Emergence of resistance to carbocyclic oxetanocin G in herpes simplex virus type 1 and genetic analysis of resistant mutants

AIM: To elucidate the potentiality of emergence of drug-resistance to carbocyclic oxetanocin G (***-G ), a new effective antiviral drug for herpetic keratitis during treatment and the mechanism of this drug resistance. METHODS: A ***-G resistant strain (***-Gr) was established by serially propagating the herpes simplex virus (HSV) -1 in African green monkey kidney (VERO) cells in the presence of ***-G. After the drug sensitiv- ity assay and the thymidine kinase (TK) activity assay, the molecular basis for the drug resistance was studied using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) analysis and PCR direct se- quencing technology. RESULTS: After the 10th passage in 10 μm ***-G, the ED50 of the ***-Gr was 17.08-fold greater than that of the original strain on the average and the TK activities of these resistant strains were extremely reduced. PCR-SSCP analysis on TK gene of the wild HSV-1 and the ***-Gr showed altered migration patterns in part 3 and part 4, while PCR-SSCP analysis on DNA polymerase gene showed no difference among the viruses. Sequence analysis revealed a deletion of G at position of 430 that caused frameshift, resulting in premature termination in the TK gene. CONCLUSION: The drug resistance to ***-G may appear during the treatment due to the deficiency of TK activity caused by a single mutation in the TK gene of HSV-1.

Chiral ligand-protected gold nanoclusters: Considering the optical activity from a viewpoint of ligand dissymmetric field

Chirality is a geometric property of a physical, chemical, or biological object, which is not superimposable on its mirror image. Its significant presence has led to a strong demand in the development of chiral drugs, sensors,catalysts, and photofunctional materials. In recent years, chirality of nanoscale organic/inorganic hybrids has received tremendous attention owing to potential applications in chiral nanotechnology. In particular, with the recent progress in the syntheses and characterizations of atomically precise gold nanoclusters protected by achiral thiolates, atomic level origins of their chirality have been unveiled. On the other hand, chirality or optical activity in metal nanoclusters can also be introduced via the surface chiral ligands, which should be universal for the nanosystems. This tutorial review presents some optically-active metal(gold) nanoclusters protected by chiral thiolates or phosphines, and their chiroptical(or circular dichroism; CD) properties are discussed mostly from a viewpoint of the ligand dissymmetric field scheme. The examples are the gold nanoclusters protected by(R)-/(S)-2-phenylpropane-1-thiol,(R)-/(S)-mercaptosuccinic acid, phenylboronateD/L-fructose complexes, phosphine sulfonate-ephedrinium ion pairs, or glutathione. Some methodologies for versatile asymmetric transformation and chiroptical controls of the nanocluster compounds are also *** the dissymmetric field model as the origin of optical activity, the chiroptical responses of the gold nanoclusters are strongly associated with coupled oscillator and/or CD stealing mechanisms based on the concept of induced CD(ICD) derived from a perturbation theory, so on this basis, some characteristic features of the observed CD responses of chiral ligand-protected gold nanoclusters are presented in detail. EXPERIMENTAL STUDY AND MODELING OF PRESSURE LOSS FOR FOAM-CUTTINGS MIXTURE FLOW IN HORIZONTAL PIPE

In this study, we first sought to elucidate foam rheology to describe foam flow behavior, and then to experimentally investigate the pressure losses for both foam and foam-cuttings flow in a horizontal pipe by considering both varied foam qualities of 80%, 85% and 90% and foam velocities. Also, a two-layer numerical model to predict pressure loss was developed based on experimental observations of cuttings behavior. Results show that the foam behaves like a power-law fluid. Furthermore, and the pressure loss significantly increases as foam velocity increases, while the delivered cuttings concentration dramatically decreases. Moreover, results indicate that both the pressure loss and the delivered cuttings concentration increase with foam quality. Comparisons between the experimental results and numerical model predictions show satisfactory agreement.

BACKGROUND Carcinosarcoma(spindle cell carcinoma) of the esophagus is an extremely rare event;the etiology and origins of this neoplasm have not yet been ***-mesenchymal transition(EMT) has been associated with invasion and metastasis, and may be related to the generation of a stem cell population within this *** SUMMARY We present the case of a 61-year-old male with nausea and fever. Upper gastrointestinal endoscopy revealed the presence of type 1 and 0-Ⅱc lesions located 35 cm from the incisors toward the esophago-gastric *** esophagectomy was performed. Macroscopic analysis revealed three polypoid lesions in the abdominal esophagus that accompanied the main lesion in the lower thoracic esophagus and 0-Ⅱc lesions that spread continuously with them. Histologically, the lesions included proliferating spindle cells. Adenocarcinomatous components were detected in a section near the foot, and squamous cell carcinoma was identified in the mucosa at the base of the *** patient was diagnosed with multiple carcinosarcomas, staged at pT1b(SM3),pN1(#110, #7), cM0, Stage Ⅱ(sarcomatous metastasis to the lymph nodes).Spindle cells did not express E-cadherin but were positive for EMT markers,including zinc finger E-box-binding homeobox 1, TWIST, and snail family transcriptional repressor 2. The patient has experienced no recurrence at 5 years and 2 mo after *** This report suggests that multiple sarcomatous tumors may be generated from primary squamous cell carcinoma via mechanisms related to EMT.

Testing of Rice Stocks for Their Survival of Winter Cold

Rice cultivation is considered to be initiated by vegetative propagation of sprout from wild perennial stocks. To test whether any presently cultivated rice cultivar can survive the winter cold or not, rice stocks of several cultivars including indica and japonica types were placed in a shallow pool from October to April in 2015–2016 and 2016–2017. During the coldest period of the winter, the bases of the stocks were placed 5–6 cm below the surface of water, where temperatures ranged from 3 ℃ to 5 ℃, while the surface was frozen for two or three times and covered with snow for a day. Only one cultivar, Nipponbare, a japonica type, survived the winter cold and regenerated sprouts in the end of April or early May. A possibility to develop perennial cultivation of rice or perennial hybrid rice is discussed.

Computer-generated holograms for 3D display(Invited Paper)

We have been studying various types of computer-generated holograms for three-dimensional （3D） displays both for a real-time holographic video display and a hard copy, or a printed hologram. For the hard copy output, we have developed a direct fringe printer, which is achieved to print over 100 gigapixels computer-generated hologram with 0.44μm pitch. In this paper, we introduce our recent progresses on the rainbow hologram, the cylindrical holograms, and the disk hologram for 3D display.
