular adhesion molecule-1, vascular cell adhesion molecule-1, and endothelial cell selectin,in a dose-dependent manner. Pre-treatment with EXS significantly inhibited translocation and transcriptional activity of nuclear factor-κB(NF-κB) increased by TNF-α. EXS also significantly inhibited formation of intracellular reactive oxygen species(ROS). Moreover, the vascular protective effects of EXS were linked to up-regulation of heme oxygenase-1(ho-1) and nuclear factor E2-related factor-2(Nrf-2) expression. EXS-induced ho-1 expression was significantly decreased in SnPP(ho-1 inhibitor)-and ho-1 siRNA-treated cells, whereas an increase was found in cobalt protoporphyrin IX(CoPP)(ho-1 inducer)-treated cells. In addition, pretreatment with EXS increased ho-1 and Nrf-2 expression under TNF-α stimulation with or without N-acetyl-L-cysteine. Furthermore, the inhibitory effects of EXS on TNF-α-induced vascular inflammation were partially reversed in SnPP-and of ho-1siRNA-treated cells but increased by ***: These results suggest that EXS may have important implications for prevention of vascular complications associated with vascular inflammation by inhibition of the NF-κB/ROS pathway and activation of the Nrf-2/ho-1 pathway.

Predicting the Rainfall-Triggered Landslides in a Forested Mountain Region Using TRIGRS Model

Landslides are natural disasters which can pose a serious threat to human and property in many areas around the world. The Transient Rainfall Infiltration and Grid-based Regional Slope-stability (TRIGRS) model was used to investigate the rainfall-induced shallow landslides in a forested mountain region, Korea. Various input data for TRIGRS model include time-varying rainfall, topographic characteristics, soil depth, material strength, and hydraulic properties. A series of calculations were conducted in determining the slope stability over the Jangheung region in Korea during the storm occurred on August 6, 1998. The results show that TRIGRS model captured about 64.1% of landslides that were extracted from the IKONOS2 imageries. The model demonstrated how the factor of safety changed with time during a storm considering both the transient and spatial responses of pore water pressure in its slope stability calculation.

High prevalence of osteoporosis in patients with gastric adenocarcinoma following gastrectomy

AIM： To evaluate the prevalence and predictive factors of osteoporosis in patients with gastric adenocarcinoma alter gastrectomy.METhoDS： The study included 133 patients diagnosed with gastric adenocarcinoma but who did not undergo prior diagnostic work-up for osteoporosis. Bone mineral density （BMD） was measured by dual-energy X-ray absorptiometry （DXA） and vertebral deformity was assessed by plain X-rays. We evaluated the effects of age, sex, body mass index （BMI）, anemia, back pain, vertebral deformity, tumor staging, reconstruction type, and past medical history to determine predictive factors of osteoporosis in these patients.RESULTS： The prevalence of osteoporosis in the lumbar spine was 38.3% （male, 28.9%; female, 54.0%）, and 15.0% in the femoral neck （male, 10.8%; female, 22.0%）. The vertebral deformity rate was 46.6% （male, 43.4%; female, 52.0%）. Age, BMI and hemoglobin correlated with BMD （P 〈 0.01）. In males, anemia and age 〉 64 years were independent predictive factors of osteoporosis in multivariate analysis. In females, back pain was an independent factor for osteoporosis.CONCLUSION： The results of this study revealed that prevalence of osteoporosis and vertebral bone deformity rate were high in gastric cancer patients, regardless of post-gastrectomy duration and operation type. Early diagnosis and a proper management plan must be established in these patients.

Effect of Fe_2P in LiFePO_4/Fe_2P composite on electrochemical properties synthesized by MA and control of heat condition

In order to control the size and distribution of the high conductive Fe2P in LiFePO4/Fe2P composite, two different cooling rates (Fast: 15 ℃·min-1, Slow: 2 ℃·min-1) were employed after mechanical alloying. The discharge capacity of the fast cooled was 83 mAh·g-1 and the slow cooled 121 mAh·g-1. The particle size of the synthesized powder was examined by transmission electron microscopy and distribution of Fe2P was characterized using scanning electron microscopy (SEM). In addition, two-step heat treatment was carried out for better distribution of Fe2P. X-ray diffraction (XRD) and Rietveld refinement reveal that LiFePO4/Fe2P composite consists of 95.77% LiFePO4 and 4.33% of Fe2P.

BACKGROUND The roots of Achyranthes japonica Nakai(AJN),called“Useul-puli,”has been traditionally used to control pain and improve dysfunction in osteoarthritis patients in South *** SUMMARY We described 3 patients diagnosed with herbal medicine induced interstitial lung disease after consuming boiled the roots of *** were referred to our hospital because of the modified Medical Research Council grade 4 *** computed tomography showed bilateral ground-glass opacities with patchy *** treatment with systemic glucocorticoid therapy and discontinuation of the roots of AJN,their symptoms improved,and almost all ground-glass opacities and patchy consolidations on chest radiography and chest computed tomography *** We present three cases of interstitial lung disease induced by the roots of AJN.

Cholesterol is a critical lipid for all mammalian cells,ensuring proper membrane integrity,fluidity,and biochemical *** evidence indicates that macrophages rapidly and profoundly reprogram their cholesterol metabolism in response to activation signals to support host defense ***,our understanding of the molecular details underlying how and why cholesterol homeostasis is specifically reshaped during immune responses remains less well *** review discusses our current knowledge of cellular cholesterol homeostatic machinery and introduces emerging concepts regarding how plasma membrane cholesterol is partitioned into distinct *** then discuss how proinflammatory signals can markedly reshape the cholesterol metabolism of macrophages,with a focus on the differences between MyD88-dependent pattern recognition receptors and the interferon signaling *** also discuss recent work investigating the capacity of these proinflammatory signals to selectively reshape plasma membrane cholesterol *** examine how these changes in plasma membrane cholesterol metabolism influence sensitivity to a set of microbial pore-forming toxins known as cholesterol-dependent cytolysins that specifically target cholesterol for their effector *** also discuss whether lipid metabolic reprogramming can be leveraged for therapy to mitigate tissue damage mediated by cholesterol-dependent cytolysins in necrotizing fasciitis and other related *** expect that advancing our understanding of the crosstalk between metabolism and innate immunity will help explain how inflammation underlies metabolic diseases and highlight pathways that could be targeted to normalize metabolic homeostasis in disease states.

Acheivement of Nano-Scale SiGe Layer with Discrete Ge Mole Fraction Profile Using Batch-Type HVCVD

The strained Si grown on the relaxed SiGe-on-insulator C-MOSFET's is a promising device for the future system LSI devices with the design rule of sub-micron. The achievement of the discrete Ge mole fraction in the SiGe layer is a key engineering in low-temperature SiGe epitaxial growth using HVCVD. The pre-flow of GeH4 gas enhanced the Ge mole fraction and SiGe layer thickness. In addition, the Ge mole fraction and SiGe layer thickness increases with the gas ratio of GeH4/SiH4 + GeH4, process temperature, and gas flow time. however, the haze was produced if the Ge mole fraction is above 22wt%. The discrete-like Ge mole fraction with 22 wt% in 10 nm SiGe layer was obtained by the pre-flow of GeH4 for 10 s, the mixture gas ratio of GeH4/SiH4 + GeH4 of 67%, and the gas flow time for 150 s at the process temperature of 550 C.
