Circulating tumor DNA dynamics analysis in a xenograft mouse model with esophageal squamous cell carcinoma

BACKGROUND It remains unclear which factors,such as tumor volume and tumor invasion,influence circulating tumor DNA(ctDNA),and the origin of ctDNA in liquid biopsy is always *** use liquid biopsies clinically,it will be very important to address these *** To assess the origin of ctDNA,clarify the dynamics of ctDNA levels,assess ctDNA levels by using a xenograft mouse after treatment,and to determine whether tumor volume and invasion are related to ctDNA *** Tumor xenotransplants were established by inoculating BALB/c-nu/nu mice with the TE11 cell *** of mice were injected with xenografts at two or four sites and sacrificed at the appropriate time point after xenotransplantation for ctDNA *** of ctDNA was performed by droplet digital PCR,using the human telomerase reverse transcriptase(hTERT)*** Mice given two-site xenografts were sacrificed for ctDNA at week 4 and week *** hTERT was detected at week 4,but it was detected at week ***,in four-site xenograft mice,hTERT was detected both at week 4 and week *** experiments revealed that both tumor invasion and tumor volume were asso ciated with the detection of *** resection experiments,hTERT was detected at resection,but had decreased by 6 h,and was no longer detected 1 and 3 d after *** We clarified the origin and dynamics of ctDNA,showing that tumor volume is an important *** also found that when the tumor was completely resected,ctDNA was absent after one or more days.

Combined treatment of pancreatic cancer xenograft with 90Y-ITGA6B4-mediated radioimmunotherapy and PI3K/m TOR inhibitor

AIM To investigate the therapeutic effect of combined integrin α6β4-targeted radioimmunotherapy(RIT) and PI3 K/m TOR inhibitor BEZ235 in a pancreatic cancer *** Phosphorylation of Akt, m TOR, the downstream effectors eukaryotic initiation factor 4 E binding protein 1(4 EBP1) and S6 ribosomal protein(S6) were evaluated in Bx PC-3 human pancreatic cancer cells treated with Yttrium-^(90)(^(90) Y) labeled anti-integrin α6β4 antibody(ITGA6 B4) and BEZ235 by western blotting. The cytotoxic effect of BEZ235 was investigated using a colony formation assay. Therapeutic efficacy enhancement by oral BEZ235 administration was assessed using mice bearing Bx PC-3 xenograft tumors. Tumor volume measurements and immunohistochemical analyses(cell proliferation marker Ki-67, DNA damage marker p-H2 AX and p-4 EBP1 staining) of tumors were performed for evaluation of combined treatment with ^(90) Y-ITGA6 B4 plus BEZ235, or each arm *** We found that phosphorylation of Akt(p-Akt), 4 EBP1(p-4 EBP1) and S6(p-S6) was inhibited by BEZ235. Colony formation in Bx PC-3 cells was additively suppressed by the combination of ^(90) Y-ITGA6 B4 and BEZ235. Pretreatment with BEZ235 before ^(90) Y-ITGA6 B4 exposure resulted in significant reduction of cells plating efficiency(PE)(0.54 ± 0.11 vs 2.81 ± 0.14 with 185 k Bq/m L ^(90) Y-ITGA6 B4 exposure, P < 0.01; 0.39 ± 0.08 vs 1.88 ± 0.09 with 370 k Bq/m L ^(90) Y-ITGA6 B4 exposure, P < 0.01) when 5 × 10~3 cells per dish were plated. In vivo, the combined treatment with ^(90) Y-ITGA6 B4 plus BEZ235 enhanced the inhibition of tumor growth and statistically significant differences of relative tumor volume were observed for 27 d after the treatment start date when compared with the ^(90) Y-ITGA6 B4 single injection treatment(1.03 ± 0.38 vs 1.5 ± 0.15 at Day 27, P < 0.05), and for 41 d when compared with the BEZ235 treatment alone(1.8 ± 0.7 vs 3.14 ± 1.19 at Day 41, P < 0.05). Characteristics of non-magnetic nanoparticles in magnetically fluidized bed by adding coarse magnets

The fluidization behavior of SiO2, ZnO and TiO2 non-magnetic nanoparticles was investigated in a magnetically fluidized bed （MFB） by adding coarse magnets. The effects of both the amount of coarse magnets and the magnetic field intensity on the fluidization quality of these nanoparticles were investigated. The results show that the coarse magnets added to the bed lead to a reduction in the size of the aggregates formed naturally by the primary nanopartieles. As the macroscopic performances of improved fluidization quality, the bed expansion ratio increases whilst the minimum fluidization velocity decreases with increasing the magnetic field intensity, but for TiO2 nanoparticles there exists a suitable magnetic field intensity of 0.059 6 T. The optimal amounts of coarse magnets for SiO2, ZnO and TiO2 non-magnetic nanoparticles are 40%, 50% and 60% （mass fraction）, respectively. The bed expansion results analyzed by the Richardson-Zaki scaling law show that the exponents depend on both the amount of coarse magnets and the magnetic field intensity.

The results of numerical simulation of the mosaic embedding iron ore sintering （MEBIOS） process according to the model presented in the first part of the study are introduced. The main objective of the second part is the elucidation of key factors influencing the process efficiency, particularly the sintering completeness, velocity of the heat wave propagation and maximum temperature achievable in the pellet during the wave propagation. Numerical results reveal that the mass fraction of coal, diameter of coal particles and air inlet velocity are of prime importance in determining efficiency of the MEBIOS sintering process.

Leaching of phosphorus from incinerated sewage sludge ash by means of acid extraction followed by adsorption on orange waste gel

Ashes from sewage sludge incineration have a high phosphorus content, approximately 8% （W/W）, which indicates a potential resource of the limiting nutrient. Incineration of sewage sludge with subsequent recovery of phosphorus is a relatively new sludge treatment technique. In this article, the leaching of phosphorus by using sulfuric acid as well as hydrochloric acid by means of several batch experiments was presented. At the same time a selective recovery of phosphorus by adsorption was also discussed. The effects of acid concentration, temperature and time on extraction were studied. The phosphorus leaching increased with the increase in acid concentration and temperature. Kinetic studies showed that the complete leaching of phosphorus took place in less than 4 h. Selective adsorption of phosphorus by using orange waste gel provided a hint for recovery of this natural resource, which eventually could meet the ever-increasing requirement for phosphorus. The overall results indicated that the incinerated sewage sludge ash can be treated with acid to efficiently recover phosphorus and thus can be considered a potentially renewable source of phosphorus.

Diurnal Variation in the Vertical Profile of the Raindrop Size Distribution for Stratiform Rain as Inferred from Micro Rain Radar Observations in Sumatra

The diurnal variation in the vertical structure of the raindrop size distribution(RSD)associated with stratiform rain at Kototabang,West Sumatra(0.20°S,100.32°E),was investigated using micro rain radar(MRR)observations from January 2012 to August *** with the MRR data,the RSD from an optical disdrometer and vertical profile of precipitation from the Tropical Rainfall Measuring Mission were used to establish the microphysical characteristics of diurnal *** during 0000–0600 LST and 1800–2400 LST had a lower concentration of small drops and a higher concentration of large drops when compared to rainfall during the daytime(0600–1800 LST).The RSD stratified on the basis of rain rate(R)showed a lower total concentration of drops and higher mass-weighted mean diameter in 0000–0600 LST and1800–2400 LST than in the *** the daytime,the RSD is likely governed by a riming process that can be seen from a weak bright band(BB).On the other hand,during 0000–0600 LST and 1800–2400 LST,the BB was stronger and the rainfall was associated with a higher concentration of midsize and large drops,which could be attributed to more active aggregation right above the melting layer with minimal *** variation in the vertical profile of RSD led to a different radar reflectivity(Z)–R relationship in the rain column,in which Z during the periods 0000–0600 LST and1800–2400 LST was larger than at the other times,for the same R.

Influence of sulfuric-acid-bath pretreatment and soaked in sulfuric acid on surface morphology and photocatalytic activity of titania coatings

Titania(TiO2) photocatalyst coatings have been fabricated by the low-cost approach of sulfuric-acid-bath pretreatment(SAP)followed soaked in sulfuric acid(SA) at room temperature then oxidated in air. The influence of the SAP and soaked conditions on the surface morphology and photocatalytic activity of TiO2 on Ti coatings was investigated. With different SAP conditions,the surface morphologies of the TiO2 on Ti coatings clearly show the formed porous-like structure. With higher SA concentration,the porous-like structure becomes obviously. With extending soaked time,the porous-like structure tends to disappear. Raman spectroscopy reveals that the formed TiO2 coatings are with mixed-phase of anatase and rutile. Compared with those of SA concentration and SAP time,the influence of the soaked time on the phase transformation is obvious. Notably,the photocatalytic activity of TiO2 on Ti coatings had been efficiently enhanced by extending the soaked time,compared with those of higher SA concentration and longer SAP time. The enhanced photocatalytic activity of TiO2 on Ti coatings could be related with the changed surface morphology,mixed-phase of anatase and rutile,and formed hydroxyl groups.
