Background:To compare the efficacy of Ex-PRESS implantation versus trabeculectomy combined with phacoemulsification.Methods:A retrospective 12-month study on patients with coincident primary open-angle glaucoma(POAG)and cataract.The patients underwent combined phacoemulsification and Ex-PRESS implant(Phaco-ExPRESS,n=35)or phacotrabeculectomy(Phaco-Trab,n=35).The morphological structures of the filtering bleb were examined by slitlamp,anterior segment optical coherence tomography(AS-OCT)and in vivo confocal microscopy(IVCM).Complete success was defined as postoperative intraocular pressure(IOP)he use of anti-glaucoma medication.Qualified success was defined as postoperative IOPhe data were collected preoperatively and postoperatively at 2 weeks,1 month,3 months,6 months,and 12 months.Results:No significant difference in the variables such as age,IOP and perimetry was found between the groups of Phaco-ExPRESS and Phaco-Trab.At the one-year postoperative visit for filtering blebs,Phaco-ExPRESS increased the mean area of epithelial microcysts significantly from 0.10±0.05 to 0.20±0.09μm^(2)perμm^(2),while Phaco-Trab decreased the mean area significantly from 0.08±0.04 to 0.04±0.06μm^(2)perμm^(2).Notably,the hyperreflective dots detected by IVCM decreased by 84.9%in Phaco-ExPRESS but increased by 36.3%in Phaco-Trab.The hyperreflective dots were further identified as neutrophil-and monocyte-like cells.The number of these cells were negatively correlated with the microcysts area(r=−0.7,Phe grade of connective tissue(r=0.5,Phe filtering blebs,Phaco-ExPRESS produced a higher complete success rate(84.9%vs.41.2%,Phe number of anti-glaucoma medications(Phen compared with those in Phaco-Trab.However,the qualified success showed no significant difference between the two groups(100.0%vs.91.2%,P=0.24).Conclusions:At the one-ye

Impacts of pancreatic exocrine insufficiency on gut microbiota

Pancreatic exocrine insufficiency(PEI)can be induced by various kinds of diseases,including chronic pancreatitis,acute pancreatitis,and post-pancreatectomy.The main pathogenetic mechanism of PEI involves the decline of trypsin synthesis,disorder of pancreatic fluid flow,and imbalance of secretion feedback.Animal studies have shown that PEI could induce gut bacterial overgrowth and dysbiosis,with the abundance of Lactobacillus and Bifidobacterium increasing the most,which could be partially reversed by pancreatic enzyme replacement therapy.Clinical studies have also confirmed the association between PEI and the dysbiosis of gut microbiota.Pancreatic exocrine secretions and changes in duodenal p H as well as bile salt malabsorption brought about by PEI may affect and shape the abundance and composition of gut microbiota.In turn,the gut microbiota may impact the pancreatic exocrine acinus through potential bidirectional crosstalk.Going forward,more and higher-quality studies are needed that focus on the mechanism underlying the impact of PEI on the gut microbiota.

Tb^(3+)-nucleic acid probe-based label-free and rapid detection of mercury pollution in food

Mercury is a threatening pollutant in food,herein,we developed a Tb^(3+)-nucleic acid probe-based label-free assay for mix-and-read,rapid detection of mercury pollution.The assay utilized the feature of light-up fluorescence of terbium ions(Tb^(3+))via binding with single-strand DNA.Mercury ion,Hg^(2+)induced thymine(T)-rich DNA strand to form a double-strand structure(T-Hg^(2+)-T),thus leading to fluorescence reduction.Based on the principle,Hg^(2+)can be quantified based on the fluorescence of Tb^(3+),the limit of detection was 0.0689μmol/L and the linear range was 0.1-6.0μmol/L.Due to the specificity of T-Hg^(2+)-T artificial base pair,the assay could distinguish Hg^(2+)from other metal ions.The recovery rate was ranged in 98.71%-101.34%for detecting mercury pollution in three food samples.The assay is low-cost,separation-free and mix-to-read,thus was a competitive tool for detection of mercury pollution to ensure food safety.

In the past 5 years,there has been significant research interest in the intrinsic magnetic topological insulator family compounds MnBi2+2nTe4+3n（where n=0,1,2…）.In particular,exfoliated thin films of MnBi2Te4have led to numerous experimental breakthroughs,such as the quantum anomalous Hall effect,axion insulator phase and high-Chern number quantum Hall effect without Landau levels.However,despite extensive efforts,the energy gap of the topological surface states due to exchange magnetic coupling,which is a key feature of the characteristic band structure of the system,remains experimentally elusive.The electronic structure measured by using angle-resolved photoemission （ARPES） shows significant deviation from ab initio prediction and scanning tunneling spectroscopy measurements,making it challenging to understand the transport results based on the electronic structure.This paper reviews the measurements of the band structure of MnBi2+2nTe4+3nmagnetic topological insulators using ARPES,focusing on the evolution of their electronic structures with temperature,surface and bulk doping and film thickness.The aim of the review is to construct a unified picture of the electronic structure of MnBi2+2nTe4+3ncompounds and explore possible control of their topological properties.

Micromechanics of Thermal Conductive Composites:Review,Developments and Applications

Micromechanics investigations of composites with fiber-shaped reinforcement are extensively applied in the engineering design and theoretical analysis of thermal composites in the aerospace engineering and high-tech industry.In this paper,a critical review of various classical micromechanics approaches is provided based on the classification framework and the development of micromechanics tools.Several numerical micromechanics tools have been developed to overcome limitations through exactly/approximately solving the internal governing equations of microstructures.The connections and limitations of those models are also investigated and discussed,based on which three recently developed numerical or semi-analytical models are explained,including finite-element micromechanics,finite-volume direct averaging micromechanics,and locally exact homogenization theory,as well as machine learning tools.Since it is almost inevitable to mention the interfacial effects on thermal behavior of fibrous composites,we review the new mathematical relations that interrupt the original continuity conditions due to the existence of interphase/interface within unit cells.Generally speaking,the interphase/interface is demonstrated to play a significant role in influencing the effective coefficients and localized thermal fields.The present work also briefly reviews the application of micromechanics tools in emerging engineered woven composites,natural fibrous composites,and ablative thermal protection composites.It is demonstrated that sophisticated micromechanics tools are always demanded for investigating the effective and localized responses of thermal fibrous composites.

Gut microbial interactions based on network construction and bacterial pairwise cultivation

Association networks are widely applied for the prediction of bacterial interactions in studies of human gut microbiomes.However,the experimental validation of the predicted interactions is challenging due to the complexity of gut microbiomes and the limited number of cultivated bacteria.In this study,we addressed this challenge by integrating in vitro time series network(TSN) associations and cocultivation of TSN taxon pairs.Fecal samples were collected and used for cultivation and enrichment of gut microbiome on YCFA agar plates for 13 days.Enriched cells were harvested for DNA extraction and metagenomic sequencing.A total of 198 metagenome-assembled genomes(MAGs) were recovered.Temporal dynamics of bacteria growing on the YCFA agar were used to infer microbial association networks.To experimentally validate the interactions of taxon pairs in networks,we selected 24 and 19 bacterial strains from this study and from the previously established human gut microbial biobank,respectively,for pairwise co-cultures.The co-culture experiments revealed that most of the interactions between taxa in networks were identified as neutralism(51.67%),followed by commensalism(21.67%),amensalism(18.33%),competition(5%) and exploitation(3.33%).Genome-centric analysis further revealed that the commensal gut bacteria(helpers and beneficiaries) might interact with each other via the exchanges of amino acids with high biosynthetic costs,short-chain fatty acids,and/or vitamins.We also validated 12 beneficiaries by adding 16 additives into the basic YCFA medium and found that the growth of 66.7% of these strains was significantly promoted.This approach provides new insights into the gut microbiome complexity and microbial interactions in association networks.Our work highlights that the positive relationships in gut microbial communities tend to be overestimated,and that amino acids,short-chain fatty acids,and vitamins are contributed to the positive relationships.

A Predictive Energy Management Strategies for Mining Dump Trucks

The plug-in hybrid vehicles(PheV)technology can effectively address the issues of poor dynamics and higher energy consumption commonly found in traditional mining dump trucks.Meanwhile,plug-in hybrid electric trucks can achieve excellent fuel economy through efficient energy management strategies(EMS).Therefore,a series hybrid system is constructed based on a 100-ton mining dump truck in this paper.And inspired by the dynamic programming(DP)algorithm,a predictive equivalent consumption minimization strategy(P-ECMS)based on the DP optimization result is proposed.Based on the optimal control manifold and the SOC reference trajectory obtained by the DP algorithm,the P-ECMS strategy performs real-time stage parameter optimization to obtain the optimal equivalent factor(EF).Finally,applying the equivalent consumption minimization strategy(ECMS)realizes real-time control.The simulation results show that the equivalent fuel consumption of the P-ECMS strategy under the experimentally collected mining cycle conditions is 150.8 L/100 km,which is 10.9%less than that of the common CDCS strategy(169.3 L/100 km),and achieves 99.47%of the fuel saving effect of the DP strategy(150 L/100 km).

Establishment of a humanized mouse model using steady-state peripheral blood-derived hematopoietic stem and progenitor cells facilitates screening of cancer-targeted T-cell repertoires

Background:Cancer-targeted T-cell receptor T(TCR-T)cells hold promise in treating cancers such as hematological malignancies and breast cancers.However,approaches to obtain cancer-reactive TCR-T cells have been unsuccessful.Methods:here,we developed a novel strategy to screen for cancer-targeted TCR-T cells using a special humanized mouse model with person-specific immune fingerprints.Rare steady-state circulating hematopoietic stem and progenitor cells were expanded via three-dimensional culture of steady-state peripheral blood mononuclear cells,and then the expanded cells were applied to establish humanized mice.The human immune system was evaluated according to the kinetics of dendritic cells,monocytes,T-cell subsets,and cytokines.To fully stimulate the immune response and to obtain B-cell precursor NAML-6-and triple-negative breast cancer MDA-MB-231-targeted TCR-T cells,we used the inactivated cells above to treat humanized mice twice a day every 7 days.Then,human T cells were processed for TCRβ-chain(TRB)sequencing analysis.After the repertoires had been constructed,features such as the fraction,diversity,and immune signature were investigated.Results:The results demonstrated an increase in diversity and clonality of T cells after treatment.The preferential usage and features of TRBV,TRBJ,and the V–J combination were also changed.The stress also induced highly clonal Science and Technology,Grant/Award Number:2021C03010;Zhejiang Provincial Natural Science Foundation of China,Grant/Award Numbers:LTGY24H080003,LY21H080004 expansion.Tumor burden and survival analysis demonstrated that stress induction could significantly inhibit the growth of subsequently transfused live tumor cells and prolong the survival of the humanized mice.Conclusions:We constructed a personalized humanized mouse model to screen cancer-targeted TCR-T pools.Our platform provides an effective source of cancer-targeted TCR-T cells and allows for the design of patient-specific engineered T cells.It therefore has the potential to greatly benefi

Single crystals of a bismuth-based coordination polymer(CP)with carboxyl-thiol ligands,[Bi(C_(8)H_(2)O_(4)S_(2))(C2H8N)]n(Bi-DSBDC-DMA,DMBDC=2,5-disulfur-1,4-dicarboxylate,DMA=dimethylamine),have been successfully synthesized.X-ray diffraction analysis reveals that Bi-DSBDC-DMA possesses a layered structure,with two-dimensional(2D)Bi-DSBDC networks alternating with layers composed of dimethylamine ions.This material demonstrates semiconducting properties,featuring an optical bandgap of 2.2 eV and an electrical conductivity of 2×10^(-8) S/cm.Furthermore,electrodes based on this material exhibit a capacity of 250 mAh/g after 200 cycles for lithium-ion storage.

The estrogen signaling system is a crucial regulator of metabolicandphysiologicalprocesses.However,abnormal activation of estrogen signaling may play a role in breast cancer initiation and progression.Crucial to this pathway is the interaction between estrogen receptor alpha(ERa)and various co-transcription activators.1 Although numerous studies have investigated ER coregulators,the protein-protein interaction networks of ERa are not fully understood.Recent research has shown that high chromodomain helicase DNA-binding 4(CHD4)expression is linked to poor prognosis in various cancers.2,?In this study,we demonstrated that both CHD4 and ERαcontribute to breast cancer progression while providing evidence of the regulatory processes and functional interplay between these two proteins.
