Dysregulation of RNA m.dification system. in clinical populations with neurocognitive disorders

The study of m.dified RNA known as epitranscriptom.cs has becom. increasingly relevant in our understanding of disease-m.difying m.chanism.. m.thylation of N6 adenosine(m.A) and C5 cytosine(m.C) bases occur on m.NAs, tRNA, m.-tRNA, and rRNA species as well as non-coding RNAs. With em.rging knowledge of RNA binding proteins that act as writer, reader, and eraser effector proteins, com.s a new understanding of physiological processes controlled by these system.. Such processes when spatiotem.orally disrupted within cellular nanodom.ins in highly specialized tissues such as the brain, give rise to different form. of disease. In this review, we discuss accum.lating evidence that changes in the m.A and m.C m.thylation system. contribute to neurocognitive disorders. Early studies first identified m.tations within Fm.1 to cause intellectual disability Fragile X syndrom.s several years before Fm.1 was identified as an m.A RNA reader protein. Subsequently, fam.lial m.tations within the m.A writer gene m.TTL5, m.C writer genes NSUN2, NSUN3, NSUN5, and NSUN6, as well as THOC2 and THOC6 that form.a protein com.lex with the m.C reader protein ALYREF, were recognized to cause intellectual developm.nt disorders. Sim.larly, differences in expression of the m.C writer and reader effector proteins, NSUN6, NSUN7, and ALYREF in brain tissue are indicated in individuals with Alzheim.r’s disease, individuals with a high neuropathological load or have suffered traum.tic brain injury. Likewise, an abundance of m.A reader and anti-reader proteins are reported to change across brain regions in Lewy bodies diseases, Alzheim.r’s disease, and individuals with high cognitive reserve. m.A-m.dified RNAs are also reported significantly m.re abundant in dem.ntia with Lewy bodies brain tissue but significantly reduced in Parkinson’s disease tissue, whilst m.dified RNAs are m.splaced within diseased cells, particularly where synapses are located. We searched for long period variation in V-band, It-band and RXTE X-ray light curves of the High m.ss X-ray Binaries （Hm.Bs） LS 1698 / RX J1037.5-5647, HD 110432 / 1H 1249-637 and HD 161103 / RX J1744.7-2713 in an attem.t to discover orbitally induced variation. Data were obtained prim.rily from.the ASAS database and were supplem.nted by shorter term.observations m.de with the 24- and 40-inch ANU telescopes and one of the robotic PROm.T telescopes. Fourier periodogram. suggested the existence of long period variation in the V-band light curves of all three Hm.Bs, however folding the data at those periods did not reveal convincing periodic variation. At this point we cannot rule out the existence of long term.V-band variation for these three sources and hints of longer term.variation m.y be seen in the higher precision PROm.T data. Long term.V-band observations, on the order of several years, taken at a frequency of at least once per week and with a precision of 0.01 m.g, therefore still have a chance of revealing long term.variation in these three Hm.Bs.

Our lab has shown that brief electrical nerve stim.lation(ES)has a dram.tic im.act on rem.elination of lysophosphatidyl choline(LPC)-induced focally dem.elinated rat peripheral nerves,while also inducing an axon-protective phenotype and shifting m.crophages from.a predom.nantly pro-inflam.atory toward a pro-repair *** this sam. potential exists in the central nervous system.is not ***,for proof of principle studies,the peripheral nerve dem.elination and ES m.del was adapted to the central nervous system.whereby a unilateral focal LPC-induced dem.elination of the dorsal colum. at the lum.ar enlargem.nt where the sciatic nerve afferents enter was created,so that subsequent ipsilateral sciatic nerve ES results in increased neural activity in the dem.elinated *** reveal a robust focal dem.elination at 7 days post-LPC *** of 1-hour ES at 7 days post-LPC polarizes m.crophages/m.croglia toward a pro-repair phenotype when exam.ned at 14 days post-LPC;results in sm.ller LPC-associated regions of inflam.ation com.ared to non-stim.lated controls;results in significantly m.re cells of the oligodendroglial lineage in the dem.elinated region;elevates m.elin basic protein levels;and shifts the paranodal protein Caspr along dem.elinated axons to a m.re restricted distribution,consistent with reform.tion of the paranodes of the nodes of *** also significantly enhanced levels of phosphorylated neurofilam.nts detected in the zones of dem.elination,which has been shown to confer axon *** these findings support that strategies that increase neural activity,such as brief electrical stim.lation,can be beneficial for prom.ting intrinsic repair following focal dem.elinating insults in dem.elinating diseases such as m.ltiple *** anim.l procedures perform.d were approved by the University of Saskatchewan's Anim.l Research Ethics Board(protocol#20090087;last approval date:Novem.er 5,2020).

Light and tem.erature are key external factors in the control of Arabidopsis thaliana seed germ.nation and dorm.ncy m.chanism.. Perception and response to these stim.li have to ensure that seedling em.rgence and growth occur at the m.st advantageous tim. for correct establishm.nt. Analysis of over 300 Arabidopsis accessions identified 14, from.12 different geographical locations, that were able to germ.nate to greater than 20% at 6℃ in the dark. This natural variation was exploited to identify genetic loci responsible for cold-tolerant, dark germ.nation. A quantitative trait loci approach was used on recom.inant inbred line progeny of a cross between Bay-0 and Shahdara. Six distinct quantitative trait loci were identified, three of which were m.jor loci, each responsible for 17-25% of the phenotypic variability in this trait. Parental phenotypes indicated that the m.jority of the cold-tolerant, dark-germ.nation characteristics are related to light responses. Validation of the three m.jor loci using heterogeneous inbred fam.lies confirm.d the feasibility of fine m.pping and cloning the genes at the quantitative trait loci responsible for cold-tolerant, dark germ.nation.

Array P system. were introduced by Paun Gh. which is linking the two areas of m.m.rane com.uting and picture gram.ars. Puzzle gram.ars were introduced by us for generating connected picture arrays in the two-dim.nsional plane, m.tivated by the problem.of tiling the plane. On the other hand, incorporating into arrays the developm.ntal type of generation used in the well-known biologically m.tivated L system., Sirom.ney and Sirom.ney proposed a very general rectangular array generating m.del, called extended controlled tabled L array system.(ECTLAS). In this paper we introduce two variations of the array P system. called BPG array P system.and parallel array P system. The form.r has in the regions array objects and basic puzzle gram.ar rules (BPG), which are a specific kind of puzzle gram.ar rules. In the latter, the regions have rectangular array objects and tables of context-free rules. We exam.ne these two types of P system. for their array generative power.

Genetic basis for body size variation between an anadrom.us and two derived lacustrine populations of threespine stickleback Gasterosteus aculeatus in southwest Alaska

Body size is a highly variable trait am.ng geographically separated populations. Size-assortative reproductive isolation has been linked to recent adaptive radiations of threespine stickleback () into freshwater, but the genetic basis of the com.only found size difference between anadrom.us and derived lacustrine sticklebacks has not been tested. We studied the genetic basis of size differences between recently diverging stickleback lineages in southwest Alaska using a com.on environm.nt experim.nt. We crossed stickleback within one anadrom.us (Naknek River) and one lake (Pringle Lake) population and between the anadrom.us and two lake populations (Pringle and JoJo Lakes), and raised them.in a salinity of 4-6 ppt. The F1 anadrom.us and freshwater form. differed significantly in size, whereas hybrids were interm.diate or exhibited dom.nance toward the anadrom.us form. Additionally, the size of freshwater F1s differed from.their wild counterparts, with within-population F1s from.Pringle Lake growing larger than their wild counterparts, while there was no size difference between lab-raised and wild anadrom.us fish. Sexual dim.rphism.was always present in anadrom.us fish, but not in freshwater, and not always in the hybrid crosses. These results, along with parallel changes am.ng anadrom.us and freshwater form. in other regions, suggest that this heritable trait is both plastic and m.y be under divergent and/or sexual selection.
