Direct evidence for dynamics of cell heterogeneity in watercored apples: turgor-associated metabolic modifications and within-fruit water potential gradient unveiled by single-cell analyses

Watercore is a physiological disorder in apple(Malus×domestica Borkh.)fruits that appears as water-soaked tissues adjacent to the vascular core,although there is little information on what exactly occurs at cell level in the watercored apples,particularly from the viewpoint of cell water *** combining picolitre pressure-probe electrospray-ionization mass spectrometry(picoPPESI-MS)with freezing point osmometry and vapor pressure osmometry,changes in cell water status and metabolisms were spatially assayed in the same *** the watercored fruit,total soluble solid was lower in the watercore region than the normal outer parenchyma region,but there was no spatial difference in the osmotic potentials determined with freezing point ***,a disagreement between the osmotic potentials determined with two methods has been observed in the watercore region,indicating the presence of significant volatile compounds in the cellular fluids *** the watercored fruit,cell turgor varied across flesh,and a steeper water potential gradient has been established from the normal outer parenchyma region to the watercore region,retaining the potential to transport water to the watercore ***-specific analysis using picoPPESI-MS revealed that together with a reduction in turgor,remarkable metabolic modifications through fermentation have occurred at the border,inducing greater production of watercore-related volatile compounds,such as alcohols and esters,compared with other *** alcohols including ethanol have low reflection coefficients,it is very likely that these molecules would have rapidly penetrated membranes to accumulate in apoplast to *** addition to the water potential gradient detected here,this would physically contribute to the appearance with high tissue transparency and changes in colour ***,it is concluded that these spatial changes in cell water relations are closely associated with watercore symptom

Advantage of autologous blood transfusion in surgery for hepatocellular carcinoma

AIM: To evaluate the significance of autologous blood transfusion (AT) in reducing homologous blood transfusion (HT) in surgery for hepatocellular carcinoma (HCC). METHODS: The proportion of patients who received HT was compared between two groups determined by the time of AT introduction; period A (1991-1994, n = 93) and period B (1995-2000, n = 201). Multivariate logistic regression analysis was performed in order to identify independent significant predictors of the need for HT. We also investigated the impact of AT and HT on long-term postoperative outcome after curative surgery for HCC. RESULTS: The proportion of patients with HT was significantly lower in period B than period A (18.9% vs 60.2%, P < 0.0001). Multivariate logistic regression analysis identified AT administration as a significant independent predictor of the need for HT (P < 0.0001). Disease-free survival in patients with AT was comparable to that without any transfusion. Multivariate analysis identified HT administration as an independent significant factor for poorer disease-free survival (P = 0.0380). CONCLUSION: AT administration significantly decreased the need for HT. Considering the postoperative survival disadvantage of HT, AT administration could improve the long-term outcome of HCC patients.

Pigmented Villonodular Synovitis of the Spine: Two Case Reports and a Literature Review

Introduction: Pigmented villonodular synovitis (PVNS) of the spine is a rare condition, with only a limited number of cases documented in the medical literature. In this study, we review imaging findings of two cases of PVNS arising in the spine. Case 1: A 52-year-old male presented with right thigh pain of unclear etiology. His condition subsequently deteriorated and he was referred to our hospital. MRI revealed a spinal tumor with high intensity on T1-weighted imaging (T1WI) and iso-intensity on T2WI in the right L2/3 intervertebral foramen. The tumor exhibited heterogeneous enhancement on contrast-enhanced MRI. Computed tomography (CT) myelography showed the presence of an epidural tumor within the right L2/3 intervertebral foramen, situated externally to the spinal canal and compressing the dura mater. Notable scalloping was observed in the posterior margin of the L2 vertebral body on CT images. Case 2: A 78-year-old male presented with a complaint of muscle weakness of the left upper limb. His gait gradually deteriorated, which led to suspicion of cervical myelopathy. A tumor was observed on plain MRI and exhibited low to iso-intensity on T1WI and high intensity on T2WI. The tumor was in the left C7/T1 intervertebral segment and the spinal cord was compressed from the left side. CT showed destruction of the medial aspect of the left C7 vertebral arch. Conclusion: In spinal PVNS, the signal intensity on MRI is dependent on the timing of hemorrhage within the tumor, and there are no distinctive features. However, if an extradural tumor is suspected and CT shows bone erosion, scalloping or osteolytic changes, PVNS should be considered as a differential diagnosis.

Fresh frozen plasma transfusion does not affect outcomes following hepatic resection for hepatocellular carcinoma

AIM:To investigate whether fresh frozen plasma (FFP) transfusion affects outcomes following hepatic resection for hepatocellular carcinoma (HCC) in terms of liver function,postoperative complications and cancer ***:We retrospectively compared the incidence of postoperative complications between 204 patients who underwent hepatectomy for HCC with routine FFP transfusion in an early period (1983-1993,Group A) and 293 with necessity for FFP transfusion during a later period (1998-2006,Group B),and also between two subgroups of Group B [22 patients with FFP transfusion (Group B1) and 275 patients without FFP transfusion (Group B2)].Additionally,only in limited patients in Group B1 and Group B2 with intraoperative blood loss≥ 2000 mL (Group B1 ≥ 2000 mL and Group B2 ≥ 2000 mL),postoperative complications,liver function tests,and cancer prognosis were ***:No mortality was registered in Group B,compared to 8 patients (3.9%) of Group *** incidence of morbidity in Group B2 [23.2% (64/275)] was not significantly different from Group B1 [40.9% (9/22)] and Group A [27.0% (55/204)].The incidence of complications and postoperative liver function tests were comparable between Group B1 ≥ 2000 mL vs Group B2 ≥ 2000 *** prognosis did not correlate with administration of FFP,but with tumor-related ***:The outcome of hepatectomy for HCC is not influenced by FFP *** suggest FFP transfusion be abandoned in patients who undergo hepatectomy for HCC.

This work aims to study the dependence of the antibacterial activity on the crystal plane of Cu. The generation of reactive oxygen species (ROS) on the thin film of Cu with grains oriented in the plane (100) was evaluated by chemiluminescence (CL). The authors proposed the generation mechanism of these three ROS on the outermost surface consisting of Cu2O thin film, CuO layer and bulk Cu.

We describe a polarization rotator based on a parallel-core structure consisting of a silicon nanowire waveguide and a silicon-nitride *** 60-μm-long rotator provides a polarization extinction ratio of more than 10 dB with excess loss of less than 1 *** addition,the extremely wide bandwidth of more than 150 nm expected from calculations is confirmed in experiments.A study of the fabrication tolerance of our rotator indicates that fabrication error of around 25 nm is allowable.

Liver transplant practices in the era of normothermic machine perfusion in the United States

BACKGROUND Normothermic liver machine perfusion(NMP)is a novel technology used to preserve and evaluate the function of liver *** To assess NMP utilization in liver transplant(LT)*** All adult deceased-donor LT recipients between January 2021 and September 2023 in the United States were *** including discard rates,survival,preservation time and timing of surgery were compared between two groups:NMP vs *** Between 2021 and 2023,NMP was utilized in 1493(6.3%)of all LTs in the United *** to non-NMP group,NMP group had lower allograft discard rate(6.5%vs 10%,P<0.001),older recipients’age(median:47 vs 42 years,P<0.001),and higher utilization of donors from donation after circulatory death(DCD)(55%vs 11%,P<0.001).NMP group also had longer distances between recipient and donor hospitals(median:156 vs 138 miles,P<0.001),longer preser-vation time(median:12.2 vs 5.8 hours,P<0.001),and more daytime reperfusion(74%vs 55%,P<0.001).Post-transplant survival outcomes were comparable between the two *** a subgroup analysis of NMP,recipients in the long preservation time(≥8 hours)group had higher daytime reperfusion(78%vs 55%,P<0.001)and similar post-transplant survival when compared to the short preservation time(<8 hours)*** The utilization of NMP is associated with lower discard rates and increased DCD organs for *** allows for prolonging the preservation time and increased occurrence of daytime LT,without any impact on the survival outcomes.

Emergence of resistance to carbocyclic oxetanocin G in herpes simplex virus type 1 and genetic analysis of resistant mutants

AIM: To elucidate the potentiality of emergence of drug-resistance to carbocyclic oxetanocin G (***-G ), a new effective antiviral drug for herpetic keratitis during treatment and the mechanism of this drug resistance. METHODS: A ***-G resistant strain (***-Gr) was established by serially propagating the herpes simplex virus (HSV) -1 in African green monkey kidney (VERO) cells in the presence of ***-G. After the drug sensitiv- ity assay and the thymidine kinase (TK) activity assay, the molecular basis for the drug resistance was studied using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) analysis and PCR direct se- quencing technology. RESULTS: After the 10th passage in 10 μm ***-G, the ED50 of the ***-Gr was 17.08-fold greater than that of the original strain on the average and the TK activities of these resistant strains were extremely reduced. PCR-SSCP analysis on TK gene of the wild HSV-1 and the ***-Gr showed altered migration patterns in part 3 and part 4, while PCR-SSCP analysis on DNA polymerase gene showed no difference among the viruses. Sequence analysis revealed a deletion of G at position of 430 that caused frameshift, resulting in premature termination in the TK gene. CONCLUSION: The drug resistance to ***-G may appear during the treatment due to the deficiency of TK activity caused by a single mutation in the TK gene of HSV-1.

Chiral ligand-protected gold nanoclusters: Considering the optical activity from a viewpoint of ligand dissymmetric field

Chirality is a geometric property of a physical, chemical, or biological object, which is not superimposable on its mirror image. Its significant presence has led to a strong demand in the development of chiral drugs, sensors,catalysts, and photofunctional materials. In recent years, chirality of nanoscale organic/inorganic hybrids has received tremendous attention owing to potential applications in chiral nanotechnology. In particular, with the recent progress in the syntheses and characterizations of atomically precise gold nanoclusters protected by achiral thiolates, atomic level origins of their chirality have been unveiled. On the other hand, chirality or optical activity in metal nanoclusters can also be introduced via the surface chiral ligands, which should be universal for the nanosystems. This tutorial review presents some optically-active metal(gold) nanoclusters protected by chiral thiolates or phosphines, and their chiroptical(or circular dichroism; CD) properties are discussed mostly from a viewpoint of the ligand dissymmetric field scheme. The examples are the gold nanoclusters protected by(R)-/(S)-2-phenylpropane-1-thiol,(R)-/(S)-mercaptosuccinic acid, phenylboronateD/L-fructose complexes, phosphine sulfonate-ephedrinium ion pairs, or glutathione. Some methodologies for versatile asymmetric transformation and chiroptical controls of the nanocluster compounds are also *** the dissymmetric field model as the origin of optical activity, the chiroptical responses of the gold nanoclusters are strongly associated with coupled oscillator and/or CD stealing mechanisms based on the concept of induced CD(ICD) derived from a perturbation theory, so on this basis, some characteristic features of the observed CD responses of chiral ligand-protected gold nanoclusters are presented in detail. We believe that various kinds of origins of chirality found in ligand-protected gold nanoclusters may provide models for understanding those of many related nanomater
