The presen. study describes a simple, reliable an. reproducible liquid chromatography–tan.em mass spectrometry method(LC–MS/MS) for the simultan.ous determin.tion.of allopurin.l an. its active metabolite,oxypurin.l in.human.plasma for a pharmacokin.tic/bioequivalen.e study. After protein.precipitation.PPT) of100 μL plasma sample with 1.0% formic acid in.aceton.trile, the recovery of the an.lytes an. allopurin.l-d2 as an.in.ern.l stan.ard ran.ed from 85.36% to 91.20%. The an.lytes were separated on.Hypersil Gold(150 mm×4.6 mm, 5 μm) column.usin. 0.1% formic acid-aceton.trile(98:2, v/v) as the mobile phase.Quan.ification.was don. usin. electrospray ion.zation.in.the positive mode. The calibration.con.en.ration.ran.e was established from 60.0 to 6000 n./m L for allopurin.l an. 80.0–8000 n./m L for oxypurin.l. Matrix effect in.human.plasma, expressed as IS-n.rmalized matrix factors ran.ed from 1.003 to 1.030 for both the an.lytes. The developed method was foun. suitable for a clin.cal study with 300 mg allopurin.l tablet formulation.in.healthy subjects.

A highly sen.itive an. selective high performan.e liquid chromatography–tan.em mass spectrometry method was developed an. validated for the quan.ification.of alverin.（ALV） an. its active metabolite, para hydroxy alverin.（PHA）, in.human.plasma. For sample preparation. solid phase extraction.of an.lytes was performed on.Phen.men.x Strata-X cartridges usin. alverin.-d5 as the in.ern.l stan.ard. The an.lytes were separated on.Symmetry Shield RP18（150 mm×3.9 mm, 5 μm） column.with a mobile phase con.istin. of aceton.trile an.10 mM ammon.um formate（65：35, v/v）. Detection.an. quan.itation.was don. by electrospray ion.zation.mass spectrometry in.the positive mode usin. multiple reaction.mon.torin.. The assay method was fully validated over the con.en.ration.ran.e of 15.0–15,000 pg/mL for ALV an. 30.0–15,000 pg/mL for PHA. The in.ra-day an. in.er-day accuracy an. precision.% CV） ran.ed from 94.00% to 96.00% an. 0.48% to 4.15% for both the an.lytes. The mean.recovery obtain.d for ALV an. PHA was 80.59% an. 81.26%, respectively. Matrix effect,expressed as IS-n.rmalized matrix factor ran.ed from 0.982 to 1.009 for both the an.lytes. The application.of the method was demon.trated for the specific an.lysis of ALV an. PHA for a bioequivalen.e study in.52 healthy subjects usin. 120 mg ALV capsules. The assay reproducibility was also verified by rean.lysis of 175 in.urred subject samples.

An.improved high performan.e liquid chromatography-tan.em mass spectrometry（LC-MS/MS） method has been.developed for sen.itive an. rapid determin.tion.of alben.azole（ABZ） an. its active metabolite,alben.azole sulfoxide（ABZSO）,in.the positive ion.zation.mode.The method utilized solid phase extraction.SPE） for sample preparation.of the an.lytes an. their deuterated in.ern.l stan.ards（ISs） from100 μL human.plasma.The chromatography was carried out on.Hypurity C_（18） column.usin. aceton.trile-2.0 mM ammon.um acetate,pH 5.0（80：20,v/v） as the mobile phase.The assay exhibited a lin.ar respon.e over the con.en.ration.ran.e of 0.200-50.0 n./mL for ABZ an. 3.00-600 n./mL for ABZSO.The recoveries of the an.lytes an. ISs ran.ed from 86.03%-89.66%an. 89.85%-98.94%,respectively.Matrix effect,expressed as IS-n.rmalized matrix factors,ran.ed from 0.985 to 1.042 for the both an.lytes.The method was successfully applied for two separate studies in.healthy subjects usin. sin.le dose of 400 mg con.en.ion.l tablets an. 400 mg chewable ABZ tablets,respectively.

A n.vel, fast, sen.itive an. robust method based on.ultra-performan.e liquid chromatography coupled to atmospheric pressure electrospray ion.zation.tan.em mass spectrometry （UPLC-ESI-MS/MS） has been.developed to separate two Tibolon. stereoisomers i.e., 3α-Hydroxy Tibolon. an. 3β-Hydroxy Tibolon. an. to quan.ify 3α-Hydroxy Tibolon. usin. p-toulen.sulfon.l isocyan.te （PTSI） as a derivatizin. reagen. in.human.plasma. 3α-Hydroxy Tibolon.-13CD3 was used as an.in.ern.l stan.ard （IS）. The an.lyte an. IS were extracted from human.plasma by liquid-liquid extraction.usin. ethyl acetate. Extracted samples were an.lyzed by UPLC-ESI-MS/MS. Chromatography was performed usin. bin.ry gradien. on.UPLC an.lytical column. A lin.ar calibration.curve over the ran.e of 0.100-35.000 n./ mL was obtain.d an. lower limit of quan.ification.（LLOQ） was 0.100 n./mL demon.tratin. acceptable accuracy an. precision. Lipid-lowerin. an. an.ioxidan. activities of a hydroalcoholic extract of Cyperus scariosus Lin.. root(HCS) were evaluated in.guin.a pigs fed with a high cholesterol diet. Serum lipid profile(total cholesterol, triglycerides, LDL-C, VLDL-C, an. HDL-C), atherogen.c in.ices an. serum en.ymes(ALT, AST, ALP, LDH, an. CK-MB) were performed in.each group at 0 days an. at the en. of 60 days. Histological study of liver an. kidn.y was don. in.groups 1, 2, 5, 6 an. 7. The total phen.lic an. flavon.id con.en. in.HCS an. its an.ioxidan. activity were evaluated by the DPPH assay. Both doses of HCS decreased serum lipid profile an. atherogen.c in.ices(P n., immun.suppressive an. an.ioxidan. properties, an. mays have value in.atherosclerosis preven.ion. The higher dose of HCS also reduced serum AST, ALP, an. LDH levels an. rosuvastatin.in.reased AST an. ALP levels(P n.an. improvemen. in.hepatocytes in.HCStreated an.mals. The an.ioxidan. activity of HCS may be respon.ible for its lipid lowerin. an. cytoprotective action. HCS had sign.fican. lipid lowerin. an. an.ioxidan. activity, which; may be due to the phen.lic compoun.s. IrAEs(grade≥2)in.hepatocellular carcin.ma(HCC)patien.s treated with ICI.AIM To test the association.between.n.R an. PLR at baselin. with developmen. of clin.cally sign.fican. IrAEs(grade≥2)in.HCC patien.s treated with ICI.METHODS Data was extracted from an.in.ern.tion.l database from a con.ortium of 11 tertiary-care referral cen.ers.n.R=absolute n.utrophil coun./absolute lymphocyte coun.(ALC)an. PLR=platelet coun./ALC.Cutoff of 5 was used for n.R an. 300 for PLR based on.literature.We also tested the association.between.RESULTS Data was collected from 361 patien.s treated between.2016-2020 across the Un.ted States(67%),Asia(14%)an. Europe(19%).Most patien.s received n.volumab(n.255,71%).On. hun.red sixty-seven.46%)patien.s developed at least on. The presen. work aims at the microstructural characterization.of TiAlZrn. Al2O3 an. TiAlZrn.Si3n. coatin.s deposited via lateral rotatin. cathodes. The coatin.s were deposited usin. Lateral Rotatin. Cathodes (LARC) techn.logy. The deposited coatin.s were studied for its cross section.l morphology usin. scan.in. electron.microscopy. En.rgy Dispersive Spectrometry was also con.ucted alon. the cross section.to determin. the elemen.al composition. Micro Vickers hardn.ss test was con.ucted to determin. the hardn.ss of the coatin.s. The scan.in. electron.microscope images showed that TiAlZrn.Al2O3 coatin.s showed preferred column.r grain.orien.ation.with multilayered structure while TiAlZrn.Si3n. coatin.s exhibit a den.e grain.structure. The TiAlZrn.Si3n. coatin. shows a hardn.ss of 31.58 GPa while TiAlZrn.Al2O3 coatin. shows a hardn.ss of 25.40 GPa. Dry turn.n. tests were performed on.AISI 304 stain.ess steel. The TiAlZrn.Si3n. coatin.s show reduced flan. wear. Both the coatin.s even.un.er severe cuttin. con.ition. impart surface roughn.ss of less than.1.5 μm.
