Potential Secretory Transporters and Biosynthetic Precursors of Biological Nitrification Inhibitor 1,9-Decanediol in Rice as Revealed by Transcriptome and Metabolome Analyses

Biological nitrification inhibitors(BNIs)are released from plant roots and inhibit the nitrification activity of microorganisms in soils,reducing NO_(3)^(‒)leaching and N2O emissions,and increasing nitrogenuse efficiency(NUE).Several recent studies have focused on the identification of new BNIs,yet little is known about the genetic loci that govern their biosynthesis and secretion.We applied a combined transcriptomic and metabolomic analysis to investigate possible biosynthetic pathways and transporters involved in the biosynthesis and release of BNI 1,9-decanediol(1,9-D),which was previously identified in rice root exudates.Our results linked four fatty acids,icosapentaenoic acid,linoleate,norlinolenic acid,and polyhydroxy-α,ω-divarboxylic acid,with 1,9-D biosynthesis and three transporter families,namely the ATP-binding cassette protein family,the multidrug and toxic compound extrusion family,and the major facilitator superfamily,with 1,9-D release from roots into the soil medium.Our finding provided candidates for further work on the genes implicated in the biosynthesis and secretion of 1,9-D and pinpoint genetic loci for crop breeding to improve NUE by enhancing 1,9-D secretion,with the potential to reduce NO_(3)^(‒)leaching and N2O emissions from agricultural soils.

Ammonium(NH) toxicity is a significant agricultural problem globally,compromising crop growth and productivity in many areas.However,the molecular mechanisms of NH4+ toxicity are still poorly understood,in part due to a lack of valuable genetic resources.Here, a novel Arabidopsis mutant,amos2(ammonium overly sensitive 2),displaying hypersensitivity to NH in both shoots and roots,was isolated.The mutant exhibits the hallmarks of NH toxicity at significantly elevated levels:severely suppressed shoot biomass,increased leaf chlorosis,and inhibition of lateral root formation.Amos2 hypersensitivity is associated with excessive NH accumulation in shoots and a reduction in tissue potassium(K),calcium(Ca),and magnesium(Mg).We show that the lesion is specific to the NH4 ion,is independent of NH metabolism,and can be partially rescued by elevated external K.The amos2 lesion was mapped to a 16-cM interval on top of chromosome 1,where no similar mutation has been previously mapped.Our study identifies a novel locus controlling cation homeostasis under NH stress,and provides a tool for the future identification of critical genes involved in the development of NH toxicity.

The formation of root system architecture is greatly dependent on the environmental conditions such as nutrients.Ammonium(NH) is an important nitrogen source for higher plants.However,excess NH shows a strong suppression on root system development. Here,with local nutrition supply device,we revealed that NH-mediated inhibition of root elongation is primarily through root tip,whereas NH-mediated reduction of lateral root number is a result of shoot contact with NH.Furthermore,NH inhibition of root growth was shown to target primarily cell elongation,which was directly linked to excessive NH efflux in the root elongation zone,but not related to auxin pathway.This NH efflux is regulated by GDP-mannose pyrophosphorylase(GMPase).On the other hand, NH-induced reduction of lateral root formation results from less lateral root emergence rather than lateral root initiation,which is caused by the decreased transport of shoot-derived auxin to roots,via the auxin influx carrier AUX1.Taken together,these results suggest regulations of primary root elongation and lateral root formation by NH is through local and systemic two independent pathways.Our findings provide a novel insight to regulations of root development by environmental nutrient factors.

Liver stiffness in pregnant women with intrahepatic cholestasis of pregnancy:A case control study

BACKGROUND Intrahepatic cholestasis of pregnancy(ICP)is a rare but severe complication for both the mother and the unborn child.The diagnosis is primarily based on elevated serum levels of bile acids.In a large ICP cohort,we here study in detail liver stiffness(LS)using transient elastography(TE),now widely used to noninvasively screen for liver cirrhosis within minutes.AIM To specifically explore LS in a large cohort of women with ICP compared to a control group with uncomplicated pregnancy.METHODS LS and hepatic steatosis marker controlled attenuation parameter(CAP)were measured in 100 pregnant women with ICP using TE(Fibroscan,Echosens,Paris,France)between 2010 and 2020.In 17 cases,LS could be measured postpartum.450 women before and 38 women after delivery with uncomplicated pregnancy served as control group.Routine laboratory,levels of bile acids and apoptosis marker caspase-cleaved cytokeratin 18 fragment(M30)were also measured.RESULTS Women with ICP had significantly elevated transaminases but normal gammaglutamyl transferase(GGT).Mean LS was significantly increased at 7.3±3.0 kPa compared to the control group at 6.2±2.3 kPa(P<0.0001).Postpartum LS decreased significantly in both groups but was still higher in ICP(5.8±1.7 kPa vs 4.2±0.9 kPa,P<0.0001),respectively.In ICP,LS was highly significantly correlated with levels of bile acids and M30 but not transaminases.No correlation was seen with GGT that even increased significantly after delivery in the ICP group.Bile acids were mostly correlated with the liver apoptosis marker M30,LS and levels of alanine aminotransferase,aspartate aminotransferase,and bilirubin.In multivariate analysis,LS remained the sole parameter that was independently associated with elevated bile acids.CONCLUSION In conclusion,LS is significantly elevated in ICP which is most likely due to toxic bile acid accumulation and hepatocyte apoptosis.In association with conventional laboratory markers,LS provides additional non-invasive information to rapidly identify women at risk for ICP.

Acute septic arthritis in children is an orthopaedic emergency.A delay in diagnosis and inappropriate treatment can result in devastating damage to the joint with lifelong disability as a consequence.The clinical presentation can be a diagnostic challenge,especially in young children.A recent systematic review showed that joint tenderness and fever are important signals of septic arthritis.Ultrasound is helpful in detecting the presence of a joint effusion.Plain radiographs may show bone changes but magnetic resonance imaging is the most reliable imaging study for detecting concomitant osteomyelitis.The diagnosis of acute septic arthritis is highly suggestive when pus is aspirated from the joint,in case of a positive culture or a positive gram stain of the joint fluid,or if there is a white blood-cell count in the joint fluid of more than 50000/mm3.Staphylococcus aureus is the most commonly cultured organism.Recent systematic reviews have identified the most effective drainage techniques,including needle aspiration,arthroscopy and arthrotomy,depending on the affected joint.After the drainage procedure it is important to monitor the clinical and laboratory outcomes.Additional drainage procedures may be necessary in select cases.

Background:There exist few maximal oxygen uptake(VO_(2max))non-exercise-based prediction equations,fewer using machine learning(ML),and none specifically for older adults.Since direct measurement of VO_(2max)is infeasible in large epidemiologic cohort studies,we sought to develop,validate,compare,and assess the transportability of several ML VO_(2max)prediction algorithms.Methods:The Baltimore Longitudinal Study of Aging(BLSA)participants with valid VO2_(max)tests were included(n=1080).Least absolute shrinkage and selection operator,linear-and tree-boosted extreme gradient boosting,random forest,and support vector machine(SVM)algorithms were trained to predict VO_(2max)values.We developed these algorithms for:(a)the overall BLSA,(b)by sex,(c)using all BLSA variables,and(d)variables common in aging cohorts.Finally,we quantified the associations between measured and predicted VO_(2max)and mortality.Results:The age was 69.0±10.4 years(mean±SD)and the measured VO_(2max)was 21.6±5.9 mL/kg/min.Least absolute shrinkage and selection operator,linear-and tree-boosted extreme gradient boosting,random forest,and support vector machine yielded root mean squared errors of 3.4 mL/kg/min,3.6 mL/kg/min,3.4 mL/kg/min,3.6 mL/kg/min,and 3.5 mL/kg/min,respectively.Incremental quartiles of measured VO_(2max)showed an inverse gradient in mortality risk.Predicted VO_(2max)variables yielded similar effect estimates but were not robust to adjustment.Conclusion:Measured VO_(2max)is a strong predictor of mortality.Using ML can improve the accuracy of prediction as compared to simpler approaches but estimates of association with mortality remain sensitive to adjustment.Future studies should seek to reproduce these results so that VO_(2max),an important vital sign,can be more broadly studied as a modifiable target for promoting functional resiliency and healthy aging.

Comparison of direct fecal smear microscopy,culture,and polymerase chain reaction for the detection of Blastocystis sp.in human stool samples

Objective:To compare the sensitivity and specificity of direct fecal smear microscopy,culture,and polymerase chain reaction in the detection of Blastocystis sp.in human stool.Methods:Human stool samples were collected from a community in San Isidro,Rodriguez,Rizal,Philippines.These samples were subjected to direct fecal smear microscopy,culture and polymerase chain reaction to detect the presence of Blastocystis sp.Results:Of the 110 stool samples collected,28(25%)were detected positive for the presence of Blastocystis sp.by two or more tests.Culture method detected the highest number of Blastocystis-positive stool samples(n=36),followed by PCR of DNA extracted from culture(n=26),PCR of DNA extracted from stool(n=10),and direct fecal smear(n=9).Compared to culture,the sensitivity of the other detection methods were 66.7%for PCR from culture and 19.4%for both PCR from stool and direct fecal smear.Specificity of the methods was high,with PCR from culture and direct fecal smear having97.3%,while PCR from stool at 95.9%.Conclusions:In this study,in vitro culture is the best method for detecting Blastocystis sp.in human stool samples.

Avian sibling cannibalism: Hoopoe mothers regularly use their last hatched nestlings to feed older siblings

Sibling cannibalism is relatively common in nature,but its evolution in birds and certain other vertebrates with extended parental care had been discarded.Here,however,we demonstrate its regular occurrence in two European populations of the Eurasian hoopoe(Upupa epops)and explore possible adaptive and non-adaptive explanations.Results showed that sibling cannibalism was more frequently detected in Spain(51.7%)than in Austria(5.9%).In these two populations,the hoopoes laid similar clutch sizes,resulting in similar fledging production,but hatching failures were more frequent in the northern population.Consequently,having more nestlings condemned to die in the southern population may explain the higher incidence of sibling cannibalism.In accordance with this interpretation,hatching span and failure,but not breeding date,explained the probability of sibling cannibalism in the Spanish hoopoes,while all three variables predicted brood reduction intensity.Furthermore,experimental food supply reduced the probability of sibling cannibalism,but not the intensity of brood reduction.Finally,females allocated fewer resources to the smallest nestlings when they were going to starve,but not necessarily when they were going to be used as food for their siblings.These results suggest that hoopoes produce extra eggs that,in the case of reduced hatching failure and food scarcity,produce nestlings that are used to feed older siblings.These findings provide the first evidence that sibling cannibalism occurs regularly in a bird species,thus expanding our evolutionary understanding of clutch size,hatching asynchrony,parent-offspring conflict,infanticide,and sibling cannibalism in the animal kingdom.

Genetic Testing in Professional Football:Perspectives of Key Stakeholders

Purpose Genetic research in football is currently in it’s infancy but is growing rapidly.However,the practical application of genetic testing in football and the views concerning its use are unknown.Thus,the purpose of this study was to assess the current practical application of genetic testing in professional football and provide an insight into the perspectives of key stakeholders(i.e.,coaches,practitioners,players).Methods In total,122 participants completed an online anonymous survey.This consisted of 21 multiple choice and Likert scale questions,with the option of providing an explanation for each response.Results Findings revealed genetic testing is rarely utilised by key stakeholders(10%)or their respective organisations(14%).However,three quarters(75%)had the opinion that genetic testing will have great utility in the future.The majority(72%)believed genetic testing should be used for athlete development and injury risk,whilst 35%believed that genetic testing should be utilised for talent identification purposes.However,most key stakeholders viewed their own(89%)and their col-leagues’(79%)knowledge related to genetic testing as insufficient;mainly due to ineffective current communication methods(91%).Most believed educational workshops are required(71%),whilst nearly all(91%)were interested in developing their expertise on the utility of genetic testing.Conclusion Genetic testing is rarely used within professional football,although key stakeholders anticipate that it will be utilised more in the future.As such,educational support may prove valuable in improving key stakeholder knowledge and the practical application of genetic testing in professional football.

A Systematic Review of the Genetic Predisposition to Injury in Football

Purpose Synthesise genetic association studies investigating injury involving football players to identify which genetic variants have the most empirical evidence to date.Methods A comprehensive search of the PubMed,SPORTDiscus,and MEDLINE databases until March 11th 2022 identi-fied 34 studies.Inclusion criteria:primary investigations,included football players,examined the association of a genetic variant with injury,and were published in English.Risk of bias was assessed using the Newcastle–Ottawa Scale.A narrative synthesis summarised results.Results There were 33 candidate gene studies and one genome-wide study,with 9642 participants across all studies(range=43–1311;median=227).Ninety-nine polymorphisms were assessed within 63 genes.Forty-one polymorphisms were associated with injury once.Three polymorphisms had their specific allelic associations with injury replicated twice in independent cohorts:ACTN3(rs1815739)XX genotype was associated with an increased susceptibility to non-contact muscle injuries,ACAN(rs1516797)G allele was associated with increased susceptibility to anterior cruciate ligament(ACL)injuries,and VEGFA(rs2010963)CC genotype was associated with an increased susceptibility to ACL and ligament or tendon injuries.However,several methodological issues(e.g.,small sample sizes,cohort heterogeneity,and population stratification)are prevalent that limit the reliability and external validity of findings.Conclusion At present,the evidence base supporting the integration of genetic information as a prognostic or diagnosis tool for injury risk in football is weak.Future participation of organisations in international consortia is suggested to combat the current methodological issues and subsequently improve clarity concerning the underlying genetic contribution to injury susceptibility.
