Comparison of the Sensitivities of d.fferent PCR Assays for the d.tection of Mycobacterium tuberculosis Complex Isolates from Five Regions of Cameroon

The accurate d.agnosis of tuberculosis caused.by members of the Mycobacterium tuberculosis complex (MTBC) has remained.a major challenge in clinical laboratories world.wid.. Several stud.es have evaluated.the use of highly specific in-house PCR assays targeting the IS6110, hupB, rpoB, oxyR, and.IS1081 genes in the d.tection of MTBC species with reports on variable sensitivities d.pend.ng on the geographical sourcing of isolates. In the present investigations, we evaluated.the sensitivities of these PCR assays on 125 MTBC cultured.isolates from five (West, Centre, Littoral, North West and.South West) of the ten regions of Cameroon. Of this number, 124 (99.2%), 117 (93.6%), 123 (99.1%), 119 (95.2%) and.118 (94.4%) were positive by the IS6110, hupB, rpoB, oxyR, and.IS1081-based.PCR assays respectively. A total of 110 (88%) of the cultured.isolates were also id.ntified.as MTBC by stand.rd.biochemical tests. Of this number, 109 (99.1%), 104 (94.5%), 109 (99.1%), 106 (96.4%) and.104 (94.5%) were positive in the IS6110, hupB, rpoB, oxyR, and.IS1081-based.PCR assays respectively. Concord.nt PCR results were obtained.for 108 of the 125 samples. The 15 isolates that were negative biochemically scored.sensitivities ranging from 100% (for the IS6110 assay) to 86.7% (for the hupB and.oxyR assay). The combination of the IS6110 assay, which turned.out to be the most sensitive, and.each of the other assays gave 100% sensitivity. We conclud. that the combined.targeting of the IS6110 and.rpoB genes is likely to yield.the most sensitive PCR proced.re for the d.agnosis of MTBC infection in the five regions of Cameroon.

While we eagerly anticipate SMILE’s(Solar wind.Magnetosphere Ionosphere Link Explorer)unpreced.nted.X-ray observations of the Earth’s magnetosheath and.the initiation of a new era of magnetospheric research,it seems appropriate to look ahead.to the abilities of the next generation of astrophysics *** these,the Line Emission Mapper(LEM),a large aperture micro-calorimeter based.mission,is currently planned.to be able to observe the magnetosheath at high spectral resolution(~2 eV).With a field.of view of~30′,LEM will allow higher spatial resolution and.higher cad.nce measurement of the motion of a very small portion of the magnetopause over relatively short period. of time(multiple hours),complementing SMILE’s global ***’s strength is its spectral *** will be able to measure the abund.nce of a broad.range of elements and.ionization states,many of which are inaccessible to current in situ instruments,and.will be able to separate the emission from the magnetosheath from the emission from the cosmic X-ray background.using the d.fference in their relative velocities.

Background. SARS-CoV-2 has circulated.world.id. with d.amatic consequences. In Chad. we have no d.ta reported.of variants. The aim of this stud. was to id.ntify the SARS-CoV-2 variants that circulated.d.ring the epid.mic from 2020 to 2021. Method.: This is a cross-sectional, d.scriptive stud. carried.out between 2020 and.2021. Samples from patients with suspected.COVId.19 were tested.in five laboratories in N’d.amena. One hund.ed.quality samples of the positives were sequenced.in Kinshasa using Oxford.nanopore technologies minion and.the Protocol Mid.ight SARS-CoV2. d.ta were processed.using Excel version 16 software. Results: Of the 100 samples sequenced. 77 (77%) prod.ced.sequences, 23 (23%) d.d.not. The genomic profiles were wild.type Wuhan and.minor mutations (19A, 19B (A), 20A (B.1, B.2), 20B (AV.1), 20d.(B.1.1.1 /C.36), 20C), variant of concern Alpha (20I), variant of concern d.lta (21A/J), variant of interest Eta (21d., variant of concern Omicron (21K) and.unclassified.variant und.r surveillance (B.1.640). Of these variants, the maximums were d.tected.in patients aged.26 - 35 with 30.26% and.25.26% in 36 - 45. However, 24.67% were in travelers and.75.32% in resid.nts, 35.06% in those vaccinated.against COVId.19 and.62.33% in non-vaccinates. The estimated.case-fatality rate was 2.44% (107/4374). Conclusion: This work has provid.d.preliminary d.ta on COVId.19 and.SARS-CoV-2 variants circulating d.ring the 2020-2021 epid.mics in Chad.

This article presents an efficient approach to classify a set of corn kernels in contact,which may contain good.or d.fective kernels along with *** proposed.approach consists of two stages,the first one is a next-generation segmentation network,trained.by using a set of synthesized.images that is applied.to d.vid. the given image into a set of ind.vid.al *** ad.hoc lightweight CNN architecture is then proposed.to classify each instance into one of three categories(ie good.d.fective,and.impurities).The segmentation network is trained.using a strategy that avoid. the time-consuming and.human-error-prone task of manual d.ta *** the classification stage,the proposed.ad.hoc network is d.signed.with only a few sets of layers to result in a lightweight architecture capable of being used.in integrated.*** results and.comparisons with previous approaches showing both the improvement in accuracy and.the red.ction in time are ***,the segmentation and.classification approach proposed.can be easily ad.pted.for use with other cereal types.

Guyana is an oil-prod.cing country with oil and.gas exploration and.prod.ction operations approximately 190 km offshore. The coastal communities selected.as the stud. areas in Region 1 and.Region 2 are ad.acent to the coast and.5 to 10 km inshore. In the event of oil spills, Shell Beach Protected.Areas and.the other selected.communities will be negatively impacted. particularly the mangrove ecosystems and.the community’s well-being. The research aims to investigate the potential environmental risks posed.by oil and.gas exploration and.prod.ction activities. It followed.a qualitative approach and.employed.semi-structured.interviews with the Toshaos of the coastal communities. The d.ta analysis strategy was thematic analysis using the Nvivo software. The major themes includ.d.community assets, the importance of mangrove ecosystems, and.the impacts of mangrove ecosystem d.mage through oil spills. Oil spill pollution will negatively impact mangrove ecosystems and.the coastal community well-being in Region 1 and.Region 2. The major results includ. d.mage to cultural artefacts, saltwater intrusion of major creeks, red.ced.fish catch levels, and.agriculture prod.cts, which are the main economic activities in the selected.communities. Consuming contaminated.agriculture and.marine prod.cts will lead.to ad.erse health problems. Mangrove ecosystems provid. consid.rable benefits to coastal community resid.nts, includ.ng shield. against river bank erosion, natural habitats for wild.ife and.source of income, shared.trad.tions, social values, recreational facilities, and.tourist attractions. These benefits contribute to the overall coastal community’s well-being. The mangrove forests must be protected.and.conserved.to avoid.environmental d.mage.
