Rabbit Annulus Fibrosus Cell Apoptosis Induced by Mechanical Overload via a Mitochondrial Apoptotic Pathway

In order to investigate the apoptotic pathway of rabbit annulus fibrosus(AF) cells induced by mechanical overload,an experimental air-pressure model was established in this study to pressurize the rabbit AF cells in *** were randomly divided into five groups in which the cells were exposed to a continuous pressure of 1.1 MPa for different lengths of time(0,5,12,24 and 36 h).The cell proliferation and apoptosis were detected by cell counting kit-8(CCK-8) assay and flow cytometry;the alterations in mitochondrial membrane potential were measured by fluorescence microscopy and fluorescence spectrophotometer;the activities of caspase-8 and 9 were determined by *** results showed that after the cells were subjected to the pressure for 24 or 36 h,the cell proliferation was inhibited;the ratio of cell apoptosis was increased;the mitochondrial membrane potential was decreased;the activity of caspase-9 was enhanced;no activity changes were observed in *** results suggested that treatment with a pressure of 1.1 MPa for more than 24 h can lead to the proliferation inhibition and the apoptosis of rabbit AF cells in vitro,and the mitochondrial-dependent pathway is implicated in the pressure-induced AF cell apoptosis.

Identification of <i>JAK2</i>(V617F) Mutation in Myeloproliferative Neoplasms by Using Allele Specific Polymerase Chain Reaction (AS-PCR)

Myeloproliferative neoplasms (MPNs) are a group of clonal haematopoietic stem cell disorders characterized by the proliferation of one or more myeloid cell lineages. According to WHO classification, the Janus associated kinase 2 (JAK2) V617F mutation is one of the major diagnostic criteria in BCR-ABL1 negative myeloproliferative neoplasms. The aim of this study is to detect the JAK2 (V617F) mutation in patients with myeloproliferative neoplasms to get accurate diagnosis and proper management. A total of 90 clinically diagnosed MPN patients attending to Department of Clinical Haematology, Yangon General Hospital were enrolled in this study. The mean age was 53.4 ± 14 years which ranged from 16 to 81 years old and male and female ratio was 2.4:1. The identification of JAK2 (V617F) point mutation was found to be positive in 44/90 MPN patients (48.9%). According to MPN subtypes, the JAK2 mutation positivity was found in 19 out of 46 polycythemia vera patients (41.3%), 17 out of 25 essential thrombocythemia patients (68%), 8 out of 15 primary myelofibrosis patients (53.3%), 0 of 4 others myeloproliferative neoplasms (0%). Confirmation of each of nine JAK2 mutation positive and negative samples was done by Sanger sequencing. The arterial or venous thrombotic attack was found in 32/44 JAK2 mutation positive cases (72.7%) and 12/44 JAK2 mutation negative cases (27.3%). The association between thrombotic attack and presence of JAK2 mutation was statistically significance with p = 0.000. The diagnosis of myeloproliferative neoplasms mainly relies on the molecular genetics according to WHO classification. The Allele specific PCR reaction is sensitive, simple test and relatively cost-effective. Therefore, the identification of JAK2 (V617F) somatic point mutation by AS-PCR should be implemented as

Challenges in early phase of implementing the 1-3-7 surveillance and response approach in malaria elimination setting:A field study from Myanmar

Background:The National Plan for Malaria Elimination(NPME)in Myanmar(2016–2030)aims to eliminate indigenous Plasmodium falciparum malaria in six states/regions of low endemicity by 2020 and countrywide by *** achieve this goal,in 2016 the National Malaria Control Program(NMCP)implemented the“1-3-7”surveillance and response *** study aims to identify the barriers to successful implementation of the NPME which emerged during the early phase of the“1-3-7”approach ***:A mixed-methods study was conducted with basic health staff(BHS)and Vector Born Disease Control Program(VBDC)staff between 2017 and 2018 in six townships of six states/regions targeted for sub-national elimination by 2020.A self-administered questionnaire,designed to assess the knowledge required to implement the“1-3-7”approach,was completed by 544 *** analysis was performed for quantitative findings and thematic analysis was conducted for qualitative findings using *** ***:Although 83%of participants reported performing the key activities in the“1-3-7”surveillance and response approach,less than half could report performing those activities within 3 days and 7 days(40 and 43%,respectively).Low proportion of BHS correctly identified six categories of malaria cases and three types of foci(22 and 26%,respectively).In contrast,nearly 80%of respondents correctly named three types of case detection *** cited challenges included‘low community knowledge on health’(43%),‘inadequate supplies’(22%),and‘transportation difficulty’(21%).Qualitative data identified poor knowledge of key surveillance activities,delays in reporting,and differences in reporting systems as the primary *** dominant perceived barrier to success was inability to control the influx of migrant workers into target jurisdictions especially in hard-to-reach *** with township medical officers and the NMCP team leaders further highlighted the necessity o
