Mice have frequently been used to model human diseases involving immune dysregulation such as autoimmune and inflammatory *** models help elucidatethe mechanisms underlying the disease and in the development of novel ***,if mice are deficient in certain cells and/or effectors associated with human diseases,how can their functions be investigated in this species?Mucosal-associated invariant T(MAIT)cells,a novel innate-like T cell family member,are a good *** cells are abundant in humans but scarce in laboratory *** cells harbor an invariant T cell receptor and recognize nonpeptidic antigens vitamin B2metabolites from bacteria and *** studies have shown that MAIT cells play a pivotal role in human diseases such as bacterial infections and autoimmune and inflammatory *** cells possess granulysin,a human-specific effector molecule,but granulysin and its homologue are absent in ***,MAIT cells show poor proliferation in *** overcome these problems and further our knowledge of MAIT cells,we have established a method to expand MAIT cells via induced pluripotent stem cells(iP SCs).In this review,we describe recent advances in the field of MAIT cell research and our approach for human disease modeling with iP SCderived MAIT cells.

Dear Editor,Adrenocortical carcinomas and adrenal cysts,including pseudocysts,are uncommon tumors[1,2].No consistent approach to surgery for large adrenal tumors including cysts,has been established,and especially when malignancy is suspected,consideration is still given to the option of *** robot-assisted surgery,we successfully performed the resection of a giant adrenal pseudocyst and a giant adrenocortical carcinoma,as reported previously[3],underlining the advantages of robot-assisted surgery for large adrenal tumors.

During vascular development, procambial and cambial cells give rise to xylem and phloem cells. Because the vascular tissue is deeply embedded, it has been difficult to analyze the processes of vascular development in detail. Here, we establish a novel in vitro experimental system in which vascular development is induced in Arabidopsis thaliana leaf-disk cultures using bikinin, an inhibitor of glycogen synthase kinase 3 proteins. Transcriptome analysis reveals that mesophyll cells in leaf disks synchronously turn into procambial cells and then differentiate into tracheary elements. Leaf-disk cultures from plants expressing the procambial cell markers TDRpro：GUS and TDRpro：YFP can be used for spatiotemporal visualization of procambial cell formation. Further analysis with the tdr mutant and TDIF （tracheary element differentiation inhibitory factor） indicates that the key signaling TDIF-TDR-GSK3s regulates xylem differentiation in leaf-disk cultures. This new culture system can be combined with analysis using the rich material resources for Arabidopsis including cell-marker lines and mutants, thus offering a powerful tool for analyzing xylem cell differentiation.

Deciphering Starch Quality of Rice Kernels Using Metabolite Profiling and Pedigree Network Analysis

The physiological properties of rice grains are immediately obvious to consumers. High-coverage metabolomic characterization of the rice diversity research set predicted a negative correlation between fatty acid and lipid levels and amylose/total starch ratio （amylose ratio）, but the reason for this is unclear. To obtain new insight into the relationships among the visual phenotypes of rice kernels, starch granule structures, amylose ratios, and metabolite changes, we investigated the metabolite changes of five Japonica cultivars with various amylose ratios and two knockout mutants （e 1, a Starch synthase Ilia （SSIIla）-deficient mutant and the SSIIla/starch branching enzyme （BE） double-knockout mutant 4019） by using mass spectrometry-based metabolomics techniques. Scanning electron microscopy clearly showed that the two mutants had unusual starch granule structures. The metabolomic compositions of two cultivars with high amylose ratios （Hoshiyutaka and Yumetoiro） exhibited similar patterns, while that of the double-knockout mutant, which has an extremely high amylose ratio, differed. Rice pedigree network analysis of the cultivars and the mutants provided insight into the association between metabolic-trait properties and their underlying genetic basis in rice breeding in Japan. Multidimensional scaling analysis revealed that the Hoshiyutaka and Yumetoiro cultivars were Indica-like, yet they are classified as Japonica subpopulations. Exploring metabolomic traits is a powerful way to follow rice genetic traces and breeding history.

Heterogeneity in supraglacial debris thickness and its role in glacier mass changes of the Mount Gongga

In the Tibetan Plateau, many glaciers have extensive covers of supraglacial debris in their ablation zones, which affects glacier response to climate change by altering ice melting and spatial patterns of mass loss. Insufficient debris thickness data make it difficult to analyze regional debris-cover effects. Maritime glaciers of the Mount Gongga have been characterized by a substantial reduction in glacier area and ice mass in recent decades. The thermal property of the debris layer estimated from remotely sensed data reveals that debris-covered glaciers are dominant in this region, on which the proportion of debris cover to total glacier area varies from 1.74% to 53.0%. Using a physically-based debris-cover effect assessment model, we found that although the presence of supraglacial debris has a significant insulating effect on heavily debris-covered glaciers, il accelerates ice melting on -10.2% of total ablation zone and produces rapid wastage of -25% of the debris-covered glaciers, leading to the similar mass losses between the debris-covered and debris-free glaciers. Widespread debris cover also facilitates the development of active terminus regions. Regional differences in debris-cover effects are apparent, highlighting the im- portance of debris cover for understanding glacier mass changes in the Tibetan Plateau and other mountain ranges around the world.

Influence of rolling parameters on dynamically recrystallized microstructures in AZ31 magnesium alloy sheets

Conventional rolling experiments via the embedded pin in rolling sheet method were carried out at different reduction rates,starting rolling temperatures,and rolling speeds,and the effects of rolling parameters(i.e.,temperature,equivalent strain,and rolling time)on dynamically recrystallized(DRX)microstructures of AZ31 alloy during hot rolling were studied *** temperature-strain dependence of the high-angle grain boundary fraction(HAGB%)was examined through electron backscattered *** showed that as-rolled microstructures with high HAGB%may be obtained under average rolling temperatures of 270-320℃,equivalent strains higher than 0.8,and a rolling speed of 246 mm/*** results may be related to the DRX kinetics and dynamic recovery which are controlled by deformation temperature and ***%decreased with increasing rolling time(decreasing rolling speed),which is attributed to dynamic recovery,and the recrystallized grain size decreased as rolling time ***,further increases in rolling time increased average grain sizes but decreased mean subgrain sizes;these results are attributed to increases in the low-angle grain boundary(LAGB)length per unit area with rolling *** formation was controlled by dynamic recovery,which consistently follows polygonization or formation of new subgrains inside larger grains;hence,average subgrain sizes decreased with the rolling *** effect of dynamic recovery on HAGB and LAGB formation and their related mechanisms over a wide range of strains and temperatures were discussed in detail.

Hydrochloric acid leaching, chloride evaporation, acetic acid leaching, and biological leaching were evaluated and compared as processes of heavy metal removal for municipal solid waste incineration fly ash(MSWFA). Six factors, namely, energy consumption, process efficiency, process handling, process cost estimation, cost reduction potential, and study progress, were used in order to find out their advantages and disadvantages and to help develop a better recovery process of heavy metals from MSWFA in terms of treatment of the waste material. Hydrochloric acid leaching process was found to be most balanced among the evaluated processes. It showed superiority on energy consumption, process cost estimation, and study progress. On the other hand, despite of its excellency in process efficiency, chloride evaporation process was most unfavorable mainly due to heavy energy dependence. Biological process, with huge potential of cost reduction, was concluded to be the second best process.

Effect of roselle calyx extract on in vitro viability and biofilm formation ability of oral pathogenic bacteria

Objective: To investigate the effect of the roselle calyx extract(RCE)(Hibiscus sabdariffa L.) on the in vitro viability and biofilm formation ability of oral pathogenic bacteria. Methods:RCE was prepared by soaking roselle calyx powder with ethyl alcohol for 24 h at room temperature. After centrifugation, the extract was lyophilized. Then, the extract was dissolved in phosphate-buffered saline, the p H was adjusted, and the extract was aseptically filtered. We used Streptococcus mutans, Streptococcus sanguinis, Lactobacillus casei, Actinomyces naeslundii, Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum, Porphyromonas gingivalis and Prevotella intermedia in this study. The antibacterial activity of the RCE was determined by treating the cells of these bacteria with the extract for 10 or 20 min at room temperature. The minimum inhibitory concentration(MIC) and minimum bactericidal concentration was determined using the micro dilution method, and the effect of the RCE on the ability to form biofilm was determined using a polystyrene micro plate assay. In addition, we used the WST-1 assay to determine the cytotoxicity of the RCE on HGF, Ca9-22 and KB cells. Results: The RCE had antibacterial activity against oral bacteria used in this study. In particular, most significant antibacterial activity was observed against Fusobacterium nucleatum, Prevotella intermedia and Porphyromonas gingivalis. The MIC and minimum bactericidal concentration were 7.2 mg/m L–28.8 mg/m L and 14.4 to >57.6 mg/m L. The RCE had an inhibitory effect on biofilm formation at the MIC and sub-MIC levels. In addition, the RCE had low cytotoxic effects on HGF, Ca9-22 and KB cells. Conclusions: Thus, our results indicate that the RCE may be used for preventing oral diseases.
