Effect of ALDH2 Genetic Polymorphism on the Adaptive Change in Alcohol Metabolism Due to Continuous Moderate Alcohol Consumption in Humans

Few studies have assessed the difference in adaptive changes of alcohol metabolism in the case of chronic alcohol consumption pertaining to the genetic polymorphism of aldehyde dehydrogenase 2 (ALDH2) in humans. To evaluate the influences of ALDH2 genotypes on changes in alcohol metabolism due to continuous alcohol intake, we conducted an intervention study by setting a continuous drinking period between two abstinence periods. The subjects in this study comprised 20 - 25-year-old males, including 15 males carrying ALDH2*1/*1 and 16 carrying ALDH2*1/*2 genotypes. Following the abstinence period of 4 weeks (from day 1 to day 28), all subjects drank commercially available beer (500 ml) every evening for 6 weeks (from day 30 to day 71) and subsequently abstained from drinking again for 4 weeks (from day 73 to day 100). The next morning, after the end of each period, drinking tests (DTs) were performed on each subject (DT1 on day 29, DT2 on day 72, and DT3 on day 101) to examine alcohol metabolism. The subjects drank shochu (a distilled alcoholic beverage), with an ethanol dose of 0.32 g/kg, within 20 minutes after overnight fasting. The alcohol elimination rate in subjects with ALDH2*1/*1 genotype was significantly higher during DT2 after the drinking period as compared with those at both DT1 and DT3 after the abstinence periods, whereas the elimination rate in subjects with ALDH2*1/*2 genotype did not change significantly during 3 DTs. However, blood acetaldehyde levels significantly decreased in subjects with both ALDH2 genotypes during DT2 as compared with those during DT1. The physiological subjective responses to alcohol also significantly decreased during DT2 in subjects with ALDH2*1/*2 genotype. Moreover, serum lipids, gamma-glutamyltransferase (GGT), and uric acid concentrations also varied between subjects with different ALDH2 genotypes due to continuous drinking. Synthesis of Chiral Schiff Base Metal Complex Inducing CD and Elucidation of Structure of Adsorption on Surface of Gold Nanoparticles

We have prepared supramolecular systems of chiral Schiff base Ni(II), Cu(II), Zn(II) complexes and colloidal gold nanoparticles (AuNP) of 10 nm diameters. They demonstrated that direct adsorption of chiral Schiff base metal complex on the surface of AuNP owing to observation of clear induced CD spectra for the first time. We observed and discussed induced CD bands on AuNP from chiral Schiff base Ni(II), Cu(II), Zn(II) complexes.

目前治疗晚期胃癌的方法为开腹胃切除及D2淋巴结清扫并在术后行S-1或卡倍他滨加奥沙利铂辅助化疗。然而III期胃癌患者的预后并不理想。为了提高生存率,需要加大化疗药物的剂量。鉴于患者接受二联或三联化疗的顺应性差,新辅助化疗将是一个充满前景的方法。目前在东亚国家,正进行多项III期临床试验用于评估新辅助化疗效果。另一方面,手术治疗方法已倾向于行腹腔镜手术。在日本和韩国已经进行了多项III期临床试验评估腹腔镜辅助远端胃切除(LADG)在早期和晚期患者中的疗效。因此,未来标准治疗肿瘤位于中、下1/3晚期胃癌的候选方案由综合治疗组成,包括新辅助化疗及随后的LADG。只要新辅助化疗是按标准实施,随后的腹腔镜胃切除术的可行性、安全性及长期生存率是有保障的。在这个背景下,我们实施了一项随机II期临床试验比较新辅助化疗后LADG和开腹远端胃切除术(ODG)治疗胃癌的疗效。

Search for Signature Inversion in the πi_(13/2) vi_(13/2) Band in Odd-Odd ^(178I)r

The search for the ri_(13/2) vi_(13/2 ) band in ^(178)Ir has been conducted through the ^(152)Sm(^(31) P, 5nγ)^( 178)Ir reaction and the excitation functions, x-γ and γ-γ-t coincidence measurements. Five rotational bands have been newly identified. The low-spin signature inversion in the πi13/2 vi13/2 band has been confirmed by the observations of linking transitions and signature crossing at I = 25.5 h. The inversion phenomenon in rh11/2 vi_(13/2), rh9/2 vi_(13/2) and ri_(13/2) vi13/2 structures in ^(178)Ir provides a unique testing ground for different theoretical interpretations.

Investigation of Patients’ Mood on Their Distance from the Pharmacist during Medication Instruction

Background: Pharmacists must adjust their distance from patients to facilitate communication during interviews and gain their trust. The distance between the patients and the pharmacists varies depending on many factors, such as gender, posture and the patients’ mood. Only a few of these papers have actually measured and validated distance with patients. In this study, we validated our method of assessing mood and measuring distance before beginning a survey with patients. Methods: We measured comfortable interpersonal distance among men and women using an ecological scenario, in which a pharmacist approaches the subject, and the subject is asked to stop the pharmacist at the distance he/she feel comfortable with. Five pharmacists and 33 subjects participated in the study. The Japanese version of the Brief Mood Questionnaire Checklists (BMC-J) was used to quantify the subject’s mood for the day, and then the distance from the pharmacist that the subjects considered comfortable was measured at the bedside. The relationship between the mood and distance obtained was examined. Results: The comfortable distance of subjects was influenced by gender, posture, and mood. The shortest distance was 94.7 ± 11.1 cm (mean ± SD), for the male subjects versus the female pharmacists in the sitting position. The distance of male subjects shorted when they had positive emotions and lengthened when they were worried. Female subjects maintained a long distance from both male and female pharmacists when they had positive emotions and a short distance when they were worried. Conclusion: Findings show that the distance changes depending on the subjects’ mood at the time of measurement. It was found that the present measurement method can be used to determine the psychological state of the patient and measure the comfort distance at that time, and can be used as a simple method to examine these relationships. Assessment of Japanese healthy children’s nutritional status using Waterlow classification

Malnutrition is associated with an increased risk of morbidity and mortality, and may case an undesirable effect on the growth and development in children. There have been several reports about the prevalence of malnutrition in hospitalized children, but reports about malnutrition in a large number of healthy children are quite limited. The aim of this study was to evaluate the prevalence of likelihood of malnutrition in Japanese healthy children. We retrospectively reviewed data of height and weight at a regular health check in 7517 healthy children (age 3 - 17;3747 boys, 3770 girls) during 2008 and 2010 in Nara city, Japan. The data were evaluated using Waterlow classification, i.e., weight-for-height (W/H) and height-for-age (H/A). The prevalence of under-nutrition, as estimated by the values of W/H less than 90% and H/A less than 95%, were 20.1% and 8.3%, respectively, and this prevalence of malnutrition risk tended to vary substantially with age. The peak levels of the prevalence were found to be at around 12 years old for both sexes in W/H, and at around 11 years old for boys and at around 10 years old for girls in H/A. We have clearly demonstrated the existence of certain percentages of the likelihood of malnutrition at different ages in healthy children. These findings suggest that when we want to evaluate the nutritional status using Waterlow classification in hospitalized children, we should be careful by taking such reference values into account.

Individual Differences in Blood Alcohol Concentrations after Moderate Drinking Are Mainly Regulated by Gastric Emptying Rate Together with Ethanol Distribution Volume

Blood alcohol concentration (BAC) differs greatly among individuals, even when people of the same sex and age drink alcohol under the same drinking conditions. In this study, we investigated the main factors involved in the internal reg-ulation of individual differences in BAC, focusing on the alcohol dehydrogenase 1B (ADH1B) genotype, blood acetal-dehyde concentration (BAcH), amount of habitual alcohol consumption, pharmacokinetic parameters of BAC, distribution volume of ethanol (Vd), and gastric emptying rate (GER) under the same drinking conditions. Twenty healthy Japanese males aged between 40 and 59 years old and having the aldehyde dehydrogenase 2 (ALDH2) genotype of ALDH 2*1/*2 were recruited for this study. The subjects were given 0.32 g ethanol/kg body weight in the form of commercially available beer (5%, v/v). The results showed that BAC-max differed greatly among individuals with a more than two-fold variation. When the BAC-time curve was compared among ADH1B genotypes (ADH1B*1/*1, *1/*2, and *2/*2), there were no differences in BAC among the genotypes. Although BAcH, monthly alcohol consumption, elimination rate of blood ethanol (β value) and ethanol disappearance rate from the body (EDR) can affect BAC, all of them had no correlations with BAC-max. However, Vd (liter/kg), ΔPlasma glucose concentration (ΔPGC = PGC30 min ? PGC0 min) and the serum concentration of gastric inhibitory polypeptide (GIP) did correlate with BAC-max. Model 2 in multiple linear regression analysis showed the optimal model for Vd and GIP with positive correlations with BAC-max. As GIP and ΔPGC are both reflected by gastric emptying rate (GER), we concluded that the individual differences in BAC after moderate drinking are mainly regulated by GER together with Vd. These findings demonstrate that together with body water content, the gastrointestinal tract plays an important role in the regulation of individual differences in BAC, involving first pass metabolism of ethanol.

＜正＞According to the statistic of Japan Coast Guard, in 2003 the numbers of marine accidents in Japan’s waters were 2500 or more. The marine accidents mostly occurred within 3 nautical miles from the coastal line or in the port were collision or stranding. Meanwhile 75% of the marine accidents were caused by human error, due to insufficient lookout. To deal with this problem, we suggest the application of image-processing techniques to automate the lookout system. The purpose of this system is to prevent oversight of lookout, therefore the marine accidents can be minimized and the safety and efficiency of maritime vessel traffic can be assured. We proposed a detection method of moving ships through a navigational image sequence taken by cameras installed at ship’s bridge. Since the image is influenced by roll and pitch, the general image processing technique, such as Background Difference method cannot be used. Moreover, in the images classification sea waves appear as noise. So we proposed a novel technique to eliminate the influence of roll, pitch and sea waves in the images. Since the application of the video camera causing insufficient exposure to light during the night, or the detection of ships was difficult under rain condition, this system used the infrared camera together with the video camera. We verified the advantage of using the multi-sensor camera system composed of the video camera with CCD （Charge Coupled Device） sensor and the infrared camera with bolometer sensor under rain condition in the twilight at Tokyo Bay.

Aqueous Components of Tomato Accelerate Alcohol Metabolism by Increasing Pyruvate Level

Consumption of food while drinking alcohol has been suggested to play important roles in alleviating the physiological and pharmacological influences of alcohol. Vegetables are believed to provide health benefits, but there is little evidence for their influence on the effects of alcohol consumption. The present study aimed to investigate the effect of a common vegetable, tomato, on alcohol metabolism. In a randomized, controlled, crossover study with12 Japanese healthy men aged between 24 and 56 years, drinking tomato juice containing 5% (v/v) alcohol (TJAlc) significantly attenuated the elevation of blood ethanol level and subsequently increased the level of acetate compared with a water-based alcoholic beverage with an equal dose of alcohol (0.4 g/kg body weight). Significantly higher levels of blood pyruvate and lactate were also observed in subjects who had consumed TJAlc compared with those consuming the water-based beverage. Additionally, a biphasic alcohol effects scale method showed that subjective feelings for alcohol-induced stimulant effects were significantly enhanced by drinking TJAlc. Animal experiments using male Sprague Dawleyrats suggested that the effect on blood biomarkers was attributable to the serum fraction of tomato (TS), which largely consisted of aqueous compounds, but not lipophilic compounds such as the carotenoid lycopene. Furthermore, it was suggested the TS possibly included potent compound(s) in addition to alanine, glutamine, and citric acid, all of which have previously been reported to affect alcohol metabolism. Administration of TS clearly increased the activity of NAD (H)-dependent enzymes such as lactate-(LDH), alcohol-, and aldehyde-dehydrogenase in rat liver cytosols. These findings suggest that aqueous compound(s) in tomato promote alcohol metabolism, probably through increasing pyruvate level, enhancing LDH activity, and improving the ratio of NAD to NADH.

Organic and Inorganic Pigments from Commercial Inks Used in Eyebrows’ Microblading: Approach for Assessing the Safety Use by Bacterial Reverse Mutation Assay

Background and Objective: The aesthetic products’ safety should not be neglected to the detriment of the market. This study aimed to evaluate the mutagenic potential of commercial inks. It was formulated with organic (P1) and inorganic (P2) pigments for eyebrows microblading using a validated method by regulatory agencies, the Salmonella/microsome assay, to assure the safety of use. Methods: The assay was carried out in three steps: preliminary toxicity, medium without (-S9), and presence (+S9) of metabolic activation. The strains, auxotrophic to histidine (His-) TA97a, TA98, TA100, and TA102, were exposed to both types of inks, in triplicate, compared to negative and positive controls. Data were statistically analyzed, and values with mutagenic index ≥ 2.0 were indicative of mutagenicity. Results: The inks with organic (P1) and inorganic (P2) pigments were not toxic to TA98 and TA100 S. typhimurium tester strains, even at concentrations applied in humans. Both inks were not mutagenic either in the absence or presence of metabolic activation in the tested concentrations, including that applied in humans. The assay showed that P1 and P2 were not direct (-S9) or indirect (+S9) mutagens as commercially formulated. Conclusions: These results indicate that applying these inks on organisms with microsomal enzymes, including humans, is safe since no compound in the inks became more toxic to induce the bacterial reverse mutation.
