Melatonin Alleviates Acute Gouty Inflammation In Vivo and In Vitro

The aim of this study was to identify the effects of melatonin on acute gouty inflammation and to investigate the underlying mechanisms.We found significantly lower serum melatonin levels in gout patients in the acute phase than in those in the remission phase or in normal individuals.The mRNA expression of melatonin receptor 2(MT2)was also lower in gout patients than in normal individuals.To verify the in-vivo role of melatonin,a gouty arthritis model was established by intraarticular injection of monosodium urate(MSU,1 mg)crystals into the paws of C57BL/6 mice.Joint inflammation in the mouse model was evaluated by measuring the thickness of the right paw/left paw,and the inflammation index was determined by examining infiltrating neutrophils with haematoxylin and eosin(H&E)staining.Melatonin was found to reduce both paw thickness and the inflammation index in the mouse model,and melatonin also reduced the mRNA levels of interleukin-1 beta(IL-1β),IL-6 and NLR family pyrin domain containing 3(NLRP3)inflammasome.To mimic gouty inflammation in vitro,mouse peritoneal macrophages were stimulated with lipopolysaccharides(LPS)plus MSU.Melatonin was revealed to reduce IL-1βsecretion by stimulated macrophages.The mRNA expression levels of IL-1βand IL-6 were also inhibited by melatonin.Western blot analysis showed that the expression of NLRP3,caspase-1 and pro-IL-1βwas also inhibited by melatonin.In conclusion,our study demonstrated that melatonin alleviated gouty inflammation in vivo and in vitro,and the underlying mechanism may involve inhibiting the assembly of the NLRP3 inflammasome.

Cardiovascular events in hyperuricemia population and a cardiovascular benefit-risk assessment of urate-lowering therapies:a systematic review and meta-analysis

Background:Hyperuricemia and gout have become public health concerns;many important guidelines have recommended xanthine oxidase inhibitors(XOIs)as the first-line urate-lowering therapies(ULTs)to treat chronic gout with hyperuricemia.However,whether treating hyperuricemia and gout with ULTs modifies cardiovascular risks remains controversial.The aim of this study was to assess the incident risk of cardiovascular(CV)events(CVE)in hyperuricemia population,assess the cardiovascular benefit-risk of ULTs in hyperuricemia patients with or without gout in diverse cardiovascular risk sub-groups,and specify the safety of different ULTs.Methods:We searched PubMed,Embase,the Cochrane Library,Wanfang,Chongqing VIP(CQVIP,***),and China National Knowledge Infrastructure Database for prospective cohort studies and randomized controlled trials(RCTs)in English and Chinese.Potential medications included XOIs,and uricosurics.RCTs were divided into sub-groups analysis based on blinding status and patients’history of CV diseases.Risk ratios(RRs)were calculated and were reported with corresponding 95%confidence intervals(CIs)by fixed-effects or random-effects model.Results:Seven prospective cohort studies and 17 RCT studies were included.The risks of both major adverse cardiovascular events(MACE)(RR=1.72,95%CI 1.28-2.33)and CVE(RR=1.35,95%CI 1.12-1.62)were higher in the hyperuricemia population than non-hyperuricemia one.In seven RCT studies where XOIs were compared with no-treatment or placebo,the results of five low CV risk studies showed that XOIs lowered the risks of both MACE(RR=0.35,95%CI 0.20-0.62)and CVE(RR=0.61,95%CI 0.44-0.85);whereas two high CV risk studies showed that XOIs lowered the risk of CVE(RR=0.69,95%CI 0.54-0.88)rather than MACE(RR=0.62,95%CI 0.29-1.35).In nine RCT studies where the cardiovascular safety between febuxostat and allopurinol were compared,no statistical difference was found in the risk of MACE or CVE.Conclusions:The hyperuricemia population does have a higher incidence of CVE,and the results suggeste

Remaining useful life(RUL)prediction for bearing is a significant part of the maintenance of urban rail transit trains.Bearing RUL is closely linked to the reliability and safety of train running,but the current prediction accuracy makes it difficult to meet the re-quirements of high reliability operation.Aiming at the problem,a prediction model based on an improved long short-term memory(ILSTM)network is proposed.Firstly,the variational mode decomposition is used to process the signal,the intrinsic mode function with stronger representation ability is determined according to energy entropy and the degradation feature data is constructed com-bined with the time domain characteristics.Then,to improve learning ability,a rectified linear unit(ReLU)is applied to activate a fully connected layer lying after the long short-term memory(LSTM)network,and the hidden state outputs of the layer are weighted by attention mechanism.The Harris Hawks optimization algorithm is introduced to adaptively set the hyperparameters to improve the performance of the LSTM.Finally,the ILSTM is applied to predict bearing RUL.Through experimental cases,the better perfor-mance in bearing RUL prediction and the effectiveness of each improving measures of the model are validated,and its superiority of hyperparameters setting is demonstrated.

Temperature-responsive polymers have garnered significant attention due to their ability to respond to external stimuli.In this work,dual temperature-responsive block copolymers are synthesized via reversible addition-fragmentation chain transfer polymerization(RAFT)polymerization utilizing zwitterionic monomer methacryloyl ethyl sulfobetaine(SBMA) and N-isopropyl acrylamide(NIPAAm) as monomers.The thermal responsive behaviors can be easily modulated by incorporating additional hydrophobic monomer benzyl acrylate(BN) or hydrophilic monomer acrylic acid(AA),adjusting concentration or pH,or varying the degree of polymerization of the block chain segments.The cloud points of the copolymers are determined by UV-Vis spectrophotometry,and these copolymers exhibit both controlled upper and lower critical solu bility temperatures(LCST and UCST) in aqueous solution.This study analyzes and summarizes the influencing factors of dual temperature responsive block copolymers by exploring the effects of various conditions on the phase transition temperature of temperature-sensitive polymers to explore the relationship between their properties and environment and structure to make them more selective in terms of temperature application range and regulation laws.It is very interesting that the introduction of poly-acrylic acid(PAA) segments in the middle of di-block copolymer PSBMA_(55)-b-PNIPAAm_(80) to form PSBMA_(55)-b-PAA_(x)-b-PNIPAAm_(80) results in a reversal of temperature-responsive behaviors from 'U'(LCST UCST) type,while the copolymer PSBMA_(55)-b-P(NIPAAm_(80)-co-AA_(x)) not.This work provides a clue for tuning the phase transition behavior of polymers for manufacture of extreme smart materials.

Lanthanide complex-based materials with magnetic and luminescent properties have received a great deal of attention,owing to their importantly potential applications in diverse fields.In this work,the 9-anthracenecarboxylic acid(9-Haca)as bridging ligand was applied in combination with optically activeπ-conjugated 2,4,6-tri(2-pyridyl)-1,3,5-triazine(tptz)to construct a series of new di-nuclear and tetranuclear lanthanide 9-anthracenecarboxylates[La_(2)(9-aca)_(6)(tptz)_(2)(μ-H_(2)O)]·0.25H_(2)O(1),[Ln_(2)(9-aca)_(6)(tptz)_(2)(H_(2)O)_(2)]·0.5AQ·CH_(3)COOH_(2)H_(2)O[Ln=Sm(2a),Eu(2b),Gd(2c),Tb(2d),Dy(2e),Tm(2f);AQ=9,10-anthracenedione]and[Ln_(4)(9-aca)_(10)(tptz)_(2)(CH3COO)2]·H_(2)O[Ln=Eu(3a),Gd(3b),Dy(3c)].1 displays a di-nuclear centrosymmetric moiety[La_(2)(9-aca)_(6)(tptz)_(2)(μ-H_(2)O)]built up from two complex[La(9-aca)_(2)(tptz)]groups bridged by one H_(2)O molecule and two syn-syn-μ2-η1:η1-9-aca-bridging ligands,while 2a-2f are isostructural and co mprise another di-nuclear centrosymmetric moieties[Ln_(2)(9-aca)_(6)(tptz)_(2)(H_(2)O)_(2)]based on two complex[Ln(9-aca)_(2)(tptz)(H_(2)O)]groups connected by two syn-syn-μ2-η1:η1-9-aca-bridging ligands,3a-3c are isostructural and show tetra-nuclear centro symmetric moieties[Ln4(9-aca)10(tptz)2(CH3COO)2]containing three types of coordination modes of 9-aca-ligands.The present compounds offer the only examples of lanthanide 9-anthracenecarboxylates decorated by optically active tptz.Magnetic investigations of 2a,2c-2f,and 3b-3c indicate that 3c is typical of slow relaxation of the magnetization,while 2c and 3b show significant magnetocaloric effect(MCE)with the value of-ΔSm of 5.26 J/(kg·K)at 4 K and 15.65 J/(kg·K)at 2 K(ΔH=7 T),respectively.The luminescent properties were also studied and reveal that the characteristic luminescent properties of the 7r-conjugated aromatic ligands are introduced into magnetic lanthanide 9-anthracenecarboxylates,simultaneously exhibiting slow magnetic relaxation(or MCE)and luminescent properties.

Erratum to: Melatonin Alleviates Acute Gouty Inflammation In Vivo and In Vitro

The authors examined the original data of their work and noticed some mistakes.The ELISA kit was bought from IBL Co.Germany instead of Abeam Co,UK.The anti-NLRP3,anti-IL-lp and anti-caspase-1 primary antibodies were from CST Co,USA.There were 31 acute gout patients and 25 gout patients in remission who met the American Rheumatism Society(ACR)diagnostic criteria of gout in this study,and 7 normal individuals were recruited as controls.The authors sincerely apologize for the mistakes and confirm that the changes do not affect the scientific conclusion of the published work.

Investigation of wake-induced vibration interference between two staggered wave-cone cylinders at subcritical Reynolds number

The wake induced vibration(WIV)of a one-and two-degree-of-freedom(1DOF,2DOF)downstream wave-cone cylinder(WCC)behind a stationary equal-size upstream wave-cone cylinder in the staggered arrangement is numerically investigated at subcritical Reynolds number of 3900 by using shear stressed transfer(SST)k-ωturbulence model.The streamwise pitch ratios(P/Dm)vary from 4 to 6 with a fixed incident angleα=8°.Experimental measurements were also performed for the validation of the present numerical models.It is found that the largest vibration amplitude in crossflow direction occurred at P/Dm=4,Ur=8 with small difference of streamwise vibration at P/Dm=4,6.Different from single wavy-cone cylinder(SWCC),the downstream flexible one of a pair staggered WCCs got larger vibration amplitude during phase switching stage instead of in-phase stage.The upstream wake will suppress the triple frequency of main frequency in the power spectra density(PSD)functions of Cl but stimulate the double one of that.An intriguing vibration mechanism happened in all 2DOF cases where the trajectory of downstream WCC is a significant ellipse rather than a figure of 8.The transformation of phase switching and the variation of the main frequency of drag coefficient(Cd)can be explained by the vortex-shedding modes of downstream WCC.

helicobacter pylori(H.pylori)infection is a threat to human health.The lipopolysaccharide(LPS)O-antigen holds promise for developing vaccines.It is meaningful to explore the immunological activity of oligosaccharides with different lengths and frameshifts for antigen development.herein,a glycan library of H.pylori O2 O-antigen containing eight fragments is constructed.After screening with anti-H.pylori O2 LPS sera and patients’sera by glycan microarray,the disaccharide HPO2G-2b and trisaccharide HPO2G-3a show strong antigenicity and then are separately conjugated with carrier protein CRM197.Both glycoconjugates elicit a robust immunoglobulin G(IgG)immune response in rabbits.The anti-HPO2G-3a IgG antibodies possess a much stronger binding affinity with the LPS and bacteria of H.pylori O2 than the anti-HPO2G-2b IgG antibodies.There is no cross-reaction between both sera IgG antibodies with LPS and bacteria of H.pylori O1,O6,and E.coli.The results demonstrate the trisaccharide HPO2G-3a is a promising candidate for H.pylori vaccine development.

An efficient synthesis of the electrophilic reagent,S-(1,3-dioxoisoindolin-2-yl)O,O-diethyl phosphorothioate(SDDP)is described.Moreover,the synthetic applications of SDDP wherein the transfer of the SP(O)(OEt)_(2) moiety occurs were investigated.In this manner,SDDP underwent facile SP(O)(OEt)_(2) transfer with electron-rich substrates such as ketones,indoles,and thiols to formα-phosphorothiolated ketones,3-phosphorothiolated indoles and S-phosphorothiolated thioethers,respectively.

Difference in microstructural characteristics of stamp-charged coke and gravity-charged coke with approximate macroscopic performance

The microstructural characteristics including optical texture,porosity and pore structure and chemical structure of stamp-charged coke(SCC)and gravity-charged coke(GCC)with similar conventional macro-indicators were investigated,and the properties including micro-strength,reactivity of coke matrix and that after alkali enrichment were comparatively studied by various characterization methods.The anisotropic structure of SCC is composed of high content of fine mosaic texture,while the content of medium mosaic texture,coarse texture and fibrous texture is low.The statistical average shows that the fine mosaic average of SCC(24.89%)is 3.78 times the GCC average(6.58%),and the coarse mosaic average(1.24%)is only about 1/3 of the GCC average(3.43%).The porosity of SCC is lower than that of GCC,but tamping process does not lead to the fact that the number of closed pores of SCC is significantly lower than that of GCC.Although the structure of SCC is compact,its pore number is large and the pore wall is thin.Pores of coke with diameter less than 150 nm seem unaffected by tamping process.The aromatic structure of SCC was less ordered than that of GCC,which was speculated to be related to the addition of more low metamorphic coal in coking.The microscopic strength and structural strength of SCC are lower than those of GCC.The reactivity of coke matrix is affected by the specific surface area,but it is not the determining factor of its macro-reactivity.The improvement in dissolution reactivity of coke after potassium enrichment is independent of coke type.
