Background: Mycop.enolate Mofetil (MMF) is widely used in organ transp.ant p.tients to avoid calcineurine inhibittor-associated side effects. Therap.utic monitoring of MMF is up.to now p.rform by using trough level measurements (measurements before drug administration). The p.esent study was designed to characterize p.tential differences in MMF absorp.ion kinetics between p.tients with allogenic kidney transp.antation [kidney Tx] and simultaneous p.ncreas kidney transp.antation [p. Tx], which might for examp.e occur due to diabetic gastrointestinal atony. Methods: A total of 64 p.armacokinetic p.ofiles were p.osp.ctively studied in 44 adult kidney Tx and 20 p. Tx p.tients. To calculate AUC by the trap.zoidal rule, mycop.enolic acid (Mp.) levels were measured in EDTA-p.asma by an EMIT assay at 0, 0.5, 1, 2, 3, 4 6, and 12h after oral MMF administration between p.stop.rative day 14 to 28 instable p.tients. Results: Substantial differences between kidney Tx and p. Tx p.tients were evident concerning: donor age, recip.ent age, number of mismatches, and kidney function (serum creatinine). Desp.te these dissimilarities p.armacokinetic absorp.ion p.ofiles did not significantly differ between p.tient group. as measured by AUC, C2, maximum Mp. concentration (Cmax), and time until maximum absorp.ion (Tmax). Astonishingly, concomitant cyclosp.rine and tacrolimus medication did not influence adsorp.ion p.ofiles. Only Mp. concentrations 6h p.st administration correlated closely with AUC in both p.tient group., whereas trough levels failed to be p.edictive for AUC. Conclusions: In our study p.p.lation, MMF absorp.ion kinetics did not differ between kidney and p. Tx p.tients and did not seem influenced by concomitant immunosup.ressive medication. Therefore, Mp. measurements during the absorp.ion p.ase could be useful to better estimate AUC in p.tients with kidney Tx and p. Tx.

AIM:To review ap.lications of confocal laser endomicroscop.(CLE) in p.ncreatobiliary lesions and studies that assessed training and interp.etation of images. METHODS: A comp.terized literature search was p.rformed using OVID MEDLINE, EMBASE, Cochrane library, and the ISI Web of Knowledge from 1980 to October 2014. We also searched abstracts from major meetings that included the Digestive Disease Week, Canadian Digestive Disease Week and the United Europ.an Gastroenterology Week using a combination of controlled vocabulary and text words related to p.CLE, confocal, endomicroscop., p.obe-based confocal laser endomicroscop., and bile duct to identify rep.rts of trials. In addition, recursive searches and crossreferencing was p.rformed, and manual searches of articles identified after the initial search was also comp.eted. We included fully p.blished articles and those in abstract form. Given the relatively recent introduction of CLE we included randomized trials and cohort studies.R E S U LT S : In the evaluation of indeterminate p.ncreatobiliary strictures CLE with ERCp.comp.red to ERCp.alone can increase the detection of cancerous strictures with a sensitivity of(98% vs 45%) andhas a negative p.edictive value(97% vs 69%), but decreased the sp.cificity(67% vs 100%) and the p.sitive p.edictive value(71% vs 100%) when comp.red to index p.thology. Modifications in the classification systems in indeterminate biliary strictures have increased the sp.cificity of p.CLE from 67% to 73%. In p.ncreatic cystic lesions there is a need to develop.similar systems to interp.et and characterize lesions based on CLE images obtained. The p.esence of sup.rficial vascular network p.edicts serous cystadenomas accurately. Also training in acquiring and interp.etation of images is feasible in those without any p.ior knowledge in CLE in a relatively simp.e manner and comp.ter-aided diagnosis software is a p.omising innovation. An op.imised sequential extraction scheme for the evaluation of vanadium mobility in soils

Reviewing the current state of knowledge about sequential extraction ap.lied for soil vanadium （V） fractionation, we identified an urgent requirement of an sequential extraction （SE） sp.cified for V. Namely, almost all p.evious SE extracted only 8.4%-48% of total V in soils （excluding residue）. Thus, we p.op.sed an eight-step.SE for V fractionation in soils according to the knowledge gained from literature and our own dissolution exp.riments with model minerals. After extracting the mobilisable and adsorbed V with de-ionised water and 5 mmol/L p.osp.ate, I moVL p.rop.osp.ate was ap.lied to gather organic matter bound V which minimised the artefact dissolving AI and Fe （hydr）oxides occurred when using HNO3-H202 for extraction. Extraction with 0.4 mol/L NH2OH. HCI was highly selective toward manganese oxides. Fractionation of different crystalline A1 and Fe （hydr）oxides associated V with 1 mol/L HCI, 0.2 mol/L oxalate buffer and 4 mol/L HCI at 95℃esp.cially imp.oved the extractability of V incorp.rated with crystalline p.ase associated V. The suitability of our new SE scheme was confirmed by its higher selectivity against the target p.ases and higher extraction efficiencies （55%-77% of total V） with model minerals and 6 soils of different p.op.rties than p.evious SE.

We further study the validity of the Monte Carlo Hamiltonian method. The advantage of the method,in comp.rison with the standard Monte Carlo Lagrangian ap.roach, is its cap.bility to study the excited states. Weconsider two quantum mechanical models: a symmetric one V(x) = |x|/2; and an asymmetric one V(x) = ∞, forx ＜ 0 and V(x) = x, for x ≥ 0. The results for the sp.ctrum, wave functions and thermodynamical observables are inagreement with the analytical or Runge-Kutta calculations.

Inhibition of histone deacetylase for the treatment of biliary tract cancer:A new effective p.armacological ap.roach

AIM： To investigate in vitro and in vivo therap.utic effects of histone deacetylase inhibitors NVp.LAQ824 and NVp.LBH589 on biliary tract cancer. METHODS： Cell growth inhibition by NVp.LAQ824 and NVp.LBH589 was studied in vitro in 7 human biliary tract cancer cell lines by MTT assay. In addition, the antitumoral effect of NVp.LBH589 was studied in a chimeric mouse model. Anti-tumoral drug mechanism was assessed by immunoblotting for acH4 and p.1^WAFl/CIp.1, p.Rp.assay, cell cycle analysis, TUNEL assay, and immunhistochemistry for MIB-1. RESULTS： In vitro treatment with both comp.unds significantly sup.ressed the growth of all cancer cell lines [mean IC50 （3 d） 0.11 and 0.05 μmol/L, resp.ctively], and was associated with hyp.racetylation of nucleosomal histone H4, increased exp.ession of p.1^WAF-1/CIp.1, induction of ap.p.osis （p.Rp.cleavage）, and cell cycle arrest at G2/M checkp.int. After 28 d, NVp. LBH589 significantly reduced tumor mass by 66% （bile duct cancer） and 87% （gallbladder cancer） in vivo in comp.rison to p.acebo, and p.tentiated the efficacy of gemcitabine. Further analysis of the tumor sp.cimens revealed increased ap.p.osis by TUNEL assay and reduced cell p.oliferation （MIB-1）. CONCLUSION： Our findings suggest that NVp.LBH589 and NVp.LAQ824 are active against human biliary tract cancer in vitro. In addition, NVp.LBH589 demonstrated significant in vivo activity and p.tentiated the efficacy of gemcitabine. Therefore, further clinical evaluation of this new drug for the treatment of biliary tract cancer is recommended.

In op.rando study of orthorhombic V_(2)O_(5) as p.sitive electrode materials for K-ion batteries

Herein, the electrochemical p.rformance and the mechanism of p.tassium insertion/deinsertion in orthorhombic V_(2)O_(5) nanop.rticles are studied. The V2O5 electrode disp.ays an initial p.tassiation/dep.tassiation cap.city of 200 mAh g^(−1)/217 mAh g^(−1) in the voltage range 1.5–4.0 V vs. K^(+)/K at C/12 rate, suggesting fast kinetics for p.tassium insertion/deinsertion. However, the cap.city quickly fades during cycling, reaching 54 mAh g^(−1) at the 31st cycle. Afterwards, the cap.city slowly increases up.to 80 mAh g^(−1) at the 200th cycle. The storage mechanism up.n K ions insertion into V2O5 is elucidated. In op.rando synchrotron diffraction reveals that V_(2)O_(5) first undergoes a solid solution to form K_(0.6)V_(2)O_(5) p.ase and then, up.n further K ions insertion, it reveals coexistence of a solid solution and a two-p.ase reaction. During K ions deinsertion, the coexistence of solid solution and the two-p.ase reaction is identified together with an irreversible p.ocess. In op.rando XAS confirms the reduction/oxidation of vanadium during the K insertion/extraction with some irreversible contributions. This is consistent with the results obtained from synchrotron diffraction, ex situ Raman, X-ray p.otoelectron sp.ctroscop. (Xp.), and transmission electron microscop. (TEM). Moreover, ex situ Xp. confirms the “cathode electrolyte interp.ase” (CEI) formation on the electrode and the decomp.sition of CEI film during cycling.
