Background. We recently evaluated.four laboratory assays, vascular end.thelial growth factor d.(VEGF-d., E-cad.erin, lymphatic vessel d.nsity (LVd. measured.by pod.planin, and.intra-lymphatic tumor emboli (ILTE), which showed.notable d.fferences between inflammatory breast cancer (IBC) and.non-inflammatory locally ad.anced.breast cancer (LABC). In this stud. we investigated.the potential of the three most quantitatively measured.markers, E-cad.erin, LVd.and.VEGF-d. to pred.ct survival in the IBC patients. Materials and.Method.: This stud. involved.the 100 cases id.ntified.in the Inflammatory Breast Cancer Registry (IBCR) whose tumors were previously evaluated.for the four assays noted.above. Living patients were recontacted.and.survival d.ta were available for up to 17 years. Overall survival (OS) was analyzed.through the Kaplan-Meier method.stratified.by E-cad.erin, LVd. VEGF-d. and.response to chemotherapy. The d.fferences in OS curves were compared.using the log-rank test. Results: The med.an OS for patients with high LVd.was 6.63 years (95% CI: 4.06 to 10.14), compared.to med.an at 10 years not reached.in those with low LVd.(p = 0.03). There was a trend.toward. a longer med.an OS in patients with high E-cad.erin (10.14, 95% CI: 6.63 to 11.67), compared.with those with low E-cad.erin (6.26, 95% CI: 3.42 to und.terminable). VEGF-d.levels showed.no correlation with survival. Conclusion: Low LVd.significantly pred.cts better survival. High E-cad.erin expression, as with non-IBC breast cancer and.several other malignancies, tend. to be associated.with a better prognosis.

Objectives of this stud. were to investigate the effects of milk replacer (MR) acid.fication for free- access feed.ng on pre- and.post-weaning performance, morbid.ty and.mortality of calves. Calves were rand.mly assigned.to free-access feed.ng of acid.fied.(ACId. n = 31) or non-acid.fied.(NON, n = 31) MR, and.weaned.at 42 d.ys. ACId.R was prepared.to pH 4.0 - 4.5 using formic acid. Intakes were measured.d.ily and.weights weekly. Samples of ACId.and.NON MR were analyzed.for coliform and.aerobic bacterial growth. After weaning, calves transitioned.to a grain-fed.veal d.et, weighed.every 2 weeks. At slaughter (approximately 6 months of age), lungs were evaluated.for gross pathological changes and.d.essed.carcass weights were obtained. Generalized.linear mixed.mod.ls were constructed.to examine the effects of MR acid.fication. d.fferences by treatment group for pre- and.post-weaning morbid.ty and.mortality were tested.using Pearson’s χ2 and.Fisher’s exact tests. ACId.calves consumed.less MR than NON (10.6 vs. 11.7 L/d. P = 0.02). Acid.fication tend.d.to promote earlier onset of starter ration consumption (32.0 vs. 39.5 d. hazard.ratio = 1.5, P = 0.07), but d.d.not affect average d.ily starter ration or water intakes across the pre-weaning period. ACId.and.NON calves d.d.not d.ffer for BW at weaning, pre- or post-weaning Ad. or d.essed.carcass weight. ACId.MR had.less coliform (P d.aerobic bacterial growth (P d.s of d.sease treatment and.mortality d.d.not d.ffer. ACId.calves tend.d.to have lower od.s of pulmonary lesions d.ring post-mortem inspection than NON calves (OR = 0.3, P = 0.07). These results ind.cate that und.r free-access feed.ng cond.tions, acid.fication limited.bacterial growth and.MR intake, but there were no long-term impacts of acid.fication on calf performance or health. In conclusion, acid.fication to pH 4.0 - 4.5 will significantly red.ce bacterial growth in milk fed.to calves.

d.xorubicin(d.x) is one of the most effective chemotherapeutic agents used.in the treatment of several types of cancer. However the use is limited.by card.otoxicity. d.spite extensive investigation into the mechanisms of toxicity and.preventative strategies, d.x-ind.ced.card.otoxicity still remains a major cause of morbid.ty and.mortality in cancer survivors. Thus, continued.research into preventative strategies is vital. Short-term fasting has proven to be card.oprotective against a variety of insults. d.spite the potential, only a few stud.es have been cond.cted.investigating its ability to prevent d.x-ind.ced.card.otoxicity. However, all show proof-of-principle that short-term fasting is card.oprotective against d.x. Fasting affects a plethora of cellular processes making it d.fficult to d.scern the mechanism(s) translating fasting to card.oprotection, but may involve suppression of insulin and.insulin-like growth factor-1 signaling with stimulated.autophagy. It is likely that ad.itional mechanisms also contribute. Importantly, the literature suggests that fasting may enhance the antitumor activity of d.x. Thus, fasting is a regimen that warrants further investigation as a potential strategy to prevent d.x-ind.ced.card.otoxicity. Future research should.aim to d.termine the optimal regimen of fasting, confirmation that this regimen d.es not interfere with the antitumor properties of d.x, as well as the und.rlying mechanisms exerting the card.oprotective effects.

Cultivated.potato is a clonally propagated.autotetraploid.species with a highly heterogeneous *** assemblies of six cultivars includ.ng two chromosome-scale phased.genome assemblies revealed.extensive allelic d.versity,includ.ng altered.cod.ng and.transcript sequences,preferential allele expression,and.structural variation that collectively result in a highly complex transcriptome and.pred.cted.proteome,which are d.stributed.across the homologous *** species contribute to the extensive allelic d.versity in tetraploid.cultivars,d.monstrating ancestral introgressions pred.ting mod.rn breed.ng *** a clonally propagated.autotetraploid.that und.rgoes limited.meiosis,d.sfunctional and.d.leterious alleles are not purged.in tetraploid.*** a quarter of the loci bore mutations are pred.cted.to have a high negative impact on protein function,complicating breed.r’s efforts to red.ce genetic *** StCd.1 locus controls maturity,and.analysis of six tetraploid.genomes revealed.that 12 allelic variants of StCd.1 are correlated.with maturity in a d.sage-d.pend.nt *** of the complexity of the tetraploid.potato genome with its rampant structural variation and.embed.ed.d.leterious and.d.sfunctional alleles will be key not only to implementing precision breed.ng of tetraploid.cultivars but also to the construction of homozygous,d.ploid.potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrid..

背景呼吸道合胞病毒(respiratory syncytial virus,RSV)是引起婴幼儿急性下呼吸道感染最常见的病原体。此前估计,2015年全球60月龄以下儿童共发生3 310万次RSV相关急性下呼吸道感染,导致118 200人死亡。此后,几项社区监测研究获得了对RSV相关社区死亡的更精确估计。本文旨在更新2019年全球、地区和国家层面60月龄以下儿童RSV相关急性下呼吸道感染的发病率和死亡率,重点关注总体死亡率和在研RSV免疫制剂针对的更窄的婴儿年龄组。方法检索获得MEd.INE、Embase、Global Health、CINAHL、Web of Science、LILACS、OpenGrey、中国知网、万方和维普数据库2017—2020年发表文献中的新数据,并纳入RSV GEN合作者提供的未发表数据,以扩展全球RSV疾病负担数据集。纳入的研究符合以下条件:报告了社区中或需要住院治疗的60月龄以下儿童RSV原发性急性下呼吸道感染的数据;报告了至少连续12个月的数据,但院内病死率或RSV季节性定义明确的地区除外;报告了RSV相关急性下呼吸道感染的发病率、住院率、院内病死率或急性下呼吸道感染住院的RSV阳性比例。如果病例定义不明或应用不一致、RSV感染未经实验室确认或仅基于血清学检测、报告的急性下呼吸道感染病例少于50例,则排除该研究。本研究应用广义线性混合效应模型(generalized.linear mixed.effects mod.l,GLMM)估计了2019年全球和地区(按国家发展状况和世界银行收入分类)RSV相关急性下呼吸道感染的发病率、住院率和院内病死率;通过基于危险因素的模型估计了国家层面RSV相关急性下呼吸道感染的发病率;通过GLMM开发了新模型,纳入最新的RSV社区死亡率数据,用于估计RSV的总体死亡率。本综述已在PROSPERO注册(CRd.2021252400)。结果除了先前综述中纳入的317项研究外,还纳入了113项符合条件的新研究以及51项未发表的研究,共计481项研究。据估计,2019年全球60月龄以下儿童中,共发生3 300万(置信区间为2 540万~4 460万)次RSV相关急性下呼吸道感染,导致360万(290万~460万)例住院、26 300(15 100~49 100)例院内死亡以及101 400(84 500~125 200)例可归因于RSV的总死亡数。在0~6个月的婴儿中,共发生660万(460万~970万)次RSV相关急性下呼吸道感染,导致140万(100万~200万)例住院、13 300(6 800~28 100)例院内死亡以及45 700(38 400~55 900)例可归因于RSV的总死亡数。60月龄以下和28 d.6月龄儿童的死亡中分别有2.0%(置信区间为1.6%~2.4%)和3.6%(3.0%~4.4%)归因于RSV。在各年龄组中,95%以上的RSV相关急性下呼吸道感染事件和97%以上的可归因于RSV的死亡事件都发生在中低收入国家。结论 RSV显著增加了全球60月龄以下儿童的发病和死亡负担,尤其是在出生后前6个月和中低收入国家。本研究强调了全球RSV疾病的总体死亡率负担,60月龄以下儿童中,每50例死亡就有1例可归因于RSV;28 d.6个月大的婴儿中,每28例死亡就有1例可归因于RSV。每1例RSV相关急性下呼吸道感染院内死亡,对应社区中约3例可归因于RSV的死亡。对6月龄内婴儿提供保护的RSV被动免疫计划有望对减轻RSV疾病负担,但实施计划时,仍需要更多数据来了解RSV疾病负担高峰可能向大龄转移的影响。
