An examination of the population dynamics of syngnathid fishes within Tampa Bay,Florida,USA

Seagrass ecosystems worldwide have been declining, leading to a decrease in associated fish populations, especiallythose with low mobility such as syngnathids (pipefish and seahorses). This two-year pilot study investigated seasonal patterns indensity, growth, site fidelity, and population dynamics of Tampa Bay (FL) syngnathid fishes at a site adjacent to two marinas underconstruction. Using a modified mark-recapture technique, fish were collected periodically from three closely located sites thatvaried in seagrass species (Thalassia spp., Syringodium spp., and mixed-grass sites) and their distance from open water, but hadconsistent physical/chemical environmental characteristics. Fish were marked, photographed for body size and gender measurements,and released the same day at the capture site. Of the 5695 individuals surveyed, 49 individuals were recaptured, indicatinga large, flexible population. Population density peaks were observed in July of both years, with low densities in late winter andlate summer. Spatially, syngnathid densities were highest closest to the mouth of the bay and lowest near the shoreline. Sevenspecies of syngnathid fishes were observed, and species-specific patterns of seagrass use emerged during the study. However,only two species, Syngnathus scovelli and Hippocampus zosterae, were observed at high frequencies. For these two species, bodysize decreased across the study period, but while S. scovelli's population density decreased, H. zosterae's increased. Aim: To develop an algorithm for use in population-based studies to assess testicular function by measurements of totalurinary follicle stimulating hormone (FSH). Methods: Total concentrations of urinary FSH were measured in a groupof 44 men at the University of California, Davis (UCD) and were compared to FSH measurements in serum. On thebasis of these and other published data, a urinary FSH value of >2 ng/mg creatinine (Cr) was selected as the cutoffpoint to identify men with elevated serum FSH ( > 12 IU/L) or low sperm counts ( < 20 million/mL). Results: Thesensitivity and specificity of this algorithm for detecting elevated serum FSH in a group of 58 agricultural workers in thePeople's Republic of China were 100 % and 50 %, respectively. Immunophilin-like FKBP42/TWISTED DWARF1 Interacts with the Receptor Kinase BRI1 to Regulate Brassinosteroid Signaling in Arabidopsis

Mutation of the immunophilin-like FK506-binding protein TWlSTED DWARF1 （FKBP42/TWD1） causes dwarf and twisted-organ phenotypes in Arabidopsis. However, the function of FKBP42 is not fully understood at the molecular level. Using genetic, physiological, and immunological experiments, we show here that FKBP42/TWD1 is necessary for brassinosteroid （BR） signal transduction. The twdl mutant showed reduced BR sensitivity in growth responses and activation of the BZR1 transcription factor. However, twdl showed normal responses to an inhibitor of BIN2/GSK3, suggesting that twdl has a defect upstream of BIN2 in the BR signaling pathway. In vitro and in vivo assays showed that TWD1 interacts physically with the kinase domains of the BR receptor kinases BRI1 and BAK1. TWD1 is not required for normal localization of BRI1-GFP to the plasma membrane or for activation of the flagellin receptor kinase FLS2. Our results suggest that FKBP42/TWD1 plays a specific role in the activation of BRI1 receptor kinase.

Assessing support alternatives for longwall gateroads subject to changing stress

Longwall gateroad entries are subject to changing horizontal and vertical stress induced by redistribution of loads around the extracted *** stress changes can result in significant deformation of the entries that may include roof sag,rib dilation,and floor *** operators install different types of supports to control the ground response and maintain safe access and ventilation of the longwall *** paper describes recent research aimed at quantifying the effect of longwall-induced stress changes on ground stability and using the information to assess support *** research included monitoring of ground and support interaction at several operating longwall mines in the U.S.,analysis and calibration of numerical models that adequately represent the bedded rock mass,and observation of the support systems and their response to changes in *** models were then used to investigate the impact of geology and stress conditions on ground deformation and support response for various depths of cover and geologic *** research results were summarized in two regression equations that can be used to estimate the likely roof deformation and height of roof yield due to longwall-induced stress *** information is then used to assess the ability of support systems to maintain the stability of the *** application of the method is demonstrated with a retrospective analysis of the support performance at an operating longwall mine that experienced a headgate roof *** method is shown to produce realistic estimates of gateroad entry stability and support performance,allowing alternative support systems to be assessed during the design and planning stage of longwall operations.

A method for quantifying bias in modeled concentrations and source impacts for secondary particulate matter

Community Multi-Scale Air Quality （CMAQ） estimates of sulfates, nitrates, ammonium, and organic carbon are highly influenced by uncertainties in modeled secondary formation processes, such as chemical mechanisms, volatilization, and condensation rates. These compounds constitute the majority ofPM2.5 mass, and reducing bias in estimated concentrations has benefits for policy measures and epidemiological studies. In this work, a method for adjusting source impacts on secondary species is developed that provides estimates of source contributions and reduces bias in modeled concentrations compared to observations. The bias correction adjusts concentrations and source impacts based on the difference between modeled concentrations and observations while taking into account uncertainties at the location of interest; and it is applied both spatially and temporally. We apply the method over the US for 2006. The mean bias for initial CMAQ concentrations compared to observations is -0.28 （OC）, 0.11 （NO3）, 0.05 （NH4）, and -0.08 （SO4）. The normalized mean bias in modeled concentrations compared to observations was effectively zero for OC, NO3, NH4, and SO4 after applying the secondary bias correction. Ten-fold cross-validation was conducted to determine the performance of the spatial application of the bias correction. Cross-validation performance was favorable; correlation coefficients were greater than 0.69 for all species when comparing observations and concentrations based on kriged correction factors. The methods presented here address model uncertainties by improving simulated concentrations and source impacts of secondary particulate matter through data assimilation. Secondary-adjusted concentrations and source impacts from 20 emissions sources are generated for 2006 over continental US.

Depletion of BBS Protein LZTFL1 Affects Growth and Causes Retinal Degeneration in Mice

Bardet-Biedl syndrome（BBS） is a heterogeneous disease characterized by deficiencies in various organs that are caused by defects in genes involved in the genesis, structural maintenance, and protein trafficking of cilia. Leucine zipper transcription factor-like 1（LZTFL1）has been identified as a BBS protein（BBS17）, because patients with mutations in this gene exhibit the common BBS phenotypes. In this study, we generated a knockout mouse model to investigate the effects of LZTFL1 depletion. Lztfl1 knockout mice were born with low birth weight, reached similar weight to those of wild-type mice at 10 weeks of age, and later gained more weight than their wild-type counterparts. LZTFL1 was localized to the primary cilium of kidney cells, and the absence of LZTFL1 increased the ciliary localization of BBS9. Moreover, in the retinas of Lztfl1 knockout mice, the photoreceptor outer segment was shortened, the distal axoneme of photoreceptor connecting cilium was significantly enlarged, and rhodopsin was targeted to the outer nuclear layer. TUNEL assay showed that many of these abnormal photoreceptor cells in Lztfl1 knockout mice underwent apoptosis. Interestingly, the absence of LZTFL1 caused an abnormal increase of the adaptor protein complex 1（AP1） in some photoreceptor cells. Based on these data, we conclude that LZTFL1 is a cilium protein and regulates animal weight and photoreceptor connecting cilium function probably by controlling microtubule assembly and protein trafficking in cilia.

The role of apoptosis in the development and function of T lymphocytes

Apoptosis plays an essential role in T cell biology. Thymocytes expressing nonfunctional or autoreactive TCRs are eliminated by apoptosis during development. Apoptosis also leads to the deletion of expanded effector T cells during immune responses. The dysregulation of apoptosis in the immune system results in autoimmunity, tumorogenesis and immunodeficiency. Two major pathways lead to apoptosis: the intrinsic cell death pathway controlled by Bcl-2 family members and the extrinsic cell death pathway controlled by death receptor signaling. These two pathways work to- gether to regulate T lymphocyte development and function.
