Evaluation of thermostable endoglucanase in Paenibacillus lautus strain BHu3 for yield enhancement

In the present study,Paenibacillus lautus strain BHu3 isolated from landfill soil was evaluated for the presence of potential endoglucanases which are the first candidate of cellulase enzyme system to act on ***-silico analysis revealed high potential thermostable endoglucanases which can efficiently interact with *** most potent and thermostable endoglucanase(locus tag ***30_18280)belonged to glycosyl family-5 and had interaction energy of−12.981 kcal/mol for the best docked cluster containing three out of ten docking conformations,and Tm value of 73.3℃.MD simulation of 100 ns proved highly stable binding interactions of CPZ30_18280 endoglucanase with cellulose with root mean square deviation(RMSD)values ranging from 0.15 to 0.30 *** interactions with characteristic active site residues(tyrosine,tryptophan and aspartate)of glycosyl family-5 endoglucanases were ***,to enhance the production of endoglucanases,the fermentation conditions were optimized employing approaches like one factor at a time(OFAT)and response surface methodology(RSM).Maximum activity of endoglucanase was determined at 60℃.The optimized condi-tions for enhanced production of endoglucanase(10.15 u/mL)were pH 6.63,yeast extract conc.3.44 g/L,wheat bran 3.59%,and inoculum size 2.65%.Hence,*** strain BHu3 has enormous potential to synthesize highly efficient thermostable endoglucanases under optimized regime using ***,it could find immense industrial applications including large scale cellulose conversion to bioethanol.

In this paper we developed a fuzzy inventory model for deteriorating items with time dependent demand rate. Shortages are allowed and completely backlogged. The backlogging rate of unsatisfied demand is assumed to be a decreasing exponential function of waiting time. The demand rate, deterioration rate and backlogging rate are assumed as a triangular fuzzy numbers. The purpose of our study is to defuzzify the total profit function by signed distance method and centroid method. Further a numerical example is also given to demonstrate the developed crisp and fuzzy models. A sensitivity analysis is also given to show the effect of change of the parameters.

Introduction Radon,a known carcinogen,is the second largest cause of lung carcinoma as per the World Health *** primary purpose of the present research is to examine and compare the concentrations of radon gas(^(222)Rn)in different sources of water in the Lar and Ganderbal tehsils of the district of Ganderbal,Jammu and *** groundwater ^(222)Rn concentrations indicate higher indoor radon concentrations and long-term radon exposure is extremely harmful to human health as it increases the risk of developing lung *** A scintillation-based radon monitor called the smart RnDuo was used to quantify the amount of radon activity present in water samples collected from different locations of Lar and Ganderbal regions of district *** Average radon(^(222)Rn)concentration of 1.84 BqL^(−1) was found in tap waters,3.73 BqL^(−1) in surface waters,16.56 BqL^(−1) in borewells,and 20.82 BqL^(−1) in *** higher radon concentration in groundwater is because of the direct and prolonged interaction of uranium-rich rocks and soil with the ***,monitoring and regulating radon levels in these water sources is crucial to ensure public health and safety.

Embryo-to-lamella transition of grain boundary twins in magnesium

A combined experimental and computational analysis is performed to investigate the less commonly studied embryo-to-lamella transition of deformation twins in magnesium. This work aims to understand the structural variables controlling the embryo-to-lamella transition from grain boundaries. Statistical analysis of hundreds of early-stage twins in the lightly deformed microstructure reveals a prevailing wedge shape,with a much thicker base along the grain boundary(GB) where they originate and a thinner tip terminating in the crystal. The analysis also shows that the GB base is super thick and identifies a minimum GB twin thickness among all early-stage twins that is about one micron. A crystal plasticity-based full-field model is employed to calculate the driving forces to migrate the boundary of a three-dimensional GB twin embryo. The stress analysis, considering a full range of embryo shapes and neighboring grain orientations, indicate that the twin embryo is most likely going to form a wedge shape when it first propagates. The calculations predict that the thickness of the embryo at the GB needs to be significantly larger than its length into the crystal in order to propagate into the crystal. The analysis finds that the more aligned the twin embryo variant is with basal slip in the neighboring grain, the thinner the twin embryo needed for propagation.

The analysis of spliced column has been carried out to detect optimum location of providing splices in the *** the present work,static and dynamic(free vibration)analyses of spliced column have been done by randomising the location of splicing.A symmetrical four storey steel framed building has been modelled,analysed and designed for loads(dead,live and earthquake loads)recommended by Indian Codal provisions using *** critical column at each floor level is identified based on axial force(AF),bending moment(BM)and shear force(SF).The total 16 models of spliced columns have been designed and then modelled in a 3D CAD Design tool(SOLIDWORKS)and then imported in the finite element tool(ANSYSWorkbench 14.0)for detailed *** variation of stress,strain and deflection of the spliced column are shown in the form of ***,the modal analysis is performed to determine the natural *** results of static and dynamic analyses are compared for each modelled spliced column to obtain the optimum location for providing splices in the *** dynamic analysis of spliced column is of utmost importance in the region where dynamic loadings like earthquake,cyclones *** more frequent,and mere static analysis does not account for the safety of the *** study will help the engineers to select directly the optimum size and location of the splices in the column of a steel framed building.

Recently,software defined networking(SDN)is a promising paradigm shift that decouples the control plane from the data *** can centrally monitor and control the network through softwarization,i.e.,*** controllers are a necessity of current SDN based *** multiple controllers in an optimum way is known as controller placement problem(CPP).Earlier,solutions of CPP only concentrated on propagation latency but overlooked the capacity of controllers and the dynamic load on switches,which is a significant factor in real *** this paper,we develop a novel optimization algorithm named varna-based optimization(VBO)and use it to solve *** the best of our knowledge,this is the first attempt to minimize the total average latency of SDN along with the implementation of TLBO and Jaya algorithms to solve CPP for all twelve possible *** experimental results show that TLBO outperforms PSO,and VBO outperforms TLBO and Jaya algorithms in all scenarios for all topologies.

Diagnosis of Helicobacter pylori : What should be the gold standard?

Since the discovery of Helicobacter pylori (H. pylori) in 1983, numerous detection methods for the presence of the bacterium have been developed. Each one of them has been associated with advantages and disadvantages. Noninvasive tests such as serology, up>13up>C urea breath test (uBT) and stool antigen tests are usually preferred by the clinicians. Serology has its own limitation especially in endemic areas while up>13up>C uBT is technically very demanding. The stool antigen detection method, although specific, is usually associated with poor sensitivity. The up>13up>C uBT is believed to be specific, but with present revelation of the fact that stomach is colonized by many other urease producing bacteria makes it questionable. Histology, culture, rapid urease test and polymerase chain reaction (PCR) are the tests which are carried out on antral biopsies collected by invasive means. Histology has been proposed to be very sensitive and specific but the question is how by simply looking the morphology of the bacteria in the microscope, one can claim that the curved bacterium is exclusively H. pylori. Rapid urease test (RuT), the doctor’s test, is also challenged because the presence of other urease producing bacteria in the stomach cannot be denied. Moreover, RuT has been reported with poor sensitivity specially, when density of the bacterium is low. Isolation of H. pylori is essential to investigate its growth requirements, antibiotic susceptibility testing, studying virulence factor to develop vaccine and many more explorations. It has also got several disadvantages i.e., special condition for transporting, media, incubation and few days waiting for the colonies to appear, apart from the speed essentially needed to process the specimens. Till date, majority of the microbiological laboratories in the world are not equipped and trained to isolate such fastidious bacterium.
