IL28B polymorphism and cytomegalovirus predict response to treatment in Egyptian HCV type 4 patients

AIM:To test whether the status of positive cytomegalovirus(CMV) DNA detection adds to the predictive value of IL28B and to further categorize C/T allele ***:This study included 166 chronic hepatitis C(CHC) patients who received combined interferon and ribavirin therapy for 48 wk,84 spontaneous hepatitis C virus(HCV) resolvers who were positive for IgG anti-HCV antibody and negative for HCV RNA,and 100 healthy subjects who were negative for both HCV antibodies and RNA as *** DNA from peripheral blood was used for IL28B rs.12979860 single nucleotide polymorphism(SNP) and CMV DNA detection.A 139 bp fragment containing IL28B SNP was amplified in all subjects by polymerase chain reaction using a specifically designed *** the IL28B rs.12979860 SNP was detected by restriction fragment length polymorphism(RFLP) *** presence of CMV DNA was tested by amplification of the gB1 gene using nested polymerase chain *** role of CMV and IL28B rs.12979860 SNP genotypes in determining the response rate to combined interferon therapy and clinical status of patients were statistically ***:Current data showed that 67% of patients carrying the IL28B 12979860 C/C allele had a sustained viral response(SVR) while the genotypes C/T and TT were associated with lower SVR rates,50% and 48%,*** rates for the C/C allele were lower than other HCV genotypes and/or other *** CC was associated with the response to interferon(P = 0.025).Genotype C/C was reduced from 48% in controls to 14% in CHC patients suggesting its protective role against progression to *** majority of spontaneously cleared subjects(86%) were C/C,confirming its protective *** C/T allele was present in 71% of CHC patients compared with 38% of controls,so the use of IL28B SNP genotyping only in these patients may be of little value as a predictor of *** reactivation occurred in 40% of CHC ***-infection with

3D finite element analysis of composite noise barrier con- structed of polyurethane products

This paper presents a numerical investigation on the structural performance of an innovative noise barrier consisting of poly-block, rigid polyurethane foam （RPF） and polyurea. The mechanical characteristics of RPF as well as the flexural resistance of the proposed wall system （poly-wall） were established and presented in another study. The experimental results are used in the current study to develop, calibrate and verify 3D finite element （FE） models of the wall system. The components of the poly-wall including steel rebars, poly-blocks and RPF cores were simulated and then verified using the results of experiments conducted on the wall components. The results of numerical analysis exhibited a satisfactory agreement with the experimental outcomes for the entire wall system. The verified numerical models were then used to conduct a parametric study on the performance of poly-wall models under uniform wind load and gravity load. The findings of the current study confirmed that the structural performance of poly-wall is satisfactory for noise barrier application. Simulation techniques for improvement of the numerical analysis of multi-martial 3D FE models were discussed.

Comparison of bacterial community structures in two systems of a sewage treatment plant using PCR-DGGE analysis

The combination of PCR amplification of 16S rRNA genes with denaturing gradient gel electrophoresis （DGGE） analysis was used to reveal the compositions and dynamics of bacterial communities in a sewage treatment plant with two systems, i.e., an anoxic- anaerobic-aerobic system （inverted A2O） and an anaerobic-anoxic-aerobic one （conventional A2O） over a period from February to July 2009, during which both systems experienced serious sludge bulking problems. The DGGE patterns showed that there were many common bands in both systems, suggesting the high similarity of bacterial communities of the two systems. Meanwhile, the moving window correlation analysis showed that the two systems experienced different microbial community structure changes during the period, which might be related with the different situations of the occurrence and disappearance of sludge bulking, as being reflected by sludge volume index （SVI） values. Major bands of DGGE patterns of sludge samples were further sequenced. Phylogenetic affiliation indicated that the majority of the sequences obtained were affiliated with Actinobacteria, Firmicutes, Bacteroidetes/Chlorobi group and α- and β-Proteobacteria. Two sequences showed high similarities to typical filamentous bacteria Microthrix parvicella and Nostocoida limicola I, indicating that these bacterial species have been involved in the sludge bulking problems.

Optimizing the intraocular lens formula constant according to intraocular lens diameter

AIM:To determine whether the different diameters of a specific intraocular lens(IOL)have significantly different optimized SRK/T A constants and whether these new A constants can improve refractive ***:Data were collected prospectively from Jan.2011 to Dec.2012 on all patients undergoing routine cataract surgery at a district general hospital in the *** were divided into three groups according to the size of the Akreos AO MI60 IOL used.A constants for the SRK/T formula were optimized according to the size of the *** optimized A constants were then used to select future refractive ***:A total of 2398 cataract operations were performed during the study period of which 1131 met the inclusion *** three optimized A constants for the different sized IOLs were 118.98,119.13,*** difference between them was highly significant(P≤0.0001).Two optimized A constants for three sizes of IOL led to an improvement in refractive outcomes(from 93.4%to 94.6%of refractive outcomes within 1.00 D of predicted spherical equivalent).The optimized A constant for the largest IOL was based on a small number of cases and was not ***:Optimizing the A constant for the three distinct sizes of the Bausch&Lomb Akreos MI60 lens lead to three significantly different A *** our practice,using two different optimized A constants for three different sized IOLs give the least refractive error,however,using three optimized A constants may give better results with a larger dataset.

Ophthalmic photographer virtual clinics in medical retina

AIM: To determine whether ophthalmic photographers(OPs) can conduct virtual clinics(VCs) in medical retina appropriately and whether this increases clinic ***: Three OPs underwent a training programme to learn how to assess and manage macular edema secondary to diabetes or retinal vein occlusion. The 300 consecutive patients over a 7-month observation period in 2018 were assessed in VCs by both OPs and medical retina consultants. The degree of agreement in treatment decisions between doctors and photographers, as well as adverse events, was recorded. The change in number of patients seen in VCs over two years was also ***: There was 100% agreement in management decisions between doctors and photographers during the 300-patient observation period. No adverse events were recorded. In 2017, 572 patients were seen in VCs by doctors. After OP clinics were introduced in 2018, this number increased by 24% to 709 patients seen by both photographers and doctors. There was a significant increase in mean number of patients seen per month between 2017(47.7±11.7) and 2018(59.1±14.4;P=0.045, 95%CI-22.5 to-0.296).CONCLUSION: OPs can manage certain medical retina patients in VCs appropriately and can increase clinic capacity significantly for particular conditions.

Hydraulic head difference at two sides of suspended waterproof curtain during multi-grade dewatering of excavation

This paper proposes an approach to calculate the head difference at two sides of suspended waterproof curtains during multi-grade *** seepage during the dewatering process can be subdivided into three regions:(i)seepage in pit,(ii)seepage between cur-tains,and(iii)seepage outside the *** flow rate of the first region is equal to the pumping rate,and the flow rate of the second and third regions can be obtained by numerical analysis.A numerical model is established to simulate the seepage in the second and third regions and its performance is validated by using the measured data of a series of field *** flow rate of each region is then used to derive formulae for the head difference in conventional dewatering,which can be used to determine the head difference at two sides of each waterproof curtain during multi-grade *** proposed formula expresses the head difference as a function of the relative depth of the curtain inserted into the confined aquifer,the thickness of the aquifer,the distance between two curtains,and the anisotropy of the hydraulic conductivity of the *** proposed numerical approach is further validated by using data derived from numerical *** validation results demonstrated that the predictions of the proposed approach are acceptable and convenient.

electrodeposited Ni and Ni-Co alloys using cysteine and conventional ultrasound waves

Ni-Co alloys were electroplated from sulphate electrolyte using addition agents including sodium gluconate, boric acid and eysteine on copper foil by the galvanostatic technique and ultrasound waves. The chemical composition, surface morphologies, crystalline structure and hardness of the Ni-Co alloys were studied using energy dispersive spectroscopy, scanning electron microscope, X-ray diffraction and Vickers testing method, respectively. The effect of current density and addition agents on the microstructure and morphology of Ni-Co alloys were examined. The appropriate concentration of additives and ultrasound waves were found to produce fine and smooth crystals leading to higher hardness of Ni-Co alloys. The microhardness of the Ni-Co alloys was varied between 4860-7530 HV. The surface morphology of coatings was changed from granular to fine due to using of gluconate, boric acid, cysteine and ultrasound waves. The mechanical properties of nanocrystalline Ni-Co alloys showed an increase of the hardness with the growing of Ni content in the alloy. The X-ray diffraction studies indicated that nanocrystalline structure was face-centred cubic for pure Ni and Ni-Co alloys with Co content in the range of 1-75 wt.%. A hexagonal closed-package structure was obtained for pure Co and Ni-Co alloys with the cobalt content with range of 75-99 wt.%.

Soil enzymatic activities and microbial community structure with different application rates of Cd and Pb

This study focused on the changes of soil microbial diversity and potential inhibitory effects of heavy metals on soil enzymatic activities at different application rates of Cd and/or Pb. The soil used for experiments was collected from Beijing and classified as endoaquepts. Pots containing 500 g of the soil with different Cd and/or Pb application rates were incubated for a period of 0, 2, 9, 12 weeks in a glasshouse and the soil samples were analyzed for individual enzymes, including catalase, alkaline phosphatase and dehydrogenase, and the changes of microbial community structure. Results showed that heavy metals slightly inhibited the enzymatic activities in all the samples spiked with heavy metals. The extent of inhibition increased significantly with increasing level of heavy metals, and varied with the incubation periods. The soil bacterial community structure, as determined by polymerase chain reaction- denaturing gradient gel electrophoresis techniques, was different in the contaminated samples as compared to the control. The highest community change was observed in the samples amended with high level of Cd. Positive correlations were observed among the three enzymatic activities, but negative correlations were found between the amounts of the heavy metals and the enzymatic activities.

Numerical modeling of time-dependent deformation and induced stresses in concrete pipes constructed in Queenston shale using micro-tunneling technique

Effects of time-dependent deformation（TDD） on a tunnel constructed using the micro-tunneling technique in Queenston shale（QS） are investigated employing the finite element method. The TDD and strength parameters of the QS were measured from tests conducted on QS specimens soaked in water and lubricant fluids（LFs） used in micro-tunneling such as bentonite and polymer solutions. The numerical model was verified using the results of TDD tests performed on QS samples, field measurements of some documented projects, and the closed-form solutions to circular tunnels in swelling rock. The verified model was then employed to conduct a parametric study considering important micro-tunneling design parameters, such as depth and diameter of the tunnel, in situ stress ratio（K;）, and the time lapse prior to replacing LFs with permanent cement grout around the tunnel. It was revealed that the time lapse plays a vital role in controlling deformations and associated stresses developed in the tunnel *** critical case of a pipe or tunnel in which the maximum tensile stress develops at its springline occurs when it is constructed at shallow depths in the QS layer. The results of the parametric study were used to suggest recommendations for the construction of tunnels in QS employing micro-tunneling.

Impacts of produced water origin on bacterial community structures of activated sludge

The purpose of this study was to reveal how activated sludge communities respond to influent quality and indigenous communities by treating two produced waters from different origins in a batch reactor in succession. The community shift and compositions were investigated using Polymerase Chain Reaction–denaturing gradient gel electrophoresis（PCR–DGGE） and further16 S ribosomal DNA（r DNA） clone library analysis. The abundance of targeted genes for polycyclic aromatic hydrocarbon（PAH） degradation, nah Ac/phn Ac and C12O/C23 O, was tracked to define the metabolic ability of the in situ microbial community by Most Probable Number（MPN） PCR. The biosystem performed almost the same for treatment of both produced waters in terms of removals of chemical oxygen demand（COD） and PAHs. Sludge communities were closely associated with the respective influent bacterial communities（similarity 〉 60%）, while one sludge clone library was dominated by the Betaproteobacteria（38%） and Bacteriodetes（30%）and the other was dominated by Gammaproteobacteria（52%）. This suggested that different influent and water quality have an effect on sludge community compositions. In addition, the existence of catabolic genes in sludge was consistent with the potential for degradation of PAHs in the treatment of both produced waters.
