Rock inhibitor may compromise human induced pluripotent stem cells for cardiac differentiation in 3D

Cardiomyocytes differentiated from human induced pluripotent stem cells(iPSCs)are valuable for the understanding/treatment of the deadly heart diseases and their drug ***,the very much needed homogeneous 3D cardiac differentiation of human iPSCs is still ***,it is discovered surprisingly that Rock inhibitor(RI),used ubiquitously to improve the survival/yield of human iPSCs,induces early gastrulation-like change to human iPSCs in 3D culture and may cause their heterogeneous differentiation into all the three germ layers(i.e.,ectoderm,mesoderm,and endoderm)at the commonly used concentration(10μm).This greatly compromises the capacity of human iPSCs for homogeneous 3D cardiac *** reducing the RI to 1μm for 3D culture,the human iPSCs retain high pluripotency/quality in inner cell mass-like solid 3D ***,the beating efficiency of 3D cardiac differentiation can be improved to more than 95%in~7 days(compared to less than~50%in 14 days for the 10μm RI condition).Furthermore,the outset beating time(OBT)of all resultant cardiac spheroids(CSs)is synchronized within only 1 day and they form a synchronously beating 3D construct after 5-day culture in gelatin methacrylol(GelmA)hydrogel,showing high homogeneity(in terms of the OBT)in functional maturity of the ***,the resultant cardiomyocytes are of high quality with key functional ultrastructures and highly responsive to cardiac *** discoveries may greatly facilitate the utilization of human iPSCs for understanding and treating heart diseases.

Small RNAs in pollen

In plants, each pollen mother cell undergoes two rounds of cell divisions to form a mature pollen grain, which contains a vegetative cell(VC) and two sperm cells(SC). As a companion cell, the VC carries the SCs to an ovule by germinating a pollen tube. In-depth sequencing analyses of mature pollen showed that micro RNAs(mi RNAs) and short interfering RNAs(si RNAs) are present in both the VC and SCs. Additionally, epigenetically-regulated transposable elements(TEs) are reactivated in the VC and these TE m RNAs are further processed into 21-nt epigenetically reactivated si RNA(easiR NA) in SCs, which prevent 24-nt si RNA accumulation and sequester mi RNA loading. Small RNAs are thought to move from the VC to SCs, where they regulate gene expression and reinforce TE silencing. here, we summarize current knowledge of the biogenesis and function of mi RNAs, si RNAs, and easi RNAs in pollen, emphasizing how these different small RNAs coordinately contribute to sperm cell formation and TE silencing.

magnesium hydride(mgH_(2))is the most feasible and effective solid-state hydrogen storage material,which has excellent reversibility but initiates decomposing at high temperatures and has slow kinetics ***,zinc titanate(Zn_(2)TiO_(4))synthesised by the solid-state method was used as an additive to lower the initial temperature for dehydrogenation and enhance the re/dehydrogenation behaviour of mgH_(2).With the presence of Zn_(2)TiO_(4),the starting temperature for the dehydrogenation of mgH_(2)was remarkably lowered to around 290℃–305℃.In addition,within 300 s,the mgH_(2)–Zn_(2)TiO_(4)sample absorbed 5.0 wt.%of H_(2)and 2.2–3.6 wt.%H_(2)was liberated from the composite sample in 30 min,which is faster by 22–36 times than as-milled mgH_(2).The activation energy of the mgH_(2)for the dehydrogenation process was also downshifted to 105.5 k J/mol with the addition of Zn_(2)TiO_(4)indicating a decrease of 22%than as-milled mgH_(2).The superior behaviour of mgH_(2)was due to the formation of mg Zn_(2),mgO and mgTiO_(3),which are responsible for ameliorating the re/dehydrogenation behaviour of mgH_(2).These findings provide a new understanding of the hydrogen storage behaviour of the catalysed-mgH_(2)system.

Real-variable characterizations of anisotropic product musielak-Orlicz Hardy spaces

Let A :=(A_1, A_2) be a pair of expansive dilations and φ : R^n×R^m×[0, ∞) → [0, ∞) an anisotropic product musielak-Orlicz function. In this article, we introduce the anisotropic product musielak-Orlicz Hardy space H~φ_A(R^n× R^m) via the anisotropic Lusin-area function and establish its atomic characterization, the g-function characterization, the g_λ~*-function characterization and the discrete wavelet characterization via first giving out an anisotropic product Peetre inequality of musielak-Orlicz type. moreover, we prove that finite atomic decomposition norm on a dense subspace of H~φ_A(R^n× R^m) is equivalent to the standard infinite atomic decomposition norm. As an application, we show that, for a given admissible triplet(φ, q, s), if T is a sublinear operator and maps all(φ, q, s)-atoms into uniformly bounded elements of some quasi-Banach spaces B, then T uniquely extends to a bounded sublinear operator from H~φ_A(R^n× R^m) to B. Another application is that we obtain the boundedness of anisotropic product singular integral operators from H~φ_A(R^n× R^m) to L~φ(R^n× R^m)and from H~φ_A(R^n×R^m) to itself, whose kernels are adapted to the action of A. The results of this article essentially extend the existing results for weighted product Hardy spaces on R^n× R^m and are new even for classical product Orlicz-Hardy spaces.

Surface flashover of spacers is a key factor limiting the application of HVDC GIS/GIL,while the charge accumula-tion on the surface of the spacer could have a potential adverse effect on the surface flashover *** paper discusses the laws regarding distribution patterns of surface charges and the related *** field-dependent property is discussed in detail to comprehensively illustrate the charge transport mecha-nism and explain the research differences regarding different surface charge patterns obtained by previous *** addition,the main surface charge control methods for epoxy resin are summarized and *** potential research directions of charge control methods and key points in manufacturing of spacers used in HVDC GIS/GIL are also explored.

The morphology and size of Pt-based bimetallic alloys are known to determine their electrocatalytic performance in reactions relevant to fuel ***,we report a general approach for preparing Pt-m(m=Fe,Co and Ni)bimetallic nano-branched structure(NBs)by a simple high temperature solution-phase ***-prepared Pt-m NBs show a polycrystalline structure and are rich in steps and kinks on the surface,which promote them favorable bifunctional catalytic properties in acidic electrolytes,specifically in terms of the oxygen reduction reaction(ORR)and methanol oxidation reaction(mOR).Specially,Pt-Co NBs/C catalyst shows 6.1 and 5.3 times higher in specific activity(SA)and mass activity(mA)for ORR than state-of-the-art commercial Pt/C catalysts,***,it exhibits a loss of 4.0%in SA and 14.4%in mA after 10,000 cycles of accelerated durability tests(ADTs)compared with the initial *** addition,we also confirmed the superior mOR activity of Pt-Co NBs/C catalyst in acidic *** Pt-m NBs with other alloying metals,the ORR and mOR activities are both higher than commercial catalysts and are in the sequence of Pt-Co/C>Pt-Fe/C>Pt-Ni/C>commercial Pt/C(or PtRu/C).The improved activities and durability can benefit from the morphological and compositional *** synthesis approach may be applied to develop bifunctional catalysts with enhanced ORR and mOR properties for future fuel cells designs.

Comparison of changes in land use and land cover in China and the USA over the past 300 years

Comparative studies of changes in land use and land cover between different countries over a long time-scale are helpful in understanding the processes and driving forces of these changes in different situations. We compared and analyzed the temporal and spatial characteristics of cropland, forest and grassland between China＇s mainland and the conterminous USA over the past 300 years. We found that, over the past 300 years, the area of cropland showed an overall increasing trend both in China and in the USA. In China, the area of cropland increased by about 79.46×104 km2 from 1661 to the 1980s, and the per capita cropland decreased from 0.37×104 m2 in 1685 to 0.10×104 m2 in 2000. For the USA, the area of cropland increased by 190.87×104 km2 between 1700 and 1950. The per capita cropland area of each time slice for the past 300 years was greater than 0.60×104 m2 and the maximum value was 2.01×104 m2 in 1890. As a result of the expansion in land reclamation, the forest and grassland areas of the USA have decreased by about 136.98×104 km2 and 136.98×104 km2, respectively, over the past 300 years; the corresponding values for China are about 89.73×104 km2 and 40.00×104 km2, respectively. In terms of the spatial patterns, the expansion in cropland in China mainly occurred in the border areas （including the northeast, southwest and Inner mongolia） and in the hilly areas. Cropland expansion in the USA mainly occurred in the mid-west regions. In China, population growth is the most important driving factor in changes in land use and land cover, whereas in the USA, government policy plays the most significant role.

Software visualization and visual editing are important and practical techniques to improve the development of complex software systems.A challenge when applying the two technologies is how to realize the correspondence,a bidirectional relationship,between the data and its visual representation *** many tools and frameworks have been developed to support the construction of visual tools,it is still complicated and error-prone to realize the bidirectional *** this paper,we propose a model-driven and bidirectional-transformation-based framework for data visualization and visual *** approach mainly focuses on 1)how to define and manage graphical symbols in the model form and 2)how to specify and implement the bidirectional relationship based on the technique of bidirectional model ***,a prototype tool and four case studies are presented to evaluate the feasibility of our work.

Neurons in Primary motor Cortex Encode Hand Orientation in a Reach-to-Grasp Task

It is disputed whether those neurons in the primary motor cortex（m1） that encode hand orientation constitute an independent channel for orientation control in reach-to-grasp behaviors. here, we trained two monkeys to reach forward and grasp objects positioned in the frontal plane at different orientation angles, and simultaneously recorded the activity of m1 neurons. Among the 2235 neurons recorded in m1, we found that 18.7% had a high correlation exclusively with hand orientation, 15.9% with movement direction, and 29.5% with both movement direction and hand orientation. The distributions of neurons encoding hand orientation and those encoding movement direction were not uniform but coexisted in the same region. The trajectory of hand rotation was reproduced by the firing patterns of the orientation-related neurons independent of the hand reaching direction. These resultssuggest that hand orientation is an independent component for the control of reaching and grasping activity.

Iterative Noncoherent Block Detection of Coded mPSK for Cooperative Relay Systems

maximum likelihood(mL) noncoherent block detection techniques are investigated for block-coded mPSK modulation in cooperative decode-and-forward relay systems over slow fading channels.A decision-directed iterative Viterbi algorithm(IVA) is derived for a suboptimal mL noncoherent *** results show that the IVA can approach the error performances of the exhaustive detection method but at a lower complexity.
