Comparative Investigation of Alternative Negative Staining Reagents in Bacterial Morphological Study

Negative staining is an effective method that can be used for electron microscopic study to observe fine structural morphology without destruction of bacterial structure. Although uranium acetate is used worldwide as a general dyeing solution, it is extremely difficult to use it by a new purchase at a research institution because it falls under the nuclear regulation substance in Japan. Therefore, we examined alternative reagents for negative staining that could replace uranium acetate through bacterial observation with an electron microscope. Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Streptococcus pyogenes were examined by four stain reagents (phosphotungstic acid (PTA), EMstainer, TI blue, and uranium acetate). Pre cultured bacteria were stained with each stain reagents on a copper grid, washed with PBS, and observed with a transmission electron microscope. In the comparison between bacterial structures, the cell wall structure and bacterial flagella could be observed well in the order of PTA, EMstainer, and uranium acetate. With TI blue staining, flagella could be observed very poorly. In comparison between bacteria, gram negative bacteria such as Escherichia coli and Pseudomonas aeruginosa, could be observed well as compared with gram positive cocci such as Staphylococcus aureus and Streptococcus pyogenes. The uranium acetate looked very coarse in background particles. Since crystals tend to precipitate, TI blue also required filtering, and electron beams were absorbed by the agglomerated crystals, and the frequency of electronic burning occurred high frequency. In this study, there was clear difference in the observation conditions depending on the type of bacteria and the kind of the staining reagents. Especially, it was confirmed that good negative staining features of Pseudomonas aeruginosa by electron microscope were obtained by PTA and EMstainer staining. LicT Modulates Biofilm Formation of Streptococcus pyogenes

Streptococcus pyogenes frequently causes purulent infections in humans. Biofilm formation is an important virulence property of S. pyogenes because of decreased susceptibility of bacteria to antibiotic treatment. Biofilm is composed of various types of matrix including glycocalyx which is an important exocellular matrix material related to bacterial sugar metabolism. A putative antiterminator protein, LicT (Spy0571), is one of the components of the glucose-independent β-gluco-side-specific phosphotransferase system (PTS). Although the PTS, a carbohydrate metabolic system, may play a role in biofilm formation, the relationship between LicT and biofilm formation has not yet been elucidated. Here, we evaluated whether LicT affected biofilm formation in modified chemically defined medium (CDMM) supplemented with glucose or β-glucoside:salicin. We created licT- and licT-complemented mutant strains from S. pyogenes 1529. Although the licT mutant strain tended to have higher growth rate than wild-typestrain in CDMM with glucose, it had a significant lower growth rate than the wild-type strain in CDMM with salicin. In addition, the licT mutant exhibited lower biofilm formation in CDMM containing salicin than the wild-type strain by 96 well plate analysis and confocal laser scanning microscopic analysis. Our results suggest that LicT plays an important role in biofilm formation of S. pyogenes.

Admission and Traffic Control Schemes Suitable for QoS Applications in WLAN Systems

This paper proposes the use of admission and traffic control schemes for real-time applications. The admission control scheme determines the admission of high-priority real-time applications such as voice and video streams in terms of their bandwidth utilization time (medium time), whereas the traffic control scheme maintains the communication quality of applications permitted admission by restricting other traffic. Owing to the use of contention-based access, a conventional scheme without admission control will degrade the communication quality when the number of terminals using high-priority applications increases. Moreover, only the capabilities (i.e., frame and sequence procedures) of admission control are defined in the IEEE 802.11e standard;the detailed usage in terms of the application characteristics is not specified, and it may be difficult to achieve a sufficient level of quality of service (QoS). The proposed schemes achieve the optimum QoS for actual services. The software used in the proposed schemes was implemented into hardware at the access point, and was evaluated experimentally. Based on the evaluation results, excellent performances with high QoS applications were obtained.

Study of High Power ICRF Antenna Design in LHD

In large helical device （LHD）, antenna loadings are different for minority ion cyclotron heating （MICH with 38.47 MHz） and mode-converted ion Bernstein wave heating （MC-IBW with 28.4 MHz）, and it is necessary to improve antenna loading with low heating efficiency to avoid arching on transmission line. To design a new ion cyclotron range of frequencies （ICRF） antenna in LHD, calculation for a simple antenna model is conducted using three-dimensional electrical magnetic code （high frequency structure simulator, HFSS） for an water loading as an imaginary plasma with low heating efficiency. At resonant frequencies, antenna loading is sensitive to strap width, and resonant frequencies are strongly related to strap height. There is no differences of RF current profile on the strap surface between resonant frequency and non-resonant frequency. The strap should be perpendicularly placed against the magnetic field line, since Faraday-shield angle will lead to a decrease in the effective antenna height.

Relationship between Nitrogen Atmospheric Deposition, Discharge and Concentration, and Monthly Change of Those in a River

The hypothesis that the product of discharge and concentration of nitrogen (N) in river water is equal to the atmospheric deposition was verified in the mountainous basin of the Tedori River in Japan. To verify this relationship, long-term data?are required to eliminate the effect of short-term variation of the N components. The basin has very high mountains, including Mount Hakusan (2702 maltitude), which is covered with deep snow in winter. Therefore, limited data were used for the estimation of the deposition of the entire basin by assuming a linear relationship of altitude. As a result, it was found that the estimated N concentration coincided well with observed concentrations at six sites—the Shiramine and Kuwajima (upper stream), Nakajima (lower stream) and Dainichi dam, Tedori dam and Senami sites (middle stream). The seasonal variation of N concentrations was low in the snowmelt period and high in autumn through to winter. This was not due to the larger discharge in snowmelt season as it was also found that N deposition was high in winter and low in spring, which indicated a clear relationship between N concentration and monthly atmospheric deposition including N storage in snow pack.

Quantitative Analysis of the Rate of Geochemical Weathering of Sulfur from Sedimentary Rocks Using Atmospheric Deposition, Concentration and River Discharge Data

Quantitative analysis of the rate of geochemical weathering of sulfur (S) from sedimentary rocks (GeoS) was conducted using concentration (Cs) and discharge (Qs) data from the Tedori River and atmospheric deposition (AtdepS) in the basin. First, S fluxes were calculated using 16 years of Cs and Qs data. The annual average discharge of S (TotalS) was estimated at 8597 ton·year-1 (117.3 kg·ha-1·year-1). Of this, 1331 ton·year-1 was AtdepS (18.2 kg·ha-1·year-1) and another 7266 ton·year-1 was GeoS (99.1 kg·ha-1·year-1). Monthly changes in TotalS were investigated, which showed that GeoS was highest in summer, because of the air temperature, while AtdepS peaked in winter because of seasonal wind. Using Qs and AtdepS corrected for altitude, TotalS, AtdepS and GeoS were estimated at six sites, and among these sites we found that the TotalS per unit area values were random, depending on the site characteristics. In particular, the discharge from the Kuwajima site was remarkably high suggesting that the sedimentary rocks at this site had higher pyrite content than at the other sites. Finally, we also assessed the relationship between the characteristics of sedimentary rocks and GeoS in a range of rivers in the Hokuriku Region, and found that there was a close relationship between concentrations of SO42- greater than 10 mg·l-1 and sedimentary rocks containing the pyrite group. In addition, we estimated that the influence of GeoS was present when the concentration of SO42- in river water was greater than 2 - 3 mg·l-1 in the Hokuriku region.

Long-Term Assessment of Nitrogen Pollution Load Potential for Groundwater by Mass Balance Analysis in the Tedori River Alluvial Fan Area, Japan

To evaluate the nitrogen pollution load in an aquifer, a water and nitrogen balance analysis was conducted over a thirty-five year period at five yearly intervals. First, we established a two-horizon model comprising a channel/soil horizon, and an aquifer horizon, with exchange of water between the aquifer and river. The nitrogen balance was estimated from the product of nitrogen concentration and water flow obtained from the water balance analysis. The aquifer nitrogen balance results were as follows: 1) In the aquifer horizon, the total nitrogen pollution load potential (NPLP) peaked in the period 1981-1990 at 1800 t·yr-1;following this the NPLP rapidly decreased to about 600 t·yr-1 in the period 2006-2010. The largest NPLP input component of 1000 t·yr-1 in the period 1976-1990 was from farmland. Subsequently, farmland NPLP decreased to only 400 t·yr-1 between 2006 and 2010. The second largest input component, 600 t·yr-1, was effluent from wastewater treatment works (WWTWs) in the period 1986-1990;this also decreased markedly to about 100 t·yr-1 between 2006 and 2010;2) The difference between input and output in the aquifer horizon, used as an index of groundwater pollution, peaked in the period 1986-1990 at about 1200 t·yr-1. This gradually decreased to about 200 t·yr-1 by 2006-2010. 3) The temporal change in NPLP coincided with the nitrogen concentration of the rivers in the study area. In addition, nitrogen concentrations in two test wells were 1.0 mg·l-1 at a depth of 150 m and only 0.25 mg·l-1 at 50 m, suggesting gradual percolation of the nitrogen polluted water deeper in the aquifer.

Standard chemoradiotherapy(CRT) for local advanced rectal cancer(LARC) rarely induce rectal perforation. Here we report a rare case of rectal perforation in a patient with LARC in the midst of preoperative CRT. A 56-year-old male was conveyed to our hospital exhibiting general malaise. Colonoscopy and imaging tests resulted in a clinical diagnosis of LARC with direct invasion to adjacent organs and regional lymphadenopathy. Preoperative 5-fluorouracil-based CRT was started. At 25 d after the start of CRT, the patient developed a typical fever. Computed tomography revealed rectal perforation, and he underwent emergency sigmoid colostomy. At 12 d after the surgery, the remaining CRT was completed according to the original plan. The histopathological findings after radical operation revealed a wide field of tumor necrosis and fibrosis without lymph node metastasis. We share this case as important evidence for the treatment of LARC perforation in the midst of preoperative CRT.

目的医疗照射的最优化在世界范围内都是辐射防护所关心的重要问题之一,国际辐射防护委员会和国际原子能机构推荐建立显像剂使用的剂量参考水平（DRLs）作为控制医疗照射最优化的工具。因此,根据最新调查资料构建DRLs已成为各核医学相关学会和组织的迫切需要。为构建此DRLs,本研究在全日本范围内调查了核医学成人用显像剂的实际使用剂量。方法调查在2014年11月25日至2015年1月16日间进行,使用调查表方式调查了全日本1249个医疗单位各种显像剂的使用情况,调查结果通过网络反馈收集。结果 516个医疗单位反馈了调查结果,反馈率41%。具体显像包括:99Tcm-MDP和99Tcm-HMDP的骨显像,99Tcm-HM-PAO、99Tcm-ECD和123I-IMP的脑血流显像,99Tcm-Tetrofosmin、99Tcm-MIBI和201Tl-Cl的心肌灌注显像,18F-FDG(自产18F或配送的18F)的肿瘤PET显像。以上显像的第75百分剂量分别是为932、937、763、775、200、831、818、180、235和252 MBq。日本90%以上的医疗单位所使用的显像剂剂量在上述剂量值中位数的50%范围内。很多医疗机构采用包装说明书、文本或指南推荐的剂量,比如740 MBq(99Tcm-MDP和99Tcm-HMDP骨显像)、740 MBq(99Tcm-ECD和99Tcm-HM-PAO脑血流显像)和740 MBq(99Tcm-Tetrofosmin和99Tcm-MIBI心肌灌注显像)等。99Tcm-MDP和99Tcm-HMDP（用于骨显像）、99Tcm-HM-PAO（用于脑血流显像）、99TcmTetrofosmin和99Tcm-MIBI（用于心肌灌注显像）等许多显像剂的使用剂量均在欧盟DRLs内,且大多数在日本使用的显像剂剂量均未超过美国核医学与分子影像学会的标准使用剂量上限。结论本调查结果表明,在日本所使用的各种核医学显像剂的剂量比较集中,各医疗机构均能按照文本和指南要求的剂量越来越严格地使用显像剂。此外,日本显像剂的使用剂量均在欧盟和美国核医学与分子影像学会所规定的使用剂量范围内。
