Morphometry and subpopulation structure of Holstein bull spermatozoa： variations in ejaculates and cryopreservation straws

Sperm quality is evaluated for the calculation of sperm dosage in artificial reproductive programs. The most common parameter used is motility, but morphology has a higher potential as a predictor of genetic quality. Morphometry calculations from CASA-Morph technology improve morphological evaluation and allow mathematical approaches to the problem. Semen from 28 Holstein bulls was collected by artificial vagina, and several ejaculates were studied. After general evaluation, samples were diluted, packaged in 0.25 ml straws, and stored in liquid nitrogen. Two straws per sample were thawed, and slides were processed and stained with Diff-Quik. Samples were analyzed by a CASA-Morph system for eight morphometric parameters. In addition to the ＂classical＂ statistical approach, based on variance analysis （revealing differences between animals, ejaculates, and straws）, principal component （PC） analysis showed that the variables were grouped into PC1, related to size, and PC2 to shape. Subpopulation structure analysis showed four groups, namely, big, small, short, and narrow from their dominant characteristics, representing 31.0%, 27.3%, 24.1%, and 17.7% of the total population, respectively. The distributions varied between animals and ejaculates, but between straws, there were no differences in only four animals. This modern approach of considering an ejaculate sperm population as divided into subpopulations reflecting quantifiable parameters generated by CASA-Morph systems technology opens a new view on sperm function. This is the first study applying this approach to evaluate different ejaculates and straws from the same individual. More work must be done to improve seminal dose calculations in assisted reproductive programs.

Effect of Substrate Temperature on Structural and Photoluminescence Properties of ZnO：SiOx Composites

Using zinc oxide (ZnO) powder and quartz rods as solid sources and molecular hydrogen (H2) as reactant gas, ZnO: SiOx composites were grown by hot filament chemical vapor deposition (HFCVD) technique. The absorbance spectra showed the formation of ZnO: SiOx composite in the studied temperature range. The crystalline quality of the ZnO lattice in the structure of the composite gradually lost by increasing the substrate temperature. Visible emission from blue to near-infrared region was found in the photoluminescence spectra of the composite formed at 1,100 °C. The contribution of blue, green, yellow and orange emissions present in the photoluminescence spectra of the composite formed at 1,100 °C was attributed to recombination centers produced by the presence of point defects in the structure of ZnO network. This result suggests that the ZnO in the ZnO: SiOx composite obtained at substrate temperature of 1,100 °C has an amorphous phase.

Object Detection Using SURF and Superpixels

This paper proposes a novel object detection method in which a set of local features inside the superpixels are extracted from the image under analysis acquired by a 3D visual sensor. To increase the segmentation accuracy, the proposed method firstly performs the segmentation of the image, under analysis, using the Simple Linear Iterative Clustering (SLIC) superpixels method. Next the key points inside each superpixel are estimated using the Speed-Up Robust Feature (SURF). These key points are then used to carry out the matching task for every detected keypoints of a scene inside the estimated superpixels. In addition, a probability map is introduced to describe the accuracy of the object detection results. Experimental results show that the proposed approach provides fairly good object detection and confirms the superior performance of proposed scene compared with other recently proposed methods such as the scheme proposed by Mae et al.

Experience with the surgical treatment of hepatic hydatidosis: Case series with follow-up

AIM: To describe patient characteristics and the results of surgical treatment for hepatic hydatidosis (HH) in the Hospital Hernán Henríquez Aravena from December 2001 to March 2005. METHODS: Subjects older than 16 years with suspected HH were included based on the clinical picture and radiological studies. Variables examined included clinical and laboratory aspects, outcome, features of the parasite and surgical aspects. Descriptive statistics were used, calculating central tendency, dispersion and extreme tendency. RESULTS: The series was comprised of 122 patients, with an average age of 44 ± 16.9 years. The most frequently used surgical technique was subtotal cys-tectomy in 90% of the patients, followed by hepatic resection (hepatectomy, segmentectomy and subsegmentectomy) in 5%. In 2%, a combination of subtotal cystectomy and segmentectomy was performed. In addition, 28% of the series presented complications in the postoperative period and mortality was 2%. CONCLUSION: The most frequently used surgical technique for HH was subtotal cystectomy and the morbidity and mortality rates in this Chilean series are comparable to other national and international series.

A Vaccine Is Coming but Prevention Is Still up to Us

Despite having a better understanding of the COVID-19 disease, we are facing significant challenges regarding behaviors that enforce the lack of adherence to public health recommendations including the use of face masks and vaccine acceptability. Lessons learned from the 1918 flu pandemic, highlight the importance of wearing a face mask in conjunction with adherence to other public health recommendations. There are two key factors that influence how our communities are translating scientific evidence, denialism and ignorance. Sadly, a real consequence of this pandemic among a significant number of patients with COVID-19 disease is denial. As health care providers we need to take part in addressing these issues and educate our communities especially during this time where we see a second and larger uptake of COVID-19 cases. Fighting denialism and ignorance represents a significant challenge to our society and could have a substantial positive impact on helping with vaccine acceptability and other public health recommendations.

Dynamin Regulates Autophagy by Modulating Lysosomal Function

Autophagy is a central lysosomal degradation pathway required for maintaining cellular homeostasis and its dysfunction is associated with numerous human diseases. To identify players in autophagy, we tested ~ 1200 chemically induced mutations on the X chromosome in Drosophila fat body clones and discovered that shibire （shi） plays an essential role in starvation-induced autophagy, shi encodes a dynamin protein required for fission of clathrin-coated vesicles from the plasma membrane during endocytosis. We showed that Shi is dispensable for autophagy initiation and autophagosome--lysosome fusion, but required for lysosomal/autolysosomal acidification. We also showed that other endocytic core machinery components like clathrin and AP2 play similar but not identical roles in regulating autophagy and lysosomal function as dynamin. Previous studies suggested that dynamin directly regulates autophagosome formation and autophagic lysosome reformation （ALR） through its excision activity, Here, we provide evidence that dynamin also regulates autophagy indirectly by regulating lysosomal function.

Role of microfluctuations in accommodation: a novel approach to reduce non-accommodative noise

Accommodative response and its possible role in myopia development has been explored through the study of the microfluctuations(MFs) of accommodation, which are commonly divided in high(1.0 to 2.3 Hz) and low(0.1 to 0.6 Hz) frequency components. Previous research efforts have evidenced that a certain percentage of the amplitude of MFs seems not to originate in the accommodative response. We aimed to develop and test a new approach to reduce this non-accommodative noise. For this purpose, ten healthy participants were enrolled to determine the difference between the amplitude of MFs at near and distance for each range of frequencies, which was defined as the relative amplitude of MFs. The findings support the exploration of the relative rather than absolute values of the amplitude of MFs to better understand the contribution of both accommodative and non-accommodative factors to MFs.
