Effects of Propofol on Glutamate-Induced Calcium Mobilization in Presynaptic Boutons of Rat Hippocampal Neurons

Recent reports have suggested that various general anesthetics affect presynaptic processes in the central nervous system. However, characterizations of the influence of intravenous anesthetics on neurotransmitter release from presynaptic nerve terminals (boutons) are insufficient. Because the presynaptic calcium concentration ([Ca2+]pre) regulates neurotransmitter release, we investigate the effects of the intravenous anesthetic propofol on neurotransmitter release by measuring [Ca2+]pre in the presynaptic boutons of individual dissociated hippocampal neurons. Brain slices were prepared from Sprague–Dawley rats (10 - 14 days of age). The hippocampal CA1 area was isolated with a fire-polished glass pipette, which vibrated horizontally to dissociate hippocampal CA1 neurons along with their attached presynaptic boutons. Presynaptic boutons were visualized under a confocal laser scanning microscope after staining with FM1-43 dye, and [Ca2+]pre was measured using fluo-3 AM dye. Glutamate (3 – 100 μM) administration increased [Ca2+]pre in Ca2+- containing external solution in a concentration-dependent manner. Propofol (3 – 30 μM) dose-dependently suppressed this glutamate (30 μM)-induced increase in [Ca2+]pre in boutons attached to dendrites, but not to the soma or base of the dendritic tree. The large majority of excitatory synapses on CA1 neurons are located on dendritic spines;therefore, propofol may affect glutamate-induced Ca2+ mobilization in excitatory, but not inhibitory, presynaptic boutons. Propofol may possibly have some effect on glutamate-regulated neurotransmitter release from excitatory presynaptic nerve terminals through inhibiting the increase in [Ca2+]pre induced by glutamate.

We present a step-by-step approach for constructing a framework for knowledge process analysis （KPA）. We intend to apply this framework to the analysis of own research projects in an exploratory way and elaborate it through the accumulation of case studies. This study is based on a methodology consisting of knowledge process modeling, primitives synthesis, and reflective verification. We describe details of the methodology and present the results of case studies： a novel methodology, a practical work guide, and a tool for KPA; insights for improving future research projects and education; and the integration of existing knowledge creation theories.

Applying international criminal justice fairly enhances its legitimacy, and international criminal law＇s procedural aspects also must be fair. The universal application of international criminal law in multiple forums seems to be ensured by means of the proper division of labor between national and international criminal jurisdictions.

Development and Social Implementation of Smartphone App Nige-Tore for Improving Tsunami Evacuation Drills: Synergistic Effects Between Commitment and Contingency

This research explored how we can improve tsunami evacuation behavior,which has been a major social issue since the 2011 Great East Japan Earthquake and *** introduce Nige-Tore,a smartphone app for supporting tsunami evacuation drills,which was developed within an interdisciplinary research ***-Tore serves as an effective interface tool that successfully visualizes the dynamic interactions between human actions(evacuation behavior)and natural phenomena(tsunami behavior).Drill participants can check,on their smartphone,the estimated inundation area of the approaching tsunami,along with their own current evacuation *** results of real-world trials using NigeTore show that the app is more powerful than conventional devices and methods that have been used in tsunami evacuation training,such as hazard maps and traditional drills that do not make use of any apps,because Nige-Tore provides an interface that enables commitment and contingency thinking—which at first glance appear to represent different orientations—to not only coexist but to synergize.‘‘Commitment’’(devotion or involvement)refers to the act of immersing oneself in and viewing as absolute one particular scenario or its potential to be actualized,given conditions in which infinite scenarios may be actualized,depending on the interactions between human systems and natural systems.‘‘Contingency’’thinking(an accidental or incidental state)refers to the act of relativizing and separating oneself from any particular scenario or its potential to be actualized,given the same *** synergistic effect of‘‘commitment’’and‘‘contingency’’thinking also expands people’s capacity to cope with unexpected and unforeseen events.

Scenario analysis for reduction of pollutant load discharged from a watershed by recycling of treated water for irrigation

A model in which a river model was layered on a distributed model （double-layered model） was developed to analyse the transport of water and pollutants （nitrogen, phosphorus, and BOD as organic matter） in watersheds and rivers. The model was applied to the watershed of Abragafuchi Lake, Japan, where serious water pollution has occurred over three decades, and the applicability of the model was demonstrated. Scenarios of recycling of sewage treated-water into agriculture to reduce pollutant load discharged into the lake were analysed. The results showed that irrigating paddy fields with the sewage-treated water could contribute to conserving water and reducing pollutant load, with reduction rate in BOD, nitrogen, and phosphorus ranging from 6%-36%, 16%-46%, and 18%-51%, respectively. Particularly, the results indicated that, irrigating paddy fields with the treated water during non-cropping periods and the accompanying reduction in withdrawn water from the river were more effective in reducing pollutant loads discharged into the lake. Further study is required on the effect of recycled water on crop cultivation and soil conditions for safe implementation.

Water-saving irrigation of paddy field to reduce nutrient runoff

The purpose of this work is to study the effect of a type of water-saving irrigation （WSI） on nutrient runoff of paddy field. The volume of surface drainage was maintained low by WSI. In particular, WSI effectively reduced surface drainage in rain events. Model simulation indicated that net runoff load of total nitrogen （TN） from the paddy field was increased by WSI. Meanwhile, net runoff loads of total phosphorus （TP） and total organic carbon （TOC） from the paddy field was decreased by WSI. Because ponding waters of the study fields were enriched with TP and TOC, WSI reduced runoff of these nutrients by controlling the volume of surface drainage. WSI could be considered an efficient method for reducing runoff loads and could conserve water quality in an agricultural watershed.

Prognostic factors of T4 gastric cancer patients undergoing potentially curative resection

AIM:To investigate the prognostic factors of T4 gastric cancer patients without distant metastasis who could undergo potentially curative resection. METHODS:We retrospectively analyzed the clinical data of 71 consecutive patients diagnosed with T4 gastric cancer and who underwent curative gastrectomy at our *** clinicopathological factors that could be associated with overall survival were *** cumulative survival was determined by the Kaplan-Meier method,and univariate comparisons between the groups were performed using the log-rank *** analysis was performed using the Cox proportional hazard model and a step-wise procedure. RESULTS:The study patients comprised 53 men (74.6%)and 18 women(25.4%)aged 39-89 years (mean,68.9 years).Nineteen patients(26.8%)had postoperative morbidity:pancreatic fistula developed in 6 patients(8.5%)and was the most frequent complication,followed by anastomosis stricture in 5 patients (7.0%).During the follow-up period,28 patients(39.4%)died because of gastric cancer recurrence,and 3(4.2%) died because of another disease or *** all patients,the estimated overall survival was 34.1%at 5 *** analyses identified the following statistically significant prognostic factors in T4 gastric cancer patients who underwent potentially curative resection: peritoneal washing cytology(P<0.01),number of metastatic lymph nodes(P<0.05),and venous invasion(P <0.05).In multivariate analyses,only peritoneal washing cytology was identified as an independent prognostic factor(HR=3.62,95%CI=1.37-9.57)for longterm survival. CONCLUSION:Positive peritoneal washing cytology was the only independent poor prognostic factor for T4 gastric cancer patients who could be treated with potentially curative resection.

Suppression of postprandial blood glucose elevation by buckwheat(Fagpopyrum esculentum) albumin hydrolysate and identification of the peptide responsible to the function

The objective of this study is to evaluate the suppressive effect of buckwheat-albumin hydrolysate on postprandial hyperglycemia and identify the peptide responsible to the function. Buckwheat-albumin hydrolysate was prepared by using digestive enzymes and was orally administered to rats together with soluble starch. The blood was taken from the tail vein up to 90 min after oral administration to measure blood-glucose and plasma-insulin levels. The peptide with α-amylase inhibitory activity was purified from the buckwheathydrolysate by gel-filtration chromatography, α-amylase affinity chromatography, and high performance liquid chromatography(HPLC). The amino-acid sequence of the peptide was identified by a protein sequencer and was compared with that in the buckwheat-genome database. Buckwheat-albumin hydrolysate significantly suppressed the elevation of blood glucose level 15 min after starch administration. The amino-acid sequence of the peptide with α-amylase inhibitory activity was YVEPDCGNLGCCYHC in the parental protein of molecular mass 17.8 k Da and theoretical pI 4.77. The amino-acid sequence, molecular weight, and pI of the parental protein in buckwheat albumin were different from those of α-amylase inhibitor in wheat albumin. This study suggests that the novel α-amylase inhibitor identified in buckwheat albumin is a potential candidate for a functional food material to suppress postprandial blood glucose elevation.

Study on Shock Wave and Turbulent Boundary Layer Interactions in a Square Duct at Mach 2 and 4

In this paper, the outline of the Mach 4 supersonic wind tunnel for the investigation of the supersonic internal flows in ducts was firstly described. Secondly, the location, structure and characteristics of the Mach 2 and Mach 4 pseudo-shock waves in a square duct were investigated by color schlieren photographs and duct wall pressure fluctuation measurements. Finally, the wall shear stress distributions on the side, top and bottom walls of the square duct with the Mach 4 pseudo-shock wave were investigated qualitatively by the shear stress-sensitive liquid crystal visualization method. The side wall boundary layer separation region under the first shock is narrow near the top wall, while the side wall boundary layer separation region under the first shock is very wide near the bottom wall.
