Nicotine Treatment Induces Expression of Matrix Metalloproteinases in Human Osteoblastic Saos-2 Cells

Tobacco smoking is an important risk factor for the development of severe ***,we showed that nicotine affected mineralized nodule formation,and that nicotine andlipopolysaccharide stimulated the formation of osteoclast-like cells by increasing production of macrophagecolony-stimulating factor （M-CSF） and prostaglandin E2（PGE2） by human osteoblastic Saos-2 *** thepresent study,we examined the effects of nicotine on the expression of matrix metalloproteinases （MMPs）,tissue inhibitors of matrix metalloproteinases （TIMPs）,the plasminogen activation system including thecomponent of tissue-type plasminogen activator （tPA）,urokinase-type PA （uPA）,and PA inhibitor type 1（PAI-1）,α7 nicotine receptor,and *** also examined the effect of the nicotine antagonist D-tubocurarineon nicotine-induced expression of *** expression was examined using real-time polymerase chainreaction （PCR） to estimate mRNA *** addition,expression of the MMP,TIMP,uPA,tPA,and PAI-1proteins was determined by Western blotting *** treatment caused expression of MMP-1,2,3,and 13,but not MMP-14,to increase significantly after 5 or 10 d of culture;MMP-14 expression did notchange through day *** of MMP-1 expression by nicotine treatment was eliminated bysimultaneous treatment with *** the presence of nicotine,expression of uPA,PAI-1,orTIMP-1,2,3,or 4 did not change over 14 d of culture,whereas expression of tPA increased significantly byday *** also increased expression of the α7 nicotine receptor and c-fos *** results suggestthat nicotine stimulates bone matrix turnover by increasing production of tPA and MMP-1,2,3,and 13,thereby tipping the balance between bone matrix formation and resorption toward the latter process.

Nano-precision combined process of electrolytic in-process dressing grinding and magnetic assisted polishing on optics glass material

This study is focused on the application of an effective fabrication method combining electrolytic in-process dressing(ELID) grinding and magnetic assisted polishing(MAP) to nano-precision mirror surface grinding on the optics glass-ceramic named Zerodure that is commonly used in precision optics components. The results show the variation of surface roughness after MAP processes utilizing Fe+CeO2, Fe+CeO2+diamond paste and Fe+diamond paste are applied to ELID ground surfaces. The MAP surface roughnesses for ELID ground surface roughnesses(Ra) of 52.1, 39.8 and 51.1 nm using #1200 grinding wheel are improved to 6.1, 4.6 and 1.9 nm, respectively. The surface roughness of MAP process using Fe+CeO2+diamond paste is superior to that using other processes. Moreover, it takes less than 10 min to conduct the MAP processes. The combined method suggested effectively reduces the working time to get the required surface qualities.

Near-infrared imaging survey of faint companions around young dwarfs in the Pleiades cluster

We conducted a near-infrared imaging survey of 11 young dwarfs in the Pleiades cluster using the Subaru Telescope and the near-infrared coronagraph imager. We found ten faint point sources,with magnitudes as faint as 20 mag in the K-band,with around seven *** with the Spitzer archive images revealed that a pair of the faint sources around V 1171 Tau is very red in infrared wavelengths,which indicates very low-mass young stellar ***,the results of our follow-up proper motion measurements implied that the central star and the faint sources do not share common proper motions,suggesting that they are not physically associated.

Immunotherapy for nonalcoholic steatohepatitis using the multiple cytokine production modulator Y-40138

AIM:To investigate the possible use of the multiple cytokine production modulator,Y-40138,as a novel immunotherapy in the rat nonalcoholic steatohepatitis (NASH) model. METHODS:We allocated 6-wk-old male F344 rats to choline-supplemented,L-amino acid-defined (CSAA) diet (control group),CSAA diet + Y-40138 (control + Y-40138 group),choline-def icient,L-amino acid-def ined (CDAA) diet (NASH group),or CDAA diet + Y-40138 (NASH + Y-40138 group). In each group,we measured the plasma alanine aminotransferase (ALT) levels,and the plasma and liver levels of tumor necrosis factor-α (TNF-α),interferon-γ (IFN-γ),and interleukin-10 (IL-10). Tissue specimens of phosphate buffered saline-perfused liver were subjected to hematoxylin and eosin staining,Azan staining,Sirius red staining,and immunohistochemical staining (for Kupffer cells and TNF-α). We then extracted Kupffer cells from the collagenase-perfused livers using the Percoll gradient centrifugation method,and measured the TNF-α levels in the supernatant (in vitro TNF-α production by Kupffer cells) using an enzyme-linked immunosorbent assay ***:In comparison to the NASH group,serumALT elevation was mild,production of serum and liver TNF-α and IFN-γ was inhibited,and IL-10 production was increased in the NASH + Y-40138 group. Amelioration of liver histology was also noted in the NASH + Y-40138 group. Kupffer cell immunohistochemical staining revealed no differences between groups,whereas TNF-α immunohistochemical staining showed fewer stained cells in the NASH + Y-40138 group than in the NASH group. The TNF-α levels in the in-vitro Kupffer cell culture supernatant were lower in the NASH + Y-40138 group than in the NASH ***:Administration of Y-40138 to NASH model rats reduced hepatic inflammation and suppressed fibrosis. These results indicate that the multiple cytokine production modulator,Y-40138,is promising as a novel treatment for NASH.

Innate immune reactivity of the liver in rats fed a choline-deficient L-amino-acid-defined diet

AIM： To investigate the innate immune reactivity of tumor necrosis factor-alpha （TNF-α）, Toll-like receptor 4 （TLR4）, and CD14 in the liver of non-alcoholic steatohepatitis （NASH） model rats. METHODS： Male F344 rats were fed a cholinedeficient L-amino-acid-defined （CDAA） diet. The rats were killed after 4 or 8 wk of the diet, and their livers were removed for immunohistochemical investigation and RNA extraction. The liver specimens were immunostained for TNF-α, TLR4, and CD14. The gene expressions of TNF-α, TLR4, and CD14 were determined by reverse-transcriptase polymerase chain reaction （RT-PCR）. Kupffer cells were isolated from the liver by Percoll gradient centrifugation, and were then cultured to measure TNF-α production. RESULTS： The serum and liver levels of TNF-~ in the CDAA-fed rats increased significantly as compared with the control group, as did the immunohistochemical values and gene expressions of TNF-α, TLR4, and CD14 with the progression of steatohepatitis. TNF-α production from the isolated Kupffer cells of the CDAAfed rats was elevated by lipopolysaccharide stimulation. CONCLUSION： The expressions of TNF-α, TLR4, and CD14 increased in the NASH model, suggesting that

Pollen tube guidance is controlled by multiple complex interactions with the female tissues. Here, we show that pollen tubes of Torenia fournieri are regulated by a stylar tissue in a length-dependent manner to receive and respond to attractant LURE peptides secreted from synergid cells. We developed an immunostaining method to visualize LURE peptides bound at the plasma membrane of the tip region of the pollen tube. Using this method, we found that LURE peptides bound specifically to pollen tubes growing through a cut style. The peptides also bound to pollen tubes growing through a shorter style, which were not competent to respond to these peptides. These observations suggested a possibility that acquisition of the LURE peptide reception ability and acquisition of full competency are separable pro- cesses. RNA-Seq suggested that the transcription profile of pollen tubes was affected by both the length of the style and the cultivation period, consistently with physiological changes in binding activity and LURE response ability. The data- base generated from de novo RNA-Seq of Torenia pollen tubes was shown to be useful to identify pollen tube proteins by mass spectrometry. Our studies provide insight and an effective platform for protein identification to understand pollen tube guidance.

Reversed portal flow: Clinical influence on the long-term outcomes in cirrhosis

AIM: To elucidate the natural history and the longitudinal outcomes in cirrhotic patients with non-forward portal flow(NFPF).METHODS: The present retrospective study consisted of 222 cirrhotic patients(120 males and 102 females; age, 61.7 ± 11.1 years). The portal hemodynamics were evaluated at baseline and during the observation period using both pulsed and color Doppler ultrasonography. The diameter(mm), flow direction, mean flow velocity(cm/s), and mean flow volume(m L/min) were assessed at the portal trunk, the splenic vein, the superior mesenteric vein, and the collateral vessels. The average values from 2 to 4 measurements were used for the data analysis. The portal flow direction was defined as follows: forward portal flow(FPF) for continuous hepatopetal flow; bidirectional flow for to-and-fro flow; and reversed flow for continuous hepatofugal flow. The bidirectional flow and the reversed flow were classified as NFPF in this study. The clinical findings and prognosis were compared between the patients with FPF and those with NFPF. The median follow-up period was 40.9 mo(range, 0.3-156.5 mo).RESULTS: Twenty-four patients(10.8%) demonstrated NFPF, accompanied by lower albumin level, worse ChildPugh scores, and model for end-stage liver disease scores. The portal hemodynamic features in the patients with NFPF were smaller diameter of the portal trunk;presence of short gastric vein, splenorenal shunt, or inferior mesenteric vein; and advanced collateral vessels(diameter > 8.7 mm, flow velocity > 10.2 cm/s, and flow volume > 310 m L/min). The cumulative incidence rates of NFPF were 6.5% at 1 year, 14.5% at 3 years, and 23.1% at 5 years. The collateral vessels characterized by flow velocity > 9.5 cm/s and those located at the splenic hilum were significant predictive factors for developing NFPF. The cumulative survival rate was significantly lower in the patients with NFPF(72.2% at 1 year, 38.5% at 3 years, 38.5% at 5 years) than in those with forward portal flow(8

Surgical removal of a large mobile left ventricular thrombus via left atriotomy

Left ventricular(LV) thrombus is a life-threatening complication of severe LV dysfunction. Ventriculotomy has been a commonly performed procedure for LV thrombus; however, it often further decrease LV function after surgery. We present an alternative approach to thrombectomy in order to minimize the postoperative LV dysfunction. A 37-year-old female with a postpartum cardiomyopathy found to have poor LV function and a large left ventricular apical thrombus(3 cm × 3 cm) attached to the apex by a narrow stalk. Given her severe LV dysfunction, the LV thrombus was approached via left atriotomy under cardiopulmonary bypass. The LV thrombus was easily extracted with gentle traction via the mitral valve. Postoperatively, the patient was discharged home without any embolization event or inotropic support. LV thrombectomy via left atriotomy through the mitral valve could be an alternative option for the patients with poor LV function with a mobile LV thrombus.

Overexpression of INCREASED CAMBIAL ACTIVITY, a putative methyltransferase, increases cambial activity and plant growth

Cambial activity is a prerequisite for secondary growth in plants; however, regulatory factors controlling the activity of the secondary meristem in radial growth remain elusive. Here, we identified INCREASED CAMBIAL ACTIVITY （ICA）, a gene encoding a putative pectin methyltransferase, which could function as a modulator for the meristematic activity of fascicular and interfascicular cambium in Arabidopsis. An overexpressing transgenic line, 35S：＇1CA, showed accelerated stern elongation and radial thickening, resulting in increased accumulation of biomass, and increased levels of cytokinins （CKs） and gibberellins （GAs）. Expression of genes encoding pectin methylesterases involved in pectin modification together with pectin methyltransferases was highly induced in 355：：ICA, which might contribute to an increase of methanol emission as a byproduct in 35S：ICA. Methanol treatment induced the expression of GA- or CK-responsive genes and stimulated plant growth. Overall, we propose that ectopic expression of ICA increases cambial activity by regulating CK and GA homeostasis, and methanol emission, eventually leading to stem elongation and radial growth in the inflorescence stem.

Clinical significance of white gastric crypt openings observed via magnifying endoscopy

AIM:To evaluate the relationship between Helicobacter pylori(***)-induced gastritis and white gastric mucosal crypt openings(COs)in the gastric ***:A total of 175 consecutive patients(including 69 patients with gastric cancer)were enrolled in this *** used magnifying endoscopy(ME)to observe the mucosa microsurface of the lesser and greater curvature of the gastric corpus(350 areas in all).We focused on areas with a round pit microstructure(primarily observed in non-atrophied areas)and evaluated the white openings of these gastric *** classified the whiteness of the COs as the"white-edged dark spot"type(consisting of a dark spot bordered by white);the"white"type(pure white with no dark spot);and the"dense white pit(DWP)"type(dense white,resembling a snowball).Gastritis was also histologically evaluated according to the updated Sydney ***:We detected round COs using ME in 246 of the 350 areas *** histological examination showed significantly more mononuclear cells and neutrophil infiltration in the"white"and"DWP"types than the"white-edged dark spot"type(P<0.001).Furthermore,significantly high-grade inflammation and evidence of active ***-induced gastritis was observed in the"DWP"type(P<0.001).Significant differences were observed in the whiteness of COs between ***-positive(n=139)and negative(n=36)patients(P<0.001).The sensitivity and specificity of the"white"and"DWP"types for predicting *** infection were78.5%and 81.7%,*** the patients with gastric cancer,22.5%(18/80)had"white-edged dark spots",51.3%(41/80)had"white"COs,and 26.3%(21/80)had"DWP"-type COs."DWPs"were frequently observed among patients with undifferentiated gastric cancer[45.7%(16/35)].CONCLUSION:CO whiteness detected via ME was associated with histological evidence of gastritis and helps to predict the severity of inflammation and *** activity.
