BAcKGrOUND Toll-like receptor-2(TLr2) is responsible for recognizing Helicobacter pylori(***) and activating the immune response. Polymorphisms in TLr2 may modulate gastric *** To evaluate whether the TLr2 19216 T/c(rs3804099) and TLr2-196 to-174 ins/del(rs111200466) polymorphisms contribute to gastric carcinogenesis in the Brazilian population, and to determine the influence of both polymorphisms and H. pylori infection on TLr2 mrNA *** DNA was extracted from 854 peripheral blood leukocyte or gastric tissue samples[202 gastric cancer(Gc), 269 chronic gastritis(cG), and 383 control/healthy(c)]and genotyped by allele-specific Pcr or restriction fragment length polymorphism(rFLP)-Pcr. Quantitative polymerase chain reaction by Taq Man■ assay was used to quantify TLr2 mrNA levels in fresh gastric tissues(48 Gc, 36 cG, and 14 c).rESULTS regarding the TLr2-196 to -174 polymorphism, the ins/del and del/del genotypes were associated with a higher risk of Gc by comparison with the c in all of the analyzed inheritance models(codominant, dominant, recessive, overdominant and log-additive;P rly, an increased risk was observed when comparing the Gc and cG groups [codominant(P recessive(P = 0.0260), overdominant(P contrast, TLr2 19216 T/c was associated with a protective effect in the Gc group compared to the c group [dominant(P = 0.0420) and log-additive(P =0.0300)]. regarding the association of polymorphisms with H. pylori infection,individuals infected with H. pylori and harboring the TLr2-196 to-174 ins/del polymorphism had an increased risk of gastric carcinogenesis [codominant(P =0.0120), dominant(P = 0.0051), overdominant(P = 0.0240) and log-additive(P =0.0030)], while TLr2 19216 T/c was associated with a protective effect[codominant(P = 0.0039), dominant(P rdominant(P = 0.0097) and log-additive(P = 0.0021)]. AIM: To assess the aetiological role of Helicobacter pylori (H. pylori) infection in adult patients with ironrefractory or iron-dependent anaemia of previously unknown origin. METHODS: consecutive patients with chronic irondeficient anaemia (IDA) with H. pylori infection and a negative standard work-up were prospectively evaluated. All of them had either iron refractoriness or iron dependency. response to H. pylori eradication was assessed at 6 and 12 mo from follow-up. H. pylori infection was considered to be the cause of the anaemia when a complete anaemia resolution without iron supplements was observed after eradication. rESULTS: H. pylori was eradicated in 88 of the 89 patients. In the non-eradicated patient the four eradicating regimens failed. There were violations of protocol in 4 patients, for whom it was not possible to ascertain the cause of the anaemia. Thus, 84 H. pylori eradicated patients (10 men; 74 women) were available to assess the effect of eradication on IDA. H. pylori infection was considered to be the aetiology of IDA in 32 patients (38.1%; 95%cI: 28.4%-48.8%). This was more frequent in men/postmenopausal women than in premenopausal women (75% vs 23.3%; P r of 9.8 (95%cI: 3.3-29.6). In these patients, anaemia resolution occurred in the first follow-up visit at 6 mo, and no anaemia or iron deficiency relapse was observed after a mean follow-up of 21 ± 2 mo. cONcLUSION: Gastric H. pylori infection is a frequent cause of iron-refractory or iron-dependent anaemia of previously unknown origin in adult patients.

The aim of this study was to identify three-dimensional microstructural changes of trabecular bone with age and gender, using micro-computed tomography. Human trabecular bone from two disease groups, osteoporosis and osteoarthritis was analyzed. A prior analysis of the effects of some procedure variables on the micro-cT results was performed. Preliminary micro-cT scans were performed with three voxel resolutions and two acquisition conditions. On the reconstruction step, the image segmentation was performed with three different threshold values. Samples were collected from patients, with coxarthrosis (osteoarthritis) or fragility fracture (osteoporosis). The specimens of the coxarthrosis group include twenty females and fifteen males, while the fragility fracture group was composed by twenty three females and seven males. The mean age of the population was 69 ± 11 (females) and 67 ± 10 years (males), in the coxarthrosis group, while in the fragility fracture group was 81 ± 6 (females) and 78 ± 6 (males) years. The 30 μm voxel size provided lower percentage difference for the microarchitecture parameters. Acquisition conditions with 160 μA and 60 kV permit the evaluation of all the volume’s sample, with low average values of the coefficients of variation of the microstructural parameters. No statistically significant differences were found between the two diseases groups, neither between genders. However, with aging, there is a decrease of bone volume fraction, trabecular number and fractal dimension, and an increase of structural model index and trabecular separation, for both disease groups and genders. The parameters bone specific surface, trabecular thickness and degree of anisotropy have different behaviors with age, depending on the type of disease. While in coxarthrosis patients, trabecular thickness increases with age, in the fragility fracture group, there is a decrease of trabecular thickness with increasing age. Objective: To describe the demographic and evolutionary characteristics of pregnant and postpartum women with coronavirus disease 2019 (cOVID-19) admitted to a medium-sized hospital in Brazil. Method: This is a descriptive and retrospective study, collected from medical records, from March 2020 to October 2021 in a hospital located in cuiabá (MT). results: Pregnant and puerperal women with cOVID-19 who needed hospitalization were mixed-race, from the metropolitan area, and carriers of moderate and severe forms of the disease. The primary risk condition found was overweight/obesity, and pre-gestational diabetes, hypertension, asthma, and autoimmune disease were the most prevalent comorbidities in the group. Elevated lactate dehydrogenase (LDH), c-reactive protein (crP), and D-dimer were relevant laboratory findings in this group of patients. The most frequent maternal outcomes were respiratory failure, invasive ventilatory support, thromboembolic phenomena, sepsis, and preterm labor. Maternal death occurred in 6.4% of pregnant women. Most maternal deaths were of women who lived in the interior of the state, and the minority arrived on adequate ventilatory support. Prematurity and the need for neonatal intensive care unit (NIcU) were significant complications for neonates. Stillbirth/neonatal mortality occurred in 11.0%. conclusion: The clinical conditions at hospitalization were associated with worse living conditions and lack of access to health care, resulting in increased chances of severity and worsening outcomes in this group of women and neonates.

AIM:To verify the methylation status of cDH1, DAPK, cOX2, hMLH1 and cDKN2A genes and to evaluate their association with Helicobacter pylori (H. pylori)-cagA+ and Epstein Barr virus (EBV) infections in gastric ***: Methylation-specific Pcr (MSP) assay was performed in 89 primary gastric carcinomas (intestinal and diffuse types). Microsatellite instability (MSI) analysis was performed using the BAT26 primer set and Pcr products were analyzed with the ABI PrISM 3100 Genetic Analyzer using Genescan 3.7 software (Applied Biosystems). Detection of H. pylori and genotyping were performed by Pcr, using specifi c primers for ureasec and cagA genes. The presence of EBV was assessed by in situ hybridization. Statistical analyses were performed using the χ2 or Fisher's exact ***: The most frequent hypermethylated gene was cOX-2 (63.5%) followed by DAPK (55.7%), cDH1 (51%), cDKN2A (36%) and hMLH1 (30.3%). Intestinal and diffuse adenocarcinomas showed different methylation profiles and there was an association between methylation of E-cDH1 and H. pylori-cagA+ in the intestinal adenocarcinoma type. MSI was correlated with hMLH1 methylation. There was an inverse correlation between DAPK hypermethylation and ***: We found a strong association between cDH1 methylation and H. pylori-cagA+ in intestinal-type gastric cancer, association of MSI and better prognosis and an heterogeneous cOX-2 overexpression.
