Currently,the establishment of synthetic microbial consortia with rational strategies has gained extensive attention,becoming one of the important frontiers of synthetic biology.Systems biology can offer insights into the design and construction of synthetic microbial consortia.Taking the high-efficiency production of 2-keto-L-gulonic acid(2-KLG)as an example,we constructed a synthetic microbial consortium“Saccharomyces cerevisiae-Ketogulonigenium vulgare”based on systems biology analysis.In the consortium,K.vulgare was the 2-KLG pro-ducing strain,and S.cerevisiae acted as the helper strain.Comparative transcriptomic analysis was performed on an engineered S.cerevisiae(VTC2)and a wild-type S.cerevisiae BY4741.The results showed that the up-regulated genes in VTC2,compared with BY4741,were mainly involved in glycolysis,TCA cycle,purine metabolism,and biosynthesis of amino acids,B vitamins,and antioxidant proteases,all of which play important roles in pro-moting the growth of K.vulgare.Furthermore,Vitamin C produced by VTC2 could further relieve the oxidative stress in the environment to increase the production of 2-KLG.Therefore,VTC2 would be of great advantage in working with K.vulgare.Thus,the synthetic microbial consortium"VTC2-K.vulgare"was constructed based on transcriptomics analyses,and the accumulation of 2-KLG was increased by 1.49-fold compared with that of mono-cultured K.vulgare,reaching 13.2±0.52 g/L.In addition,the increased production of 2-KLG was accompanied by the up-regulated activities of superoxide dismutase and catalase in the medium and the up-regulated oxidative stress-related genes(sod,cat and gpd)in K.vulgare.The results indicated that the oxida-tive stress in the synthetic microbial consortium was efficiently reduced.Thus,systems analysis confirmed a favorable symbiotic relationship between microorganisms,providing guidance for further engineering synthetic consortia.

Myripnois is a monotypic shrub genus in the daisy family constricted to northern China.Although wild populations of Myripnois dioica are relatively rare,this plant may potentially be cultured as a fine ornamental.In the present study,we sequenced the complete plastome of M.dioica,generating the first plastome sequences of the subfamily Pertyoideae.The plastome of M.dioica has a typical quadripartite circular structure.A large^20-kb and a small^3-kb inversion were detected in the large single copy(LSC)region and shared by other Asteraceae species.Plastome phylogenomic analyses based on 78 Asteraceae species and three outgroups revealed four groups,corresponding to four Asteraceae subfamilies:Asteroideae,Cichorioideae,Pertyoideae and Carduoideae.Among these four subfamilies,Pertyoideae is sister to Asteroideae+Cichorioideae;Carduoideae is the most basal clade.In addition,we characterized 13 simple sequence repeats(SSRs)that may be useful in future studies on population genetics.

A full understanding of adaptive genetic variation at the genomic level will help address questions of how organisms adapt to diverse climates.Actinidia eriantha is a shade-tolerant species,widely distributed in the southern tropical region of China,occurring in spatially heterogeneous environments.In the present study we combined population genomic,epigenomic,and environmental association analyses to infer population genetic structure and positive selection across a climatic gradient,and to assess genomic offset to climatic change for A.eriantha.The population structure is strongly shaped by geography and influenced by restricted gene f low resulting from isolation by distance due to habitat fragmentation.In total,we identified 102 outlier loci and annotated 455 candidate genes associated with the genomic basis of climate adaptation,which were enriched in functional categories related to development processes and stress response;both temperature and precipitation are important factors driving adaptive variation.In addition to single-nucleotide polymorphisms(SNPs),a total of 27 single-methylation variants(SMVs)had significant correlation with at least one of four climatic variables and 16 SMVswere located in or adjacent to genes,several of whichwere predicted to be involved in plant response to abiotic or biotic stress.Gradient forest analysis indicated that the central/east populations were predicted to be at higher risk of future population maladaptation under climate change.Our results demonstrate that local climate factors impose strong selection pressures and lead to local adaptation.Such information adds to our understanding of adaptive mechanisms to variable climates revealed by both population genome and epigenome analysis.

Collaborative inference(co-inference) accelerates deep neural network inference via extracting representations at the device and making predictions at the edge server, which however might disclose the sensitive information about private attributes of users(e.g.,race). Although many privacy-preserving mechanisms on co-inference have been proposed to eliminate privacy concerns, privacy leakage of sensitive attributes might still happen during inference. In this paper, we explore privacy leakage against the privacy-preserving co-inference by decoding the uploaded representations into a vulnerable form. We propose a novel attack framework named AttrL eaks, which consists of the shadow model of feature extractor(FE), the susceptibility reconstruction decoder,and the private attribute classifier. Based on our observation that values in inner layers of FE(internal representation) are more sensitive to attack, the shadow model is proposed to simulate the FE of the victim in the blackbox scenario and generates the internal representations.Then, the susceptibility reconstruction decoder is designed to transform the uploaded representations of the victim into the vulnerable form, which enables the malicious classifier to easily predict the private attributes. Extensive experimental results demonstrate that AttrLeaks outperforms the state of the art in terms of attack success rate.

An ultrafine lamellar-structured martensite steel fabricated by heavy warm rolling(HWR)has shown an excellent combination of strength and ductility.By appending tempering at 400℃to HWR,we show that the comprehensive mechanical property of a lamellar-structured low-carbon martensite steel can be further improved to reach a yield strength of~1.8 GPa,an ultimate tensile strength of~2.0 GPa and a total elongation of~9.3%.This is achieved by tempering the HWR steel from 300 to 750℃,and the optimum tempering temperature is thus obtained.We find that the tempered ultrafine lamellar martensite contains high-density nanoprecipitates dispersed within the aligned martensite laths with reduced crystallographic variations.The ultrahigh strength of the steel is rationalized as mainly the result of grain boundary strengthening and precipitation strengthening,which contribute to yield stress by 610 MPa and 440 MPa,respectively.The good ductility is believed to be closely related to the capacity of the tempered grains to accommodate dense dislocations upon plastic deformation.The present thermomechanical processing provides a feasible routine for producing steels with ultrahigh-strength and good-ductility.

Evolutionary convergence is one of the most striking examples of adaptation driven by natural selection.However, genomic evidence for convergent adaptation to extreme environments remains scarce.Here, we assembled reference genomes of two alpine plants, Saussurea obvallata(Asteraceae)and Rheum alexandrae(Polygonaceae), with 37,938 and 61,463 annotated protein-coding genes. By integrating an additional five alpine genomes,we elucidated genomic convergence underlying high-altitude adaptation in alpine plants. Our results detected convergent contractions of diseaseresistance genes in alpine genomes, which might be an energy-saving strategy for surviving in hostile environments with only a few pathogens present.We identified signatures of positive selection on a set of genes involved in reproduction and respiration(e.g., MMD1, NBS1, and HPR), and revealed signatures of molecular convergence on genes involved in self-incompatibility, cell wall modification,DNA repair and stress resistance, which may underlie adaptation to extreme cold, high ultraviolet radiation and hypoxia environments. Incorporating transcriptomic data, we further demonstrated that genes associated with cuticular wax and flavonoid biosynthetic pathways exhibit higher expression levels in leafy bracts, shedding light on the genetic mechanisms of the adaptive “greenhouse” morphology. Our integrative data provide novel insights into convergent evolution at a high-taxonomic level,aiding in a deep understanding of genetic adaptation to complex environments.

Near infrared(NIR)spectroscopy is now widely used influidized bed granulation.However,there are still some demerits that should be overcome in practice.Valid spectra selection during modeling process is now a hard nut to crack.In this study,a novel NIR sensor and a cosine distance method were introduced to solve this problem in order to make thefluidized process into"visualization".A NIR sensor wasfixed on the side of the expansion chamber to acquire the NIR spectra.Then valid spectra were selected based on a cosine distance method to reduce the influence of dynamic disturbances.Finally,spectral pretreatment and wavelength selection methods were investigated to establish partial least squares(PLS)models to monitor the mois-ture content.The results showed that the root mean square error of prediction(RMSEP)was 0.124%for moisture content model,which was much lower than that without valid spectra selection treatment.All results demonstrated that with the help of valid spectra selection treatment,NIR sensor could be used for real-time determination of critical quality attributes(CQAs)more accurately.It makes the manufacturing easier to understand than the process parameter control.

At present,Tradition Chinese Medicine(TCM)industry in China is in the stage from the empirical development to industrial production.Near infrared(NIR)spectroscopy has been widely used in the quality control of TCM's modernization with its characteristics incuding rapidness,nondestruction,simplicity,economy,and so on.In this study,as one type of a portable micro NIR spectrometer,Micro NIR 1700 was used to establish the qualitative models for identification of geographical region and authenticity of Radix codonopsis based on discriminant analysis(DA)method.Both of the DA models had better predictive ability with 100%accuracy.In addition,a method for rapid quantitative analysis of polysaccharide in Radix codonopsis was also developed based on Micro NIR 1700 spectrometer with partial last-squares(PLS)algo-rithm.In the PLS calibration model,the NIR.spectra of samples were pretreated with different preprocessing methods and the spectral region was selected with different variable selection methods as well.The performance of the final PLS model was evaluated according to correlation cofficient of calibration(Rc),correlation coefficient of prediction(Rp),root mean squared error of cross validation(RMSECV),and root mean squared of prediction(RMSEP).The values of Re,Rp,RMSECV,and RMSEP were 0.9775,0.9602,2.496,and 2.734 g/mL,respectively.This work demonstrated that micro infrared spectrometer could be more convenient and rapid for quality control of Radix codonopsis,and the presented models would be a u1seful reference for quality control of other simi lar raw materials of TCM.
