As an Executive Editor-in-Chief,I have edited this Special Issue(SI),entitled“Preimplantation genetic testing(PGT)”published in the journal of Reproductive and Developmental Medicine(RDM;ISSN:2096-2924,CN:10-1442/R),the firstever English journal in the field of reproductive medicine and developmental biology in China's Mainland.Since its launch in 2017,the RDM has been dedicated to providing a good platform for academic exchanges among scientists,both domestically and abroad.

Background： Maternal thyroid dysfunction is common during pregnancy, and physiological changes during pregnancy can lead to the overdiagnosis of hyperthyroidism and misdiagnosis of hypothyroidism with nongestation-specific reference intervals. Our aim was to compare sequential with nonsequential methods for the evaluation of thyroid function in pregnant women. Methods： We tested pregnant women who underwent their trimester prenatal screening at our hospital from February 2011 to September 2012 for serum thyroid stimulating hormone （TSH） and free thyroxine （FT4） using the Abbott and Roche kits. There were 447 and 200 patients enrolled in the nonsequential and sequential groups, respectively. The central 95% range between the 2.5th and the 97.5th percentiles was used as the reference interval for the thyroid function parameter. Results： The nonsequential group exhibited a significantly larger degree of dispersion in the TSH reference interval during the 2nd and 3rd trimesters as measured using both the Abbott and Roche kits （all P 〈 0.05）. The TSH reference intervals were significantly larger in the nonsequential group than in the sequential group during the 3rd trimester as measured with both the Abbott （4.95 vs. 3.77 mU/L, P 〈 0.001） and Roche kits （6.62 vs. 5.01 mU/L, P〈 0.004）. The nonsequential group had a significantly larger FT4 reference interval as measured with the Abbott kit during all trimesters （ 12.64 vs. 5.82 pmol/L; 7,96 vs. 4.77 pmol/L; 8.10 vs. 4.77 pmol/L, respectively, all P 〈 0.05）, whereas a significantly larger FT4 reference interval was only observed during the 2nd trimester with the Roche kit （7.76 vs. 5.52 pmol/L, P = 0.002）. Conclusions： It was more reasonable to establish reference intervals for the evaluation of maternal thyroid function using the sequential method during each trimester of pregnancy. Moreover, the exclusion of pregnancy-related complications should be considered in the inclusion criteria for thyroid function tests.

There has been a very rapid development in the area of preimplantation genetic testing(PGT)since the first baby applied PGT technology in the world was born in the last 30 years.As an alternative treatment of prenatal diagnosis,the transfer of the fetus with normal testing result has benefited many families.PGT is mainly classified into PGT for aneuploidies(PGT-A),PGT for chromosomal structural rearrangement(PGT-SR),and PGT for monogenic/single-gene defects(PGT-M).here,we reviewed the application,development,limitations,and challenges of the current PGT in reproductive medicine.

Objective:Increasing evidences have shown that prepregnancy maternal weight and gestational weight gain(GWG)may associate with offspring’s neurodevelopment.However,the effects of prepregnancy maternal overweight,obesity,and excessive GWG on offspring’s intelligence remain controversial.This meta-analysis aimed to re-assess the association between prepregnancy body mass index(BMI),GWG,and children’s intelligence.Methods:We systematically searched multiple databases,including PubMed,EMBASE,Cochrane Library,and Ovid Medline,from their inception through February 2021.Studies assessing the association between prepregnancy BMI or GWG and children’s intelligence were further screened manually before final inclusion.Cohorts that analyzed the association between prepregnancy BMI or GWG and intelligence of offspring were included,and we used the Mantel-Haenszel fixed-effects method to compute the weighted mean difference(WMD)and 95%confidence interval(CI)of each study.Results:A total of 12 articles were included in this systematic review,while six of them in the meta-analysis.There was a significant full-scale IQ reduction in children born from overweight and obese mothers,with WMDs of-3.08(95%CI:-4.02,-2.14)and-4.91(95%CI:-6.40,-3.42),respectively.Compared with control group,the WMDs for performance and verbal intelligence quotient(IQ)were decreased in overweight and obesity groups.However,we observed no association between children’s full-scale IQ and excessive GWG with WMD of-0.14(95%CI:-0.92,0.65).Conclusions:Women’s prepregnancy overweight and obesity adversely associate with children’s intelligence but no association with excessive GWG.Our study suggests that further researches focusing on the effect of prepregnancy maternal health on offspring’s intelligence development are needed.

Cadmium(Cd)bioavailability in the rhizosphere makes an important difference in grain Cd accumulation in wheat.here,pot experiments combined with 16S rRNA gene sequencing were conducted to compare the Cd bioavailability and bacterial community in the rhizosphere of two wheat(Triticum aestivum L.)genotypes,a low-Cd-accumulating genotype in grains(LT)and a high-Cd-accumulating genotype in grains(HT),grown on four different soils with Cd contamination.Results showed that there was non-significant difference in total Cd concentration among four soils.However,except for black soil,DTPA-Cd concentrations in HT rhizospheres were higher than those of LT in fluvisol,paddy soil and purple soil.Results of 16S rRNA gene sequencing showed that soil type(52.7%)was the strongest determinant of root-associated community,while there were still some differences in rhizosphere bacterial community composition between twowheat genotypes.Taxa specifically colonized in HT rhizosphere(Acidobacteria,Gemmatimonadetes,Bacteroidetes and Deltaproteobacteria)could participate inmetal activation,whereas LT rhizosphere was highly enriched by plant growth-promoting taxa.In addition,PICRUSt2 analysis also predicted high relative abundances of imputed functional profiles related to membrane transport and amino acid metabolism in HT rhizosphere.These results revealed that the rhizosphere bacterial community may be an important factor regulating Cd uptake and accumulation in wheat and indicated that the high Cd-accumulating cultivar might improve Cd bioavailability in the rhizosphere by recruiting taxa related to Cd activation,thus promoting Cd uptake and accumulation.

In the present study, we introduced point mutations into Ac_rap A which encodes a polyketide synthase responsible for rapamycin biosynthesis in Actinoplanes sp. N902-109, in order to construct a mutant with an inactivated enoylreductase(ER) domain, which was able to synthesize a new rapamycin analog. Based on the homologous recombination induced by double-strand breaks in chromosome mediated by endonuclease I-SceI, the site-directed mutation in the first ER domain of Ac_rapA was introduced using non-replicating plasmid pL YERIA combined with an I-SceI expression plasmid. Three amino acid residues of the active center, Ala-Gly-Gly, were converted to Ala-Ser-Pro. The broth of the mutant strain SIPI-027 was analyzed by HPLC and a new peak with the similar UV spectrum to that of rapamycin was found. The sample of the new peak was prepared by solvent extraction, column chromatography, and crystallization methods. The structure of new compound, named as SIPI-rapxin, was elucidated by determining and analyzing its MS and NMR spectra and its biological activity was assessed using mixed lymphocyte reaction(MLR). An ER domain–deficient mutant of Actinoplanes sp. N902-109, named as SIPI-027, was constructed, which produced a novel rapamycin analog SIPI-rapxin and its structure was elucidated to be 35, 36-didehydro-27-O-demethylrapamycin. The biological activity of SIPI-rapxin was better than that of rapamycin. In conclusion, inactivation of the first ER domain of rap A, one of the modular polyketide synthase responsible for macro-lactone synthesis of rapamycin, gave rise to a mutant capable of producing a novel rapamycin analog, 35, 36-didehydro-27-O-demethylrapamycin, demonstrating that the enoylreductase domain was responsible for the reduction of the double bond between C-35 and C-36 during rapamycin synthesis.

Hybrid model for muon tomography and quantitative analysis of image quality

Muon tomography is a novel method for the non-destructive imaging of materials based on muon rays,which are highly penetrating in natural background radiation.Currently,the most commonly used imaging methods include muon radiography and muon tomography.A previously studied method known as coinciding muon trajectory density tomography,which utilizes muonic secondary particles,is proposed to image low and medium atomic number(Z)materials.However,scattering tomography is mostly used to image high-Z materials,and coinciding muon trajectory density tomography exhibits a hollow phenomenon in the imaging results owing to the self-absorption effect.To address the shortcomings of the individual imaging methods,hybrid model tomography combining scattering tomography and coinciding muon trajectory density tomography is proposed and verified.In addition,the peak signal-to-noise ratio was introduced to quantitatively analyze the image quality.Different imaging models were simulated using the Geant4 toolkit to confirm the advantages of this innovative method.The simulation results showed that hybrid model tomography can image centimeter-scale materials with low,medium,and high Z simultaneously.For high-Z materials with similar atomic numbers,this method can clearly distinguish those with apparent differences in density.According to the peak signal-to-noise ratio of the analysis,the reconstructed image quality of the new method was significantly higher than that of the individual imaging methods.This study provides a reliable approach to the compatibility of scattering tomography and coinciding muon trajectory density tomography.

With the continuous development of deep oil and gas,minerals,geothermal resources,and other resources,there are increasingly more stringent requirements for equipment.In particular,the ultra-highpressure dynamic seals of deep mining device need to be developed.Therefore,considering the use of dynamic seals in unique deep mining environments,an ultra-high-pressure rotating combined dynamic seal was designed and developed and its sealing performance was experimentally measured and analyzed.The results show that the experimental device can operate stably under a pressure of up to150 MPa and a rotating speed of 76 r/min,and can also operate normally under a rotating speed of up to 140 r/min and a sealing pressure of 120 MPa.During the operation of the ultra-high-pressure rotating combined dynamic seal,the sealing ring does not show obvious damage,which vouches for its sealing performance.No leakage of flow and pressure was detected in the all seal structures within the sealing pressure range of 0-150 MPa.Therefore,the dynamic sealing performance of the device is intact under ultra-high-pressure conditions and can be applied in deep mining environments at a certain depth.The research and development of this device can aid future deep energy exploration and exploitation.

Piezocatalytic materials have been widely used for catalytic hydrogen evolution and purification of organic contaminants.However,most studies focus on nano-size and/or polycrystalline catalysts,suffering from aggregation and neutralization of internal piezoelectric field caused by polydomains.here we report a single crystal ZnO of large size and few bulk defects crafted by a hydrothermal method for piezocatalytic hydrogen generation from pure water.It is noteworthy that single-side surface areas of both original as-prepared ZnO and Ga-doped ZnO bulk crystals are larger than 30 cm^(2).The high quality of ZnO and Ga-doped ZnO bulks are further uncovered by high-resolution transmission electron microscope(HRTEM),photoluminescence(PL)and X-ray diffraction(XRD).Remarkably,an outstanding hydrogen production rate of co-catalyst-free Ga-doped ZnO bulk crystal(i.e.,a maximum rate of 5915μmol h^(-1) m^(-2))is observed in pure water triggered by ultrasound in dark,which is over 100 times higher than that of its powder counterpart(i.e.,52.54μmol h^(-1) m^(-2)).The piezocatalytic performance of ZnO bulk crystal is systematically studied in terms of varied exposed crystal facet,thickness and conductivity.Different piezocatalytic performances are attributed to magnitude and distribution of piezoelectric potential,revealed by the finite element method(FEM)simulation.The density functional theory(DFT)calculations are employed to investigate the piezocatalytic hydrogen evolution process,indicating a strong H_(2)O adsorption and a low energy barrier for both H_(2)O dissociation and H2 generation on the stressed Znterminated(0001)ZnO surface.
