Sex Difference in Lactate Production by Adipocytes from Lean Humans

Lactate is an anaerobic metabolite produced in the absence of an adequate O2 supply. Although for a long time considered to be a waste product of glycolysis resulting from hypoxia, lactate is, in fact, an important source of glucose and also a gluconeogenic precursor, having a role in metabolic and endocrine signaling. Lactate is produced by adipocytes and muscle cells. Objectives: In this study, we investigated the sex differences in lactate production by adipocytes isolated from lean human visceral adipose tissue. Main Methods: The experiments described were done using adipocytes isolated from adipose tissue of lean men and women. Adipocytes were isolated following Rodbell procedure, with modifications, for posterior analysis of glycerol and lactate production. Results: Morphometric analysis revealed no significant differences in the size of adipocytes from men and women (diameter: men: 172 ± 24 μm vs. women: 160 ± 16 μm, n = 4 and 10, respectively). Basal glycerol production was significantly higher in adipocytes from men compared to women (0.34 ± 0.06 vs. 0.16 ± 0.01 μmol/106 cells/60 min;mean ± SEM, n = 7 and 4, respectively;p vs. women: 0.12 ± 0.02 μmol/10 cells/60 min). However, when stimulated by norepinephrine, adipocytes from women produced more lactate than adipocytes from men. Female adipocytes also produced as much lactate as glycerol, whereas male adipocytes produced three times more glycerol than lactate. The intracellular mechanisms responsible for this sex difference in lactate production during norepinephrine-stimulated lipolysis remain to be identified.

Evidence-Based Complementary and Alternative Medicine Effects of Capybara Oil on Cardiac Remodeling of C57bl/6 Mice

Cardiovascular diseases are the main cause of morbidity and mortality in the world, and obesity and the metabolic syndrome are risk factors for its development. One of the therapies to reduce cardiovascular risk is the use of polyunsaturated fatty acids. In Brazil, a source of such acid is the oil extracted from the fat of the capybara. The objective of this work is to study the effects of the capybara oil on lipid and glucose metabolism, as well as its effects on the adipose tissue and cardiac remodeling. We assessed the effects of capybara oil treatment on body mass, lipid and carbohydrate metabolism, systolic blood pressure, adipose tissue and cardiac remodeling, and performed an ultrastructural evaluation of the myocardium in C57Bl/6 mice treated with high-fat diet. Treatment with capybara oil reduced total cholesterol and triglyceride levels, systolic blood pressure, visceral and subcutaneous adipose tissue, and adipocyte diameter. In addition, cardiac remodeling was attenuated, preserving cardiomyocytes, increasing vascularization, reducing cardiomyocyte hypertrophy and the extracellular matrix, and preserving the morphological integrity of mitochondria. Capybara oil has several beneficial effects on the cardiovascular and metabolic system, and further studies are needed to better understand its role in the prevention or treatment of cardiovascular diseases.

Objective: To identify the main chemical classes of compounds from aqueous extract of Enterolobium contortisiliquum(E. contortisiliquum) seed bark and to evaluate its antibacterial activity, as well as its potential to increase the activity of antibiotics against strains of Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia ***: Different classes of compounds in the aqueous extract of E. contortisiliquum were evaluated based on the visual changes in the coloration and the formation of precipitate after the addition of specific reagents. The antibacterial activity of the extract and its potential to increase of antibiotic activity of antibiotics drugs, gentamicin and norfloxacin was determined by using the microdilution ***: Our results demonstrated that the following secondary metabolites were presented in E. contortisiliquum seed bark: flavones, flavonols, xanthones, flavononols,chalcones, aurones, flavones and catechins. The extract itself had very low antibacterial activity against all bacterial strains tested(MIC ≥ 1024 μg/m L), but there was an increase in the antibiotic activity of gentamicin and norfloxacin when combined in the subinhibitory concentration(i.e., MIC/8).Conclusions: Our data suggests that E. contortisiliquum seed bark may be an alternative source for new drugs with the potential to increase antibiotic activity against different strains of bacteria.

We investigated the effects of formulations based on phosphite products on gas exchange and activity of antioxidant enzymes in coffee plants. Seedlings of the Mundo Novo cultivar were submitted to various treatments composed of different formulations of with potassium phosphite (ADB 120), manganese phosphite (Reforce Mn), and fungicide (PrioriXtra®). For coffee seedlings, the combination of potassium phosphite and citrus by-products, isolated or in a combination with other products elicited the antioxidant system. Besides the high activity of antioxidant enzymes, the photosynthetic rates were higher than other treatments. The better performance of coffee seedlings treated with those formulations occurred even in absence of pathogens.

The Optimization of Aspergillus ***4 Tannase Production under Submerged Fermentation

Tannase is a hydrolytic enzyme that is involved in the biodegradation of tannins and it has biotechnological potential in the pharmaceutical, chemical, food and beverage industries. Microorganisms, especially filamentous fungi, are important tannase producers. The aims of this work were to find a potential tannase producer and to improve the cultivation conditions. Three Aspergillus species (A. japonicus 246A, A. tamarii 3 and Aspergillus sp. GM4) were investigated in different culture media (Adams, Czapeck, Khanna, M5 and vogel) and inducers (1% and 2% tannic acid;1% green tea;1% methyl gallate;1% gallic acid). Aspergillus sp. GM4 and Adams medium were selected. The tannase production by Aspergillus sp. GM4 in Adams medium was induced in the presence of 2% (w/v) tannic acid and gallic acid as carbon sources, while green tea was not able to induce tannase production. The Plackett-Burman screening design was performed with the variables MgSO4, KH2PO4, yeast extract, tannic acid, agitation rate and salt solution. The variables MgSO4 and agitation rate were selected for the optimization of tannase production using a Central Composite Rotatable Design. Under optimized conditions, a 2.66-fold increase in the enzyme production was observed with small modifications in the medium composition.
